The Role of Fatty Acid Binding Protein in the Fatty Liver Induced by Alcohol or High Cholesterol Diet in Rats (알코올 및 고콜레스테롤 식이로 유도된 흰쥐의 지방간에서 지방산 결합단백질의 역할 및 특성)

  • There is a marked increase in geriatric disease, especially liver disease, due to the continuous increase in alcohol and fat consumption. Since the fatty liver, induced by alcohol or fat, is basically from abnormalities in the lipid metabolism, it is possible that fatty acid binding protein(FABP) which is related to the fatty acid metabolism may also be abnormal in these livers. FABP is a small molecular weight protein family present in cytosol in high concentration. It has been proposed as a fatty acid transfer protein and as a binding protein responsible for controlling intracellular free fatty acid concentration. In this research, we have examined the relationship between liver FABP and fatty liver induced by alcohol or high cholesterol diet. Rats were fed one of either semipurified liquid diets; control diet containing 65% carbohydrate, 20% protein, and 15% fat or high cholesterol diet containing 1%(w/w) cholesterol or alcohol diet containing 37% of alcohol instead of carbohydrate. After 5 weeks of feeding period, all rats received commercial chow diet for 5 weeks to examine recovery effect. Liver and blood samples were collected at 0, 1, 3, 5 and 10 weeks to analyze lipid compositions. FABP was purified from liver cytosol and injected to rabbit to obtain antiserum. Liver FABP amount was determined by SDS-PAGE and western blotting methods. Fatty acid binding capacity was determined by binding of 14Cpalmitate with the delipidated liver cytosol. Consumption of alcohol increased serum cholesterol, triglyceride concentration and decreased HDL-cholesterol concentration after 5 weeks. Serum apolipoprotein B concentration increased after 3 weeks and LDL-cholesterol and apolipoprotein A concentration changed after 1 week. Liver cholesterol and triglyceride concentration increased after 3 weeks. Consumption of high cholesterol diet changed liver and serum lipid composition after 3 weeks. Swiching to normal diet for 5 weeks did not normalize most of lipid composition in serum and liver except serum and liver except serum cholesterol, triglyceride and liver cholesterol. Liver cytosol FABP content and the fatty acid binding capacity decreased dramatically after 1 week with alcohol consumption. This results indicate that FABP content changes before the changes before the changes of blood or liver lipid composition, suggesting changes of FABP may cause development of the fatty liver induced by alcohol and can be used as an index of detecting a early development of fatty liver.

Activities of Enzymes Involved in Fatty Acid Metabolism in the Colon Epithelium of Piglets Fed with Different Fiber Contents Diets

  • The present study was conducted to evaluate the influence of dietary fiber on the activities of malic enzyme and citrate lyase involved in fatty acid metabolism in the colon epithelium of pigs. Thirty-six weaned 5 weeks old crossbred (Yorkshire${\times}$Swedish Landrace) piglets originating from twelve litters were randomly assigned to either a low fiber diet containing 10% non-starch polysaccharides (NSP), a control diet containing 14.7% NSP or a high fiber diet containing 20% NSP. The activity of malic enzyme in the colonic epithelium of pigs significantly (p<0.05) increased with age during the suckling-weaning transition. There was a tendency (p<0.10) of decreased malic enzyme activity in the colonic epithelium of pigs fed on the high fiber diet. At week 6, a lowered (p<0.01) activity of malic enzyme in pigs fed on the low fiber diet compared with that in pigs fed on the high fiber and the control diets. Nevertheless, there were no significant differences in the activity of citrate lyase observed either between pigs with different ages or between pigs fed with various diets. The current data suggest that piglets during the suckling-weaning transition have a limited capacity to synthesize fatty acids from carbohydrate derivatives in the coloncytes. In addition, lipogenesis in coloncytes was enhanced with age during the suckling-weaning transition. A tendency (p<0.10) to an increased capacity to utilize acetyl-CoA in coloncytes of pigs has been observed for the high fiber diet. Moreover, the present work indicated that dietary fiber resulted in a lowered rate of lipogenesis and a reduced activity of malic enzyme.

Effect of Feeding Frequency of Extruded Diets Containing Different Macro-nutrient Levels on Apparent Nutrient Digestibility in Grower Flounder Paralichthys olivaceus (영양소 함량이 다른 부상 배합사료의 공급횟수가 육성기 넙치의 영양소 소화율에 미치는 영향)

  • Two feeding trials were carried out to investigate apparent nutrient digestibility of flounder fed experimental diets containing different levels of macro-nutrients by satiation feeding rate (Exp-1) and feeding frequency (Exp-2). Triplicate groups of fish averaging 280 g were fed three experimental diets which contained different levels of carbohydrate, protein and lipid by two feeding regimes (satiation and 80% satiation) and four feeding frequencies (three meals a day, two meals a day, one meal a day and one meal every two days). Feces were collected using a fecal collection column attached to fish rearing tanks for 6 weeks. Apparent digestibilities of dry matter, protein, lipid, energy and carbohydrate were not affected by feeding satiation rate in Exp-1. Apparent protein digestibility was not affected by feeding frequency, whereas affected by dietary composition in Exp-2. Apparent protein digestibility of fish fed a high-protein diet showed a tendency to become higher compared to that of fish fed high-carbohydrate diet and high-lipid diet at the same feeding frequency. Apparent lipid digestibility was not affected by dietary composition, however, affected by feeding frequency. Apparent digestibilities of energy and carbohydrate were affected by both dietary composition and feeding frequency. Apparent digestibities of energy and carbohydrate in fish fed the high-protein diet showed a tendency to become higher compared to that of fish fed the high-carbohydrate diet and high-lipid diet at the same feeding frequency. Apparent digestibities of energy and carbohydrate tended to decrease with increasing of feeding frequency at the same dietary composition.

Combined Trial of Fish Oil and Exercise Training Prevents Impairment in Insulin Action on Glucose Transport of Skeletal Muscle Induced by High-Fat Diet in Rats

  • The purpose of the present study was to determine the preventive effects of combined interventional trial of fish oil treatment and exercise training on insulin resistance of skeletal muscle in high-fat fed rats. Male Wistar rats were randomly divided into chow diet (CD), high-fat diet (HF), high-fat diet with fish oil (FO), high-fat diet with exercise training (EX), and FO+EX groups. The rats in control group were fed chow diet containing, as percents of calories, 58.9% carbohydrate, 12.4% fat, and 28.7% protein. High-fat diet provided 32% energy as lard, 18% as corn oil, 27% as carbohydrate and 23% as casein. The fish oil diet had the same composition as the high fat diet except that 100 g menhaden oil was substituted for corn oil. Insulin sensitivity was assessed by in vitro glucose transport in the soleus muscle after diet treatment and treadmill running for 4 weeks. While the FO or EX only partially prevented insulin resistance on glucose transport and visceral obesity induced by high-fat diet, these interventions completely corrected hyperinsulinemia and hyperglycemia from the high-fat diet. The rats in the FO+EX showed normalized insulin action on glucose transport, plasma chemicals and visceral fat mass. Insulin-mediated glucose transport was negatively associated with total visceral fat mass (r=-0.734; p<0.000), plasma triglyceride (r=-0.403; p<0.05) and lepin (r=-0.583; p<0.001) concentrations with significance. Multiple stepwise regression analysis showed that only total visceral fat mass was independently associated with insulin-mediated glucose transport (r=-0.668; p<0.000). In conclusion, combined interventional trial of FO+EX recovered insulin resistance on glucose transport of skeletal muscle induced by high-fat diet. Visceral fat mass might be more important factor than plasma TG and leptin to induce insulin resistance on glucose transport of skeletal muscle in high-fat fed rats.

EFFECTS OF HIGH BY-PRODUCT DIETS CONTAINING RICE BRAN AND BEET PULP ON THE SITE AND EXTENT OF DIGESTION AND MICROBIAL SYNTHESIS IN STEERS

  • The effects of feeding rice bran and beet pulp mixtures on the site and extent of digestion and microbial synthesis in fattening steers were studied. Three Holstein steers fitted with ruminal, duodenal and ileal cannulas were fed four diets in a $4{\times}3$ Youden square design. The four diets consisted of 15% Italian ryegrass hay and 85% concentrate as a control diet which included 72% rolled barley, 20% rice bran plus 40% beet pulp, 30% rice bran plus 30% beet pulp or 40% rice bran plus 20% beet pulp. All diets provided 1.8 times digestible energy required for maintenance. The digestibility of fat in the small intestine (% of flow) showed an increase with rice bran content among the by-product diets. Digestibility of structural carbohydrate both in the rumen and the whole digestive tract decreased linearly with rice bran content. The digestibility of nonstructural carbohydrate was not affected by rice bran content, but that of nonstructural, nonstarch polysaccharides was higher in the rumen and lower in the large intestine for the by-product diets than for the control diet. A rice bran content of more than 30% in the by-product diets severely inhibits ruminal microbial synthesis and digestible energy intake in fattening steers.

The Effect of Carbohydrate Processed Snacks on Middle School Students' Emotional Intelligence (경기 일부 중학생의 당류 간식 섭취가 정서지능에 미치는 영향)

  • Adequate nutrients intake and regular eating habit are so important for the students to develop physical and mental health, and they contribute significantly for the development of intellectual capacity. The intake of simple sugar contained in the processed foods is known to have been related not only with such physical problems as obesity and diabetes but also with hyperactivity during growing period. This study analysed the effects of carbohydrate processed snacks on the middle school students' emotional intelligence and intended to demonstrate the correlation and the influence of the foods. The subjects of this study were 476 the 1st grade middle school students sampled from two public middle schools of Gyeonggi Province, Korea. Questionnaire contained 47 questions including the kinds of sugar containing processed foods, the frequency of intake, the frequency of normal diet, and the emotional test. All data obtained in this study were processed by SAS 8.2 program. 77.9% of the students had snacks more than once a day and the favored foods were in the order of bread, cookies, fruit, milk and dairy products, flour foods, ramen and ice cream. Less of the sugar contained processed snacks consumed high emotional intelligence showed(p<0.001). And low intake of carbohydrate processed snacks linked with having more regular meals and the higher levels of emotional intelligence. There was no gender difference in consuming simple sugar containing processed foods, however, out of these foods, carbonated drinks, ramen and pizza affected significantly the emotional intelligence of the male students. It was found that sugar contained processed snacks affect the eating habits like meal skipping and the emotional intelligence. It is determined that the effects of carbohydrate processed snacks on emotional intelligence can not be denied.

The Effect of high Carbohydrate and Cellulose Diets on the Growth of Albino Rate (High Carbohydrate 와 Cellulose Diet가 흰쥐의 체내대사에 미치는 영향 ( I ))

  • The present study was undertaken to investigate the metabolic problems of high carbohydrate and cellulose diets of Korean. Forty males and same number of females of Albino rats, aged $45{\pm}5$days were divided into 95% high carbohydrate (H. CHO)group, 83.8% medium carbohydrate (M. CHO)group, 50% low carbohydrate (L. CHO) group and standard (Stand). group containing 72.2% sugar. Each group was divided into two again-1.55% cellulose group and non-cellulose group, 10 rats each of eight groups in both sexes. Cellulose was added to each of non-cellulose diets in the forms of spinach powder and rice bran. After 14 weeks the rats were sacrificed for chemical analysis and the results were elucidated as follows. (1) H. CHO+Cell. group showed the lowest value in body weight gained and shrinkage of almost all organs, in contrast with this group the L.CHO group showed higher body weight gained than Stand. group. M. CHO+Cell, group showed much the same body weight gained curve as Stand. group. (2) It was observed that cellulose group showed lower F.E.R and P.E.R value than non-cellulose group comparatively. (3) Total nitrogen retention and retention rate were decreaced in H. CHP groups compared with M. CHO or L. CHO groups. (4) The amount of feces was increased due to addition of cellulose to experimental diets and in accordance with the increasing total fecal excretion of nitrogen and glucose was also increased, especially noticeable in fecal glucose excretion. (5) It was noteworthy that serum cholesterol level was decreased due to addition of cellulose in H. CHO group and L. CHO group. (6) M. CHO+Cell. group was designed to reflect the average survey data of Korean diets and there was no significant differences on body weight gained, F.E.R, P.E.R, total nitrogen retention and hematology between M. CHO+Cell. and Stand. group. Total glucose excretion was increased due to dietary cellulose in M. CHO+Cell. group, but it seemed to be no metabolic problems in this group.

Anti-Obesity Effect of a New Dietary Supplement Consisting of Hydroxycitrate, Carnitine and Red Pepper (3D-Relax Diet) in High-Fat Fed Rats

  • Anti-obesity effect of a new dietary supplement (3D-relax) in high-fat fed rats. The aim of this study was to assess the effects of 3D-relax; a proprietary formulation containing hydroxycitrate (233 mg/g), carnitine (150 mg/g) and red pepper (150 mg/g); on body weight, body fat, and serum lipids levels in rats fed a high-fat diet. Male SD 7-wk-old rats (n=8) were fed a high fat diet [52% total dietary energy (E%) from fat, 15.4 E% protein, 32.6E% carbohydrate] with or without 3D-relax administration (1 g/kg body weight/day) for 3 weeks. Administration of 3D-relax significantly reduced the increase in body weight compared to the group fed high fat without 3D-relax. Food efficiency ratio (FER) tended to be decreased with administration of 3D-relax, but was not significant. The perirenal and epididymal fat pad weights of vats administered 3D-relax were significantly lower than those of the high fat group that did not ingest 3D-relax during the 3 weeks. The oral administration of 3D-relax significantly increased HDL-cholesterol level and lowered total cholesterol level compared to those of high fat alone group. These results suggest that 3D-relax reduced body weight and fat gains, and those effects are presumably linked to its inhibitory effects on lipogenesis.

Effects of Dietary Protein on Growth and Lipid Metabolism in Growing Rats (식이단백질조성이 흰쥐의 성장과 지방대사에 미치는 영향)

  • The effect of dietary protein on growth and lipid levels of plasma and liver was studied in weanling male rats fed diets differing protein sources and amino acid balance. Rats were devided into 9 experimental diets which were grouped into 3 categories ; 1) Simple protein category includes gluten-, soy protein isolate-, and casein-containing diet groups, 2) Supplemented category includes casein supplemented with methionine, soy protein isolate supplemented with methionine, and gluten supplemented with lysine and methionine, 3) Mixed protein category includes diet groups containing gluten (2/3), casein (1/3), soy protein isolate (2/3) and casein (1/3), and casein (1/3), soy protein isolate (1/3) and gluten (1/3). The experimental diets composed of 15% protein, 65.8% carbohydrate, 10% fat and 1% cholesterol. The body wt. gain and P.E.R. were greater in rats of supplemented and mixed protein groups than simple protein groups. No statistical differences were found in plasma cholesterol among gluten, soy protein isolate and casein groups. Consumption of diets supplemented with limiting amino acid to gluten or soy protein isolate reduced the plasma cholesterol level by 23.2% and 34.2% respectively. However there was no difference between casein and the supplemented casein groups. The mixed protein groups shows relatively high plasma cholesterol concentration and low liver cholesterol levels. On the other hand gluten group showed low plasma cholesterol and high liver cholesterol levels, which means body cholesterol pool may not have been changed by the dietary protein. Feeding soy protein meal and the supplemented soy protein isolate resulted in lower plasma cholesterol, plasma triglycerides, liver cholesterol and liver triglycerides levels. This hypolipidemic effect is considered to see unique to soy protein isolate. Rats in gluten and the supplemented gluten groups showed lower plasma protein levels and a tendency of fatty liver.

Effect of High Calcium Diet on the Zinc and Copper Balance in Korean Female Adolescents (청소년기 여자에서 고칼슘 섭취가 아연과 구리 평형에 미치는 영향)

  • Intakes and excretions of zinc and copper were determined for 8 female adolescents (aged 16.4$\pm$0.5 y; body mass index 20.4$\pm$1.3kg/$m^2$; body fat 33.3$\pm$2.5%; bone mineral density of lumbar spine in L2-L4; 0.96$\pm$0.08g/$\textrm{cm}^2$) when they consumed diets basal and high in calcium for 6 days each. All subjects consumed a basal Ca diet containing 800 mg, Korean RDA level of the subjects, and a high Ca diet containing 1200mg, RDA plus 2 SDs of calcium intake. The diets provided 58% of energy intake as carbohydrate, 25% as fat, and 17% as protein. Food, urine and fecal samples were collected during the last 3 days of each feeding period and were assayed. Mean daily intakes on the basal and high calcium diets, respectively, were 6.57 and 6.37 mg for zinc and 910 and 812 $\mu\textrm{g}$for copper. Fecal excretion of copper and zinc in relation to intake was significantly greater on the high calcium than on the basal calcium diet. Hence, apparent absorption rate was significantly lowered from 98.7% on the basal calcium diet to 97.9% on the high calcium diet for zinc from 66.3% to 56.4% for copper, respectively. Urinary loss of copper was not detectable but that of zinc was 0.38mg on the basal diet and 0.47mg on the high calcium diet. Copper retention was 899$\pm$105$\mu\textrm{g}$/day on the basal calcium diet and 792$\pm$20.8$\mu\textrm{g}$/day on the high calcium diet, and zinc retention was 3.95$\pm$0.91mg/day and 3.11$\pm$0.89mg/day. Thus, copper and zinc retention was significantly decreased on the high calcium diet (p<0.05). Summarizing the results, apparent absorption and retention of zinc and copper were significantly decreased by calcium supplementation. Therefore, it is suggested that interactions among minerals should be considered in determining RDA.
