Objectives In the past, studies have been shown that Forsythiae Fructus (FF), Ulmi Cortex (UC) and JwaGwiEum (JGE) are effective in treating allergic reactions. However, no report shows the difference in effects of FF, UC and JGE in allergic rhinitis (AR). Therefore, the purpose of this study was to differentiate the effects of FF, UC and JGE in AR induced by ovalbumin (OVA) in mice. Methods From this experiment, the effects of FF, UC and JGE were several - changes in body weight, hematologic changes such as WBC, RBC, Hb and PLT counts, immunological changes such as levels of histamine, IgE, IL-4 and expressions of iNOS & COX-2 mRNA. Moreover, histological change of nasal mucosa was also investigated. Results FF administration group and JGE administration group significantly inhibited level of IgE compared to the control group. Also, FF administration group, UC administration group and JGE administration group inhibited IL-4 and expressions of iNOS & COX-2 mRNA. In histological assessment, FF administration group, UC administration group and JGE administration group showed reduced amount of subepithelial edema, desquamated mucosa and hyperplasia of basement membrane from OVA.
This study was carried out to investigate effect of lipid metabolism in Viscum album lectin on rats. The lectin was purified by sepharose 4B affinity chromatography and gel filtration using sephadex G-150 with plant material from Viscum album collected in Mt. Duk Yui. After 72 h of $CCl_4$ injection (in olive oil, 1:1, 2 mg/kg) there was a significant increase in serum total cholesterol and triglycerige levels relative to the control group. However, treatment of both Viscum album and purified lectin were significantly decreased lipid parameters against the $CCl_4$-induced. Histological observation basically supported the result obtained from serum lipid assay. The livers of rats challenged with $CCl_4$ produced a marked increase of cytoplasmic vacuoles in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered olive oil alone did not alter the normal hepatic architecture. Histological observation of the liver section in rats treated 72 h with either Viscum album purified lectin or $CCl_4$-induced liver lipogenesis showed decreased numbers of cytoplasmic vaculoes and necrotic cells. The normal hepatic architectural pattern was observed in Hematoxylin-eosin stain. These results suggest that Viscum album lectin has a possible protective effect of lipid metabolim in rats.
Objectives: This study was designed to investigate the protective effects of Atractylodis Rhizoma pharmacopuncture (ARP) against acute gastric mucosal lesions induced by compound 48/80 in rats. Methods: The ARP was injected in Joksamni (ST36) and Jungwan (CV12) 1 hr before treatment with compound 48/80. The animals were sacrificed under anesthesia 3 hrs after treatment with compound 48/80. The stomachs were removed, and the amounts of gastric adherent mucus, gastric mucosal hexosamine, thiobarbituric acid reactive substances (TBARS), xanthine oxidase (XO), and superoxide dismutase (SOD) were measured. Also, histological examination were performed. Results: Gastric adherent mucus, gastric mucosal hexosamine and histological defects of gastric mucosa declined significantly after ARP treatment. Changes in gastric mucosal TBARS were also reduced by ARP treatment, but this result was not statistically significant. ARP treatment did not change the XO and the SOD activities. Conclusions: ARP showed protective effects for acute gastric mucosal lesions induced by compound 48/80 in rats. These results suggest that ARP may have protective effects for gastritis.
Purpose: The quality of implant surface is one of the factors that influence wound healing of implant site and subsequently affect osseointegration. The objective of modification of the surface properties of an implant is to affect the biological consequence. The purpose of this study is to evaluate the biologic response of osseous tissue to anodized implants. Materials and Methods: Two machined titanium implants for control group were installed in a tibia of each rabbit and two anodized implants for test group were installed in the other tibia of each rabbit. At the moment the implants were installed, resonance frequency analysis (RFA) values were measured. After healing periods of 1, 2, 3, and 7 weeks, the implants were uncovered and RFA values were measured again. Removal torque was measured for one implant in the test group and one implant in the control group. Histological evaluation was executed in the other implants. Results: Both of test group and control group have the tendency of greater RFA change rate and removal torque value as healing periods became longer, but were statistically insignificant (P>0.05). However, in the case of the same healing period, the test group tended to have greater RFA change rate and removal torque than the control group (P<0.05). More active new bone formation from endosteal surface was noted on the anodized surface than machined surface in specimen after 1 week. There were no significant differences between the test group and control group in histological evaluations. Conclusion: In summary, the anodized surface showed slightly favorable results and it is postulated that it may facilitate improved stability in bone.
The development and change of the cartilage canals ralated to chondrocyte growth and the major histological pattern were investigated in the calcaneus of the Korean native cattle fetuses from CRL 28mm to 175mm. The results as follows; 1. The cartilage canal first appeared in the caudal subperichondral region of the sustentaculum tali of the CRL 28mm fetus, and well developed after the period. 2. The three major cartilage canals, one entered from the head of calcaneous into where achilles tendon insert and the others from the caudal region of sustentaculum tali and the calcaneal sulcus, growth distinctively. The latters participated in the ossification center prior to the former. 3. The number of cells in this canals and the size increased synchronously with chondrocytic hypertrophy. In the intermediate and deep canals, the chondrogenic change showed distinctinvely. Our results demonstrated that the cartilage canals from the lower part were important the formation of the ossification center, and the growth and development of canals associated with the hypertropic changes of chondrocytes in the calcaneus of the Korean native cattle fetus.
A number of experimental studies have been carried out in order to clarify the question as to how temporomandibular joint adapt to the changes of mandibular movement and occlusal equilibration. Recently, the studies on the interrelations between anatomical structure of temporomandibular joint and the state of occlusion have been actively performed in dentistry particularly in prosthodontic field. Author performed extraction of unilateral mandibular molars in 30 mature male rats, and observed histological changes of temporomandibular joint through the light microscope. Following results were obtained. 1. The loss of unilateral teeth gave rise to the changes in the location of condylar head, that is, interior displacement of condylar head in the extraction side and upper displacement in the non-extraction side. 2. Articular disk was compressed by the interior surface of condylar head, resulting in its extension below the condylar neck in the extraction side, and the histological arrangement of the compressed area showed irregular feature. 3. The extension of articular disk below the condylar neck was accompanied with the contraction of muscle fibers which were originated from the articular disk. 4. The cartilage layer of articular fossa to the exterior of the extraction side showed hypertropy. 5. Early in the experiment, the inernal extremity of condylar head of extract ion side showed bone resorption, and cartilage layer of condylar head showed hypertropy. At 12 weeks after experiment, the condylar surface showed flattened, and the cartilage layer of condylar head was replaced by the compact bone. 6. The articular disk showed the formation of pannus in the extraction side as well as in the non-extraction side. 7. The occlusal disturbance due to unilateral missing teeth has brought about the non-inflammatory retrogressive change and osteoarthrotic change late in the experiment.
The eggshell, which is a complex and highly ordered structure, is very important factor for food safety and egg marketing. This study investigated the changes in eggshell structure and shell components in relationship to hen age. For this study, we examined the histological change of the endometrium of the 30-, 60-, and 72-wk-old commercial layers, and analyzed the ultrastructure and ionic composition of their eggshells. The results showed that histological deformation, fibrosis, atrophy and elimination of micro-villi in the uterus endometrium were found through microscopic observation that was associated with increasing hen age. Concentration of blood-ion components such as $Ca^{2+}$, $Na^+$, $K^+$, and $Cl^-$ ions did not change with age. Along with the results from the ultrastructure analysis of the eggshell, the palisade layer ratio and the density of mammillary knobs were significantly decreased in older hens. In addition, the type B mammillary knobs were frequently observed with increasing hen age. In the mineral element assay from the eggshell, $Ca^{2+}$, $S^{2-}$, and $Co^{2+}$ significantly decreased with increasing hen age, whereas $Na^+$, $K^+$, and $V^{2+}$ significantly increased. Therefore, the damages of endometrial tissue inhibit the processes of ion transmission and the crystallization of eggshell formation, resulting in a large and non-uniform mammillary knob formation. This means the conditions of endometrial cells affect the formation of the eggshell structure. In conclusion, hen aging causes the weakness of the eggshell and degrades the eggshell quality.
Objective : The aim of this study is to investigate the preventive effect of Injinchunggan-tang (YJCGT) & Injinysaryung-san (YJSRS) on MCD-diet-induced NASH in A/J mice. Methods : A/J mice were divided into 4 groups: Normal group (normal diet without any treatment). Control group (MCD diet only), YJCGT group (MCD diet with YJCGT), and YJSRS group (MCD diet with YJSRS). After 5 weeks, body weight, liver weight, biochemical parameters for liver function test, histological changes, and real-time PCR were assessed. Results : Mice lost body weight with the MCD diet and the YJCGT and YJSRS groups lost less than the control group, though showed no statistical significance. Liver weights were decreased by the MCD diet, but not significantly. In the liver function test, all the values were increased with the MCD diet, though some did not show significance. Alp and ALT levels were significantly less increased by YJCGT compared to the normal (p<0.05). All values were decreased or increased compared to the control by treatment though showed no significance possibly due to insufficient sample numbers. In histological findings of the livers. MCD-diet induced severe fatty change and collagen accumulation in the livers, but this fatty change was reduced in the YJCGT and YJYRS groups and fibrogenesis was inhibited significantly with p<0.05 and p<0.01, respectively. In real-time PCR analysis, YJCGT and YJYRS showed inhibitory effect on liver fibrogenesis by reducing associated gene expressions caused by MCD diet. Conclusion : YJCGT and YJSRS are considered to be possible candidates for the treatment of patients with NASH and/or liver fibrosis.
Background: The purpose of this study was to evaluate the change of food intake after different dosages of botulinum toxin A (BTX) injection in the animal model. Additionally, the dimensional and histological change at 14 days after BTX injection was also evaluated. Methods: The comparative study was performed using the BTX injection model in rats (n = 5 for each group). Group 1 was the saline-injected group. Group 2 was the 5-unit BTX-injection group to each masseter muscle. Group 3 was the 10-unit BTX-injection group to each masseter muscle. Food intake rates and body weight were checked daily before and after BTX injection until 10 days. All animals were sacrificed at 14 days after BTX injection, and the specimens underwent hematoxylin and eosin stain and immunohistochemical staining for myosin type II (MYH2). Results: The recovery of food intake in groups 2 and 3 decreased significantly compared with group 1 from day 2 to day 7 and day 9 after injection (p < 0.05). The BTX-treated masseter muscles were significantly smaller than those in group 1 (p = 0.015). The immunohistochemical findings demonstrated that the expression of MYH2 was significantly higher in group 3 compared to groups 1 and 2 (p < 0.001). Conclusions: BTX injection to the masseter muscle in rats demonstrated short food-intake-rate reduction with recovery until 10 days after injection. The thickness of the masseter muscle and MYH2 expression were significantly changed according to the injected dose of BTX.
This study was performed to investigate the effect of aluminum compound on the aluminum contents and histological change in brain tissue of rats. Seventy five male Sprague-Dawley strains were divided into five groups consisting of the control, 250 ppm $AlCl_3$ group, 500 ppm $AlCl_3$ group, 250 ppm $Al_2(SO_4)_3$ group, 500 ppm $Al_2(SO_4)_3$ group and kept on the diet for 2 weeks. The weight gain was increased by administration of $AlCl_3$ but decreased by administration of $Al_2(SO_4)_3$ as compared to control group. The aluminum contents in brain tissue of each group; 250 ppm $AlCl_3$ group, 500 ppm $AlCl_3$ group, 250 ppm $Al_2(SO_4)_3$ group and 500 ppm $Al_2(SO_4)_3$ group were 64.63, 102.21, 132.64 and 180.41 ppm, respectively. Aluminum accumulation in brain tissue was higher with administration of $Al_2(SO_4)_3$ than with administration of $AlCl_3$. In $AlCl_3$ administration group, multiple small intracytoplasmic granules and microvacuole were seen in large pyramidal cells of cortex and granulovacuolar degeneration. In $Al_2(SO_4)_3$ administration group revealed pollagis pallor, cellular pyknosis, microcavitation resulted from edema in deeper cortical layers were observed. Blue-pigmentation which represents the accumulation of aluminum was noted In granulovacuolar degeneration site in $Al_2(SO_4)_3$ administration group.
