Optimal Hot Water Extraction Conditions of Mixed Herbs Extract Mixture Using Response Surface Methodology

  • Human needs energy to maintain metabolisms, and these energy sources were uptake foods or nutritions. The most effective source was known for glucose among the nutrients, furthermore the glucose is an important source of energy for blood cells and control brain maintenences cells. But as food is plentiful and eating habits become more westernized, fast food and irregular meal times by works. Nowadays, diabetes were rapidly increased by malnutriton and obesity. Diabetes was the sixth highest on the list of causes of death in Korea, released by the Statistics Korea in 2015, which is considered a serious social problem for adult diseases. Therefore, this study aims to establish the optimal hot water extraction conditions of mixed herbs extract mixture compounds that are effective in diabetes. The independent factors were extraction temperature (X1: $40-120^{\circ}C$), extraction time (X2: 2-10 hrs.), and the ratio of water to sample (X3: 40-200 mg/mL). Their effects were assessed on dependent variables of the extract properties, which included soluble solid contents, Brix of sample extract, total polyphenols content, total flavonoids content and DPPH Radical scavenging activity. As a result, the content of total polyphenol content was the highest in No.12(6 hrs, $120^{\circ}C$, 67 mg/mL) and the highest total flavonoid contents was found in No.16(6 hrs, $80^{\circ}C$, 40 mg/mL). DPPH Radical scavenging activity showed the highest activity in No.7(8 hrs, $100^{\circ}C$, 100 mg/mL).

백지를 포함하는 한약재 복합 에탄올 추출물이 피부 항노화 및 미백에 미치는 영향 (Effect of ethanol extract from mixture including Angelicae Dahuricae Radix on Dermal Anti-aging and Whitening)

  • Objective : Herbal medicinal mixture (JMB) are consisted of Caryophylli Flos, Aucklandiae Radix, and Angelicae Dahuricae Radix. Each of these herbal medicines has studied on anti-aging effect in vitro. So this study was conducted to investigate efficacy and potency of JMB extract on dermal anti-aging and whitening. Methods : The JMB was extracted at room temperature by 80% ethanol. Collagenase and elastase inhibition activity in JMB ethanol extract were determined at 10, 50, 100, 500, 1000 mg/ml concentrations by colorimetric method. The toxic range of JMB ethanol extract was evaluated using MTT assay. Also, The inhibitory effect of JMB ethanol extract on tyrosinase activity and melanin contents in mouse melanoma cell line (B16F10 cell) was identified at 50, 100, 200 ㎍/㎖ levels by spectrometric assay. In each analysis, EGCG (epigallocatechin gallate) and Kojic acid were used as positive controls, respectively. Results : The elastase and collagenase inhibitory activity of JMB ethanol extract increased dose dependently. Also, The MTT assay showed that JMB, up to 400 ㎍/㎖ concentration, exhibited no toxic effect to the B16F10 cell. And following the JMB ethanol extract treat, cellular melanin contents and tyrosinase activity were dose-dependently decreased compared to those of control. Conclusion : These results suggest that JMB ethanol extract has effects to inhibitory activity on dermal wrinkle enzyme and melanogenesis. Therefore, JMB has applicable benefits for development of materials or products to have whitening and anti-aging functions on skin.

  • Objective : This study was conducted to observe the anti-hyperlipidemic and anti-inflammatory effects, as well as the metabolic benefits, of pear extract and herbal drug mixture (Lycii Fructus, Coicis Semen, Alimatis Rhizoma, and Astragali Radix) on rats fed with a high fat diet. Methods : The animals used were male rats and the control group was fed a high fat diet only. The experimental groups were divided into four. Exp I group was fed a high fat diet with a mixture of pear extract and 3% Lycii Fructus; Exp II group was fed a high fat diet with a mixture of pear extract and 3% Coicis Semen; Exp III group was fed a high fat diet with a mixture of pear extract and 3% Alimatis Rhizoma; and Exp IV group was fed a high fat diet with a mixture of pear extract and 3% Astragali Radix for 4 weeks. Results : The body weight gain increased in all groups, but attenuated gradually in the experimental groups compared to the control group. The food intakes were significantly lower in Exp I and Exp III groups than the control group. The concentrations of serum total cholesterol (TC), HDL-cholesterol, LDL-cholesterol, and triglyceride (TG) were significantly higher in Exp II than in the control group, and lower in Exp III group than in the control group. Also the concentration of serum free fatty acid was significantly lower in the Exp III group than in the control group. In inflammatory activities, the Exp II group was significantly lower than the control group. Conclusion : The results indicated that Exp III group (administered a mixture of pear extract and Alimatis Rhizoma) most efficiently reduced fat accumulation and body weight, while the Exp II group (administered the mixture of pear extract and Coicis Semen) had the highest elevated lipid metabolism and immune activity.

정향 및 목향과 백지를 혼합한 한약재 에탄올 추출물의 항산화 효능 (Anti-oxidant effect of ethanol extract from mixture including Caryophylli Flos, Aucklandiae Radix and Angelicae Dahuricae Radix)

So this study was conducted to investigate efficacy and potency of JMB on anti-oxidation. Methods : The JMB was extracted at room temperature by 80% ethanol. And total polyphenol contents, total flavonoid contents in JMB ethanol extract were determined by colorimetric method. Also, DPPH, ABTS free radical scavenging capacity and reducing power of JMB ethanol extract were measured at 100, 500, 1000, $5000{\mu}g/m{\ell}$ concentrations by spectrometric assay. Positive control was used BHA (butylated hydroxyanisole). Results : The total polyphenol contents and total flavonoid contents of the extract were 55.38 mg/TAEg, 513.72 mg/RUEg, respectively. Also, DPPH free radical scavenging capacity and reducing power of JMB ethanol extract in treated concentrations (100, 500, 1000, $5000{\mu}g/m{\ell}$) increased dose dependently. In particular, DPPH free radical scavenging capacity of JMB ethanol extract at 500, 1000, $5000{\mu}g/m{\ell}$ was similar to positive control (BHA) at high concentration (50, $100{\mu}g/m{\ell}$). ABTS free radical scavenging capacity of JMB ethanol extract at 500, 1000, 5000 ug/ml was similar to BHA at high concentration (50, $100{\mu}g/m{\ell}$). Also, reducing power was showed that JMB ethanol extract at $5000{\mu}g/m{\ell}$ was similar to BHA at high concentration (50, $100{\mu}g/m{\ell}$). Conclusions : These results suggest that JMB ethanol extract has effects to scavenge free radicals. Therefore, JMB has potential and applicable benefits for development of materials and products to have anti-oxidation functions.

목단피((牧丹皮).도인(桃仁) 배합(配合)이 항염증(抗炎症) 작용(作用)에 미치는 영향 (Effect of Water Extract of Peonia Suffruticosa and Prunus Percica on Anti-inflammation)

  • Paeonia Suffruticosa and Prunus Persica have been used as oriental medicine for removal of fever, alleviation of pain, an anti-phlogistic effect and removal of extravasated blood. However, it has been never shown the effects of these herbal medicines on anti-inflammatory processes. This experiment was performed to show how these herbs could act as anti-inflammatory medicines at cellular level. Anti-inflammation effects of water extracts from Paeonia Suffruticosa and Prunus Persica as well as their mixture have been investigated, and the results were follows; 1) each extract slightly suppressed the expression and production of inflammatory mediators and enzymes such as NO, iNOS, IL-$1{\beta}$, and TNF-$\alpha$ in lipopolysaccharid(LPS)-stimulated RAW264.7 cells and mouse primary peritoneal macrophages in a dose-dependent manner. These suppressive effects, however, were synergistically increased by their mixture. 2) Each extract of Paeonia Suffruticosa and Prunus Persica insignificantly suppressed the activation and activity of NF-${\kappa}B$ in LPS-stimulated RAW264.7 cells, which controls the expression of inflammatory mediators such as NO, iNOS, IL-$1{\beta}$, and TNF-$\alpha$. However, extract mixture of Paeonia Suffruticosa and Prunus Persica suppressed effectively the activation and activity of NF-${\kappa}B$. 3) Each of Paeonia Suffruticosa and Prunus Persica induced translocation of NF-${\kappa}B$ to the nucleus from the cytosol and DNA-binding activity of nuclear NF-${\kappa}B$ in LPS-activated RAW264.7 cells. The extract mixture of Paeonia Suffruticosa and Prunus Persica showed more significant suppression of the NF-${\kappa}B$ translocation and its DNA-binding activity, as compared to those of the each extract. These results suggest that the extract mixture of Paeonia Suffruticosa and Prunus Persica may affect different control mechanisms for NF-${\kappa}B$ activation and the expression and production of NF-${\kappa}B$-dependent inflammatory mediators, indicating that this extract mixture may be useful for treatment of inflammatory diseases.

  • Objective : This study was conducted to investigate anti-obesity effects of mixed-Powder(Raphani Semen, Coicis Semen, Poria, Plantaginis Testa & Theae folium)(HP) & mixed water extract(Citri Pericarpium, Crataegi Fructus, Maydis Stigma, Allium Skin) (HE) in mice fed a high-fat diet. Method : 50 Female ICR mice were divided into six groups; Normal group (Ve), high-fat diet group (Po), Po+HP(0.6g/kg) group(HP), Po+HE(1g/kg) group(HE), Po+(HP+HE) group(HPE), Po+(HP+HE)*2 group(DHPE). For 8 weeks, these groups were fed their respective diets. Body weight, liver weight and weights of adipose tissues, GOT/GPT index changes & Blood Glucose index changes were measured respectively. Lipid profiles in serum were analyzed by kit of blood. Results : Groups HP, HE, HPE, DHPE had significantly reduced body weight, liver weight, adipose tissue weights and GOT/GPT index, blood glucose index compared to the Po group. Also, serum triglyceride and total cholesterol, low density lipoprotein, were significantly reduced when compared to the Po group. Group G1 had significantly increased high density lipoprotein levels. Conclusion : Mixed powder (HP) & mixed water extract (HE) show anti-obesity effects in mice fed a high-fat diet.

알코올을 투여한 흰쥐의 알코올 대사에 미치는 한약재 혼합추출물의 영향 (The effect of herbal mixture on alcohol metabolism in Sprague Dawley rats)

  • Objectives : Alcohol hangover is a common phenomenon which basically occurs after heavy drinking. Moreover, heavy alcohol consumption leads to acute and chronic diseases. We investigated the effect of herbal mixture (SJ) on alcohol metabolism in serum or/and liver. Methods : 5W-old Sprague Dawley rats were used for the study. To overnight fasted rats, 0.9% saline or SJ extract was administrated per os before alcohol treatment. Then, 40% alcohol was orally administrated to all rats in 30 mins. Ethanol, acetaldehyde concentrations, alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH) activities were measured by assay kits. Aspartate aminotransferase (AST) and alanine transaminase (ALT) were measured by analyzer. Results : Ethanol and acetaldehyde concentrations lowered in SJ groups compared with CON group. Especially, acetaldehyde concentration significantly decreased in SJ groups compared with CON group. AST and ALT levels tended to increase in SJ groups compared with CON group but there was no significant difference between CON group and SJ groups. ADH activity in serum was higher in SJ groups than CON group but no significant difference in liver. ALDH activity in both serum and liver showed significantly increased in SJ groups compared with CON group. Conclusions : Treatment of SJ extract showed not only reducing concentration of alcohol and acetaldehyde but also increasing activities of ADH and ALDH. These results suggest SJ may influence in alcohol metabolism via control of metabolic enzyme activities and metabolite. Therefore, SJ, herbal mixture, might have a function of preventing hangover after drinking alcohol.

유백피, 토복령 추출물 혼합물의 항산화, 항염, 항균 및 난소세포 보호효과 (Antioxidant, anti-inflammatory, antibacterial and ovoprotective effects of mixture of Ulmi cortex and Smilacis rhizoma extracts)

  • Objectives : US extract is a mixture of each extract of Ulmi cortex and Smilacis rhizoma. In this study, we investigated the antioxidant, anti-inflammatory, antibacterial, and ovoprotective effects of US extract in in vitro model to identify potential candidates for improving female reproductive function. Methods : The antioxidant activity of US extract was measured using 1,1-diphenyl- 2-picrylhydrazyl free radical and superoxide anion radical scavenging assays. The anti-inflammatory effect of US extract on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells were determined with a nitric oxide (NO) assay, enzyme linked immunosorbent assays, and western blots analysis. The antibacterial activity of US extract against vaginitis infection microorganisms were determined with disc diffusion and minimum inhibitory concentration assays. The ovoprotective effect of US extract on 4-vinylcyclohexene diepoxide (VCD)-induced ovotoxicity in CHO-K1 cells were evaluated with a cell viability assay. Result : US extract showed good antioxidant capacity and inhibited LPS-induced NO production as well as iNOS and COX-2 expression and secretion of pro-inflammatory cytokine IL-6 without affecting the cell viability. It showed significant clear zones for Staphylococcus aureus and Candida albicans but did not indicate the clear zones for Escherichia coli and Enterococcus faecium. VCD-induced ovotoxicity in CHO-K1 cells was significantly reduced by US extract pre-treatment. Conclusions : These results demonstrate that US extract has antioxidant activity, anti-inflammatory effects on the LPS-stimulated macrophages, antibacterial activity against vaginitis infection microorganisms, and protective effects on the ovarian cells against VCD-induced ovotoxicity. These findings suggest that the US extract can be used as new prescriptions, supplements, functional foods, and cosmetics for improving female reproductive function.

천궁(川芎), 머위, 황연(黃連) 추출물 조성이 생쥐의 지질대사에 미치는 영향 (Anti-hyperlipidemic Effects of the Herbal Extract Mixture of Cnidium officinale, Petasites japonicus and Coptis chinensis on Mice with Tyloxapol)

  • This study was performed to evaluate the effect of the herbal extract mixture of Cnidium officinale, Petasites japonicus and Coptis chinensis (CPC) on the lipids metabolism in mice with tyloxapol. ICR mice weighing between 30-40 g were divided into four groups: normal group, 600mg/kg tyloxapol injected group, $50\;{\mu}g/g$ CPC treated group 6h after tyloxapol injection (SAM1), and tyloxapol and CPC treated group (SAM2), respectively. Tyloxapol or CPC was injected intraperitoneally. Tyloxapol caused an elevation of total cholesterol (TC), triglycerides(TG), and LDL-cholesterol and a decrease of HDL-cholesterol. In addition, tyloxapol induced accumulation of lipid including cholesterol in both the liver and kidney. Serum levels of TC, TG and LDL-cholesterol were decreased whereas HDL-cholesterol was increased by CPC. CPC increased in HDL-cholesterol /total cholesterol ratio and lowered atherogenic index. The levels of TC, TG and LDL-cholesterol by CPC were rather lower in SAM2 than SAM1. CPC also inhibited lowering HDL-cholesterol by tyloxapol. CPC reduced lipid blots and cholesterol particles in both the deposition and size in the liver and kidney with tyloxapol. These results suggest that CPC might be expected to be beneficial for protection and treatment of hyperlipidemia by the disturbance of lipid metabolism.

당뇨 쥐에 대한 한약재복합추출물로 만든 현미율무밥의 혈당강하 효과 (Hypoglycemic Effects of Boiled rice made from Unpolished rice, Job' tear, and Extract From Medicinal Herbs Mixture on Diabetic Rat)

  • Objectives : The purpose of this study was to evaluate boiled rice made from unpolished rice, Job's Tears, and extract from medicinal herbs mixture (HGMCJE) for hypoglycemic effect on STZ-induced diabetic rats. Methods : In the response of glucose tolerance, control and diabetic rats groups was intubated with glucose and HGMCJE. Furthermore, boiled 100% polished rice (P rice), boiled unpolished rice mixed with white rice and Job's tears (polished rice:unpolished rice:Job's tears=60:25:15, UPJ-rice), and UPJ rice made from HGMCJE (HUPJ-rice) were intubated to control and diabetic rats groups, respectively. Then, blood glucose concentration, incremental blood glucose, and area under the curve (${\Delta}AUC$) were analyzed in every experimental groups, and these data were used to evaluate glycemic response. Results : When glucose and p rice were intubated in control and diabetic rats, blood glucose concentration, incremental blood glucose, and incremental response ${\Delta}AUC$ of diabetic groups were significantly increase than control groups. But administraion of a single dose of extract from medicinal herbs mixture and HUPJ-rice in control and diabetic rats inhibited the remarkable increase the level of postprandial blood and ${\Delta}AUC$ at 60, 90, and 120 min Conclusions : These results indicate that when intubation of glucose and P rice were out of control on postprandial glycemic response in diabetic rats. But postprandial glycemic response was well-modulated by administrating a single oral dose of HGMCJE and HUPJ-rice. Therefore, HGMCJE can be developed as an effective hypoglycemic agent.
