Cladosiphon okamuranus is edible brown algae cultured commercially and massively at Okinawa coast. Dietary fiber occupying most of C. okamuranus isn't digested and absorbed by digestive enzymes of human. But it is known to prevent and treat constipation as metabolic disease. This study was to investigate the effects of dietary fiber extracted from C. okamuranus on the damage of liver and the blood cholesterol level in the high fat diet-fed rats. The effects were measured by the levels of aspartate aminotransferase (ALT), alanine aminotransferase (AST), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and high density lipoprotein (HDL) in sera. The levels of ALT, AST, TC, TG and LDL were significantly reduced in the C. okamuranus-treated group compared with the high fat diet group. But HDL level was markedly increased. The results showed that the dietary fiber extracts from C. okamuranus have the inhibitive effects of cholesterol biosynthesis in the high fat diet-fed rats.

This study was performed to investigate the effects of a diet with a red yeast sweet potato supplement on fecal amount and lipid metabolism in male Sprague-Dawley rats fed a high-fat diet for 10 weeks. Rats were fed a high-fat diet (15% fat) with additional lard (7%) and cholesterol (1%) based on AIN-93G basal diet (7% fat) for 6 weeks during the first phase. In the second phase, which lasted 4 weeks, the rats divided into four experimental groups which were composed of a high-fat diet group as a control (CON), a high-fat diet with 5% white-fleshed sweet potato supplement group (WFSP), a high-fat diet with 5% red yeast sweet potato supplement group (RYSP), and a high-fat diet with 5% purple-fleshed sweet potato supplement group (PFSP). The fecal amount of group RYSP increased significantly during the second phase compared to the other groups (p<0.05). The fecal total cholesterol (TC) and triglyceride (TG) content of group RYSP were also highest among all experimental groups. The serum TC and TG were shown to have the lowest levels in the group RYSP, and LDL-cholesterol levels were significantly decreased in groups RYSP and PFSP than in group CON (p<0.05). These results indicate that supplementation with red yeast sweet potato seemed to be effective in increasing feces and fecal lipid excretion, and also in decreasing serum lipid levels in rats fed a high-fat diet.
1.Purpose This study was researched in order to investigate the effect of Hyungbangdojucksan-Gami and Kamsuchunilhwan on the obesity in rats. 2. Methods The rats were divided into 10 groups - The normal group fed with normal diet. One of the sample groups fed with high fat diet, and since fifth week had fed with Hyungbangdojucksan-Gami. The other sample groups fed with high fat diet, and since fifth week had fed with Kamsuchunilhwan. The control group fed with high fat diet, and since fifth week had fed sodium chloride. 3. Results 1. The rate of increasing weight was significantly decreased after oral administration of Hyungbangdojucksan-Gami and Kamsuchunilhwan. 2. The rate of feeding effect was not significantly decreased after oral administration of Hyungbangdojucksan-Gami and Kamsuchunilhwan. 3. After oral administration of Hyungbangdojucksan-Gami, the quantities of total cholesterol, HDL cholesterol, LDL cholesterol and triglyceride on the serum were significantly decreased but that of lipid peroxide on the serum was significantly increased. 4. After oral administration of Kamsuchunilhwan., the quantities of glucose, total cholesterol, LDL cholesterol and triglyceride on the serum were significantly decreased but those of HDL cholesterol, lipid peroxide on the serum were significantly increased. 4.Conclusion Hyungbangdojucksan-Gami and Kamsuchunilhwan have significant effects on the obesity in the rats.
This study was conducted to investigate the cholesterol lowering effect and weight reduction of autoclaved soy flour, cheonggulgang and doenjang, which are fermented using Bacillus subtilis DJI, in rats fed a high-fat/high-cholesterol diet. Male Sprague-Dawley rats weighing 250 g, were divided into five groups; a normal diet group (N), a high-fat/high-cholesterol diet group (HFC), a high-fat/high-cholesterol diet with 5% autoclaved soy flour group (HFC-S), a high-fat/high-cholesterol diet with 5% cheonggulgang group (HFC-CJ), and a high-fat/high-cholesterol diet with 5% deonjang group (HFC-DJ). The body weight gain and adipose tissues weights increased in rats fed a high-fat/high-cholesterol diet, but decreased significantly in rats fed autoclaved soy flour, cheonggulgang and doenjang, compared with the HFC group. Food intake was lower in the all HFC groups (HFC, HFC-S.HFC-CJ and HFC-DJ) than that of the N group, However, there was no differences in the food intake between HFC diet groups. Serum triglyceride level increased with a high-fat/high-cholesterol dietgroup whereas HFC-CJ and HFC-DJ groups were markedly decreased serum triglyceride levels compared with N group. Levels of serum total cholesterol and LDL-cholesterol, and level of total cholesterol in the liver were significantly lower in the HFC-S group, the HFC-CJ group and the HFC-DJ group than that of the HFC group, but exerted no significant change between the HFC groups on levels of total lipid and triglyceride in the liver. Fecal total lipid content increased in the HFC-CJ group and the HFC-DJ group than those of the HFC group and HFC-S group. The activities of heparin-releasable lipoprotein lipase (HR-LPL) and total-extractable LPL in adipose tissues decreased in the HFC-S group, the HFC-CJ group and the HFC-DJ group compared with the HFC group. These results suggest that dietary cheonggulgang and doenjang may exert cholesterol-lowering effect and prevent obesity.

This experimental study was designed to investigate the effects of Acanthopanax sessiliflorus $S_{EEM}$ leaves, stems, roots (ACL, ACS, ACR) on the change of weight and the serum total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, total lipid and phospholipid level in obese mice induced Dy high fat diet. Experimental groups were as follows ; Normal group was fed with normal diet and administered with distilled water during 7 weeks, Control group was fed with high fat diet and administered with distilled water during 7 weeks, Sample A group was fed with high fat diet and administered with ACL of 500 mg/kg/mouse/day during 7 weeks, Sample B group was fed with high fat diet and administered with ACS of 500 mg/kg/mouse/day during 7 weeks, Sample C group was fed with high fat diet and administered with ACR of 500 mg/kg/mouse/day during 7 weeks. The results were as follows ; 1. Sample A group, Sample B group and Sample C group were significantly decreased the serum total lipid level in comparison with Control group. 2. Sample A group was decreased the body weight (4 weeks, 7 weeks), the serum total cholesterol level, the serum LDL-cholesterol level, the serum triglyceride level, the serum free fatty acid and phospholipid level but increased the serum HDL-cholesterol level in comparison with Control group. 3. Sample B group was increased the serum HDL-cholesterol level but decreased the body weight (4weeks), the serum total cholesterol level, the serum free fatty acid and triglyceride level in comparison with Control group. 4. Sample B group was significantly decreased the body weight (7 weeks), the serum LDL-cholesterol and phospholipid level in comparison with Control group. 5. Sample C group was increased the serum HDL-cholesterol level but decreased the body weight (4 weeks) and the serum free fatty acid level in comparison with Control group. 6. Sample C group was significantly decreased the body weight (7 weeks), the serum total cholesterol level, the serum LDL-cholesterol and triglyceride level in comparison with Control group. According to above results, I suggest ACR is able to be used for the obesity.
We found previously that dietary high fat caused obesity, and levan supplementation to the regular diet reduced adiposity and serum lipids. In the present study, we examined the effects of levan [high-molecular-mass $\beta$-(2,6)-linked fructose polymer] supplement on the development of obesity and lipid metabolism in rats fed with high-fat diet. Thus, to determine whether the dietary levan may have the anti-obesity and hypolipidemic effects, 4-wk-old Sprague Dawley male rats were fed with high-fat diet for 6 wk to induce obesity, and subsequently fed with 0, 1, 5, or 10% levan supplemented high-fat diets (w/w) for another 4 wk. For the comparison, a normal control group was fed with AIN-76A diet. Supplementation with levan resulted in a significant reduction of high-fat-induced body weight gain, white fat (i.e., epididymal, visceral, and peritoneal fat) development, adipocyte hypertrophy, and the development of hyperinsulinemia and hyperlipidemia in a dose-dependent manner. Serum triglyceride and free fatty acid levels were greatly reduced by levan supplementation. Serum total cholesterol level was reduced, whereas the HDL cholesterol level was increased by dietary levan. The expression of uncoupling protein (UCP) was increased by dietary high fat, and was further induced by levan supplementation. The mRNA level of UCP1, 2, and 3 in brown adipose tissue (BAT) and UCP3 in skeletal muscle was upregulated in rats fed with dietary levan. In conclusion, upregulated UCP mRNA expression may contribute to suppression of development of obesity through increased energy expenditure. The present results suggest that levan supplementation to the diet is beneficial in suppressing diet-induced obesity and hyperlipidemia.
This study was attempted to investigate the effects of Duchesnea chrysantha (DC) on antioxidative activities by in vivo. Rats were divided into four experimental groups which are composed of normal diet group (N group), high fat high cholesterol diet group (HF group), high fat high cholesterol diet with 5% DC powder supplemented group (DA group) and high fat high cholesterol diet with 10% DC powder supplemented group (DB group). Supplementation of DC powder groups resulted in increased activities of hepatic glutathione peroxidase and catalase. The microsomal superoxide radical contents of the DA and DB groups were significantly reduced compared to the high fat high cholesterol diet group. The mitochondrial superoxide radical contents of the DB group were significantly reduced compared to the high fat high cholesterol diet group. Hepatic hydrogen peroxide contents in cytosol were significantly reduced 5% and 10% DC powder supplemented group. The carbonyl values contents in mitochondria and microsome of the DA and DB groups were significantly reduced compared to the HF group. Thiobarbituric acid reaction substance (TBARS) values in liver were reduced in 10% DC powder supplemented group compared to the HF group. These results suggest that DC powder may have a strong regulatory effect in the activation of the antioxidative defense system.

This study was conducted to investigate the cholesterol lowering and anti-obesity effects of an ethanol extract of broccoli sprouts (BS) in rats fed high fat diet. Male Sprague-Dawley rats weighing $150{\sim}155g$, were divided into 6 groups; a normal diet group (ND), a high fat diet group (HFD), a normal diet and BS with 200 mg/kg treated group (ND-BSL), a normal diet and BS with 400 mg/kg treated group (ND-BSH), a high fat diet and BS with 200 mg/kg treated group (HFD-BSL), and a high fat diet and BS with 400 mg/kg treated group (HFD-BSH). The body weight gain and mesenteric adipose tissue weight were increased by high fat diet, but gradually decreased to the corresponding level of ND group after administration of BS extract. The liver and epididymal adipose tissue weights of HFD group were the highest among the six groups, although the difference was not significant. Food intake was lower in high fat diet groups compared with normal diet groups. The serum ALT and AST activities that were elevated by the high fat diet were significantly decreased after BS administration. Levels of serum total cholesterol, LDL-cholesterol, triglyceride and the atherogenic index tended to be decrease in the BS administered groups compared with HFD group. However, HDL-cholesterol level in serum decreased in HFD group and markedly increased in BS administered groups. There were no differences in the contents of serum triglyceride, LDL-cholesterol and HDL-cholesterol between normal diet groups. Levels of total cholesterol and triglyceride in liver and adipose tissues were also lower in BS administrated groups than in HFD group. The activities of heparin-releasable lipoprotein lipase (HR-LPL) and total-extractable LPL (TE-LPL) in adipose tissue were increased in HFD group compared with the BS administered groups, but those of the ND-BSL group and ND-BSH group were similar to ND group. These results suggest that BS ethanol extract may exert cholesterol-lowering effect and potentially reduce lipid storage.
Obesity is an epidemic disease characterized by an increased inflammatory state and chronic oxidative stress with high levels of pro-inflammatory cytokines and lipid peroxidation. Moreover, obesity alters cholesterol metabolism with increases in low-density lipoprotein (LDL) cholesterols and triglycerides and decreases in high-density lipoprotein (HDL) cholesterols. It has been shown that mulberry leaf and fruit ameliorated hyperglycemic and hyperlipidemic conditions in obese and diabetic subjects. We hypothesized that supplementation with mulberry leaf combined with mulberry fruit (MLFE) ameliorate cholesterol transfer proteins accompanied by reduction of oxidative stress in the high fat diet induced obesity. Mice were fed control diet (CON) or high fat diet (HF) for 9 weeks. After obesity was induced, the mice were administered either the HF or the HF with combination of equal amount of mulberry leaf and fruit extract (MLFE) at 500mg/kg/day by gavage for 12 weeks. MLFE treatment ameliorated HF induced oxidative stress demonstrated by 4-hydroxynonenal (4-HNE) and modulated the expression of 2 key proteins involved in cholesterol transfer such as scavenger receptor class B type 1 (SR-B1) and ATP-binding cassette transporter A1 (ABCA1) in the HF treated animals. This effect was mainly noted in liver tissue rather than in cutaneous tissue. Collectively, this study demonstrated that MLFE treatment has beneficial effects on the modulation of high fat diet-induced oxidative stress and on the regulation of cholesterol transporters. These results suggest that MLFE might be a beneficial substance for conventional therapies to treat obesity and its complications.

The effects of Pleurotus (P.) eryngii on the weight gains, food efficiency ratios, serum and hepatic lipid concentrations were investigated in male rats fed the high fat diets. Twenty one week-old Sprague-Dawley rats were given three different types of diet for 10 weeks, respectively: a control diet (high fat; $20\%$ lard), two kinds of P. eryngii diet supplemented with $3\%\;or\;5\%$ of dry P. eryngii powder in substitutes for the same amounts of cellulose in the control diets. The body weight gains and food efficiency ratios of the rats fed $3\%\;and\;5\%$ P. eryngii diets were significantly lower than those of the rats fed the control diet. The concentrations of hepatic total lipid and triglyceride in the rats fed the P. eryngii diets were significantly lower than those in the rats fed the control diet. But the hepatic cholesterol contents of the rats fed the P. eryngii diets were similar to those of the rats fed the control diet. The concentrations in serum total cholesterol, LDL-cholesterol, and atherogenic index ratios were significantly lower in the rats fed the P. eryngii diets compared to those fed the control diet. The HDL-cholesterol/total- cholesterol ratio was significantly higher in the rats fed the P. eryngii diets compared to those fed the control diet. There were no significant difference found in the serum triglyceride, phospholipid and HDL-cholesterol concentrations among the experimental groups. These results showed that feeding of P. eryngii powder feeding decreased the total cholesterol, LDL-cholesterol and atherogenic index and increased the HDL-cholesterol/total-cholesterol ratio in serum of the rats fed high fat diet.
