  • As for the Japanese dietary style and dietary equipments, many are regarded, with the rice plant imported, as the influence of China and Korea, while traditional culture has recently changed in Japanese and Korean urban lives because of the increase of nuclear families and modernization of lives. Of 1,244 junior high school students of boys and girls in Tokyo and Seoul, we investigated the present situations of the employment of dining equipments October to November, 1987. The results obtained are as follows. More cases that each member of a family had his or her own dining equipment were found in Japan. The frequency of using chopsticks was high in both countries. Korean children, having no struggles, handled chopsticks better than Japanese children. As for the dishing-up of sidedishes, more than half of Japanese used their individual plates, while in Korean families they used whole dishes. As for the employment of dining equipments, Japanese people have established their own style because of the national character of delicacy and finicality. This kind of traditional habits were found much more in extended families than in nuclear families. In Korea, the traditional spirit and the broad-minded characteristics cherished in the continental circumstances were manifested in the employment of dining equipments and their strong national character will not easily accept the wave of modernization.

Enhanced Large-Scale Production of Hahella chejuensis-Derived Prodigiosin and Evaluation of Its Bioactivity

  • Prodigiosin as a high-valued compound, which is a microbial secondary metabolite, has the potential for antioxidant and anticancer effects. However, the large-scale production of functionally active Hahella chejuensis-derived prodigiosin by fermentation in a cost-effective manner has yet to be achieved. In the present study, we established carbon source-optimized medium conditions, as well as a procedure for producing prodigiosin by fermentation by culturing H. chejuensis using 10 L and 200 L bioreactors. Our results showed that prodigiosin productivity using 250 ml flasks was higher in the presence of glucose than other carbon sources, including mannose, sucrose, galactose, and fructose, and could be scaled up to 10 L and 200 L batches. Productivity in the glucose (2.5 g/l) culture while maintaining the medium at pH 6.89 during 10 days of cultivation in the 200 L bioreactor was measured and increased more than productivity in the basal culture medium in the absence of glucose. Prodigiosin production from 10 L and 200 L fermentation cultures of H. chejuensis was confirmed by high-performance liquid chromatography (HPLC) and liquid chromatography-mass spectrometry (LC-MS) analyses for more accurate identification. Finally, the anticancer activity of crude extracted prodigiosin against human cancerous leukemia THP-1 cells was evaluated and confirmed at various concentrations. Conclusively, we demonstrate that culture conditions for H. chejuensis using a bioreactor with various parameters and ethanol-based extraction procedures were optimized to mass-produce the marine bacterium-derived high purity prodigiosin associated with anti-cancer activity.

대학생 외국인 학습자를 위한 주제 중심의 교양 영어 수업방안 (Topic-oriented Liberal English Class Plan for Foreign Learners at University)

  • 본 연구는 외국인 유학생을 대상으로 한 교양 영어 수업의 실질적인 수업 방안을 제시하는 것이다. 외국인 학습자가 대학 학업 수준의 한국어 능력을 갖추지 못했을 경우 한국어 교수자가 진행하는 교양 영어 수업 내용을 이해하는데 어려움을 겪을 수 있다. 이를 위해 본 연구에서는 각종 미디어를 활용한 주제 중심의 참여 수업 활동을 제시하였다. 이에 대한 학습자들의 태도와 인식을 분석하기 위해 설문 조사를 실시하였다. 설문 조사 결과 주제 기반 수업에 대한 학습자들의 만족도는 높게 나타났으며(75%) 그 이유로는 배려하는 교수자의 태도, 편안한 수업 분위기, 수업에 대한 재미로 나타났다. 다양한 참여 수업 활동에 대한 학습자들의 만족도 또한 매우 높게 나타났으며 (81.9%) 그 이유로는 학습에 유익, 흥미와 동기 유발, 참여 수업의 효율성인 것으로 나타났다. 국제화 시대 외국인 유학생의 증가 추세에 발맞추어 그들에게 맞는 현실적인 수업 방안과 다양한 수업 활동의 개발이 필요하다.

향토문화 콘텐츠를 위한 디지털 편찬 관리시스템 개발에 관한 연구: "한국향토문화전자대전"의 사례를 중심으로 (A Study on Development of Digital Compilation Management System for Local Culture Contents: Focusing on the Case of The Encyclopedia of Korean Local Culture)

  • 향토문화란 한 지방의 자연환경 속에서 과거로부터 현재까지 면면히 전승되어 온 역사와 전통, 풍물과 생활, 예술과 유물 및 유적 등의 모든 유산을 의미한다. 한국학중앙연구원에서는 이런 향토문화를 디지털 콘텐츠로 제작하고 이를 이용하여 "한국향토문화전자대전"을 편찬하였다. 향토문화 콘텐츠는 기록물의 특성을 가지고 있어 출처주의, 계층목록과 같은 기록물관리체계를 따르고 있으며 이를 편찬 관리하기 위한 시스템은 기초자료, 단편적 정보 및 고급정보 등이 하나의 시스템 안에서 순환하면서 새로운 지식정보를 만들어내도록 도와주는 순환형 지식정보관리시스템을 지향한다. 순환형 지식정보관리시스템의 이용자는 이 시스템을 통해 직접적으로 자료를 수집할 수 있을 뿐만 아니라 다른 곳으로부터 데이터를 수집할 수 있으며, 나아가 수집한 데이터를 가공하여 새로운 지식 정보를 창출할 수 있다. 그러나 다양한 향토문화 콘텐츠의 구조에 포함된 의미적인 특징을 손상시키지 않고 데이터베이스를 구축하기가 매우 어렵고, 또한 이러한 작업은 장시간에 걸쳐 여러 차례의 교정 작업이 수행되어야 하므로 문서편찬, 교정, 서비스가 동시에 수행되는 시스템을 필요로 한다. 따라서 본 논문에서는 "한국향토문화전자대전"의 사례를 중심으로 고문서가 많이 포함된 향토문화 콘텐츠의 의미적인 특징을 손상시키지 않으면서, 문서의 구조정보를 표현할 수 있는 XML 기반의 디지털 편찬 관리시스템을 제시하고 본 연구에서 개발된 관리시스템에서 향토문화 콘텐츠 관리를 위해 확장된 기능을 소개한다.

신생 백서 척수후근절의 슈반세포 배양을 위한 Ara-C 분열억제제의 최적 효과에 대한 연구 (THE MOST APPROPRIATE ANTIMITOTIC TREATMENT OF ARA-C IN SCHWANN CELL-ENRICHED CULTURE FROM DORSAL ROOT GANGLIA OF NEW BORN RAT)

  • Schwann cell, one of important components of peripheral nervous system, interact with neurons to mutually support the growth and replication of embryonal nerves and to maintain the different functions of adult nerves. The Ara-C, known as an antimitotic agent, have been used to have high effectiveness in eliminating fibroblasts during Schwann cell culture period. This enrichment effect is also known to be cummulative with each successive pulse of Ara-C applied and is due to a progressive loss of fibroblasts. But the cytotoxicity by Ara-C is also cummulative and noticeable over the period. To determine the most effective application time and interval of Ara-C in the Schwann cell culture, we observed the Schwann cell purity and density with the Ara-C treatment in plain and three-dimensional culture from dorsal root ganglion of new born rat. By culturing dispersed dorsal root ganglia, we can repeatedly generate homogenous Schwann cells, and cellular morphology and cell count with mean percentages were evaluated in the plain culture dishes and in the immunostainings of S-100 and GFAP in the three-dimensional culture. The Ara-C treated cultures showed a higher Schwann cell percentage ($31.0%{\pm}8.09%$ in P4 group to $65.5%{\pm}24.08%$ in P2 group), compared with that obtained in the abscence of Ara-C ($17.6%{\pm}6.03%$) in the plain culture after 2 weeks. And in the three-dimensional culture, S-100 positive cells increased to $56.22%{\pm}0.67%$ and GFAP positive cells to $66.46%{\pm}1.83%$ in G2 group (p<0.05), higher yield than other groups with Ara-C application. Therefore, we concluded that the Ara-C treatment is effective for the proliferation of Schwann cells contrast to the fibroblasts in vitro culture, and the first application after 24 hours from cell harvesting and subsequent 2 pulse treatment (P2 group in plain culture and G2 group in three-dimensional culture) was more effective than other application protocols.

마늘의 경정배양에서 기내인경구 대량생산을 위한 2단계 배양의 도입 (Introduction of two-step culture method for multiple seed bulb development from shoot tip culture of garlic (Allium sativium L.))

  • 마늘의 경정을 기내배양하여 대량의 기내종구를 생산하기 위한 다신초의 유기 및 기내구의 비대조건을 찾고자 배지의 당의 농도를 다르게 하고 또한 생장조절물질을 첨가하여 신초의 대량발생 여부를 확인하였으며, 대형배양용기를 이용하여 대량생산을 하기 위한 tank배양에서 신초의 분화와 기내구의 발달에 관하여 검토하였다. 경정배양에서의 shoot 발생이나 기내구 형성 모두 3% sucrose 첨가 MS고체배지에서 8% 보다 양호하였으며, 생장조절물질의 효과는 2 mg/L 2iP와 0.2 mg/L IAA의 혼합처리에서 shoot 발생과 bulb의 형성수가 양호하였다. 이 처리구에서 발생한 shoot와bulb의 수는 각각 3.3 및 2.7개로 나타났다. 다신초를 유도할 수 있는 조건의 배지에서 경정을 2개월 동안 배양하여 다신초를 분화시킨 후, 20 L water tank를 이용하여 sucrose 농도를 3%와 8%로 설정한 MS액체배지에 옮겨 45일간 배양한 후에 bulb형성을 조사하였을 때 유식물체의 생육에 있어서는 3% sucrose 첨가조건에서 8% 첨가배지보다 양호하였다. 그러나 기내구의 형성이나 생장에 있어서는 sucrose의 농도가 8%일 때 보다 효과적이었다. 경정배양에서의 다신초의 초기생산에는 3% sucrose조건에서 양호한 효과를 보이고 있으므로 마늘의 인경구를 대량으로 생산하기 위해서는 다신초의 생산과 구비대 과정을 구분하는 2단계 배양법을 도입하는 것이 유리할 것이다. Tank배양시 배지에 0.2 mg/L BA, 0.02 mg/L NAA를 첨가한 처리에서 양호하였으며 NAA 농도가 높을 수록 유식물체의 생육이나 구비대의 효과는 현저하게 저하되었다.

기수산 물벼룩 Diaphanosoma celebensis의 안정 배양을 위한 배양 용기의 크기 선택 (Selection of Culture Scale for Stable Culture of an Estunrine Cladoceran Diaphanosoma celebensis)

  • 전세계의 담수역에 널리 분포하는 Daphnia나 Moins와 같은 담수산 물벼룩은 잉어, 붕어, 금붕어와 같은 담수어의 종묘 생산과정에서 먹이 생물로서 널라 이용되고 있다. 그러나, 해산어의 종묘 생산 과정에서는 안정 배양, 대량 배양이 어렵다는 이유로 로티퍼나 알테미아 이외에 널리 이용되는 먹이 생물은 거의 없다. 이 연구에서는 해산 물벼룩보다는 비교적 배양이 용이한 기수산물벼룩 D. celebensis의 배양 용기의 크기가 안정배양에 미치는 영향을 검토하였다. 종 보존 배양 수준의 40ml 배양에서는 16일간의 배양 기간중 14일째에 최고 밀도가 3.416$\pm$0.396 ind./ml에 달하였다 1,500ml와 15,000ml의 준 대량 배양에서는 각각 배양 개시후 12일째와 21일 째에 14.2$\pm$2.122 ind./ml와 2.489$\pm$1.618 ind./ml의 최고 밀도가 관찰되었다. 그리고 실험 기간중 배양의 안정도를 평가하기 위하여 실시한 세가지 배양 수량간 상대적 개체군 증식 지수 (RPGI)의 비교 결과에서는 1,500ml의 배양 용기에서 가장 안정적인 증식양상을 보였다. 뿐만 아니라, 매일 수확 가능한 기수산 물벼룩 D. celebensis의 개체수를 계산한 결과에서도 (possible harvesting number/ind./ml/day) 1,500ml의 배양 용기에서 가장 높은 3.567$\pm$0.607ind./ml/day가 수확 가능하였다. 한때 필요성에 의하여 담수산 물벼룩을 해산어의 종묘생산 과정에서 먹이생물로 이용한 시기가 있었다. 그러나, 앞으로는 기수산 물벼룩을 해수에서 배양하여 해산 자치어의 먹이생물로 이용할 수 있을 것이다.

상주지역 주부의 식생활 의식 실태 조사(연령별) (The Study on The Consciousness of Housewives Eating Habit in Sang-Ju City according to ages)

  • The purpose of this study was to know the conscious of eating habit of housewife. The data were collected from 250 housewives who were the age group of 20-60's in Sang-Ju. The survey was taken place from May to June in 2000. The result showed that there were significant differences in eating habit's consciousness according to housewife's age group: 1. As the age goes up, the housewife had less consideration herself when they purchased food and decided cooking method. 2. As the age goes down, the order in having meal was depended on conditions. But as the age goes up, they considered the order as important thing like followings; eating together, eating separately according to the sex, male first, senior first, housewives lastly. 3. The survey showed that there was distinction depending on sex in meal as age goes up. And The subject was conscious that the delicious and valuable meal served to male, senior and child before. 4. Regardless of senior, the consciousness for the skipping meal was high as the age goes down. 5. As the age goes up, female and seniors showed that leftover was not so good. 6. The consciousness that housewives were responsible for preparing the meal was high as the age goes up, but they had further difficulty in preparing meal as the age goes down. 7. In considering that male and senior should be participated in the kitchen work, they had high consciousness as the age goes down. 8. About role of cooking, the consciousness was hish in case of male as the age goes down, in case of female and housewife were high as the age goes up. 9. As the age goes up, The consciousness was high that Female must buy the food. 10. As the age goes up, they had high consciousness in considering that the meaning of meal was related to survival, that noodle and bread were not meal. And the consciousness about that eating out was not good for health was high as the age goes up.

한국전통간장의 맛과 향에 관여하는 주요 향미인자의 분석(I) - 일반특성 및 당류와 유기산 분석 - (Analysis of Significant Factors in the Flavor of Traditional Korean Soy Sauce (I) - Analysis of General Characteristics, Sugars and Organic Acids Contents -)

  • This study was carried out in order to investigate general characteristics, sugars and organic acids contents of Korean traditional soy sauce, and to find out possibility of high quality soy sauce production in a short period through high concentration soy sauce making. In this study, we prepared three different types of soy sauce, low concentration soy sauce (Chungjang), high concentration soy sauce and Kyupjang, high quality traditional Korean soy sauce. pH of soy sauce were $4.46{\sim}4.90$ and did not show difference among three samples. Titratable acidity, buffering power and total free acid content were the highest value in Kyupjang. Kyupjang showed the highest contents of salt and pure extract. As the ripening period increased, the salt content increased in Chungjang samples, but decreased in Kyupjang. Reducing sugar contents of Kyupjang, high concentration soy sauce and low concentration soy sauce were 1.13%, 0.76% and 0.53%, respectively. Free sugar in soy sauce were analyzed maltose, glucose, galactose and fructose. Total free sugar content was highest in high concentration soy sauce, however, contents of glucose and fructose were higher in Kyupjang than in Chungjang samples. Galactose was the main free sugar in Chungjang, but glucose was in Kyupjang. Among identified volatile organic acid, acetic acid was present in the highest concentration, and volatile organic acid content was highest in the high concentration soy sauce at 150 days. 20 nonvolatile organic acids were detected in Korean traditional soy sauce. Succinic acid, lactic acid and 2,5-pyridine dicarboxylic acid were the main nonvolatile organic acid in soy sauce.
