한방 닭죽 개발 및 최적 배합비율 확립 (Development of Herbal Chicken Porridge and the Establishment of Optimizing the Mixing Ratio)

  • 죽의 기능성 향상을 위하여 2:1:1:1의 배합으로 혼합한 한약재료(가시오가피, 숙지황, 산약, 백복령)와 닭가슴살 등의 재료를 사용해 한방 영양죽을 제조하였다. 한방 닭죽의 DPPH radical 소거능, SOD 유사활성, 맛의 선호도, cost는 수학적인 canonical model과 trace plot을 이용하여 분석한 결과, SOD 유사활성 이외에는 각 성분들이 독립적으로 작용하여 상호작용이 없는 선형 모델을 보여주었다. DPPH radical 소거능, SOD 유사활성, 맛의 선호도를 최대로, cost를 최소로 목적 범위를 설정하고, 각각의 canonical 계수를 이용하여 수적 최적화를 통하여 최적 배합비를 구한 결과, 한약 혼합물 101 g, 닭가슴살 38.66 g, 찹쌀 비율 41.34 g의 최적 배합비를 구하였으며, 이 배합비에 따른 맛의 선호도는 6.39, DPPH radical 소거능은 17.25 mgVCE/${\ell}$, SOD 유사활성은 2.25%, cost는 1,382.39 won으로 나타났으며, 여러 결과들 사이에 최적화된 한 값에 수렴하는 desirability는 0.714로 나타났다. 이는 한약혼합물이 많을수록 기능성 면에서는 높게 나타나지만 맛의 선호도면에서는 낮게 나타냈고, 닭가슴살의 경우에도 그 함량이 많을수록 맛의 선호도는 올라갔지만 그에 반해 cost가 높게 올라간 것을 반영한 결과로 사료된다.

국내생산 허브의 마이크로웨이브 건조 후 이화학적 특성 변화 연구 (Changes in the physicochemical properties of various herbs cultivated in Korea after microwave drying)

  • 마이크로웨이브는 식품의 표면을 투과하여 식품 내부를 매우 신속하게 가열할 수 있으므로 식품의 맛, 냄새, 조직감, 영양가 등에 대하여 최소한의 영향을 미치게 된다(Chandrasekaran 등, 2013; Datta 와 Hu, 1992). 본 실험은 이러한 기능이 있는 마이크로파를 이용한 국내에서 생산가능한 허브 5종의 건조에 따른 품질특성에 있어서의 변화를 알아보고자 하였다. 비조사구와 100 Watt와 600 Watt에서 각각 계속적인 가열건조 방법과 Pulse 방법을 이용한 마이크로파 건조방법을 사용하였으며, 실험결과 조직감 유지 및 갈변화 방지에 매우 효과적인 것으로 나타났다. 또한 CW에 의한 건조 보다는 PW를 활용한 방법이 조직감 유지 및 갈변화 방지, 탈수율에 효과적인 것으로 나타났다. 특히 허브의 총균수 사멸에 있어서 마이크로파는 다른 일반가열보다 짧은 시간 내에 미생물의 사멸효과가 있었으며, CW보다 PW가 미생물의 사멸속도가 빠른 것으로 나타났다. 따라서 마이크로파를 이용한 허브의 건조시 낮은 온도에서의 빠른 건조 발생으로 시간적 향상과 물리적 변화를 최대한 억제시키며 미생물의 성장을 억제하는 작용을 하여 허브 특유의 냄새를 보존시키는 작용으로 앞으로 국내산 허브의 건조저장기술에 마이크로파가 다양하게 이용될 수 있을 것이라 기대된다.

옥총(玉葱) 첨가 조청의 제조 및 특성 (Preparation and Characterization of Jochung, a Grain Syrup, with Onion)

  • 본 연구는 다른 지역에 비해 일교차가 큰 철원지역에서 재배되고 있는 양파를 사용하여 생산농가에서 양파의 특성상 장기간 저장으로 인해 발생되는 부패의 단점을 보완하고 가공을 통해 농가수익을 증대화 할 수 있는 방안모색과 다양한 양파 가공제품의 확대의 목적으로 연구되었다. 특히, 발효 추출액의 경우 pH, EC, 당도 결정시에 중요한 것은 우리 몸의 흡수관계에 맞추어 조절하는 것이 중요하다. pH는 용액 중 수소이온(H+)의 농도를 나타내는 지표이다. 물 또는 수용액 중에 수소이온(H+)과 수산이온(OH-)은 항상 일정한 관계를 갖고 균형을 이루고 있으며 EC는 물 또는 수용액에 녹아있는 모든 염류 또는 이온의 농도를 나타내는 지표로 전기전도도가 있는데 전도율이라고도 한다. 특정성분을 물에 용해시킬 경우 양이온과 음이온으로 전리된다. 이온의 양이 많을수록 전기의 흐름이 용이해지므로 EC값이 커질수록 수용액 내의 이온의 농도와 총량이 많어지게 된다. 전기전도도 값으로 수용액 내의 이온의 총량을 추정 할 수 있어 EC값을 이용하여 수용액의 농도관리에 활용하는 것이 일반적이다. 88℃ 양파추출액의 pH는 5.1, EC는 3,970 ㎲/cm, 당도는 8.1 Brix로 측정되었다. 최근 건강에 대한 관심이 높아지므로 설탕이 많이 가미된 서구적인 식생활보다 전통적인 건강한 먹거리 문화가 관심을 받고 있으므로 조청의 맛, 영양성에다 양파의 기능성을 더하여 품질 향상의 웰빙 제품으로 안전성을 확보하고 소비자의 기호에 맞는 다양한 조청을 개발할 필요성이 있을 것으로 사료된다.

약사법상 한약제제의 정의 중 한방원리의 의미에 대한 고찰 (Implication of Korean Medicine Principles in Herbal Medicinal Preparations on Pharmaceutical Affairs Act)

  • Objectives : In Pharmaceutical Affairs Act, herbal medicinal preparations are defined as medicines made by Korean medicine principles. But in Act, Korean medicine principles are vague. Thus, there is a request to explain what the Korean medicine principles are. The aim of this study is to suggest implications of Korean medicine principles in definition of herbal medicinal preparations. Methods : With regard for definition of Korean medicine principles, we referred to the domestic and foreign regulations and literature about the history of herbal medicinal preparations. As a result, the meaning of Korean medicine principles was historically examined from various angles. Results : Through this study, we studied the Korean medicine principles from the past medical history. Due to the broad definition of Korean medicinal principles, we tried to extract general ideas of medicine principles first. We also found that we had scientific formulations based on korean medicine principles which could be used in modern society. In the end, we found that numerous medicine principles which include ‘Four qi and five flavors theory’, ‘Processing of medicinals’, ‘Yin and yang theory’, ‘Five phases theory’, ‘Meridian entry of viscera and bowels theory’, ‘Herb Couplet Interaction theory’, ‘Sovereign, minister, assistant and courier theory’, etc. Conclusions : Innumerable principles used from the past existed. These principles were broad and could be used as modern scientific principles. Based on these facts, we illustrated details of Korean medicine principles, hope this principles be widely shared and Korean herbal medicinal preparations be further developed.

지적재산권 보호를 위한 전통의학 DB 구축에 관한 고찰 (Grope for a Summary Program about a Traditional Medical Database for Intellectual Property Protection in WIPO)

  • Objectives : In the WIPO conference about the protection of intellectual property for traditional knowledge (TK) and genetic resources (GR), there was a demand concerning the necessity of recognizing the invention of TK and GR and the foundation of a related database. Methods : In the standard of the foundation of a database, the content and resource identification standards, the technological standards and the security and transmission standards were discussed. In a decision on intellectual property, it means the protection and recognition as a technology precedent of the materials of the database. Results : Massive material on traditional medical care technology in Korea has existed in many areas. However, it is hard to make a database because it is difficult to organize and find the data. In addition, a database demanded by WIPO is not only the material of images or texts of simple technology information, but also the use of patent decisions about traditional knowledge technology and the public sharing of information in related areas. A traditional medical care database would consist of the core of a subject and the related areas. The core part is the medical herb resource and Oriental clinical technology. Conclusions : These are necessary to detail the kind of medicines and prescriptions, their use, and their processing. In the related areas, it needs to detail PIAS and clinical data in regarding the industrialization. These DB have to protect the TK and the GR of Korea.

Plant regeneration from callus of Iris odaesanensis Y. N. Lee native to Korea via organogenesis

  • Iris odaesanensis Y. N. Lee. is an important endangered and native plant belonging to the family Iridaceae in Korea. This study describes a method for rapid micropropagation of this species via from leaf, rhizome and root explants derived calli. Leaf, rhizome and root explants were cultured on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxy acetic acid (2,4-D) for callus induction. Rhizome explants yielded calli at a frequency of 72% when cultured at 1.0 mg/l 2,4-D. Calli were maintained at 1.0 mg/l 2,4-D. These calli were transferred to MS medium supplemented with 0, 0.5, 1.0, and 2.0 mg/l 2,4-D in combination with 0, 0.5, 1.0, and 3.0 mg/l BA for adventitious shoot induction. The highest number of adventitious shoot (228.9 per petri-dish) were formed at 1.0 mg/l 2,4-D and 1.0 mg/l BA. WPM medium was the best to convert calli into plantlets, where up to 98.2% of calli were regenerated into plantlets. This in vitro propagation protocol should be useful for conservation of this endangered plant.

데이터베이스를 이용한 식료찬요(食療纂要)의 통계적 연구 (A Statistical Study on Sikryo-chanryo by Applying Database)

  • 본 연구의 목적은 한국 고유의 전통지식에 기반하여 음식을 이용한 건강증진 방법에 대한 지식을 체계화하고 통계적으로 활용할 수 있는 방법에 대하여 연구하는 데 있다. 이를 위하여 한국 고유의 음식을 이용한 질병치료에 대하여 언급한 고문헌인 식료찬요(食療纂要)의 지식을 DB로 구축하였다. 데이터의 처리는 Relational Data Model을 이용하였고, 9개의 데이터 테이블을 사용하였다. 소프트웨어는 MS사의 Access 2014를 사용하였다. 그 결과, 데이터베이스 쿼리문장을 이용하여 식재료, 약재별, 증상, 병명, 효능, 변증 정보를 유기적으로 조건을 만들어 통계적 결과를 즉시 확인할 수 있었다. 따라서 이러한 Relational Data Model을 이용하여 기존 방식의 연구논문을 대체할 수 있었다.

방사선 조사 당귀(Angelica gigas)의 면역활성 안정성 (Stability in Immunomodulation Activity of Irradiated Angelica gigas Nakai)

  • Angelica gigas Nakai (danggui) is a popular herb which has been used as a blood-building decoction for recovery from weakness in the Chinese medicine. Its demand increased in functional foods and pharmaceutical industries. For its hygiene, fumigation has been used, but the use of fumigants are going to be prohibited for food processing. In order to investigate gamma irradiation technique for hygiene of danggui, the immunomodulation activity of danggui after irradiation was examined. The water extract of irradiated danggui showed a strong mitogenic effect on splenocytes in vitro to the same level of lipopolysaccharide (LPS) and phytohemagglutinin (PHA). The effect was not different from that of non-danggui. It was tested whether there was any difference between irradiated and non-irradiated danggui in effects on the secretion of antibodies and graft versus host reaction in vivo. It turned out that intraperitoneal (i.p.) administration of the extract of irradiated danggui for 4 days remarkably increased the number of antibody-secreting cells in mice injected with sheep red blood cells (SRBC). Splenomegaly, due to graft versus host reacton, was also increased after 7 days i.p. administration of the extract of danggui in mice injected with allogeneic splenocytes. In these two in vivo test, the effect were not different from those of non-irradiated danggui. These results indicated that immunomodulation activity of danggui might be preserved after irradiation. In the other experiments (data not shown), the irradiated danggui was stable in active component analysis and safe in genetic toxicity test. In further research, the stability in other physiological activity of irradiated danggui will have to be proved before practical application of irradiation for hygiene.

Ginsenoside Rg1 modulates medial prefrontal cortical firing and suppresses the hippocampo-medial prefrontal cortical long-term potentiation

  • Background: Panax ginseng is one of the most commonly used medicinal herbs worldwide for a variety of therapeutic properties including neurocognitive effects. Ginsenoside Rg1 is one of the most abundant active chemical constituents of this herb with known neuroprotective, anxiolytic, and cognition improving effects. Methods: We investigated the effects of Rg1 on the medial prefrontal cortex (mPFC), a key brain region involved in cognition, information processing, working memory, and decision making. In this study, the effects of systemic administration of Rg1 (1 mg/kg, 3 mg/kg, or 10 mg/kg) on (1) spontaneous firing of the medial prefrontal cortical neurons and (2) long-term potentiation (LTP) in the hippocampal-medial prefrontal cortical (HP-mPFC) pathway were investigated in male Sprague-Dawley rats. Results: The spontaneous neuronal activity of approximately 50% the recorded pyramidal cells in the mPFC was suppressed by Rg1. In addition, Rg1 attenuated LTP in the HP-mPFC pathway. These effects were not dose-dependent. Conclusion: This report suggests that acute treatment of Rg1 impairs LTP in the HP-mPFC pathway, perhaps by suppressing the firing of a subset of mPFC neurons that may contribute to the neurocognitive effects of Rg1.

Comparative study of Korean White Ginseng and Korean Red Ginseng on efficacies of OVA-induced asthma model in mice

  • Background: Korean ginseng is a well-known medicinal herb that has been widely used in traditional medicine to treat various diseases, including asthma. Ginseng can be classified as white ginseng (WG) or red ginseng (RG), according to processing conditions. In this study, the authors compared the efficacies of these two ginseng types in a mouse model of acute asthma. Methods: To produce the acute asthma model, BALB/c mice were sensitized with ovalbumin (OVA) and aluminum hydroxide, and then challenged with OVA. WG and RG extracts were administered to mice orally. The influences of WG and RG on airway hyperresponsiveness (AHR), immune cell distributions in bronchoalveolar lavage fluid (BALF), and OVA-specific immunoglobulin E (IgE), IgG1, and IgG2a in serum were investigated. Cytokine production by lymphocytes isolated from peribronchial lymph nodes and histopathological changes was also examined. Results: In OVA-sensitized mice, both WG and RG reduced AHR and suppressed immune cell infiltration in bronchoalveolar regions. BALF OVA-specific IgE levels were significantly lower in RG-treated OVAsensitized mice than in the OVA-sensitized control group. WG and RG also suppressed inflammatory cytokine production by peribronchial lymphocytes. Histopathological findings showed reduced inflammatory cell infiltration and airway remodeling (e.g., epithelial hyperplasia) in WG- and RG-treated OVA mice compared with OVA controls. Conclusion: In this study, WG and RG showed antiasthmatic effects in an OVA-sensitized mouse model, and the efficacies of RG were found to be better than those of WG.
