Anti-stress Effect by the Treatment of Vitamin C (Vitamin C 투여에 의한 항스트레스 효과)

  • The anti-stress effect by the treatment of vitamin C was investigated in this study. The treatment of ascorbic acid in the presence of $Cu^{2+}$ion induced strong time- and dose-dependent degradation of hitamine, and also the addition of histamine accelerated time-dependent decomposition of ascorbic acid in vitro. The treatment of ascorbic acid in $ODS^{od}/_{od}$rats, which cannot synthesize ascorbic acid, significantly decreased the urinary histamine. The protreatment of ascorbic acid, dexamethasone and promethazine inhibited the lethal effect induced by immobilization stress, but that of dimethylsulfoxide did not. The addition of ascorbic and to a culture of spleen cells of $ODS^{od}/_{od}$rats significantly increased the Con A-dependent T lymphocyte proliferation.

  • This experimental study was done to research effects of a number of extract on the anti-allergic effect. The results were obtained as follows: 1. In effect of herb-extract on compound $\frac{48}{80}$-mediated histamine release from Evans blue skin assay, Isatis Japonica Miquel, Dictamnus dasycarpus Turcz, Spirodela polyrhiza, Cimicifuga heracleifolia, Bupleurum chinense, Magnolia liliflora, Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(MtOH), Trichosanthes kirilowii, Phellodendron amurense Rupr, Schizonepeta tenuifolia Var, Betula platyphylla show considerable visible anti-allergic effect. In the result of quantification of histamine induced compound $\frac{48}{80}$, Spirodela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively. 2. In effect of Herb-Extract on compound $\frac{48}{80}$-mediated histamine release from RPMC, Spirodcla polyrhiza, Cimicifuga heracleifolia inhibit histamine release effectively. 3. In effect of Herb-Extract on anti-DNP IgE-mediated histamine release from Evansblue skin assay. Spirodela polyrhiza, Cimicifuga heracleifolia(0.1mg/ml). Forsythia koreana, Aster tataricus L., Xanthium strumarium L.(0.1mg/ml), Trichosanthes kirilowii(0.1mg/ml) show considerable visible anti-allergic effect. In the result of quantification of histamine induced anti-DNP IgE, Spiradela polyrhiza, Isatis Japonica Miquel, Trichosanthes kirilowii, Bupleurum chinense, Forsythia koreana inhibit histamine release effectively 4. In the result of genetic manifestative inhibition about the human mast cell-1(HMC-1), Cimicifuga heracleifolia has considerable effect in IL-4 in IL-5, and Tussilage farfara L. has in IL-4. According to the above results, it is suggested that several Herb-Extract have anti-allergic effect.

  • Nonvolatile amines in fermented ordinary Korean soysauce were separated and quantitated. 1. Tyramine and histamine were detected as nonvolatile amines. Both of them were increased during the fermentation in the soysauce prepared with 22.0% of salt concentration but histamine only was increased in the soysauce with 28.5% salt concentration. Rapid increase of histamine was observed after 80days of fermentation. 2. Two unidentified spots in paper chromatogram which were positive to Sakaguchi reagents were detected. 3. The amounts of tyramine and histamine in fermented ordinary Korean soysauce were much lower than those in Japanese style soysauce.

Effects of dihydrocubebin, a lignan isolated from Indonesian plant Piper cubeba, on the histamine release from rat mast cells

  • The fruits of Piper cubeba L. are used traditionally to treat respiratory disorders in Indonesia. In order to determine the compounds responsible for this activity, the fruits were extracted with nhexane followed by ethanol to give n-hexane and ethanol extracts. Based on tracheospasmolytic assay on these two extracts, the n-hexane extract was more active to inhibit trachea contraction than that of ethanol extract. Upon bioassay guided isolation of the n-hexane extract, a tracheospasmolytic active compound was isolated and identified as dihydrocubebin [(3,4),(3',4')-bis-methylenedioxy-9,9'dihydroxylignan] $(\underline{1})$. Compound $\underline{1}$ was tested further for its ability to inhibit histamine released from mast cells, using rat basophilic leukemia (RBL-2H3) cell line and rat peritoneal mast cells RPMCs) as models; and $DNP_{24}$-BSA, thapsigargin, ionomycin, compound 48/80 and PMA were used as inducers for histamine released from mast cell. The test result showed that $\underline{1}$ inhibited histamine release from RBL-2H3 cells induced by $DNP_{24}$-BSA, thapsigargin and ionomycin. In addition, $\underline{1}$ suppressed histamine release from RPMC induced by either thapsigargin or ionomycin. However, $\underline{1}$ did not inhibit histamine release from RPMC induced by either compound 48/80 or combination PMA-sub optimum dose of ionomycin. Therefore, it was concluded that the inhibitory effects of $\underline{1}$ on the histamine released from mast cells may involve mechanisms related to intracellular $Ca^{2+}$ signaling events or downstream processes of intracellular $Ca^{2+}$ signaling in mast cells.

Effect of Raw Material Freshness on Quality and Safety of Anchovy Fish Sauce (원료의 선도가 멸치액젓의 품질 및 위생안전성에 미치는 영향)

  • This study conducted to investigate the impact of raw material freshness on the quality and hygienic safety of anchovy fish sauce and suggest the importance of raw material freshness to make high-quality and hygienic anchovy fish sauce by measuring the chemical compositions and histamine. To devide the raw material's freshness levels, raw anchovy was left for 24 hours and was sampled every 4 hour(Group I-VII). As a result, the levels of VBN(volatile basic nitrogen) and histamine increased as time passed each contents were 16.29-87.65 mg/100 g and 6.14-1499.63 mg/100 g respectively. As fish sauce fermented for 18 months, the contents of VBN were 205.31-270.51 mg/100 g and histamine were 120.54-1707.22 mg/100 g, respectively. These results means the levels of VBN and histamine of raw materials are significantly associated with the hygiene of anchovy fish sauce. To investigate the reason of different contents of histamine at each fish sauce, bacteria from each groups were isolated and identified. At group V-VII those the lowest three groups, Leconostoc mesenteroides ssp. cremoris was identified and that produced the highest content of histamine 22.88-101.69 mg/kg and the others produced histamine 3.79-20.2 mg/kg. This means that fish sauce made by low freshness materials contain bacteria have strong ability to make histamine from histidine. Therefore, the freshness of raw material influences the hygiene and safety of fish sauce, and it is most important to manage the freshness of raw material to make the high quality and hygienically safe fish sauce.

Inhibitory Effects of the Extracts from Eleutherococcus senticosus Maxim. on Histamine-release from Rat's Mast Cell

  • Eleutherococcus senticosus Maxim. has been successfully used as an oriental medicine for various diseases including allergic disorders. Histamine is a major factor on various allergic responses and it is reported that histamine was released from mast cells by sensitization of allergens. In this study, ethanol extracts from E. senticosus Maxim. were prepared and the composition was analyzed by high performance liquid chromatography. The eleutheroside B as a primary effective component of E. senticosus was contained approximately 225 mg/kg in root bark extracts. The extracts were found to significantly inhibit compound 48/80-induced histamine release form mast cells in dose dependent manner. However the extracts had low cytotoxicity on the mast cells with MTT assay. These results showed that E. senticosus Maxim. extracts may be the effective materials on inflammatory disorders.

  • 평골근(平滑筋)으로 된 자궁근(子宮筋)은 자동성(自動性)을 가지고 있어서 신경지배(神徑支配)와 관계(關係)없이 근자체(筋自體)로 운동(運動)을 하게 된다. 그러나 이러한 자궁근(子宮筋)의 형태적(形態的) 및 기능적(機能的) 정상상태유지(正常狀態維持)에는 estrogen의 작용(作用)이 불가결(不可缺)한 요소(要素)로 되어 있으며 이 estrogen의 작용(作用)에 의하여 histamine의 자궁근(子宮筋)에 대한 (작용)作用이 수용체(受容體)의 어떤 기전에 의한 것인지를 알기 위하여 본(本) 연구(硏究)는 histamine과 5-hydroxytryptamine 및 이들 길항물질(拮抗物質)들의 자궁근(子宮筋) 운동성(運動性)에 대한 수축(收縮) 및 이완작용(弛緩作用)을 조사(調査)하였다. 자궁근(子宮筋)의 운동성(運動性)은 physiograph를 통(通)해 자궁수축(子宮收縮)의 빈도(頻度)와 크기를 기록하여 아래와 같은 결론(結論)을 얻었다. 1. 5-hydroxytryptamine에 대한 phenoxybenzamine의 억제작용(抑制作用)은 phenoxybenzamine의 길항성(拮抗性)의 결과(結果)이다. 2. histamine은 $H_1$-receptor를 통해서 흰쥐의 자궁평활근(子宮平滑筋)의 운동성(運動性)은 증가(增加)한다. 3. 반면 histamine은 $H_2$-receptor를 통해서는 자궁평활근(子宮平滑筋)의 운동성(運動性)을 이완(弛緩)시켰다. 4. 흰쥐의 자궁근(子官筋)에서 $H_2$-receptor 차단제(遮斷劑)가 $H_1$-receptor 차단제(遮斷劑)의 작용(作用)보다 더욱 강하였다.

Investigation into the mechanism of anti-asthmatic action of Lepidium sativum

  • We have studied the possible mechanism of anti-asthmatic action of ethanolic extracts of dried seeds of Lepidium sativum (EXLS, 400 mg/kg) using various experimental models. EXLS produced an increase in the Pre-Convulsion Dyspnoea time induced by histamine and acetylcholine aerosol, a significant reduction in the elevated leucocyte counts in the Broncho-Alveolar Lavage fluid of sensitized guinea-pigs and reduction in the paw edema volume as compared to the control rats. Treatment with EXLS also produced decrease in the elevated histamine release from the sensitized guinea-pig lungs. The anti-asthmatic anti-inflammatory responses of EXLS was supported by improvement in microscopic changes like infiltration of inflammatory cells, submucosal edema, epithelial desquamation and reduced lumen size of the bronchi. The $pD_2$ values of histamine in tracheal chain and taenia-coli were significantly greater and that in lung strip was lower in the sensitized animals as compared to control. Treatment of sensitized guinea pigs with EXLS significantly decreased $pD_2$ values of histamine in all three preparations. Our data suggest the prevention of hyper-responsiveness in bronchial smooth muscles and inhibition of the immediate hypersensitive reaction, histamine release in the lungs and the infiltration of various inflammatory cells as the possible mechanisms of anti-asthmatic activity of EXLS.

Cortex Mori Inhibits the CGG-specific IgE-Dependent Histamine Release

  • Cortex Mori, the root bark of mulbery tree has been used as an antiphlogistic, diuretic, and expectorant in herbal medicine. The purpose of this study is to evaluate chicken gamma globulin (CGG)-specific IgE-induced morphologic and functional changes in rat peritoneal mast cells (RPMC), and to determine whether Cortex Mori could inhibit the CGG-specific IgE-depeildent mast cell degranulation and histamine release from RPMC. Results are 1) the degranuration and histamine release from RPMC were not induced within 1 hour after addition of Cortex Mori alone, 2) the CGG and CGG-specific IgE-Induced degranulation from RPMC was observed within 10 minutes, 3) the histamine release from RPMC sensitised with CGG-specific IgE was induced by tile addition of CGG, 4) CGG-specific IgE-dependent degranulation rate in RPMC pretreated with Cortex Mori was significantly Inhibited, compared to that of control group without Cortex Mori pretreatment, and 5) the CGG-specific IgE-dependent histamine release from RPMC was significantly inhibited by pretreatment with Cortex Mori. These data suggest that Cortex Mori contains some substances with capabilities to inhibit CGG-specific IgE-dependent degranulation and histamine release from RPMC.
