항산화제가 시스플라틴에 의해 유발된 쥐의 뒷다리근 위축 경감에 미치는 영향 (Effects of Antioxidant on Reduction of Hindlimb Muscle Atrophy Induced by Cisplatin in Rats)

  • Purpose: The purpose of this study was to examine the effects of Cu/Zn SOD on reduction of hindlimb muscular atrophy induced by cisplatin in rats. Methods: Forty-two rats were assigned to three groups; control group, Cisplatin (CDDP) group and cisplatin with Cu/Zn SOD (CDDP-SOD) group. At day 35 hindlimb muscles were dissected. Food intake, activity, withdrawal threshold, muscle weight, and Type I, II fiber cross-sectional area (CSA) of dissected muscles were measured. Relative SOD activity and expression of MHC and phosphorylated Akt, ERK were measured after dissection. Results: Muscle weight and Type I, II fiber CSA of hindlimb muscles in the CDDP group were significantly less than the control group. Muscle weight and Type I, II fiber CSA of hindlimb muscles, food intake, activity, and withdrawal thresholds of the CDDP-SOD group were significantly greater than the CDDP group. There were no significant differences in relative SOD activities of hindlimb muscles between the CDDP-SOD and CDDP groups. MHC expression and phosphorylated Akt, ERK of hindlimb muscles in the CDDP-SOD group were significantly greater than the CDDP group. Conclusion: Cu/Zn SOD attenuates hindlimb muscular atrophy induced by cisplatin through increased food intake and activity. Increment of phosphorylated Akt, ERK may relate to attenuation of hindlimb muscular atrophy.

주기적인 운동이 뒷다리 부유에 의해 유발된 쥐의 뒷다리를 위축경감에 미치는 영향 (Effect of Periodic Low-intensity Exercise on the Hindlimb Muscles of Hindlimb Suspended Rats)

  • The purpose of this study was to determine the effect of periodic low-intensity exercise during hindlimb suspension on the mass, relative weight, myofibrillar protein content in soleus, plantaris and gastrocnemius muscles. To examine the effectiveness of periodic low-intensity exercise on mass, and myofibrillar protein content of hindlimb muscles, adult female Wistar rats were suspended(HS) and half of these rats walked on a treadmill for 45min/day(15 min every 4h) at 5m/min and a $15^{\circ}$ grade(HS-EX). Soleus wet weight was 33.51% significantly smaller(p<0.005) and relative soleus weight of hindlimb suspended rats was 31.96% smaller(p<0.005) compared with those of control rats following seven days of hindlimb suspension. Plantaris wet weight was 7.5% smaller(p<0.01) and relative plantaris weight was 11.83% smaller(p<0.05) compared with those of control rats following seven days of hindlimb suspension. Gastrocnemius wet weight was 11.31% significantly smaller(p<0.005) and relative gastrocnemius weight was 17.13% significantly smaller(p<0.005) compared with those of control rats following seven days of hindlimb suspension. Soleus wet weight while increased by relative soleus weight increased by 25.13%, 27.59% each through periodic low intensity exercise during hindlimb suspension(p<0.05, p<0.05). Plantaris wet weight and relative plantaris weight increased by 1.04%, 10.98%(p<0.05) each, and gastrocnemius wet weight and relative gastrocnemius weight increased by 1.98%, 12.02%(p<0.05) each through periodic low intensity exercise during hindlimb suspension. Wet weight of soleus, plantaris and gastrocnemius in HS-EX rats did not recover to control level. Myofibrillar protein content of soleus, plantaris and gastrocnemius was 48.24%, 40.85% and 37.33% significantly smaller(p<0.005) respectively compared with those of control rats following seven days of hindlimb suspension. Myofibrillar protein content of soleus, plantaris and gastrocnemius increased by 40.68%, 25.07% and 17.93%(p<0.005) each through periodic low intensity exercise during hindlimb suspension. Myofibrillar protein content of soleus, plantaris, and gastrocnemius in HS-EX rats did not recover to control level. The results suggest that periodic low intensity exercise can attenuate hindlimb muscle atrophy induced by hindlimb suspension.

후지체공(後肢滯空) 유발 흰쥐의 위축 근육에 대한 강활속단탕(羌活續斷湯)의 근 재생효과 (The Effect of KangwhalSokdan-tang(Jianghuoxuduan-tang) Administration on Muscle Regeneration in Atrophied Rat Muscle Induced by Hindlimb Suspension)

  • Objectives : This experimental study was designed to investigate the effect of KangwhalSokdan-tang(Jianghuoxuduan-tang) on the muscle regeneration of atrophied rat muscle by hindlimb suspension. Materials and methods : In this study, Sprague-Dawley rats weighing about 250g were subjected to hindlimb suspension and divided into total four groups: Normal group(n=6), Control group(n=6), Hindlimb non-treatment group(n=6), Hindlimb treatment group(n=6). Experiments were seperately tried two times. The first trial was studied by the following two groups; The first was normal group(n=6). The second was group(n=18) for hindlimb suspension during 2 weeks (control I group). The second trial after 2 weeks hindlimb suspension was studied by the following three groups; The third group(n=6) was expired immediately after 2 weeks hindlimb suspension. The forth group(n=6) was given free activity during 2 weeks after 2 weeks hindlimb suspension. The fifth group(n=6) was administrated of KST during 2 weeks after 2 weeks hindlimb suspension. In order to investigate degree of muscle atrophy, body weight and gastrocnemius muscle mass were compared. To analyze muscle regeneration factors(expression of IGF-1, Myogenin, MyoD), Western blot was used. Results : The results were analyzed by statistical process as follows, 1. In body weight, all hindlimb suspension groups were lower than normal group, but tendency of increase was shown in KST group compared to non-treatment group after 2 weeks hindlimb suspension. 2. In gastrocnemius muscle mass, KST group on both side was significantly higher than non-treatment group after 2 weeks hindlimb suspension. 3. In case of IGF-I, Type I of KST group was significantly increased than non-treatment group, but Type II was not shown significance. 4. There was no significantly difference in Myogenin. 5. In MyoD, Type I of KST group was significantly increased than control group, and Type II of KST group was significantly increased than non-treatment group. Conclusions : In summary, this study demonstrates that KST administration has an effect to prevent muscle atrophy and contribute muscle regeneration and proliferation. And also it is suggested that IGF-I and MyoD is major factors of myogenesis expression to KST adminstration after hindlimb suspension.

  • An abnormal female Holstein calf with ectrodactyly of right hindlimb was observed macroscopically and radiographically, and the results were summarized as follows. 1. The central and fourth tarsal bones in the left hindlimb were separated. 2. The tarsus in the right hindlimb consisted of five pieces; the talus, calcaneus, central and first tarsal bones were separated, and the second, third and fourth tarsal bones were fused. 3. The large metatarsal bone in the right hindlimb was fused rudimentary fourth metatarsal bone and well developed third metatarsal bone. 4. The phalanges of the lateral(fourth) digit were absent in the right hindlimb.

흰쥐의 후지 석고 고정, 탈신경 및 부유에 의한 근위축의 비교 (Comparison of Muscle Atrophy Induced by Cast Fixation, Denervation and Suspension of Rat Hindlimb)

  • The aim of this study was to compare features of muscle atrophy induced by cast fixation. denervation and suspension of rat hindimb. Muscle mass and glycogen of the soleus and plantaris muscles were studied after 3, 7, or 14 days of cast fixation, denervation and suspension. The results as follows: 1. Body weight of rats decreased significantly after 3 days and showed gradually increase after 7 and 14 days of hindlimb cast fixation, denervation and suspension. Particularly hindlimb suspended rats showed a rapid decrease after 3 days in body weight. 2. Relative weight of soleus and plantaris musclcs decreased significantly by hindlimb cast fixation, denervation and suspension, particularly after 7 days. The decrease rate was the lowest in suspended rats. 3. Glycogen content of soleus muscle decreased significantly after 14 days of hindlimb cast fixation, denervation and suspension. Also glycogen content of plantaris muscle decreased significantly after 14 days of hindlimb cast fixation and denervation, but not significantly after hindlimb suspension. These results indicate that suspension of hindlimb muscles causes less atropy than cast fixation or denervation, likely due to maintainment a few activities during hindlimb suspension. We concluded that the decrease in mechanical strains imposed on the muscle during inactivity was the main factor for the development of atrophy. These basic data suggest that some experimental conditions such as electrostimulation or stretching, participate in countermeasure programmes.

The Effect of Animal Physiotherapy on Balance and Walking in Dog with Sciatic Nerve Injury and Degenerative Joint Disease, Case Report

  • Objective: This study was conducted to confirm the effect of physiotherapy on the balance and walking in dog with sciatic nerve injury and degenerative arthritis of stifle joints. Design: Single case study Methods: The dog walked abnormally for six months and was administrated in S animal hospital. The dog's right hindlimb was operated for cranial cruciate ligament repair and the dog had been taking a nonsteroidal anti-inflammatory analgesic before being refered. There was severe degenerated osteoarthritis in the right hindlimb. During stance and walking, the right hindlimb was often shown partial weight bearing. The dog's left hindlimb was shown plantigrade stance and walking. The radiograph was shown an intact calcaneal tendon in the left hindlimb. In the neurologic examination, sciatic nerve injury in the left hindlimb was confirmed. The dog was treated using muscle strengthening, proprioceptive exercise, underwater treadmill and Laser therapy two, or three times a week for 3 months. At the 10th and 17th treatment, it was evaluated through stance and gait analyzer system to measure dog's balance and walking. Results: 3 months following physiotherapy, the dog's balance was improved in center of pressure(COP). And peak vertical force(PVF), vertical impulse(VI) was increased in right hindlimb and double stance was decreased. Conclusions: Physiotherapy may have improved the prognosis in this dog with severe osteoarthritis and sciatic nerve injury. This study suggested that animal physiotherapy is a valuable way to improve balance and walking.

간헐적인 낮은강도, 짧은기간의 운동부하가 뒷다리 부유쥐의 Type II근육에 미치는 영향 (Effect of intermittent low-intensity, short duration exercise on Type IImuscle of suspended rats)

  • The purpose of this study was to determine the effect of intermittent low - intensity, short duration exercise during hindlimb suspension on the mass, relative weight, myofibrillar protein content, cross-sectional area of Type I and Type II fibers and SDH activity in Type II(plantaris) muscle. To examine the effectiveness of intermittent low-intensity, short duration exercise on mass, myofibrillar protein content and fiber size, the hindlimbs of adult female Wistar rats were suspended(HS) and half of these rats walked on a treadmill for 45 min/day(9 min every 2h) at 5m /min and a 15$^{\circ}$grade (HS-EX). Plantaris wet weight was 19.67% significantly smaller(p<0.005) and relative plantaris weight was 6.25% smaller compared with those of control rats following seven days of hindlimb suspension. Plantaris wet weight and relative plantaris weight increased by 27.66%, 12.22% each through intermit-tent exercise during hindlimb suspension(p<0.005, p<0.05), moreover, plantaris wet weight and relative plantaris weight of the HS-EX rats were similar to those of control rats. Soleus wet weight and relative soleus weight decreased significantly by 31% and 22.0% in the HS rats(p<0.05). Soleus wet weight and relative soleus weight increased by 10.41%, 25.64% respectively through intermittent ex-ercise during hindlimb suspension, furthermore, soleus wet weight and relative weight of the HS-EX rats were closer to those of control rats. Myofibrillar protein content of plantaris and soleus decreased significantly by 51.49%, 59.65% each, following seven days of hindlimb suspension (p<0.005) Myofibrillar protein content of plantaris and soleus increased by 51.79%, 75.47% each with significance through intermittent exercise during hindlimb suspension(p<0.005). Myofibrillar protein content of plantaris and soleus in HS-EX rats was smaller than that of control rats. No change was observed in fiber type percentage following 1 week of hindlimb suspension or exercise during hindlimb suspension. The type I fiber cross-sectional area of both soleus and plantaris muscle was 18.72% and 41.07% lower in the HS than that of the controls (p<0.05, p<.001), that of both muscles was 6.60% and 29. 3% greater in the HS-EX than that of the HS rats. HS plus intermittent low- intensity short duration exercise resulted in Type I fiber cross-sectional area closer to the controls. Type II fiber cross-sectional area of both plantaris and soleus muscle was 22.45% and 22.58% sl nailer in the HS than in the controls, that of both muschles in the HS-EX was 14.10%, 5.78% greater than HS. Intermittent exercise during hindlimb suspension resulted in Type I, II fiber cross-sectional area closer to the control value. There was no change in SDH activity following 1week of hindlimb suspension or exercise during hindlimb suspension in the plantaris muscle. The results suggest that intermittent low intensity short duration exercise can ameliorate Type II muscular atrophy Induced by hindlimb suspension.

Glyphosate Surfactant Herbicide Toxicosis in a Dog with Hindlimb Paresis and Urinary Incontinence

  • A 4-year-old Yorkshire terrier was presented with hindlimb paresis and urinary incontinence after accidental ingestion of an herbicide. Based on neurologic examinations, decreased hindlimb proprioception with flaccid paresis were revealed. Other possible causes of the clinical signs were excluded. The clinical signs gradually improved after administration of anti-inflammatory and antioxidant therapy. This case report is the first to describe the long-term outcome of hindlimb paresis and urinary incontinence induced by glyphosate surfactant herbicide (GPSH) poisoning in a dog.

  • The abnormal male Newhampshire of five months age with three hindlimbs was examined macroscopically and radiographically The external feature included two normal wings, two normal hindlimbs and an underdeveloped extra hindlimb, which was attached to the pelvic region. The extra hindlimb consisted of an underdeveloped femur like bone, tibiotarsus likebone, tarsometatarsus like bone and three rows of digits with claw. The femur like bone was fused with pelvic girdle like bone. The bones of extra hindlimb were fused into one except one row of digit, which was attached to the tibiotarsus like bone. Three ceca and one cloaca with two vents were also observed.

  • The purpose of this study was to determine the effect of endurance training prior to occurrence of muscle atrophy on the mass, myofibrillar protein content and fiber crossectional area of atrophied hindlimb muscles of rats. Adult female Wistar rats were trained prior to occurrence of muscle atrophy induced by hindlimb suspension. Training began on the 1st day for 10min /day at 15m /min on a 0% grade, training exercise increased daily in time and intensity so that by the 4th week rats were running 60min /day, at 34m /min on a i3.5% grade. Wet weight and relative weight of soleus, plantaris and gastrocnemius muscle decreased significantly after seven days of hindlimb suspension. Wet weight and relative weight of soleus tended to increase and that of plantaris and gastrocnemius tended to decrease in the exercise group as compared to the control group. Myofibrillar protein content of soleus and gastrocnemius tended to increase and that of plantaris tended to decrease in the endurance trained group as compared to the control group. Fiber crossectional area of Type I, II fiber in soleus and plantaris muscle tended to increase in the exercise group as compared to the control group. Wet weight and relative weight of soleus. plantaris and gastrocnemius decreased significantly, myofibrillar protein content of soleus, plantaris and gastrocnemius increased in hindlimb suspended rats following endurance training as compared to the control group. There was no change in fiber type percentage and crossectional area of type I and II fiber in soleus muscle and that of type I and IIfiber in plantaris muscle decreased in the hindlimb suspended rats following endurance training as compared to the control group. Wet weight and relative weight of soleus and plantaris tended to increase, that of gastrocnemius increased significantly, myofibrillar protein content of soleus and plantaris muscle increased significantly and that of gastrocnemius tended to increase in the hindlimb suspended rats following endurance training as compared to sedentary rats following endurance training. Crossectional area of type I fiber of soleus muscle tended to increase. that of type I fiber of plantaris muscle increased significantly and that of type II fiber tended to increase in hindlimb suspended rats following endurance training as compared to sedentary rats following endurance training. The results suggest that endurance training prior to occurrence of muscle atrophy can attenuate the decrease of mass, myofibrillar protein content and fiber crossectional area induced by hindlimb suspension.
