Chitooligosaccharides의 마우스에 대한 급성 경구독성 (Acute Oral Toxicity of Chitooligosaccharides in Mice)

  • Chitosan을 Bacillus pumilus BN-262 유래의 chitosanase로 처리하여 trimer, tetramer, pentamer가 전체 올리고당 중 64.3%에 달하는 비교적 저분자의 chitooligosaccharides로 구성된 LMW-chitooligosaccharides를 얻었으며, chitosan을 Trichodema viride 유래의 cellulase로 처리하여 중합도 7 이상의 것이 전체 올리고당 중 49.3%에 달하는 상대적으로 분자량이 큰 chitooligosaccharides로 구성된 HMW-chitooligosaccharides 를 얻었다. 제조된 Chitooligosaccharides의 마우스에 대한 급성경구독성을 조사하였다. Chitooligosaccharides는 마우스에 대하여 어떠한 독성 효과도 보이지 않았으며 반수치사량은 5g/kg 이상인 것으로 추정되었다.

Enzymatic Production of High Molecular Weight Chitooligosaccharides Using Recombinant Chitosanase from Bacillus thuringiensis BMB171

  • The chitosanase gene (btbchito) of Bacillus thuringiensis BMB171 was cloned and heterologously expressed in the yeast Pichia pastoris. After purification, about 300 mg of recombinant chitosanase was obtained from the 1-1 culture medium with a specific activity of 240 units/mg. Results determined by the combined use of thin layer chromatography (TLC) and matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) mass spectrometry (MS) showed that the chitooligosaccharides (COSs) obtained by chitosan (N-deacetylated by 70%, 80%, and 90%) hydrolysis by rBTBCHITO were comprised of oligomers, with degrees of polymerization (DP) mainly ranging from trimers to heptamers; high molecular weight chitopentaose, chitohexaose, and chitoheptaose were also produced. Hydrolysis products was also deduced using MS since the COSs (n) are complex oligosaccharides with various acetyl groups from one to two, so the non-acetyl COSs (GlcN)n and COSs with more acetyls (> 2) were not detected. The employment of this method in the production of high molecular weight COSs may be useful for various industrial and biological applications, and the activity of chitosanase has great significance in research and other applications.

  • Chitosan (deacetylated form of chitin) possesses strong antibacterial activities such as antimicrobial effect, antifungal effect and the induction of plant defense response. Chitosan itself, however, has high molecular weight and viscosity as well as water-insolubility, These natures may restrict applications in various fields, especially in in vivo system. While the hydrolysates of chitosan, chitooligosaccharides (COS) are not only lower in the molecula. weight and viscosity, but also water-soluble. Thus, they would be expected more efficient absorption in vivo. Besides several documents have been reported antibacterial activities of COS against microorganisms (Kendra et al., 1989; Uchida et al., 1989). (omitted)

양식 넙치의 세균성 질병에 대한 키토산 및 키토올리고당의 항균효과 (Antimicrobial Effect of Chitosan and Chitooligosaccharides against Bacterial Diseases of Cultured Flounder)

  • 양식넙치의 세균성 질병의 원인균주인 Vibrio sp., Edwardsiella tarda 및 Streptococcus sp.를 제주산 양식 넙치로부터 직접 분리하였으며, 이들 세균에 대하여 키토산 및 분자량 크기에 따라 분획된 3종류의 키토산올리고당(HMW-COS, MMW-COS, LMW-COS)의 항균활성을 검토하였다. 키토산과 HMW-COS는 모든 세균에 대한 항균활성이 우수하였으며, 특히 Vibrio sp.과 Sfreptococcus sp.에 대해서 높은 항균활성을 나타내었다. 이들에 대한 결과는 키토산의 항균활성은 분자량에 따라 크게 의존한다는 사실을 보여주었다 E.tarda에 대한 키토산 및 HMW-COS의 항균활성은 처리시간을 증가시킴으로써 항균활성이 증가함을 보였으며, 이들의 항균기 작을 조사하기 위 한 SEM의 관찰에서, 세균들은 키토산에 의해 응집되어 서로 엉켜있는 형태를 보여주고 있었다. 이러한 현상은 키토산 및 HMW-COS가 세균의 증식을 명확하게 억제하고 있음을 보여주는 증거라 할 수 있다. 이것은 지금까지 여러 보고에서 밝혀진 키토산의 항균활성 메카니즘과 일치하는 것이라 판단된다.

Purification and Characterization of Antifungal Chitinase from Pseudomonas sp. YHS-A2

  • A strain producing a high amount of chitinase was isolated from soil, identified as Pseudomonas sp., and tentatively named Pseudomonas sp. YHS-A2. An extracellular chitinase of Pseudomonas sp. YHS-A2 was purified according to the procedure of ammonium sulfate saturation, affinity adsorption, Sephadex G-100 gel filtration and Phenyl-sepharose CL-4B hydrophobic interaction column chromatography. The molecular weight of the purified enzyme was estimated to be 55 kDa on SDS-PAGE was confirmed by active staining. Optimal pH and temperature of the enzyme are pH 7.0 and $50^{\circ}C$, respectively, and the enzyme is stable between pH 5.0 and 8.0 and below $50^{\circ}C$. The main products of colloidal chitin by the chitinase were N-acetyl-D-glucosamine and N,N'-diacetylchitobiose both of which were detected by HPLC analysis. The enzyme is supposed to be a random-type endochitinase which can degrade any position of ${\beta}$-l,4-linkages of chitin and chitooligosaccharides. The chitinase inhibited the growth of some phytopathogenic fungi, Fusarium oxysporum, Botrytis cineria, and Mucor rouxii and these antifungal effects were thought to be due to the characteristics of endochitinase.
