Ginseng-plus-Bai-Hu-Tang ameliorates diet-induced obesity, hepatic steatosis, and insulin resistance in mice

  • Background: Dietary fat has been suggested to be the cause of various health issues. Obesity, hypertension, cardiovascular disease, diabetes, dyslipidemia, and kidney disease are known to be associated with a high-fat diet (HFD). Obesity and associated conditions, such as type 2 diabetes mellitus and nonalcoholic fatty liver disease (NAFLD), are currently a worldwide health problem. Few prospective pharmaceutical therapies that directly target NAFLD are available at present. A Traditional Chinese Medicine, ginseng-plus-Bai-Hu-Tang (GBHT), is widely used by diabetic patients to control glucose level or thirst. However, whether it has therapeutic effects on fat-induced hepatic steatosis and metabolic syndrome remains unclear. Methods: This study was conducted to examine the therapeutic effect of GBHT on fat-induced obesity, hepatic steatosis, and insulin resistance in mice. Results: GBHT protected mice against HFD-induced body weight gain, hyperlipidemia, and hyperglycemia compared with mice that were not treated. GBHT inhibited the expansion of adipose tissue and adipocyte hypertrophy. No ectopic fat deposition was found in the livers of HFD mice treated with GBHT. In addition, glucose intolerance and insulin sensitivity in HFD mice was also improved by GBHT. Conclusion: GBHT prevents changes in lipid and carbohydrate metabolism in a HFD mouse model. Our findings provide evidence for the traditional use of GBHT as therapy for the management of metabolic syndrome.

장기간의 지구성 운동과 단삼식초 섭취가 고지방식 유도 비만 흰 쥐의 신체구성과 인슐린 저항성에 미치는 영향 (Effects of long-term endurance exercise and Salvia miltiorrhiza vinegar on body composition and insulin resistance in high fat diet-induced obese rats)

  • 본 연구는 단삼식초 및 지구성 운동트레이닝이 고지방식 유도 비만 흰쥐의 신체구성 및 혈중 항목들의 농도 변화에 미치는 영향을 비교분석함으로서 대사성 증후군의 개선에 도움을 줄 수 있는 효과적인 방법을 모색하고자 실시되었다. 10주령 수컷 Wistar 흰쥐 40마리를 대상으로 8주간 고지방식 처치로 비만을 유도한 후 좌업군, 운동군, 단삼식초섭취군, 운동군+단삼식초섭취 복합처치군 등으로 무선, 배정하여 총 8주간 운동 및 식이처치를 실시하였으며, 체중 및 신체구성, 식이섭취량, 경구당 부하검사 및 혈중 콜레스테롤 농도 등을 측정하였다. 8주간의 단삼식초섭취는 지구성 운동과 병행할 경우 체중감소 및 인슐린 저항성의 개선효과를 확인할 수 있었다. 그러나 단삼식초섭취만 처치할 경우 오히려 체지방량을 증가시키고 제지방량은 감소시켰으며, 인슐린 저항성을 현저하게 개선시키지 않는 것으로 나타났다. 또한 지구성 운동과의 병행 시도의 경우에도 운동효과를 배제한 부가적인 효과가 나타나지 않았으며, 혈중 지질에 있어서는 운동의 효과마저 둔화시키는 것으로 나타났다. 따라서 단삼식초섭취에 따른 항고혈당, 항비만 효과는 확인될 수 없었으며, 추후 분자생물학적 분석에 의한 기전과 관련된 원인분석을 통해서 보다 명확한 결과를 제시할 수 있을 것으로 생각된다.

단식(斷食), 재급식(再給食)과 인슈린주사(注射)에 따른 쥐의 간세포핵단백질(肝細胞核蛋白質)에 대한 전기영동상의 분포양상 비교 (Comparison of SDS Gel Electrophoretic Patterns of Rat Liver Nuclear Proteins in Response to Starvation, Refeeding and Insulin Injection)

  • 쥐를 48시간 굶긴 뒤에 위에 탄수화물 농도가 높은 먹이를 주거나 streptozotocin을 이용하여 당뇨병을 가지게 만든 쥐에 인슈린 주사를 주었을 때 쥐 간계포의 핵단백질중 가장 주요한 0.14M NaCl에 녹는 핵단백질, histones 그리고 페놀에 녹는 핵단백질의 분포 변화를 조사하고자 SDS gel 전기영동법을 이용하였다. 각 핵단백질 분획을 전기영동법으로 분리시킨 단백질의 상대량을 비교하였을 때 0.14M NaCl 추출물은 현저한 변화를 나타내었으나 histones과 페놀로 추출된 핵단백질 분획들은 분리된 단백질 상대량에 큰 변화가 없었다. 48시간 굶긴 쥐와 정상적으로 먹이를 준 쥐의 0.14M MaCl 추출 핵단백질 분획을 비교하였을때 분자량이 50,000 과 180,000 daltons 사이에 있는 적어도 5개의 핵단백질의 농도가 다른 핵단백질에 비교하여 크게 감소되었다. 반면 분자량이 36,000 daltons 단백질의 농도는 48시간 굶긴 쥐에게 다시 탄수화물 농도가 높은 먹이를 주었을 때 24시간 동안에 정상적으로 먹이를 준 쥐에서 관찰한 핵단백질 분포 양상과 비슷한 결과를 얻었다. 당뇨병을 가진 쥐에게 인슈린 주사를 준 쥐와 인슈린주사를 주지 않은 당뇨병 쥐의 0.14M NaCl추출 핵단백질 분획을 비교 조사한 결과는 굶은 쥐에게 탄수화물 농도가 높은 먹이를 준 다음에 얻은 뒤의 결과와 정상적으로 유사하였다. 여기서 얻은 실험결과들은 0.14M NaCl 추출 핵단백질 중에 있는 어떤 핵단백질의 분포 변화가 이미 알려진 인슈린 신호에 따라 조정되고 cytosol에 있는 지방합성에 관하는 효소(lipogenic enzymes)들의 유도에 관련된 세포핵 활성 조절에 영향을 끼치고 있음을 시사해 준다.

식이단백질조성이 흰쥐의 성장과 지방대사에 미치는 영향 (Effects of Dietary Protein on Growth and Lipid Metabolism in Growing Rats)

  • The effect of dietary protein on growth and lipid levels of plasma and liver was studied in weanling male rats fed diets differing protein sources and amino acid balance. Rats were devided into 9 experimental diets which were grouped into 3 categories ; 1) Simple protein category includes gluten-, soy protein isolate-, and casein-containing diet groups, 2) Supplemented category includes casein supplemented with methionine, soy protein isolate supplemented with methionine, and gluten supplemented with lysine and methionine, 3) Mixed protein category includes diet groups containing gluten (2/3), casein (1/3), soy protein isolate (2/3) and casein (1/3), and casein (1/3), soy protein isolate (1/3) and gluten (1/3). The experimental diets composed of 15% protein, 65.8% carbohydrate, 10% fat and 1% cholesterol. The body wt. gain and P.E.R. were greater in rats of supplemented and mixed protein groups than simple protein groups. No statistical differences were found in plasma cholesterol among gluten, soy protein isolate and casein groups. Consumption of diets supplemented with limiting amino acid to gluten or soy protein isolate reduced the plasma cholesterol level by 23.2% and 34.2% respectively. However there was no difference between casein and the supplemented casein groups. The mixed protein groups shows relatively high plasma cholesterol concentration and low liver cholesterol levels. On the other hand gluten group showed low plasma cholesterol and high liver cholesterol levels, which means body cholesterol pool may not have been changed by the dietary protein. Feeding soy protein meal and the supplemented soy protein isolate resulted in lower plasma cholesterol, plasma triglycerides, liver cholesterol and liver triglycerides levels. This hypolipidemic effect is considered to see unique to soy protein isolate. Rats in gluten and the supplemented gluten groups showed lower plasma protein levels and a tendency of fatty liver.

Effect of mild-intensity exercise training with capsiate intake on fat deposition and substrate utilization during exercise in diet-induced obese mice

  • [Purpose] While the anti-obesity effects of exercise and capsiate are well-observed individually, the effect of exercise with capsiate intake has not been systematically explored yet. Therefore, the purpose of this study is to investigate whether the anti-obesity effects of exercise training can be further enhanced by capsiate intake. [Methods] 8-week-old male mice were divided into 3 groups (n = 8 per group): sedentary group (SED; nontrained), exercise-trained group (EXE) and exercise-trained group with 10 mg/kg of capsiate intake (EXE+CAP). All mice were offered high-fat diet and water ad libitum. The mild-intensity treadmill training was conducted 5 times a week for 8 weeks. After 8 weeks, metabolism during exercise and abdominal fat weight were measured. [Results] Body weight and the rate of total abdominal fat were significantly less in EXE+CAP than in SED but not between EXE and SED. The average of respiratory exchange rate during exercise was significantly much lower in EXE+SED (p = 0.003) compared to the difference between EXE and SED (p = 0.025). Likewise, the fat oxidation during exercise was significantly much higher in EXE+SED (p = 0.016) compared to the difference between EXE and SED (p = 0.045). Then, the carbohydrate oxidation during exercise was significantly much lower in EXE+SED (p = 0.003) compared to the difference between EXE and SED (p = 0.028). [Conclusion] In conclusion, the anti-obesity functions of exercise training can be further enhanced by capsiate intake by increasing fat oxidation during exercise. Therefore, we suggest that capsiate could be a candidate supplement which can additively ameliorate obesity when combined with exercise.

상황버섯 열수추출물의 항산화 활성과 식후 혈당 상승 억제 효과 (Antioxidant and Anti-hyperglycemic Effects of a Sanghwang Mushroom(Phellinus linteusau) Water Extract)

  • The inhibitory activities of a water extract of Sanghwang mushroom(Phellinus linteusau)(SWE) against ${\alpha}$-glucosidases were evaluated in this study. Inhibiting these enzymes involved in the absorption of disaccharides significantly decreases the postprandial increase in blood glucose level after a mixed carbohydrate diet. Oxygen radical absorbance capacity and 1,1-diphenyl-2-picryl hydrazyl scavenging activities of the SWE were evaluated to investigate the antioxidant activity of the SWE associated with complications of long-term diabetes. Furthermore, the postprandial blood glucose lowering effect of SWE was compared to a known type 2 diabetes drug($Acarbose^{(R)}$) in a Sprague-Dawley rat model. SWE significantly reduced the blood glucose increase after sucrose loading. These results suggest that SWE, which has high ${\alpha}$-glucosidase inhibitory activity and high antioxidant activities, has the potential to contribute to a useful dietary strategy for controlling postprandial hyperglycemia.

The Quality of a Traditional Dietary Pattern in Relation to Metabolic Syndrome in Elderly South Koreans

  • Background: The most beneficial dietary pattern in managing metabolic syndrome (MetS) in the elderly has not been ascertained. The aim of this study is to classify dietary patterns and to examine associations between dietary pattern, MetS and body composition in elderly Koreans. Methods: This study was conducted among Koreans 65 years or older using data from the Korea National Health and Nutrition Examination Survey in 2009. A total of 1,567 study subjects were included. All statistical analyses were conducted using SPSS version 20.0 and dietary patterns were classified by cluster analysis. Results: There were three dietary patterns derived by cluster analysis in this study. We observed that most South Korean elderly still maintain a traditional dietary pattern. Dietary patterns were classified as balanced (31%), imbalanced (40%), or very imbalanced (30%), with the majority of subjects having an unbalanced diet pattern in which their total energy and nutrient intake was insufficient compared with the Dietary Reference Intake for Koreans. Those in the very imbalanced group had a ratio of macronutrients (carbohydrates:fats:protein) of 81.15:7.18:11.50 and a 54% higher likelihood of having hypertriglyceridemia (P=0.025) compared with those in the balanced group. Conclusion: The current findings indicate that the diets of South Korean elderly are nutritionally imbalanced, including high carbohydrate consumption, which confers a high risk hypertriglyceridemia. These findings highlight the effect of nutritional imbalance in elderly with MetS.

5주간의 고지방식이 섭취시 흰쥐의 에너지 대사 기질 이용과 식이지방에너지 평가에 관한 연구 (Effect of 5 Week Long High-Fat Diet on Energy Metabolic Substrate Utilization and Energy Content Evaluation of Dietary Fat)

  • 본 연구는 실험쥐 20마리를 사용하여 5주간의 고지방식이를 섭취시킨 후 CON군과 HF군으로 구분하여 식이조성에 따른 체중과 체구성 그리고 24시간 동안의 에너지 대사를 분석하였다. 그 결과 고지방식이로 인한 체중의 유의한 차이는 없었다. 5주간의 식이섭취량은 두 군간 유의한 차이가 있었고, 섭취열량은 약 6% 정도의 유의한 차이가 있었다. 산소섭취량(에너지소비량)은 두 군 간의 유의한 차이가 없었으나, 이산화탄소배출량은 유의한 차이를 보였다. 고지방식이군은 24시간 동안 유의하게 높은 지방산화량을 보였고 일반식이군은 탄수화물산화량이 유의하게 더 높은 것으로 나타났다. 특히 활동기에 두 군의 에너지 이용 기질에 많은 변화가 있는 것으로 확인되었다. 두 군간 소비에너지는 유의한 차이가 없었으나 섭취에너지에 비해 HF군의 복강 내 지방이 약 33% 이상 유의하게 높게 나타났다. 이상의 연구결과를 종합하여 볼 때 식이구성에 의하여 에너지이용기질이 달라지는 것을 확인할 수 있었다. 또한 5주간 섭취한 열량과 소비된 에너지 비를 비교하였을 때 고지방식이군에서 복강내 지방이 유의하게 증가한 것은 체내에서의 지방의 식이효율이 그동안 알려진 Atwater의 값(9 kcal/g)보다 더 높은 것으로 사료되어진다. 이는 탄수화물과 고지방식이섭취에서 체내에너지 저장량을 분석하였을 때 탄수화물의 열량은 4 kcal/g인데 비해 지방은 약 11.1 kcal/g으로 보고한 기존의 연구보고(21)를 지지하는 결과로 나타났다. 이러한 결과는 일반인 및 비만환자와 대사증후군 환자의 체중조절과 개선에 있어서 식이구성과 일일영양권장량의 재검토의 필요성을 시사하고 있으며 또한 전문운동선수들의 체급별 체중조절에 필요한 식단의 재구성 등 스포츠현장의 기초자료가 될 것으로 판단되어진다. 그러나 본 연구는 대사과정에 관여하는 여러 호르몬과 단백질 등의 분석이 이뤄지지 않았고, 에너지대사량과 조직의 중량만을 검토한 결과이므로 추후 체내에서의 지방산화와 관련된 호르몬 및 단백질 등의 측정과 전체 피하조직 및 근육 등에 저장된 지방의 분석을 포함한 추가연구가 필요한 것으로 보인다.

대학가 주변 편의점 식사대용 편의식의 영양 평가 (Nutritional Evaluation of Convenience Meals in Convenience Stores near the Universities)

  • Objectives: Rapid economic growth and industrial development in South Korea have led to a great change in dietary patterns, and the use of convenience foods has continuously increased. This study was performed to evaluate the energy and nutrient contents of convenience foods at convenience stores near the universities as a meal. Methods: Data was collected by visiting 22 convenience stores near some universities in Chungbuk and Seoul and by checking nutrition labels on convenience foods at the stores. Data of a total of 338 food items were collected, and divided into five groups according to the food categories; rice products (n=156), noodles (n=101), burger/sandwiches (n=62), Tteokbokkis (n=11), and dumplings (n=8). Further, rice products, noodles, and burger/sandwiches were divided into subcategories. Results: The proportion of calories from carbohydrates was high in the rice products and tteokbokki, while the rate of calories from fat was high in burger/sandwiches and dumplings. Among the rice products, the proportion of carbohydrate calories was high in a one-dish food, rice with soup, and triangular kimbap, while the proportion of calories from fat in lunch boxes was high. In the noodles category, ramyeon and spaghetti had a high percentage of fat calories, while udong had a high percentage of carbohydrate calories. The ratio of the calorie content in relation to the KDRIs for adults aged 19-29 years, lunch boxes provided about 1/3 of daily required energy. However, the amount of calories as one meal was not enough for other types of rice products except for lunch boxes. Ramyeon was high in calories, fat, and sodium, but low in protein content. The burger/sandwiches had a high percentage of fat and sodium. Conclusions: Our results showed several nutritional limitations of convenience meals in convenience stores according to the type of food. Therefore, college students should limit excessive intake of convenience meals on a regular basis in order to avoid unhealthy food intake patterns. Our results demonstrate the need for educating college students with regard to checking nutrition labels when choosing convenience meals in order to facilitate the selection of food items that contribute to a balanced diet.

고당질식이(高糖質食餌)가 정상(正常)쥐와 Alloxan 당뇨(糖尿)쥐의 혈당량(血糖量)과 혈청지질성분(血淸脂質成分)에 미치는 영향(影響) (Effects of High Carbohydrate Diets on Serum Glucose and lipid Values of Normal and Alloxan-Diabetic Rats)

  • The possible effects of 10% carbohydrate containting diets on the serum glucose and lipids values were studied with normal and alloxan - diabetic male rats for a period of 28 days. The diets were supplemented with either glucose, cornstarch or sucrose, fructose or the basal diet containing no added carbohy drates. After this period, 3 to 4 rats among the each group were killed and samples of serum, liver, spleen and kidney were collected. The others were rendered diabetic by the intraperitoneal injection of alloxan ($190mg/\frac{wt}{kg}$). Then original feeding scedule was continued for 3 days in all five diabetic groups, before the collection of tissue samples and serum. Feeding the nondiabetic rats with glucose and fructose delayed the bodyweight development relatively compared with nonsugar group. The weight deductions after alloxan injection were similar in all feeding groups. All diabetic animals exhibited increased blood glucose triglceride levels but almost unchanged total cholesterol values. Blood glucose values for nondiabetic rats were normal ranges, and then glucose feeding group was the highest. Total - cholesterol values were the highest in nondiabetic rats fed glucose, fructose and in diabetics fed sucrose, glucose. Triglyceride values were the highest in nondiabetic rats fed sucrose, and no difference in diabetic rats. Some cornstarch group exhibited fatty-livers in diabetic and nondiabetic, and more studies need.
