It has been known that ATM plays a central role in response of cells to ionizing radiation by enhancing DNA repair. We have investigated the feasibility of increasing radiosensitivity of tumor cells with the use of ATM inhibitors such as caffeine, pentoxifylline and wortmannin. Human colorectal cancer RKO.C cells and RKO-ATM cells (RKO cells overexpressing ATM) were used in the present study. The clonogenic cell survival in vitro indicated that RKO-ATM cells were markdely radioresistant than RKO.C cells. Treatment with 3 mM of caffeine significantly increased the radiosensitivity of cells, particulary the RKO-ATM cells, so that the radiosensitivity of RKO.C cells and RKO-ATM cells were almost similar. The radiation induced G2/M arrest in RKO-ATM cells was noticeably longer than that in RKO.C cells and caffeine treatment significantly reduced the length of the radiation induced G2/M arrest in both RKO.C and RKO-ATM cells. Pentoxifylline and wortmannin were also less effective than caffeine to radiosensitize RKO.C or RKO-ATM cells. However, wortmannin was more effective than caffeine against human lung adenocarcinoma A549 cells indicating the efficacy of ATM inhibitor to increase radiosensitivity is cell line dependent. For in vivo study, RKO.C cells were injected s.c. into the hind-leg of BALB/C-nuslc nude mice, and allowed to grow to 130mm3 tumor. The mice were i.p. injected with caffeine solution or saline and the tumors irradiated with 10 Gy of X-rays. The radiation induced growth delay was markedly increased by 1-2 mg/g of caffeine. It was concluded that caffeine increases radiosensitivity of tumor cells by inhibiting ATM kinase function, thereby inhibiting DNA repair, that occurs during the G2/M arrest after radiation.
Neutrons are high LET (linear energy transfer) radiation and cause more damage to the target cells than x-rays or gamma rays. The damage from neutrons is generally considered fatal to a cell and neutrons have a greater tendency to cause cell death through direct interaction on DNA. We performed experiments to measure growth delay ratio and oxygen enhancement ratio (OER) in mouse model system. We inoculated EMT-6 cells to the right hind leg of BALB-c mouse and X-rays and neutron beams were given when the average volume of tumors reached $200-300mm^3$. We irradiated 0, 11, 15.4 Gy of X-ray and 0, 5, 7 Gy of fast neutron beam at normoxic and hypoxic condition. The volume of tumors was measured 3 times per week. In x-ray experiment, growth delay ratio was 1.34 with 11 Gy and 1.33 with 15.4 Gy in normoxic condition compared to in hypoxic condition, respectively. In neutron experiment, growth delay ratio was 0.94 with 5 Gy and 0.98 with 7 Gy, respectively. The OER of neutron beam was 0.97. The neutron beam was more effective than X-ray in the control of hypoxic tumors.
Changes in quality characteristics and shelf-life of two types of grill sausage manufactured by using pork hind leg and re-pasteurized after packaging were investigated during storage at 5$^{\circ}C$ and 10$^{\circ}C$ up to 30 days. The total aerobic plate counts(APC) of N rnberger style grill sausage(NGS) and vegetable added grill sausage(VGS) were initially 3.08 and 3.57 log$\sub$10/CFU/$\textrm{cm}^2$, respectively, and gradually increased during storage time. After 30 days, the APC of NGS stored at 5$^{\circ}C$ and 10$^{\circ}C$ maintained relatively low at the levels of 5.32 and 6.58 log$\sub$10/CFU/$\textrm{cm}^2$, respectively. However, the APC of VGS exceeded the level of 8 log$\sub$10/CFU/$\textrm{cm}^2$ after 30 days at 5$^{\circ}C$ and after 20 days at 10$^{\circ}C$. pH was decreased with storage period, however it tended to increase after 30 days. The values of TBA and VBN for both sausages tended to increase with storage time, and this trend was pronounced at 5$^{\circ}C$ rather than at 10$^{\circ}C$. The purge loss was apparently increased in the VGS compared to the NGS sample as the storage period extended. According to the sensory evaluation, the marketing values of grill sausages stored at 5$^{\circ}C$ and 10$^{\circ}C$ were maintained until day 20 for NGS and day 14 for VGS, respectively.

In this study, we analyzed 24 pieces ($4{\times}3{\times}2$) of pork jerky produced using raw meat and we then randomly assigned the hind leg meat to a treatment group. In addition, the effects of storage according to aerobic plate counts and sensory evaluation on pork jerky were examined. Form the results, We found that volatile basic nitrogen (VBN) content and aerobic plate counts of pork jerky gradually increased with increased storage time, ; moreover, there was a significant difference between the treatment groups and the control group (p<0.01). However, the counts did not increase as significantly in the ginseng powder group (B), as compared to the ginger powder group (A) (p<0.05). The sensory evaluation of pork jerky peaked during the second week of storage in all treatment groups, after which these characteristics gradually decreased (p<0.05). In addition, these characteristics were generally higher in the control group, compared to the treatment groups. When comparing the ginseng powder addition group (B) and ginger powder-addition group (A), only color was significantly higher in the ginger powder group. Based on this study as well as previous studies, storage of pork jerky significantly affects the sensory evaluation of raw pork. Ginger powder may have a more beneficial effect than other treatments on the sensory evaluation. Additionally, in all treatment groups, sensory characteristics were significantly higher in the ginger treated groups than in those of the ginseng-treated groups.
Effects of spent composts of selenium-enriched mushroom (Se-SMC) on plasma glutathione peroxidase (GSH-Px) activity and selenium (Se) deposition in finishing Hanwoo (Bos taurus coreanae) steers were investigated. Twenty-five Hanwoo steers (average body weight = 613 kg, average age = 22 months) were allotted to treatments in five groups of five steers per pen for 12 weeks preceding slaughter. Treatments were SMC alone (CON; 0.1 ppm Se), 0.3 ppm (0.3 Se-SMC), 0.6 ppm (0.6 Se-SMC), 0.9 ppm (0.9 Se-SMC), and 0.9 ppm (sodium selenite; SENI) Se. During the experimental period, blood samples were taken to analyze Se concentrations and GSH-Px activities. Muscle and liver samples were collected for analyses of Se contents after slaughter. Dry matter intake and body weight gain were not affected by Se-SMC or sodium selenite supplementation. Selenium concentration in the whole blood and GSH-Px activity in plasma were linearly increased (p<0.01) with increasing levels of Se-SMC. The whole blood Se concentration of SENI treatment was significantly higher (p<0.05) than that of CON treatment from 4 weeks, whereas there was no significant difference in GSH-Px activities between both treatments at 8 and 12 weeks. Selenium content in the hind leg and liver increased linearly (p<0.05) with increasing levels of Se-SMC, but those of SENI treatments were not significantly different from CON treatments. These results suggested that Se in the Se-SMC was highly bioavailable to blood and tissues of ruminants, especially compared with Se in the sodium selenite. Therefore, Se-SMC might be used not only as an inexpensive way of providing Se for ruminants but also as another way of producing Se-fortified beef.
In 1964, Abbott and Colleagues published the world's first heterotopic heart transplantation technique in the rat. Their method established circulation by end-to-end anastomoses of the graft's aorta and pulmonary artery to the recipient's abdominal aorta and Inferior Vena Cava(IVC), respectively. In 1966, Tomita et al altered Abbott's technique by employing end-to-side rather than end-to-end anastomoses, thus eliminating the hind leg paralysis that sometimes resulted from Abbott's technique. In order to prevent postsuture hemorrhage (since 7-0 silk suture was the finest available at that time), Tomita's aortic anastomosis was done with double up-and-down continuous suture technique. A single layer continuous anstomosis effected the pulmonary artery-IVC anastomosis. The availability of Nylon monofilament suture made it possible for Ono and Lindsey to use a single layer suture technique for the aortic end-to-side anastomosis in their modified rat heart transplantation. We observed survival time between control group and Immunosuppression(Cyclosporine administration, 10mg/Kg${\times}$4 times postoperatively) group after heterotopic heart transplantation in the rat model. The cyclosporine adminstration group survived longer than the control group, thus we concluded that cyclosporine was based on Immunosuppressive drugs.
Color rings have been commonly and widely used in many ecological and ornithological studies in the field, but color ring-induced injury in passerines is poorly documented. In this report, we describe the first case of toe loss caused by a dislocated split Darvic ring in a Pale Thrush (Turdus pallidus) that was recaptured 596 days after its initial capture and release on Hong-do Island, the Republic of Korea. We also found that a left hind toe of a Light-vented Bulbul (Pycnonotus sinensis) was entrapped by a split color ring when the bird was observed 135 days after the marking at Heuksan-do Island. Based on these two cases out of the 1,900 color ringed birds, we emphasize a precautionary approach following general guidelines in the application and use of color leg rings for future studies.

This study was carried out to determine effects of the feeding length of spent mushroom composts from selenium-enriched mushroom (Se-SMC) on muscular selenium deposition in finishing Hanwoo steers. A total of 30 steers were allotted to an experiment with a 3 × 2 factorial arrangement. Treatments were 2 groups of with and without Se-SMC at three different feeding lengths (2, 4, and 6 months) by body weight (BW). And then dry matter intake, body weight gain, Se concentration and/or content in blood and/or muscle were investigated. Dry matter intake was not affected by feeding length and Se-SMC supplementation. As the feeding length was prolonged, initial and final BW was significantly lowered (p<0.01) with no Se-SMC effect. However, total BW gain was significantly increased (p<0.0001) with increasing feeding length with no effect of Se-SMC. Se-SMC supplementation significantly increased Se concentration in whole blood, but there was no effect on feeding length. Se contents in muscles, especially hind-leg, were significantly increased (p<0.05) as the feeding length was increased. However, there was no difference for Se content between four and six months feeding groups of Se-SMC and also hepatic Se content was not affected by feeding length. On the contrast, hepatic Se content for Se-SMC group within each feeding length was significantly increased (p<0.001) compared with no Se-SMC supplementation. In conclusion, our results suggested that optimum feeding length of Se-SMC as a strategy for the production of Se-fortified beef might be approximately 4 months.
Purpose: Although radiation-induced fibrosis is one of the common sequelae occurring after irradiation of skin and soft tissues, the treatment methods are not well standardized. This study aimed to establish the skin fibrosis mouse model by fractionated radiation for the further mechanism studies or testing the efficacy of therapeutic candidates. Materials and Methods: The right hind limbs of BALB/c mice received two fractions of 20 Gy using a therapeutic linear accelerator. Early skin damages were scored and tissue fibrosis was assessed by the measurement of a leg extension. Morphological changes were assessed by H&E staining and by Masson's Trichrome staining. TGF-${\beta}1$ expression from soft tissues was also detected by immunohistochemistry and PCR. Results: Two fractions of 20 Gy irradiation were demonstrated as being enough to induce early skin damage effects such as erythema, mild skin dryness, dry and wet desquamation within several weeks of radiation. After 13 weeks of irradiation, the average radiation-induced leg contraction was $11.1{\pm}6.2mm$. Morphologic changes in irradiated skin biopsies exhibited disorganized collagen and extracellular matrix fibers, as well as the accumulation of myofibroblasts compared to the non-irradiated skin. Moreover, TGF-${\beta}1$ expression in tissue was increased by radiation. Conclusion: These results show that two fractions of 20 Gy irradiation can induce skin fibrosis in BALB/c mice accompanied by other common characteristics of skin damages. This animal model can be a useful tool for studying skin fibrosis induced by radiation.
In the present study, to determine the effects of terminal sire strains, carcass characteristics, yields of retail cut, and meat quality of 425 pigs were examined. Terminal sires were divided into five strains (D1, D2, B1, B2, and BD) based on the countries of origin and breeds. The strains D1 and D2 showed high grade 1+ appearance ratios and strains B1, B2, and D1 showed high appearance ratios of carcass yield grade A. As for the real retail cut yields of meat, strain D1 showed large real retail cut yields of meat in shoulder, rib, fore leg, loin, belly, tenderloin and hind leg. Moisture contents were significantly higher in strain B1, and protein and fat contents were significantly higher for strain D1 than in the other strains (p<0.05). Shear force values were significantly higher for strain B1 than in the other strains (p<0.05). For meat color properties, the lightness were significantly higher in strains D1, D2, and BD, and the yellowness were significantly higher in strains D1, D2, and BD relative to the other strains (p<0.05). In sensory evaluation, drip of strains D1 and D2 were significantly lower while marbling and overall acceptability were significantly higher in the other four strains (p<0.05). Therefore, when considering the results of the present study comprehensively, using strains D1 and D2 that showed high grade 1+ appearance ratios, large real retail cut yields of belly, significantly higher lightness and sensory evaluation will be helpful to the incomes of producers and farm households.
