Effect of Dietary Lipid Level and Herb Mixture on Growth of Parrot Fish, Oplegnathus fasciatus (사료 지질 및 한방제 첨가가 돌돔의 성장에 미치는 영향)

  • A feeding trial was carried out to Investigate the effect of lipid level and herb mixture in the diets on growth of juvenile parrot fish, Oplegnathus fasciatus. Two plicate groups of fish averaging 4.5 g were fed four experimental diets containing herb mixture (0 and 5 g/kg diet) at each of two lipid levels $(8\%\;and\;16\%)$ for 8 months from summer to spring (water temperature, $7.2-25.4^{\circ}C)$. Weight gain improved with increasing dietary lipid level or supplementation of herb mixture during the first 4 months feeding period. Weight gain of fish fed the diet containing $16\%$ lipid level was significantly higher (P<0.05) than that of fish fed the diet containing $8\%$ lipid level. Weight gain of fish fed the diet with herb mixture was significantly higher (P<0.05) than that of fish fed the diet without herb at the $16\%$ lipid level. Feed efficiency was influenced by dietary lipid level or herb mixture during the first 4 months. On the other hand, weight gain and feed efficiency were not affected by dietary lipid level and herb mixture after 4 months until end of feeing period. Survival of fish fed the diet containing $8\%$ lipid without supplementation of herb mixture was the lowest among the groups (P<0.05). Lipid contents of liver and viscera tended to increase with increasing dietary lipid level during the first 4 months feeding period, however muscle lipid content was not influenced by dietary lipid and herb. Total cholesterol of serum was influenced by dietary lipid level during the first 4 months feeding period. These results indicate that an increase of dietary lipid level from $8\%\;to\;16\%$ and supplementation of herb mixture can improve growth performance of juvenile parrot fish when water temperature is optimum for growth such as the summer season in Korea.

Molecular authentication of Lepidii seu Descurainiae Semen by the development of matK amplification primers and analysis of sequences (matK 증폭용 primer 개발 및 염기서열 분석을 통한 정력자(葶藶子) 유전자 감별)

  • Objectives : Lepidii seu Descurainiae Semen has been frequently adulterated with the seeds of several inauthentic plant species. However, the accurate identification of these plant seeds is very difficult. To develop a reliable genetic authentication tool for Lepidii seu Descurainiae Semen, we analyzed matK sequence. Methods : To obtain the matK sequences of plant materials, genomic DNA was extracted from 24 samples and PCR amplification was carried out using matK-AF/matK-8R universal primer set and matK-LDSF/matK-LDSR primer set. For identifying species-specific nucleotides and phylogenetic analysis, matK regions were sequenced and comparatively analyzed by the ClustalW and Maximum Likelihood method. Results : We developed a new primer set to amplify matK region in Lepidii seu Descurainiae Semen and closely related plant samples. From the comparative analysis of matK sequences, we identified species-specific marker nucleotides for D. sophia, L. apetalum, L. latifolium, E. cheiranthoides, E. macilentum, and D. nemorosa, respectively. Furthermore, phylogenetic analysis revealed clear classification depending on the species. These results indicated that the matK sequence obtained a new primer set in this study was useful to identify Lepidii seu Descurainiae Semen in species level. Conclusions : We developed a primer set and identified species-specific marker nucleotides enough to distinguish authentic Lepidii seu Descurainiae Semen and adulterants at the species level based on the matK sequences. These genetic tool will be useful to prevent adulteration and to standardize the quality of Lepidii seu Descurainiae Semen.

  • Sixteen kinds of Chinese herb drugs using in digestive system have screened for antibacterial activity. It was revealed that all of the herb drug extracts showed antibacterial activity in one or more strains of microbials selected in this experiments. It is noteworthy that Aconitum trilobum showed inhibition of Klebsiella pneumoniae.

A Basic Study of Muan Onion Kimchi with Herb (무안양파김치 개발을 위한 기초연구)

  • In this study, physical properties and color were measured and sensory evaluation was conducted on the onion kimchi with herb. Onion was produced during the winter season at Muan area. It means that water content of the onion was least of all the seasons. Onion kimchi was made with various herbs and various amounts of herb was added to the onion kimchi for the selected herb. Sensory evaluation indicated that Rosemary was best of all the tested herbs. Adequate amounts of Rosemary was 1 and 2%, while 5% was not evaluated as good.
