Objective: This study was undertaken to investigate the effects of Mori folium on insulin resistance and adipose tissue inflammation in an experimental mouse model of obesity.Methods: Obesity was induced in C57BL/6 mice by feeding them a high-fat diet. The mice were divided into four groups (n=6): a normal diet, high-fat diet, high-fat diet with 40 mg of Mori folium, and high-fat diet with 800 mg of Mori folium groups. After 13 wk, the body weights, fasting blood glucose and fasting serum insulin levels, insulin resistance (homeostatic model assessment) levels, oral glucose tolerance test levels, epididymal fat and liver weights, and gene expression of tumor necrosis factor-α, interleukin-6, and interferon-γ were measured. In addition, adipose tissue macrophages were analyzed by fluorescence-activated cell sorting.Results: Mori folium significantly reduced blood glucose levels, oral glucose tolerance levels, and liver weights. It also reduced adipose tissue macrophage numbers and tumor necrosis factor receptor-α gene expression.Conclusions: These results show that Mori folium has insulin resistance reduction and anti-inflammatory effects in an experimental mouse model of obesity.
In order to control glucose concentration during fed-batch culture for antibiotic production, we applied so called “software sensor” which estimates unmeasured variable of interest from measured process variables using software. All data for analysis were collected from industrial scale cultures in a pharmaceutical company. First, we constructed an estimation model for glucose feed rate to keep glucose concentration at target value. In actual fed-batch culture, glucose concentration was kept at relatively high and measured once a day, and the glucose feed rate until the next measurement time was determined by an expert worker based on the actual consumption rate. Fuzzy neural network (FNN) was applied to construct the estimation model. From the simulation results using this model, the average error for glucose concentration was 0.88 g/L. The FNN model was also applied for a special culture to keep glucose concentration at low level. Selecting the optimal input variables, it was possible to simulate the culture with a low glucose concentration from the data sets of relatively high glucose concentration. Next, a simulation model to estimate time course of glucose concentration during one day was constructed using the on-line measurable process variables, since glucose concentration was only measured off-line once a day. Here, the recursive fuzzy neural network (RFNN) was applied for the simulation model. As the result of the simulation, average error of RFNN model was 0.91 g/L and this model was found to be useful to supervise the fed-batch culture.

UDP-glucose는 세포 성장기에 다량 생산되다가 ${\beta}$-1,3-glucan 합성하는 시기에 일정한 농도가 되며 ${\beta}$-1,3-Glucan 합성 메카니즘에서 glucose를 운반하는 중요한 역할을 하는 것으로 나타났다. 2단 연속 발효조를 이용하여 세포 성장 발효조와 ${\beta}$-1,3-glucan 생산 발효조에서 UDP-glucose 변화를 관찰하여 ${\beta}$-1,3-glucan 생산시 농도가 높음을 관찰할 수 있었다. ${\beta}$-1,3-Glucan 생산 발효조의 pH를 5.5로 조절함으로써 UDP-glucose의 농도를 증가시킬 수 있을 뿐만 아니라 ${\beta}$-1,3-glucan의 생산 속도를 최적화할 수 있었다.
The development of glucose biosensors has been attracting much attention because of their importance in monitoring glucose in the human body; such sensors are used to diagnose diabetes and related human diseases. Thanks to the high selectivity, sensitivity to glucose detection, and relatively low-cost fabrication of enzyme-immobilized electrochemical glucose sensors, these devices are recognized as one of the most intensively investigated glucose sensor types. In this work, ZnO nanofibers were synthesized using an electrospinning method with polyvinyl alcohol zinc acetate as precursor material. Using the synthesized ZnO nanofibers, we fabricated glucose biosensors in which glucose oxidase was immobilized on the ZnO nanofibers. The sensors were used to detect a wide range of glucose from 10 to 700 M with a sensitivity of $10.01nA/cm^2-{\mu}M$, indicating that the ZnO nanofiber-based glucose sensor can be used for the detection of glucose in the human body. The control of nanograins in terms of the size and crystalline quality of the individual nanofibers is required for improving the glucose-sensing abilities of the nanofibers.
인삼은 고전적으로 항당뇨 효과가 있는 것으로 보고되고 있다. Insulin-like growth factor (IGF) 역시 당뇨병성 신증의 발병 초기에 중요한 역할을 하는 것으로 알려져 있다. 이에 본 연구에서는 mesangial 세포에서 고포도당에 의한 IGF 분비에 대한 ginsenoside의 차단 효과 및 이와 관련된 신호전달계를 알아보았다. 결과는 다음과 같다. 고포도당에 의해 증가 되었던 IGF-I 및 IGF-II 분비 촉진 작용은 GTS, PD 및 PT 처리 시 차단되었으며, 세포 성장 촉진작용에서도 같은 효과를 볼 수 있었다. 아울러 고포도당에 의한 산화성 스트레스 종가, GSH 감소, AA 방출 증가 작용 및 $PGE_2$ 합성 증가 작용은 GTS 처리시 현저하게 차단되었으며 PD 및 PT 처리 시 역시 억제 되는 것으로 나타났다. 이상의 결과를 볼 때 mesangial 세포에서 ginsenoside는 산화성 스트레스 및 arachidonic acid 활성 경로를 억제하여 고포도당에 의한 IGFs 분비 작용을 차단하는 것으로 나타났다.

BACKGROUND/OBJECTIVES: This study aimed to develop healthy, appetizing high-protein snacks with enhanced isolated soy protein for diabetic patients and determine the blood glucose and insulin response after being consumed by these patients. MATERIALS/METHODS: Thirty adult patients aged between 30 and 75 years, with a ≤ 10-year history of type 2 diabetes and hemoglobin A1c of < 7.5%, were enrolled in this study. They made 3 clinical visits at one-week intervals. The control group consumed 50 g carbohydrates (white bread), whereas the test groups consumed high-protein grain (HP_G) or high-protein chocolate (HP_C) after an 8-hrs fast. Blood (2 ㎤) was drawn at 15, 30, 45, 60, 90, and 120 min before and after consumption to analyze the blood glucose and insulin concentrations. RESULTS: Compared to the commercial snacks, the developed high-protein snacks had below-average calorie, carbohydrate, and fat content and a 2.5-fold higher protein content. In diabetic patients who consumed these snacks, the postprandial blood glucose increased between 15 min and 2 h after consumption, which was significantly slower than the time taken for the blood glucose to increase in the patients who consumed the control food product (P < 0.001). Insulin secretion was significantly lower at 45 min after consumption (P < 0.05), showing that the high-protein snacks did not increase the blood glucose levels rapidly. The incremental area under the curve (iAUC), which indicated the degree of blood sugar and insulin elevation after food intake, was higher in the control group than the groups given the 2 developed snacks (P < 0.001), and there was no significant difference in insulin secretion. CONCLUSIONS: The results of the postprandial blood glucose and insulin response suggest that high-protein snacks are potential convenient sources of high-quality protein and serve as a healthier alternative for patients with type 2 diabetes, who may have limited snack product choices. Such snacks may also provide balanced nutrition to pre-diabetic and obese individuals.
Objectives: The prevalence of obesity continues rise and obesity and metabolic syndrome is a major problem in global health care. Animal models are used in the drug discovery of novel treatment for obesity. One of common models of obesity is a high fat diet induced obesity in a C5BL/6 mouse, and the development of obesity and glucose tolerance in mouse model is different according to period of diet. Therefore, this study was performed to observe the development of obesity and glucose tolerance during a high fat diet (HFD). Methods: Male C57BL/6 mice, 5 weeks of age, were fed on a standard chow diet as a normal diet (18 kcal% fat) or a HFD (60 kcal% fat) for up to 16 weeks. The various factors related with obesity and insulin resistance were measured at 8, 12, and 16 weeks. Results: The weights of body and epididymal fat were gradually increased for 8~16 weeks, however the change of hyperglycaemia and glucose tolerance have shown different with that of body weight. Blood glucose, oral glucose tolerance and insulin tolerance were increased more clearly at week 12 and 16 than week 8. Lipid accumulation of liver and body temperature were also significantly increased at week 16, compared with normal group. Conclusions: The developments of obesity and related factors were different by a HFD period in a C57BL/6 obese mice. This result suggests that the development of obesity with glucose tolerance and liver lipid may induce clearly by a HFD for 16 weeks.
이상의 결과를 종합해 볼 때, 높은 수준의 포도당 섭취는 공복혈당과 공복혈당면적을 높이고 혈중 인슐린 농도와 아티포넥틴의 수준을 낮추어 혈당조절 능력을 억제시켰다. 반면, 높은 수준의 과당 섭취는 인슐린을 요구하지 않는 과당대사의 특이성으로 인해 혈당조절에는 효과적으로 보인다. 그러나 고과당 섭취는 간 조직 및 혈중 중성지방의 농도를 높이고 염증조절복합체 구성단백질의 발현을 조절하여 전염증인자의 발현을 증가시켰다. 이는 간조직에 있어 과당이 포도당보다 높은 수준의 염증반응을 유도하여 NAFLD의 발병과 진행에 보다 유의적인 영향을 준다는 것을 보여준다. 본 연구의 제한점은 일상에서 과당이나 포도당을 단독으로 섭취하는 경우가 드물다는 점과 3주라는 짧은 중재기간에 의한 실험 결과라는 것이다. 앞으로의 연구는 단순당을 장기간 섭취했을 때 혈청과 간 조직을 포함한 다른 대사 관련조직에서 나타나는 변화에 초점을 맞출 필요가 있으며, 염증조절복합체가 염증인자의 발현을 증가시키는 기전을 구체화하는 것이 요구된다.
고농도 포도당이 뼈모세포와 치주인대세포의 세포자멸사에 미치는 영향과 그 경로를 알아보기 위하여 뼈모세포주인 MC3T3-E1 (E1) 세포와 사람 치주인대로부터 일차배양을 통해 얻은 치주인대세포를 1,000 mg/L 농도의 포도당이 포함된 배양액 (대조군)과 4,500 mg/L 농도의 포도당이 포함된 배양액 (실험군)으로 나누어 24시간과 48시간 배양하였다. 그 후, ELISA assay를 통해 p38 MAPK와 caspase-3의 발현을 평가하고 Western blot을 통해 JNK-1과 ERK-1의 발현을 평가하여 다음과 같은 결론을 얻었다. 1. 뼈모세포와 치주인대세포 모두 대조군에 비해 실험군에서 caspase-3와 p38 MAPK 발현이 증가하였다. 2. 실험군에서의 caspase-3와 p38 MAPK 발현은 뼈모세포에 비해 치주인대세포에서 더욱 크게 증가하였다. 3. 뼈모세포와 치주인대세포 모두 대조군에 비해 실험군에서 JNK-1 발현이 증가하였다. 4. 뼈모세포와 치주인대세포 모두 ERK-1 발현에는 변화가 없었다. 이상의 결과로 보아, 고혈당 조건에 의해 뼈모세포와 치주인대세포의 세포자멸사가 증가하며, 치주인대세포가 고혈당 조건에 더욱 민감하게 반응하여 세포자멸사가 크게 증가하는 것으로 생각된다. 또한 이들 세포의 세포자멸사 과정은 p38 MAPK와 JNK-1 경로가 관여하며 ERK-1 경로는 관여하지 않는 것으로 추정된다.
우수한 셀룰로오스 생산 균주인 Acetobacter xylinum BRC5의 교반배양에 의한 셀룰로오스 생산성을 향상시키기 위하여 fed-batch 배양을 하였으며, 기질공급속도, 기질 공급량 및 용존 산소의 영향을 검토하였다. 초기 glucose 양을 변화시켜 회분배양하였을 때 glucose 농도가 10 및 20 g/L인 경우 셀룰로오스 생산량은 각각 2.05와 4.10 g/L이였으며 glucose에 대한 셀룰로오스 수율 (Yp/s)은 0.21이었다. 초기 glucose 농도 40g/L일 때 셀룰로오스 수율을 향상시키기 위해서 초기 glucose 농도 20 g/L에서 회분배양을 시작하여 glucose가 gluconic acid로 완전히 전환된 시점부터 추가적으로 glucose를 공급하여 fed-batch 배양기간에 glucose 공급속도는 셀룰로오스 생산성에 큰 영향을 미쳐 20g/L의 glucose를 2.22 g/L.h의 속도로 9시간 첨가하여 fed-batch 배양한 결과 셀룰로오스 생성량이 10 g/L로 가장 우수하여 초기 glucose 농도 20 g/L로 회분배양하였을 때 비하여 약 2배 증가하였으며, $Y_{P/S}$도 0.26으로 현저히 향상되었다. 또한 동일조건으로 fed-batch 배양하면서 glucose 공급량을 증가시켜 40g/L의 glucose를 추가적으로 첨가한 경우 셀룰로오스 생산량은 10.7g/L는 거의 증가되지 않았으며, $Y_{P/S}$가 0.18로 감소하였다. 이는 셀룰로오스 농도가 증가함에 따라 산소 공급이 부족하기 때문이므로 용존산소(DOT)를 2~15% 포화범위에서 조절하여 fed-batch 배양했을 때 DOT를 10% 수준으로 유지하면서 fed-batch 배양기간에 40g/L의 glucose를 추가공급 했을 때 셀룰로오스 생성량은 15.3 g/L로 증가되었고 이때 $Y_{P/S}$는 0.26로 향상되었다. 이는 DO를 제어하지 않는 경우에 비하여 셀룰로오스 생성량이 1.5배 증가한 결과이다.
