Bacteriocins: Assay, Biochemistry, and Mode of Action

  • Bacteriocins are proteins produced by a heterogeneous group of bacteria that have a bactericidal effect on closely related organisms. Recently, bacteriocins from lactic acid bacteria and other food-related organisms have been the subject of much research because of their potential as food biopreservatives. Various modifications of agar plate diffusion assays are the most widely used methods even though the limitations of such assays are generally recognized. The ability to obtain a concentrated crude preparation on bacteriocin by optimizing production parameters greatly simplifies recovery of bacteriocin on subsequent purification steps. Some studies performed to optimize bacteriocins have been purified to homogeneity, and the amino acid sequences of many of these purified bacteriocins have been determined. Obtaining characterization data on purified bacteriocin will minimize the risk of overlapping of research and confusion on identification of these compounds. Several me-chanisms leading to cell death have been hypothesized. These include depletion of the proton motive force(PMF) across the cell membrane: RNase and/or DNase activity within the sensitive cell; and pore formation and lysis of sensitive cells at the cell membrane.

  • In the chemical industry, in the pharmaceutical industry, and in a number of other industries separation processes are necessary to separate and purify products and raw materials [1,2]. Separation processes are also widely used in other applications such as in recycling valuable materials from waste streams. Unit operations for separation processes can be classified in phase separation techniques and component separation techniques based on the nature of the feed mixtures to be separated. The former techniques are used for the separation of heterogeneous mixtures, in which the feed is already present in two or more separated phases on a micro-scale. The latter are suitable for the separation of homogeneous mixtures such as gaseous mixtures and mixtures of completely miscible liquids. tn these cases the separation into individual components is generally achieved by utilizing the differences in physico-chemical properties of components, and is much more difficult compared to phase separation techniques. Separation processes such as distillation, evaporation, liquid-liquid extraction, and crystallization belong to this class.

  • Purpose: Human tooth proteins are highly heterogeneous, comprising diverse proteins derived from a number of genes. The attempts to identify protein for activity of tooth matrix proteins have been defied by several factors. First, the amount of proteins within teeth is very small relative to many extracellular matrix proteins of other tissues. Second, the bioassay system is tedious and needed for long time. Therefore we tried to find easy techniques, which increase the product rate, and an assay of small proteins, with which amino acid sequence is possible without additional procedures. Materials and Methods: Total protein were extracted from 300 g enamel removed teeth and 600 g teeth with 4 mol/L guanidine HCl and purified by gel chromatography. Aliquot of proteins was implanted into muscle pouches in Sprague-Dawley rats for bioassay. By SDS-PAGE and membrane blotting, molecular weight of each protein was estimated and a partial amino acid sequence was obtained. Each fraction blotted on the membrane was cut out and inserted in rat ectopic model. Results: In dissociative method, total tooth proteins were obtained 1mg/ml from enamel removed teeth and 3.5 mg/ml from teeth. In SDS-PAGE, four clear bands at the sites corresponding to 66, 40, 20 and 18 kD. Especially The 66 kD band was clearly exhibited. Amino acid sequencing from tooth could be possible using PVDF membrane blotting technique. In amino acid sequencing, 66 kD protein was identified as albumin. Conclusion: Compared with conventional method for extraction of teeth protein and bioassay of proteins, the methods in this study were easy, time-saving and more productive technique. The matured tooth proteins omitting additional procedure of mechanical removal of enamel were simply analyzed using blotted PVDF membrane. This method seems to make a contribution as a technique for bioassay and amino acid sequencing of protein.

Expression of Type IV Collagen and Fibronectin in Salivary Gland Tumors (타액선 종양에서 제4형 교원질과 Fibronectin 발현)

  • Objectives: Salivary gland tumors pose considerable difficulty in diagnostic and prognostic assessment based on the histopathologic features alone. We studied the expression of type IV collagen and fibronectin in salivary gland tumors with special emphasis on the differential diagnostic significance. Materials and Methods: We did immunohistochemical stain on paraffin embedded tissues of 33 benign and 24 malignant salivary gland tumors using monoclonal antibody for type IV collagen and polyclonal antibody for fibronectin. Results: 1) Well preserved linear basement membrane-like staining of type IV collagen was detected in duct-cell-derived benign salivary gland tumors. But pleomorphic adenoma exhibited a heterogeneous pattern as focal augmentation and interruption. 2) In malignant tumors, type IV collagen was distributed in an irregular, interrupted manner or completely absent. Adenoid cystic carcinomas displayed a marked staining of the basal membrane associated substances in the pseudocysts. 3) The staining pattern of fibronectin was similar to that of type IV collagen execpt more dense in the stroma. 4) Salivary gland tumors which have a prominent myoepithelial cell component revealed a particular deposition of basement membrane materials adjacent to the myoepithelial cells. Conclusion: The study of the basal membrane substances may be helpful for differential diagnosis of benign and malignant salivary gland tumors and identifying special features of salivary gland tumors such as pseudocystic pattern of adenoid cystic carcinoma. Also we think that the myoepithelial cells contribute to the formation of basement membrane materials.

  • Landfill leachate is one of highly contaminated and heterogeneous wastewater. The leachate from initial landfill can be treated by anaerobic process because it contains biodegradable matters, particularly, volatile fatty acids (VFAs). However, the anaerobic treatment of leachate is generally required longer hydraulic retention time (HRT) than aerobic process and another treatment process to satisfy effluent concentration. Therefore the modification of conventional anaerobic treatment is needed. Two phase anaerobic membrane process (TPAMP) is an integrated membrane process to be able to separate anaerobic metabolism into two phase which are acidogenesis and methanogenesis for improvement of anaerobic treatment efficiency. In this study, the efficiency of TPAMP and conventional anaerobic treatment were compared in terms of HRT, effluent SCOD, VFAs Membrane used in TPAMP was the UF of capillary type with the surface area of $0.048m^2$. The average effluent SCOD of conventional anaerobic treatment was 1352 mg/L and the removal was 96 % at HRT 60 days, while in TPAMP, 927 mg/L and 98% at HRT 30 days.

Osteogenic Potential of the Periosteum and Periosteal Augmentation for Bone-tunnel Healing

  • Periosteum and periosteum-derived progenitor cells have demonstrated the potential for stimulative applications in repairs of various musculoskeletal tissues. It has been found that the periosteum contains mesenchymal progenitor cells capable of differentiating into either osteoblasts or chondrocytes depending on the culture conditions. Anatomically, the periosteum is a heterogeneous multi-layered membrane, consisting of an inner cambium and an outer fibrous layer. The present study was designed to elucidate the cellular phenotypic characteristics of cambium and fibrous layer cells in vitro, and to assess whether structural integrity of the tendon in the bone tunnel can be improved by periosteal augmentation of the tendon­bone interface. It was found the cells from each layer showed distinct phenotypic characteristics in a primary monolayer culture system. Specifically, the cambium cells demonstrated higher osteogenic characteristics (higher alkaline phosphatase and osteocalcin levels), as compared to the fibrous cells. Also in vivo animal model showed that a periosteal augmentation of a tendon graft could enhance the structural integrity of the tendon-bone interface, when the periosteum is placed between the tendon and bone interface with the cambium layer facing toward the bone. These findings suggest that extra care needs to be taken in order to identify and maintain the intrinsic phenotypes of the heterogeneous cell types within the periosteum. This will improve our understanding of periosteum in applications for musculoskeletal tissue repairs and tissue engineering.

Electrochemical Characteristics of Solid Polymer Electrode Fabricated with Low IrO2 Loading for Water Electrolysis

  • To maximize the oxygen evolution reaction (OER) in the electrolysis of water, nano-grade $IrO_2$ powder with a low specific surface was prepared as a catalyst for a solid polymer electrolyte (SPE) system, and a membrane electrode assembly (MEA) was prepared with a catalyst loading as low as $2mg\;cm^{-2}$ or less. The $IrO_2$ catalyst was composed of heterogeneous particles with particle sizes ranging from 20 to 70 nm, having a specific surface area of $3.8m^2g^{-1}$. The anode catalyst layer of about $5{\mu}m$ thickness was coated on the membrane (Nafion 117) for the MEA by the decal method. Scanning electron microscopy (SEM) and electrochemical impedance spectroscopy (EIS) confirmed strong adhesion at the interface between the membrane and the catalyst electrode. Although the loading of the $IrO_2$ catalyst was as low as $1.1-1.7mg\;cm^{-2}$, the SPE cell delivered a voltage of 1.88-1.93 V at a current density of $1A\;cm^{-2}$ and operating temperature of $80^{\circ}C$. That is, it was observed that the over-potential of the cell for the oxygen evolution reaction (OER) decreased with increasing $IrO_2$ catalyst loading. The electrochemical stability of the MEA was investigated in the electrolysis of water at a current density of $1A\;cm^{-2}$ for a short time. A voltage of ~2.0 V was maintained without any remarkable deterioration of the MEA characteristics.

  • The graft copolymer of chitosan with amino acid, L-lysine was synthesized by heterogeneous copolymerization and was evaluated as an artificial skin. The mechanical properties under dry and wet state, water content, water vapor transmittance rate and biodegradability were measured. The tensile strength and elongation under wet state ranged $0.3-0.5\;kg/mm^2$, 10-13%, respectively. Water vapor transmittance rate ranged $450-500\;g/m^2{\cdot}day$ like that of the normal skin. The weight loss of prepared membrane by protease IV was measured for the degree of biodegradation. The degree of biodegradation was around 15% and after 4 days it was slow. Biocompatibility was evaluated by studying the attachment of human fibroblast on the prepared membrane surface.

  • Cytologic features of inflammatory pseudotumor of the lung have not been described frequently. We report fine needle aspiration cytologic(FNAC) finding of a case of inflammatory pseudotumor misdiagnosed as adenocarcinoma in a 63-year-old man. The FNAC displayed a mixture of histiocytes, myofibroblasts, pneumocytes, and plasma cells. Some histiocytes and myofibroblasts had large nuclei with irregular nuclear membrane and prominent nucleoli, which mislead the diagnosis on adenocarcinoma on FNAC. The heterogeneous cell population is the unique cytologic features of inflammatory pseudotumor, which are helpful to distinguish it from other circumscribed benign and malignant lesions. Familiarity with these features is essential to avoid misdiagnosis and possible overtreatment.

  • The purpose of this study is to propose a topological design principles and to analyze the space of digital architecture applying topological invariant expressive characteristics. As this study is based on topology as a science of true world's pattern, we intented to explain the concepts and provide some methods of low-level and hyperspace topological invariant Properties. Four major aspects are discussed. Those are connection theory, boundary concept, homotopy group, knot Pattern theory as topological invariant properties. Then we intented to make understand topological characteristics of the Algorithms, luring machine, cellular automata, string theory, membrane, DNA and supramolecular chemistry. In fine, the topological invariant properties of the digital architecture as genetic algorithms based on self-organization and heterogeneous networks of interacting actors can be analyzed and used as a critical tool. Therefore topology can be provided endless possibilities for architecture, designers and scientists intended in expressing the more complex and organic patterns of nature as life.
