Effects of Dendropanax morbifera Leaf Extracts on Lipid Profiles in Mice Fed a High-Fat and High-Cholesterol Diet (황칠나무 잎 추출물이 고지방·고콜레스테롤 식이를 급여한 마우스의 지질 개선 효과에 미치는 영향)

  • The purpose of this study was to determine the effects of dried Dendropanax morbifera leaf extracts on lipid profiles of mice fed a high-fat and -cholesterol diet (HFCD). ICR mice were divided into six groups based on mice fed AIN-93G diet (Normal), HFCD (Control), HFCD+100 mg/kg/d of D. morbifera leaf aqueous extract (DA-100), HFCD+200 mg/kg/d of D. morbifera leaf aqueous extract (DA-200), HFCD+100 mg/kg/d of D. morbifera leaf ethanol extract (DE-100), or HFCD+200 mg/kg/d of D. morbifera leaf ethanol extract (DE-200) for 7 weeks. The final body weights of mice fed D. morbifera extracts were all lower than those of the control group. Mice treated with D. morbifera extracts showed significantly reduced plasma and hepatic triglyceride (TG), total cholesterol (TC), and low-density lipoprotein cholesterol levels, along with increased plasma high-density lipoprotein cholesterol level. Fecal TG level was higher in DE-100 and DE-200 groups and TC level was significantly higher in the DA-200 and DE-200 groups. Relative liver weight, spleen weight, and testicle fat weight in mice treated with D. morbifera were reduced compared to the control group. Plasma insulin, aspartate transaminase, and alanine transaminase levels of experimental groups were also lower than those of the control group. All mice treated with D. morbifera extracts had lower malondialdehyde (MDA) content and higher superoxide dismutase (SOD) activity than the control group. Particularly MDA levels of the DA-200 and DE-200 groups and SOD levels of the DE-200 group were identical to levels of the normal group. These results suggest that D. morbifera extracts have lipid improvement effects in mice fed a HFCD.

  • This study investigated the effects of fermented black raspberry (BR) and red ginseng (RG) extract co-treatment on lipid metabolism and obesity in rats fed with a high fat/high cholesterol diet (HFHCD) for 12 weeks. Compared to the corresponding values in rats fed with a HFHCD, total cholesterol and low-density lipoprotein (LDL)-cholesterol and triglyceride levels decreased whereas high-density lipoprotein (HDL)-cholesterol levels increased in rats treated with fermented BR and RG extracts. These extracts significantly increased the expression of HMG-CoA reductase, LDL receptor, and sterol regulatory-element-binding protein-2 (SREBP-2) mRNA, but decreased the mRNA expression of SREBP-1. Additionally the serum levels of leptin and fatty acid synthase were decreased. Moreover, supplementation with fermented BR and RG effectively increased fecal cholesterol excretion. These results suggest that fermented BR and RG extracts might be effective at preventing hypercholesterolemia and obesity.

Effect of Naringin on Major Biochemical Parameters in Sera of Rats Fed High Fat and Cholesterol Diet (나린진이 고지방 및 고콜레스테롤 식이 랫드의 혈청 성분 농도의 변화에 미치는 영향)

  • This study was performed to investigate the changes of the serological lipid-related parameters of the rats when they were fed with the high fat diets supplemented with or without naringin for five weeks. Twenty-four Sprague-Dawley male rats($272.2{\pm}7.2$ g of body weight) were randomly divided into three groups(eight rats per each group) : control(C) group and two treatment groups. Rats in the C group were fed with the high-fat diet containing 15% lard, 1% cholesterol and 0.5% sodium cholate(w/w) which was modified from the formula of the American Institute of Nutrition-76(AIN-76) diet. Rats in treatment groups were fed with above diet supplemented with 0.1% naringin(N-0.1) or 0.2% naringin(N-0.2) on the weight to weight basis, respectively. The supplementation of naringin did not induce any significant difference on the final body weight, gain of body weight, the amount of feed intake and the feed efficiency of rats in between control and treatment groups. In addition the levels of glucose, total protein, albumin, globulin and albumin/globulin(A/G) ratio in sera of rats showed no significant differences between control and treatment groups. The levels of total cholesterol(TC) and low density lipoprotein-cholesterol(LDL-C)in sera of rats in both N-0.1 and N-0.2 groups were significantly lower than in C group(p<0.05). The levels of high density lipoprotein-cholesterol(HDL-C) were significantly higher in both N-0.1 and N-0.2 groups than in C group(p<0.05). The values of atherogenic index(AI) were significantly lower in both N-0.1 and N-0.2 groups than in C group(p<0.05). The levels of triglyceride in sera of rats showed no significant differences between control and treatment groups. The values of AST and ALT were significantly lower in both N-0.1 and N-0.2 groups than in C group(p<0.05). Therefore the supplementation of naringin to high fat diet in rats reduced effectively the serum lipid levels such as TC and LDL-C and AI which were regarded as to cause the cardiovascular diseases, and moreover it elevated the HDL-C value effectively which was regarded to protect cardiovascular diseases.

  • For a probiotic yoghurt it is desirable to utilize lactic acid bacteria with a high survival rate and beneficial function to human beings. We have examined a variety of lactic acid bacteria to assess the acid and bile tolerance and antibiotic resistance. In addition, an in vitro culture experiment was performed to evaluate their ability to reduce cholesterol levels in the growth medium. Thirteen strains were selected from in vitro cholesterol assays and fed to Sprague-Dawley rats with a high-cholesterol diet. Among the 13 strains tested, 8 strains were shown to reduce serum cholesterol levels significantly after 24 days of administration in vivo. Rats were fed lyophilized yoghurt powder fermented with a combination of 3 selected strains: Bifidobacterium infantis AM-220, Lactobacillus AM-245, and Styeptococcus MA-1. The levels of total cholesterol and low density lipoprotein were significantly lower (p$\leq$0.05) in rats fed the yoghurt powder compared with control group. These studies suggest that yoghurt fermented with appropriately selected lactic acid bacteria may have a anticholesterolemic effect.

Prevalence and risk factors of gallbladder polyps among adults on Jeju Island according to genders

  • Gallbladder polyps (GBPs) may be a cause of gallbladder cancer. The known risk factors for GBPs are male gender, obesity, metabolic syndrome, and dyslipidemia. Especially, male gender has been known that it strongly affects on the prevalence of GBPs. Therefore, this study aimed to investigate risk factors affecting the prevalence of GBPs among adults on Jeju Island according to genders. We examined 5,574 subjects who visited a single health-screening center at Jeju National University Hospital between January 2015 and December 2019. Univariate and multiple logistic regression analysis were performed to identify risk factors affecting the prevalence of GBPs. The prevalence of GBPs were 8.9% in male subjects and 8.1% in female subjects, respectively. Multivariate analysis revealed that ≥60 year age (odds ratio [OR] 0.659; P=0.027) and aspartate aminotransferase >32 IU/L (OR 0.658; P=0.009) in male subjects and high-density lipoprotein-cholesterol ≥60 mg/dL (OR 0.514; P=0.013) in female subjects were independent factors affecting the prevalence of GBPs. The prevalence of GBPs in men (8.9%) is comparable to that in women (8.1%) on Jeju Island. Age <60 years and the higher level of aspartate aminotransferase in men and the normal or lower high-density lipoprotein-cholesterol in women were independent risk factors of GBPs on Jeju Island.

Relationship between Albuminuria and Uric Acid to High-Density Lipoprotein Cholesterol Ratio in Korean Adults (대한민국 성인에서 알부민뇨와 요산 대 고밀도 지단백 콜레스테롤 비율의 관련성)

  • This study assesses the urine microalbumin to urine creatinine ratio (uACR) with uric acid and the association with the high-density lipoprotein cholesterol (UA/HDL-C) ratio in Korean adults. Data from the 2019 Korean National Health and Nutrition Examination Survey was procured and included 5,845 adults aged 20 years or older. Several key findings were obtained in the present study. After adjusting for the related variables (age, gender, hypertension, obesity, type 2 diabetes mellitus, and chronic kidney disease), the uACR was found to be positively associated with the quartiles of the UA/HDL-C ratio (P=0.002). Moreover, after adjusting for related variables, the odds ratio (OR) of albuminuria (uACR≥30.0 mg/g) was not significant in quartile 2 (Q2) (OR, 1.015; 95% confidence interval [CI], 0.757~1.361) and Q3 (OR, 1.090; 95% CI, 0.811~1.465) but was significantly higher in Q4 (OR, 1.416; 95% CI, 1.044~1.921) as compared to Q1 of the UA/HDL-C ratio. We conclude that since the UA/HDL-C ratio is positively associated with albuminuria in Korean adults, an increased UA/HDL-C ratio could be applied as a useful measure to assess albuminuria in Korean adults.

Impact of Obesity on Cardiometabolic Disease Risk Factor Clusters among Korean Young Adults (젊은 성인에서 비만이 심혈관대사질환 위험인자 군집에 미치는 영향)

  • Background: This study investigated the association between obesity and cardiometabolic disease clusters in Korean adults aged 19-39 years. Methods: We included 3,785 participants (1,767 men and 2,018 women) aged 19-39 years who participated in the Korea National Health and Nutrition Examination Survey (2020-2022). The risk factors for cardiometabolic disease were defined as follows: high blood pressure; hypertriglyceridemia; hyperuricemia; and high low-density lipoprotein cholesterol, low high-density lipoprotein cholesterol, high fasting plasma glucose, elevated alanine aminotransferase levels. Results: The prevalence of participants with ≥ 1, ≥ 2, and ≥ 3 cardiometabolic disease risk factors was 95.2%, 84.2%, and 65.6% in men and 90.0%, 70.5%, and 43.0%, in women with class 2-3 obesity, respectively. Compared to those for young adults with underweight/normal weight, the odds ratios (ORs) and 95% confidence intervals (CIs) for the clustering of cardiometabolic risk factors were 17.26 (8.32-35.80) for ≥ 1; 19.43 (12.18-31.00) for ≥ 2; and 22.93 (14.15-37.15) for ≥ 3 factors in men with class 2-3 obesity and 14.67 (8.00-26.91) for ≥ 1; 20.88 (12.15-35.88) for ≥ 2; and 36.26 (20.20-65.09) for ≥ 3 factors in women with class 2-3 obesity. Conclusion: Although the patients were young, the prevalence and ORs for cardiometabolic disease risk factor clusters in young adults with obesity were prominently high. Prevention and management of obesity in young Korean adults are urgently needed at both individual and public levels.

Correlations among Anthropometric Measurements, Serum Lipid Levels and Nutrient Intake in Female University Students

  • The purpose of this study was to investigate the correlations among the anthropometry, serum lipid levels and nutrient intake in Korean female university students. The subjects were 119 female students at a university located in Incheon. This study was conducted using a self-administered questionnaire. Anthropometric data were measured and blood lipid levels were analyzed. Nutrient intake collected from 3 day-recalls was analyzed by the Computer Aided Nutritional Analysis Program. The data were analyzed by SPSS 10.0 program. Average age, height and weight of the subjects were 20.9 years, 160.1cm and 54.3kg, respectively. Average serum TG (triglyceride), total cholesterol, HDL-C (high density lipoprotein-cholesterol) and LDL-C (low density lipoprotein-cholesterol) levels of the subjects were 69.47mg/dl, 146.85 mg/dl, 50.49mg/dl and 82.52mg/dl, respectively. Average AI (atherogenic index) of the subjects was 2.03, which was in the normal range based on risk values. Average intake of most nutrients except protein, vitamin B$_1$, vitamin C and phosphorus were lower than the Korean RDA. Especially calcium and iron intakes of the subjects were under 65% of the Korean RDA. Serum TG, total cholesterol and LDL-C levels were negatively correlated with DBP (diastolic blood pressure). HDL-C/LDL-C and HDL-C/total cholesterol were positively correlated with height. Age was positively correlated with phosphorus intake. DBP of the subjects was positively correlated with calcium and iron intakes. Serum TG level was positively correlated with total cholesterol, HDL-C, LDL-C and AI, while negatively correlated with HDL-C/total cholesterol. Total cholesterol level was positively correlated with HDL-C, LDL-C and AI, while negatively correlated with HDL-C/LDL-C, HDL-C/total cholesterol. HDL-C level was positively correlated with LDL-C, HDL-C/LDL-C and HDL-C/total cholesterol, while negatively correlated with AI. LDL-C level was negatively correlated with HDL-C/LDL-C and HDL-C/total cholesterol, while positively correlated with AI HDL-C/LDL-C ratio was positively correlated with HDL-C/total cholesterol and AI. HDL-C/total cholesterol was negatively correlated with AI. Fat intake was positively correlated with total cholesterol, HDL-C level, and vitamin B$_2$ intake was positively correlated with TG, HDL-C/LDL-C. Therefore, nutrition education is necessary for female university students to promote the lipid profile and to optimize the nutritional status. (J Community Nutrition 4(3) : 151∼158, 2002)

Anti-cholesterol Effects and Molecular Mechanism Study of Mixture of Atractylodes Macrocephala and Amomum Villosum Extracts (백출과 양춘사 추출 혼합물의 항콜레스테롤 효과 및 기전 연구)

  • Atractylodes macrocephala (AM) and Amomum villosum (AV) are the most common herbs in Korean Medicine to treat digestive diseases. In this study, we investigated the cholesterol lowering effects of mixtures of AM and AV extracts on high cholesterol diet (HCD) induced dyslipidemia mouse model. We classified animals into six different groups; Group 1: Normal diet, Group 2: HCD, Group 3: AV extracts : AM extracts (1:1) (200 mg/kg) + HCD, Group 4: AV extracts : AM extracts (1:2) (200 mg/kg) + HCD, Group 5: AV extracts : AM extracts (1:3) (200 mg/kg) + HCD, Group 6: Simvastatin 40 mg/kg + HCD. After 4 weeks of oral administration of respective drugs, we checked body, liver and epididymal fatweights along with liver and serum triacylglyceride (TG) concentration, total and low density lipoprotein (LDL) cholesterol in serum. Moreover, 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase (HMGCR), LDL receptor (LDLR), and sterol regulatory element-binding protein 2 (SREBP2) were detected by RT PCR or western blot analysis. The overall results showed that mixtures of AM and AV extracts inhibited HCD-induced increases of total cholesterol and LDL cholesterol in serum. Those effects seem to be caused by AM and AV extracts through inhibition of HMGCR expression. And thus blood cholesterol is induced into the liver by increasing LDLR expression, which is regulated by SREBP2 transcrption factor. The cholesterol lowering effects and mechanism of mixtures of AM and AV extracts was similar to the statin. We have identified the potential mixtures of AM and AV extracts as a new treatment for dyslipidemia.

Corn silk extract improves cholesterol metabolism in C57BL/6J mouse fed high-fat diets

  • BACKGROUNG/OBJECTIVES: Corn silk (CS) extract contains large amounts of maysin, which is a major flavonoid in CS. However, studies regarding the effect of CS extract on cholesterol metabolism is limited. Therefore, the purpose of this study was to determine the effect of CS extract on cholesterol metabolism in C57BL/6J mouse fed high-fat diets. MATERIALS/METHODS: Normal-fat group fed 7% fat diet, high-fat (HF) group fed 25% fat diet, and high-fat with corn silk (HFCS) group were orally administered CS extract (100 mg/kg body weight) daily. Serum and hepatic levels of total lipids, triglycerides, and total cholesterol as well as serum free fatty acid, glucose, and insulin levels were determined. The mRNA expression levels of acyl-CoA: cholesterol acyltransferase (ACAT), cholesterol 7-alpha hydroxylase (CYP7A1), farnesoid X receptor (FXR), lecithin cholesterol acyltransferase (LCAT), low-density lipoprotein receptor, 3-hyroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA reductase), adiponectin, leptin, and tumor necrosis factor ${\alpha}$ were determined. RESULTS: Oral administration of CS extract with HF improved serum glucose and insulin levels as well as attenuated HF-induced fatty liver. CS extracts significantly elevated mRNA expression levels of adipocytokines and reduced mRNA expression levels of HMG-CoA reductase, ACAT, and FXR. The mRNA expression levels of CYP7A1 and LCAT between the HF group and HFCS group were not statistically different. CONCLUSIONS: CS extract supplementation with a high-fat diet improves levels of adipocytokine secretion and glucose homeostasis. CS extract is also effective in decreasing the regulatory pool of hepatic cholesterol, in line with decreased blood and hepatic levels of cholesterol though modulation of mRNA expression levels of HMG-CoA reductase, ACAT, and FXR.
