오이 추출물의 항돌연변이 및 항미생물 효과 (Antimutagenic and Antimicrobial Effect of Cucumber (Cucumis sativus) Extracts)

  • 이 연구에서는 오이 methanol 추출물과 methanol 추출물의 분획물에 대한 항돌연변이 효과와 항미생물 효과를 조사하였다. Ames 실험계를 이용한 항돌연변이 효과는 Salmonella typhimurium TA100에서 간접 돌연변이원인 aflatoxin B$_1$에 대해 methanol 추출물은 1.25~5.0%에서 11~70%의 항돌연변이 활성을 나타냈었고, 직접 돌연변이원인 MNNG에 대해서는 46~85%의 항돌연변이 활성을 나타내었다. 또한 methanol 추출물을 분획하여 얻은 각 용매별 획분 중에서는 aflatoxin B$_1$에 대해서는 hexane 획분이, MNNG에 대해서는 butanol획분이 89% 및 95%로 가장 큰 항돌연변이 효과를 나타내었다. 항미생물 효과의 조사에서는 오이 metanol 추출물이 실험균주 11종 중 8종의 세균에 대하여 항균활성을 나타내었다. 이 중 K. pneumonia KCTC 2208, P. aeruginosa KCTC 2004가 clear zone 13 mm로서 가장 항균 활성에 민감한 균종으로 나타났다. 각 용매별 획분 중에서는 hexane 획분은 V. parahaemolyticus KCTC 2471에 대하여 항균효과를 보였고, chloroform과 ethyl acetate 획분에서도 약하게 항균 효과를 보였다 Methanol추출물의 농도에 따른 균증식억제 효과의 결과로는 V. parahaemolyticus가 가장 낮은 최소증식억제농도를 보여 500 ppm으로 나타났다. Methanol 추출물의 살균효과는 P. aeruginosa에서 1%의 농도가 0.5%의 농도에 비하여 약 10배정도의 빠른 살균효과를 보였다. 즉 초기 균수의 90%균을 살균하는데 1%와 0.5%의 농도에서 각각 26분, 250분이 소요되는 것으로 예측되었다.

  • 계피로부터 항균성 물질은 ethanol로 추출하고 다시 여러 가지 유기용매로 분획하여 항균물질의 분획별 항균성과 안정성을 조사하고 균종별 항균효과를 검토한 결과를 요약하면 다음과 같다. 1. 계피의 ethanol 추출물을 n-haxane, chloroform, ethyl acetate, n-butanol 등으로 분획한 결과 n-hxane 분획물의 항균력이 가장 우수하였다. 2. 계피의 n-hexane 분획물은 균종별로 차이는 다소 있으나 최저증식억제농도는 세균과 효모의 경우 각각 약 $320~640\;\mu\textrm{g}과\;160~320\;\mu\textrm{g}$, 그리고 곰팡이의 경우 $40~80\;\mu\textrm{g}$으로 나타나 세균보다는 곰팡이에 대한 항균력이 큰 것으로 나타났다. 3. 항균성 물질은 pH4.0~10.0 범위에서 항균력의 변화가 거의 없었다. 4. 항균성 물질을 $100^{\circ}C$에서 60분 또는 $121^{\circ}C$에서 15분간 열처리한 후에도 항균력은 거의 감소하지 않았다. 5. 계피추출물에 대하여 Ames test를 실시하여 돌연변이원성을 조사한 결과 plate당 $320\;\mu\textrm{g}$까지 첨가하여도 돌연변이능이 나타나지 않았으며, 오히려 $benzo(\alpha)pyrene$의 변이원능을 억제하는 항돌연변이원성을 가지는 것으로 나타났다.

쑥 추출물의 Tyrosinase 효소활성에 미치는 억제효과 (Inhibitory Effect of Mugwort Extracts on Tyrosinase Activity)

  • Melanin 생합성에 관련된 tyrosinase 저해활성올 생 쑥과 건조 쑥 용매 추출물로부터 검색하였다. 쑥은 ethanol, hexane, chloroform, ethyl acetate 용매 및 열수 추출을 하였고, 생 쑥의 ethanol 추출물의 ethyl acetate 분획과 건조 쑥의 ethan야 추출물의 hexane 분획에서 회수올은 각각 15.2%와 15.5%로 가장 높았다. 각 용매 추출물의 tyrosinase 활성 저해율은 건조 쑥 추출불보다 생 쑥 추출울이 더 높았고, 생 쑥의 ethanol 추출물의 chloroform 분획과 hexane 분획에서 각각 98.9%와 9 96.7%로 큰 저해활성을 가졌다.

Chemical Composition of Pinus koraiensis Seed and Its Biological Activity

  • This study was to investigate the chemical composition and biological activities of Pinus Koraiensis seed. The oil, moisture, ash, crude protein, and carbohydrate contents of P. Koraiensis seed were 58.21, 7.84, 1.56, 14.26, and 18.13%, respectively. The ratios of essential amino acid and nonessential amino acid against total amino acids were 36.6 and 60.3%, respectively. The saponification value of seed oil was 166.8 mg KOH/g oil. Among various fatty acids, the linoleic acid content was the highest, 35.5%, which was approximately 72.6% of polyunsaturated fatty acid. The ethylacetate extract of P. Koraiensis seed had the highest DPPH radical scavenging activity (62.8%) at 7.0 mg/mL, followed by hexane extract, methanol extract, and hot water extract. The maximum nitrite scavenging activity was obtained 59.3% at pH 1.2. The total phenolic concentration of ethylacetate extract was 98.7 mg/g, approximately 4.8 folds higher than that of the hot water extract. The maximum inhibition activities of elastase using ethylacetate extract and collagenase using hexane extract were 58.8 and 40.7%, respectively. These results indicate that P. koraiensis seed extract could be applied to present the possibilities of industrial applications for the developments of cosmetics.

Comparison of methods for proanthocyanidin extraction from pine (Pinus densiflora) needles and biological activities of the extracts

  • Flavonoids are known to be effective scavengers of free radicals. In particular, proanthocyanidins are flavonoids that possess cardiovascular protection, antioxidative activities, and immunomodulatory activities. Here, we evaluated proanthocyanidin contents in the total polyphenolic compounds of pine needle extracts prepared by hot water, ethanol, hexane, hot water-hexane (HWH), and hot water-ethanol (HWE). Analysis of each extract indicated that the ethanol extract contained the highest proanthocyanidin concentration. The HWH and hexane extracts also contained relatively high concentrations of proanthocyanidin. On the other hand, proanthocyanidin content analyses out of the total polyphenolic compounds indicated that the HWH extract contained the highest content. These results suggest that HWH extraction is a suitable method to obtain an extract with a high level of pure proanthocyanidins and a relatively high yield. The HWH extract possessed superior activity in diverse antioxidative analyses such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferrous ion chelating (FIC), and ferric-ion reducing power (FRAP) assays. In addition, upon assessing the effects of the pine needle extracts on macrophages (Raw 264.7 cell), the HWH extract exhibited the highest activity. In this study, we discerned an efficient extraction method to achieve relatively pure proanthocyanidins from pine needles and evaluated the biological functions of the resulting extract, which could potentially be used for its efficacious components in functional food products.

Antifertility Activity of Dictamnus albus Root Bark

  • The preliminary antifertility test of Dictamnus albus(Rutaceae) showed that a methanol extract of root bark decreased fertility in rats when administered orally on days $1{\sim}10$ postcoitum. Hexane, chloroform, butanol and aqueous fractions from the methanol extract, were tested to produce the result that only the hexane fraction exhibited positive activity. The hexane fraction was subfractionated into acidic, alkaloidal and neutral subtractions and tested. Neutral subtraction showed positive activity. From the neutral subtraction, fraxinellone was isolated as an active principle. The timed-dosing experiments revealed that antifertility activity of fraxinellone appeared to be exerted after tubal exit of the ova to prevent implantations.

  • 80% MeOH extract of black rices fractionated with n-hexane, EtOAc, and n-BuOH. Copper-induced LDL oxidation inhibition assay and ACAT inhibition assay examined with fractions. n-Hexane and EtOAc fractions showed high inhibition activity. We divided n-hexane fraction into three sub-layers(H1 - H3) and EtOAc into eight sub-layers(E1 - E8). H1, H2, E6, and E7 are showed higher inhibition activity than standard in Lp-PLA2 inhibition assay and H1 and E6 are showed higher ACAT inhibition activity than standard.

정제들깨기름의 산화안정성에 미치는 흑미 추출물의 영향 (Effect for Oxidation Stability of Refined Perilla Oil Use in Extract of Black Rice (Oryza sativa L.))

  • Proximate compositions of black rice and antioxidative effects of its methanol extract on refined perilla oil were investigated. Black rice seeds were composed of water 12.4%, crude protein 7.9%, crude fat 2.2%, crude fiber 1.0%, ash 1.4% and N-free extract 75.1%. The hexane and 80% methanol fractions in yields were higher than 80% ethanol and chloroform. In AOM(active oxygen method) test, antioxidative effects of black rice added to the refined perilla oil was increased with dose-dependent fashion in concentration of the black rice extracts. Among several extracts tested, 80% aqueous methanol extract showed the highest antioxidative activity, followed by hexane, chloroform, in that order. The peroxide value and TBA value for their antioxidation stability were also lower than that of control. This result was similar to that of AOM test.

용매의 분획에 따른 겨자 물추출물의 항균성 (Antimicrobial Activities in the Water Extract of Mustard Seed Fractionated by Solvents)

  • The water extract of mustard seed was fractionated by solvents with an increase of polarity, and antimicrobial activities of each extracts were examined, and volatile compounds of each extract were identified by GC-MS. When the water extract was fractionated with solvents, the antimicrobial activities were high in the order of chloroform, ethylacetate, hexane, butanol and aqueous layer. In chloroform fraction, 16 volatile compounds, including 2 isothiocyanates such as 3-isothiocyanato- 1-propene and 4-isothiocyanato- 1-butane, 1 nitrile and 4 acids were identified, their contents were higher than other fractions. Twelve, 10, 4 and 7 volatile compounds were identified in ethylaceate, hexane, butanol and water fractions, respectively. The volatile compounds were considerably less in the fractions of butanol and water than others.

  • Saccharomyces Cerevisiae was cultured in the malt wort added 0%, 0.001%, 0.01%, 0.1% Chloroform - Methanol and Hexane extract of Ganoderma Lucidum. The more the quantity of Ganoderma Lucidum extract were added the more total alcohol quantity were produced during the culture. Especially higher alcohol contents were more increased about 2.2 times 0.1% added group than on control group 120 hr cultured. Ethanol product was more increased on Hexane extract added group than on Chloroform - Methanol extract added group.
