당뇨환자를 위한 새로운 식이요법제에 관한 연구 (Studies on a New Alimentotherapy for Diabetic Patients)

  • 상엽과 상심자, 구기자, 인삼 등은 당뇨에 효능이 있는 것으로 널리 알려진 약재들이다. 본 실험에서는 기본 실험을 통하여 상엽, 상심자, 구기자, 인삼, 차가버섯의 최적 배합비와 최적의 추출방법으로 엑기스 형태로 만들어진 소당엑스$^{TM}$와 이를 특허 출원한 방법으로 백미에 코팅한 소당미$^{TM}$를 사용하여 시험한 결과 Alloxan으로 당뇨를 유도한 마우스, 정상인, 그리고 당뇨환자 모두에서 혈당이 저하되었음을 확인하였다. 따라서 소당엑스$^{TM}$와 소당미$^{TM}$는 당뇨환자의 혈당 조직 및 합병증 예방에 효과적이므로 당뇨식이요법제로서 이용될 수 있는 건강 식품으로 사료된다.

Effects of Garlic and Thyme Extracts on Growth Performance and Carcass Characteristics of Broiler Chicks

  • A total of 800 one-day-old chickens (Cobb 500) were obtained from a local hatchery and allocated randomly to five dietary treatments. This study conducted for 42 d. Treatment groups included 1) CON (control), 2) T0.3 (0.3% of thyme extract), 3) T0.6 (0.6% of thyme extract), 4) G0.3 (0.3% of garlic extract) and 5) G0.6 (0.6% of garlic extract). Thyme extract included $90{\mu}g/ml$ Carvacrol and 400 mg/ml thymol while garlic extract included 100 mg/ml elastin. There were no significant effects among the treatments on weight gain, feed intake and feed conversion ratio over the entire trial. Also, no significant differences were observed between CON group and other treatments in plasma parameters (cholesterol, triglyceride and HDL) and hematocrit. Sex of chickens had no effect on blood parameters. Supplementation of herb extracts did not affect carcass characteristics including carcass yield, breast, thigh and abdominal fat. There were no significant differences observed in carcass characteristics between males and females fed with different levels of herb extracts. In conclusion, supplementation of garlic and thyme extracts as feed additives at two levels of 3 and 6% in the present study did not improve performance and carcass traits of broiler chicks.

한방 소갈약 추출물 및 복합물에 의한 In vivo 및 In vitro 지질과산화 저해효과 (In vivo and In vitro Anti-lipid Peroxidative Effect of the Extract Complex of Korean Anti-thirst Drugs)

  • In Oriental medicine, the prescriptions composed of several herb medicines have been used. It is still unclear how the sum of several extracts of anti-thirst drugs represents the anti-lipid peroxidative action. Three anti-thirst herb medicines, Kalopanax pictus (K), Pueraria thunbergiana (P) and Rhus verniciflua (R), were extracted with MeOH and $H_2O$, respectively, and the former one was fractionated into the resultant EtOAc extract. Each extract was reconstituted to give KPR311, KPR131 and KPR113 where, for example, KPR311 represents the complex of K-P-R {3:1:1 (w/w/w)} of the three extracts. The order of the inhibitory effect in bromobenzene-induced lipid peroxidation in rats was as follows: EtOAc extract>$H_2O$ extract>MeOH extract. Extract complexes were found to be more potent than the extracts of individual crude drugs. The KPR131 of EtOAc extract was found to be the most potent among the tested samples. These anti-lipid peroxidative effects were also supported by the decrease of aniline hydroxylase activity and aminopyrine N-demethylase activity, on the other hand by the increase of epoxide hydrolase activity. All the tested samples were assayed in vitro antioxidative effects such as DPPH assay, ADP/NADPH/Fe$^{3+}$ assay and ascorbic acid/Fe$^{2+}$ assay. The EtOAc extracts also showed the most significant antioxidative effects. These results suggest that the sum of anti-thirst drugs could reflect the effects of respective crude drugs.s.s.

3가지 약용 허브 추출물에 함유된 유기산 검색 및 조성 비교 (The Screening and Pattern Comparison of Organic Acids in 3 Kinds of Medicinal Herbal Extracts)

  • Gas chromatography를 이용한 profiling과 신속한 동정을 허브류에 존재하는 유기산의 분석에 적용하여 보았다. 흡착제로 Chromosorb P와 용출 용매로 diethyl ether를 사용하는 고체상 추출법을 이용하여 허브추출물로부터 유기산을 분리한 후, tert-butyldimethylsilyl 유도체로 만들어 극성이 다른 DB-5와 DB-17 컬럼으로 동시분석하였다. 두 컬럼에서 분리된 유기산 피크들을 dual capillary column system의 retention index(RI) library 탐색과 RI specta 비교로 신속하고 정확하게 동정할 수 있었으며 이들을 GC-MS로 확인하였다. 허브추출물들의 유기산 분석 결과 fatty acid, dioic acid, hydroxy acid, aromatic acid등 24가지의 유기산이 동정되었고 각 허브의 종류에 따라 특징적인 유기산 GC-profile을 나타내었다. 각 허브추출물의 GC profile의 유기산 조성만을 단순화시킨 RI spectra로 표현한 결과, 각 허브의 유기산 패턴의 분별이 매우 용이하였다.

한약재발효액의 항생제 다제내성균에 대한 항균활성 및 항산화활성 (Antibacterial Activities of Fermented Sayuksan Ingredient Extracts for Multidrug-resistant Strains)

  • Sayuksan has been widely applied to treat a variety of diseases such as acute hepatitis, gastritis, and colitis. Sayuksan consists of medicinal herbs such as Glycrrhizae uralensis Fischer, Paeonia lactiflora Pallas, Bupleurum falcatum Linne, and Poncirus trifoliata Rafinesqul. Methanol extracts (1 mg/mL) from the four kinds of medicinal herbs did not show antibiotic activities against general test strains and multi-drug resistant strains. The antibacterial activity of fermented medicinal herbs extracts with Lactobacillus spp. strain was confirmed as Gram-positive bacteria which are higher than Gram-negative bacteria. Extracts of Glycrrhizae uralensis Fischer fermented with Lb. casei KCTC 3109 displayed inhibitory diameters of 16 mm against Pseudomonas aeruginosa P01828. Superoxide dismutase (SOD)-like activity of the medicinal herb extracts was not determined, but the extract of Paeonia lactiflora Pallas fermented with six strains of Lactobacillus spp. had the highest antioxidant activity. SOD-like activity of Paeonia lactiflora Pallas extracts fermented by Lb. brevis KCTC 3498 was $41.4{\pm}0.8%$, which was the highest antioxidant activity among the fermented extracts with the other medicinal herbs.

황금, 산조인, 백출 추출물의 생리활성 (Biological Activities of Extracts from Scutellaria baicalensis, Zizyphus jujuba and Atractylodes macrocephala)

  • Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of extracts from Scutellaria baicalensis, Zizyphus jujuba and Atractylodes macrocephala. Methods : Three kinds of medicinal herbs were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidant activity and growth inhibition activity against cancer cells. Results : Electron donating abilities (EDA) of the water and ethanol extract from Zizyphus jujuba and Atractylodes macrocephala were over than 87% at 1,000 ${\mu}g/mL$, respectively. The highest nitrite scavenging abilities (NSA) of the water and ethanol extracts from Scutellaria baicalensis were 68.9 and 79.2% at 1,000 ${\mu}g/mL$, respectively. Atractylodes macrocephala extracts had shown the highest growth inhibition activity against both of MDA and A549 cells and the water extract had higher activity than ethanol extract. Conclusions : These results suggest that the extracts of medicinal herbs used for this experiment had specific biological activity and they can be used as natural antioxidant to prevent oxidative damage in normal cells.

길경, 황기와 오미자 혼합추출물의 NO 억제활성과 Hyaluronidase 억제활성 효과 (Inhibition of Nitric Oxide Production and Hyaluronidase Activities from the Combined Extracts of Platycodon grandiflorum, Astragalus membranaceus, and Schisandra chinensis)

  • 본 연구에서는 천연식물자원으로부터 항염증, 항산화 및 항관절염에 활성을 갖는 유용한 물질을 검색하기 위해 문헌 조사 및 기존의 연구결과를 통하여 선정된 53가지의 생약재로부터 NO와 HAse(hyaluronidase)의 억제활성 여부를 측정하였다. 측정 결과 억제효과가 우수하다고 확인된 황기(Astragalus membranaceus)와 오미자(Schisandra chinensis), 길경(Platycodon grandiflorum)을 최종 시료로 선정하였고, 이를 단독으로 사용하기보다는 유효성분의 항염증 및 항산화 효능에 대한 상승효과를 알아보기 위하여 혼합추출물의 최적조건을 확인하였다. 각 소재들을 1:2:1로 혼합한 혼합추출물에서 다양한 추출조건에 따라 단일 생약재 추출물에 비해 2배에서 4배까지의 수율 증가를 보였으며, NO 억제활성 효과도 증가한 것을 확인할 수 있었다. 수율과 NO 억제활성을 고려할 경우 혼합추출물의 최적 추출 조건은 황기, 오미자, 길경의 혼합비율 1:2:1, 추출시간 12시간, 추출온도 $30^{\circ}C$, 혼합 속도 50 rpm, 추출용매로 ethyl acetate이었다. 위의 최적 조건을 적용한 혼합추출물은 HAse 저해 활성 실험에서 최초 추출물(not grinded, 95% EtOH, 24시간, 실온, 0 rpm)보다 약 16% 정도의 저해 상승효과가 나타났다. 이는 길경과 황기, 오미자의 상승작용 때문으로 사료된다. 이를 통해 황기와 오미자, 길경의 혼합추출물이 항염증, 항산화 및 항관절염에 활성을 갖는 유용한 물질로 사용 가능할 것이라 사료된다.

국내 천연물 항 당뇨 실험연구의 체계적 논문 고찰 - 2000년 ~ 2010년 (Systematic Reviews of Current Domestic Studies of Herbaceous Plants on Anti-diabetes - since 2000)

  • This study tried to integrate the traditional oriental medical theories and results of experimental studies of herbaceous plants on anti-diabetes. And I tried to analyze recent experimental study trend on the anti-diabetic herb. I searched anti-diabetic herb studies on 4 korean databases and 10 korean journals by keywords, 'diabetes', 'blood glucose', 'glycometabolism', 'pancreatic ${\beta}$-cell', etc. In order to see detail review, searching was performed from 2000 to 2010. And I searched 125 study cases concerning anti-diabetic herb and 72 varieties herbaceous plants used in study of anti-diabetes. and I analyzed the choice motives of each herb for anti-diabetic study, the extract methods and anti-diabetic evaluation contents. And I analyzed anti-diabetic herbs from a traditional oriental medical point of view. When the researchers chose herb for anti-diabetic experiment, just 8.8% of the choice was based on the oriental medical evidences. I found that 60.6% of the herb shown to be effective in diabetes experimentally had oriental medical theory-based Properties(性). There were studies with whole plants(16.8%), aqueous extract(45.6%), methanol extract(8.0%), ethanol extract(8.0%) and comparative studies of more than 2 types of extracts or various fractions(18.4%). The most frequent experimental diabetic models was diabetic mouse induced by streptozotocin(STZ)(87.8%). And there were db/db mouse(6.7%), ob/ob mouse(1.1%), etc. 33.6% of all studies just measured hematological indices of diabetes, and 66.4% researches analyzed details. To improve herbaceous plants study on diabetes, we oriental medical scientists have to integrate the oriental medical theories and results of experimental studies.
