This study was carried out to investigate the supplementary effects of ${\gamma}$-oryzanol extracted from rice bran on lipid metabolism in diabetic mice. We supplied 2 kinds of experimental diets (CO without and GO with 0.2% ${\gamma}$-oryzanol) to diabetic mice for 8 weeks. Diet intake, body weight, organ weight, contents of serum and hepatic lipid profiles, and fecal lipid levels were measured. Though there was no significant difference in diet intake, body weight change and organ weight between experimental groups, the concentration of serum total cholesterol and hepatic total lipid, total cholesterol and HMG-CoA reductase activity was significantly lower in GO group treated with 0.2% ${\gamma}$-oryzanol of diet than CO group after supplementary period of experimental diets. And total lipid, triglyceride, total cholesterol, and bile acid levels excreted to feces were significantly higher in GO group than CO group. These results suggest that ${\gamma}$-oryzanol decrease the serum and hepatic lipid levels by lowing HMG-CoA reductase activity or increasing the contents of fecal lipid in diabetic KK mice.
Objectives: The objective of the present study was to investigate the lipid and the antioxidant regulatory potential of a multigrain diet in laboratory animals with reference to lipid profiles, tissue lipid peroxidation and antioxidant status. Methods: Two types of diets, with or without addition of cholesterol, were used in the study - a commercial diet and a formulated multigrain diet (with Sorghum vulgare, Avena sativa, Pennisetum typhoideum, Oryza sativa, Eleusine coracana and Zea mays grains). After a 10-week period of feeding the diets to albino rats the plasma, liver and fecal lipid profiles and the hepatic and renal antioxidant status of the animals that were fed the commercial and the formulated diets (with and without cholesterol addition) were assessed. Results: The commercial diet supplemented with cholesterol elevated the levels of plasma total lipids, total cholesterol, triglycerides, low-density lipoprotein cholesterol (LDL-C), and very low-density lipoprotein cholesterol (VLDL-C), as well as the atherogenic index (AI). The high-density lipoprotein cholesterol (HDL-C) content and the antioxidant profiles (total ascorbic acid, superoxide dismutase, catalase, glutathione peroxidase reduced glutathione) declined along with increases in lipid peroxidation. The formulated diet (with and without addition of cholesterol) was found to be more efficient than the commercial diet in controlling plasma, hepatic and fecal lipid profiles, as well as hepatic and renal lipid peroxidation and antioxidant status, than of the hypercholesteremic animals. Conclusion: The multigrain diet used in the present study is effective in countering the hyperlipidemia and oxidative stress caused by high cholesterol intake.
콜레스테롤을 0.5% 수준으로 첨가하여 고콜레스테롤혈증을 유발시킨 상태에서 0.2% 클로로겐산을 병합 투여한 식이를 흰쥐에게 2주간 자유 급여시켜 혈청과 간장의 지질농도, 혈당치 및 뇨 단백질량에 미치는 영향을 검토하였다. 동물 실험군은 콜레스테롤과 클로로겐산을 무첨가한 정상 식이군, 콜레스테롤을 0.5% 수준으로 첨가한 콜레스테롤 식이군 및 콜레스테롤 식이군에 클로로겐산을 0.2% 수준으로 첨가한 클로로겐산 병합투여군으로 나누었다. 정상 식이군에 비해 콜레스레롤 식이군에서 혈청 총 콜레스테롤 농도의 유의적 증가는 클로로겐산 병합투여군에서 유의적으로 감소되었으며, 동시에 동맥경화 지수도 감소되었다. 간장의 지질 농도는 각 식이군에 의한 유의적인 차이는 없었다. 혈당치는 정상 식이군에 비해 콜레스테롤 식이군 및 클로로겐산 병합투여군에서 증가되었다. 뇨 단백질양은 정상 식이군과 콜레스테롤 식이군 간에는 유의적인 차이가 없었으나, 클로로겐산 병합 투여군에서는 유의적으로 증가하였다 이상의 결과로부터, 클로로겐산은 콜레스테롤 식이에 의한 고콜레스테롤혈증을 개선시키고 동맥경화 지수도 감소시켰다.
There is an increasing interest in curcumin (Curcuma longa L.) as a cardiovascular disease (CVD) protective agent via decreased blood total cholesterol and low-density lipoprotein-cholesterol (LDL-cholesterol) level. The aim of this study was to investigate further the potential mechanism in the hypocholesterolemic effect of curcumin by measuring cholesterol 7a-hydroxylase (CYP7A1), a rate limiting enzyme in the biosynthesis of bile acid from cholesterol, at the mRNA level. Male Sprague-Dawley rats were fed a 45% high fat diet or same diet supplemented with curcumin (0.1% wt/wt) for 8 weeks. The curcumin diet significantly decreased serum triglyceride (TG) by 27%, total cholesterol (TC) by 33.8%, and LDL-cholesterol by 56%, respectively as compared to control group. The curcumin-supplemented diet also significantly lowered the atherogenic index (AI) by 48% as compared to control group. Hepatic TG level was significantly reduced by 41% in rats fed with curcumin-supplemented diet in comparison with control group (P < 0.05). Conversely, the curcumin diet significantly increased fecal TG and TC. The curcumin diet up-regulated hepatic CYP7A1 mRNA level by 2.16-fold, compared to control group p (P < 0.05). These findings suggested that the increases in the CYP7A1 gene expression may partially account for the hypocholesterolemic effect of curcumin.
본 연구는 새송이 버섯을 첨가하여 만든 어묵이 체내 지질 대사와 효소 활성에 미치는 영향을 알아보고자 수컷 Sprague-Dawley 흰쥐 30마리를 대상으로 고콜레스테롤(1%)첨가 기본식이, 30% 일반어묵 첨가 식이와 40% 새송이 버섯 첨가 오징어 어묵 식이를 급여하였다. 평균 식이 섭취량은 대조군, 일반 어묵군, 새송이 버섯 첨가 오징어 어묵군에 따른 각 군 간의 유의적인 차이는 볼 수 없었다. 체중 증가량과 식이효율은 대조군에 비해 실험군 모두 유의적으로 감소하는 경향을 보였다. 혈장 중성지질과 총 콜레스테롤, 동맥경화지수(A.I), GOT, GPT 수준은 대조군이 새송이 버섯 첨가 오징어 어묵군에 비해 유의적으로 높게 나타났다. HDL-콜레스테롤의 경우 새송이 버섯 첨가 오징어 어묵군이 대조군과 일반 어묵군보다 낮게 나타났고, 간조직의 콜레스테롤 및 중성 지방 농도는 유의적으로 차이가 없는 것으로 나타났다. 혈장과 간조직 TBARS 수준은 대조군보다 새송이 버섯 첨가 오징어 어묵군에서 유의적으로 낮은 수치를 보였다. 항산화 효소 활성도에서 SOD활성도는 대조군과 일반 어묵과의 유의적인 차이가 없었으며, CAT활성도는 대조군보다 새송이 버섯 첨가 어묵이 유의적으로 높게 나타났으며. GPx활성도 역시 유의적으로 높게 나타났다. 이 결과들을 종합해 보면, 고콜레스테롤혈증 흰쥐에서의 새송이 버섯 첨가 오징어 어묵은 전반적인 지질대사 개선과 함께 SOD, CAT, GPx와 같은 항산화 효소의 활성을 증가시켜 항산화 방어계에도 긍정적인 효과를 나타냄으로써 앞으로 고콜레스테롤 혈증 예방 등의 혈관 순환기 계통 질병을 예방하기 위한 식품으로써의 활용이 기대된다.

Background: The renal function of individuals is one of the reasons for the variations in therapeutic response to various drugs. Patients with renal impairment are often exposed to drug toxicity, even with drugs that are usually eliminated by hepatic metabolism. Previous study has reported an increased plasma concentration of indoxyl sulfate and decreased plasma concentration of $4{\beta}$-hydroxy (OH)-cholesterol in stable kidney transplant recipients, implicating indoxyl sulfate as a cytochrome P450 (CYP) inhibiting factor. In this study, we aimed to evaluate the impact of renal impairment severity-dependent accumulation of indoxyl sulfate on hepatic CYP3A activity using metabolic markers. Methods: Sixty-six subjects were enrolled in this study; based on estimated glomerular filtration rate (eGFR), they were classified as having mild, moderate, or severe renal impairment. The plasma concentration of indoxyl sulfate was quantified using liquid chromatography-mass spectrometry (LC-MS). Urinary and plasma markers ($6{\beta}$-OH-cortisol/cortisol, $6{\beta}$-OH-cortisone/cortisone, $4{\beta}$-OH-cholesterol) for hepatic CYP3A activity were quantified using gas chromatography-mass spectrometry (GC-MS). The total plasma concentration of cholesterol was measured using the enzymatic colorimetric assay to calculate the $4{\beta}$-OH-cholesterol/cholesterol ratio. The correlation between variables was assessed using Pearson's correlation test. Results: There was a significant negative correlation between MDRD eGFR and indoxyl sulfate levels. The levels of urinary $6{\beta}$-OH-cortisol/cortisol and $6{\beta}$-OH-cortisone/cortisone as well as plasma $4{\beta}$-OH-cholesterol and $4{\beta}$-OH-cholesterol/cholesterol were not correlated with MDRD eGFR and the plasma concentration of indoxyl sulfate. Conclusion: Hepatic CYP3A activity may not be affected by renal impairment-induced accumulation of plasma indoxyl sulfate.
The study was designed to observe the effect of dietary calcium and fats on plasma cholesterol level, hepatic microsomal fluidity and HMG-CoA reductase activity as well as the excretion of fecal bile acids and neutral sterols in 1, 2-dimethylhydrazine(DMH)-treated rats. Male Sprague Dawley rats, at 7 weeks of age, were divided into 2 groups, 0.3% and 1.0% Ca levels and each group again subdivided into 2 groups of corn oil and perilla oil. Each rat was intramuscularly infused with DMH for 6 weeks to give total dose of 180mg/kg body weight and also fed experimental diet containing 15%(w/w) different fit and Ca(0.3% or 1.0%) for 20 weeks. High dietary calcium(1.0%) did not significantly influence on plasma cholesterol as well as hepatic microsomal fluidity and HMG CoA reductase activity, but significantly reduced the excretion of total bile acid per gram of faces and increased the excretion of total neutral sterol. However, high dietary Ca reduced the excretion of secondary bile acid(deoxycholic and lithocholic acids) which was known as promoter for colon cancer. Perilla oil rich in n-3 ${\alpha}$-linolenic acid significantly decreased plasma cholesterol by increasing hepatic microsomal fluidity compared with corn oil, but did not influence on HMG CoA reductase activity. Perilla oil did not influence on fecal excretion of total and primary bile acids, but reduced the excretion of secondary bile acids. Therefore, it could be recommended to consume more fish product and food rich in calcium and use more perilla oil in meal preparation to prevent from coronary hear disease and colon cancer especially when high fit diet has been practiced. BACKGROUND/OBJECTIVES: Nobiletin (NOB), a citrus flavonoid, is reported to have beneficial effects on cardiovascular and metabolic health. However, there is limited research investigating the effect of long-term supplementation with low-dose NOB on high-cholesterol diet (HCD)-induced hypercholesterolemia and non-obese nonalcoholic fatty liver disease (NAFLD). Therefore, we investigated the influence of NOB on hypercholesterolemia and NAFLD in HCD-fed mice. SUBJECTS/METHODS: C57BL/6J mice were fed a normal diet (ND) or HCD (35 kcal% fat, 1.25% cholesterol, 0.5% cholic acid) with or without NOB (0.02%) for 20 weeks. RESULTS: HCD feeding markedly reduced the final body weight compared to ND feeding, with no apparent energy intake differences. NOB supplementation suppressed HCD-induced weight loss without altering energy intake. Moreover, NOB significantly decreased the total cholesterol (TC) levels and the low-density lipoprotein (LDL)/very-LDL-cholesterol to TC ratio, and increased the high-density lipoprotein-cholesterol/TC ratio in plasma, compared to those for HCD feeding alone. The plasma levels of inflammatory and atherosclerosis markers (C-reactive protein, oxidized LDL, interleukin [IL]-1β, IL-6, and plasminogen activator inhibitor-1) were significantly lower, whereas those of anti-atherogenic adiponectin and paraoxonase were higher in the NOB-supplemented group than in the HCD control group. Furthermore, NOB significantly decreased liver weight, hepatic cholesterol and triglyceride contents, and lipid droplet accumulation by inhibiting messenger RNA expression of hepatic genes and activity levels of cholesterol synthesis-, esterification-, and fatty acid synthesis-associated enzymes, concomitantly enhancing fatty acid oxidation-related gene expression and enzyme activities. Dietary NOB supplementation may protect against hypercholesterolemia and NAFLD via regulation of hepatic lipid metabolism in HCD-fed mice; these effects are associated with the amelioration of inflammation and reductions in the levels of atherosclerosis-associated cardiovascular markers. CONCLUSIONS: The present study suggests that NOB may serve as a potential therapeutic agent for the treatment of HCD-induced hypercholesterolemia and NAFLD.

Guinea pigs were fed a hypercholesterolemic diet (0.25 g/100 g cholesterol) and randomly allocated either to a Control group (n = 9) or to a Lutein (0.1 g/100 g) group (n = 10) for 12 weeks to evaluate oxidative stress and inflammation in both liver and eyes. Malondialdehyde (MDA) concentrations and inflammatory cytokines were measured as well as hepatic nuclear factor-kappaB (NF-${\kappa}B$) binding. Lutein concentrations were greater in eyes ($P$ < 0.01) and liver ($P$ < 0.001) in the Lutein group. All guinea pigs had high concentrations of hepatic cholesterol as well as high plasma ALT and AST levels indicative of liver injury. However, the Lutein group had 43% lower hepatic free cholesterol than the Controls ($P$ < 0.05). Hepatic MDA and MDA in the eye were lower in the Lutein compared to the Control group ($P$ < 0.05). Hepatic tumor necrosis factor-${\alpha}$ was 32% lower in the Lutein group ($P$ < 0.05). Lastly, the Lutein group presented lower NF-${\kappa}B$ DNA binding activity than the Control group ($P$ < 0.001). These results suggest that in the presence of high cholesterol, lutein exerts both antioxidant and anti-inflammatory effects, which can be explained by attenuated NF-${\kappa}B$ DNA binding activity. Furthermore, results also suggest that lutein accumulates in the eyes of guinea pigs to protect against oxidative stress.
본 실험을 통해 뚜렷한 경향을 보여주는 몇가지 결과를 요약하면 다음과 같다. 식이vitamin E의 과량 첨가는 쥐의 혈청에 반영되어 나타났으며, 식이중의 cholesterol은 이 증가 효과를 감소시키는 경향을 보여주었다. 식이 cholesterol첨가로 인해 쥐의 혈청 cholesterol농도는 영향을 받지 않은 반면, 간의 cholesterol농도는 증가되었다. 과량 vitamin E와 cholesterol 두가지를 식이에 첨가하였을 경우 (IV-ECh IV'-ECh) 간의 cholesterol농도는 식이에 cholesterol만을 첨가한 경우 (III-Ch, III'-Ch)보다 더욱 높게 나타났다. 그리고 vitamin E만을 첨가한 II-E와 II'-E군에서는 간의 cholesterol농도를 낮추어주는 경향을 보였다. 간의 vitamin A 농도는 쥐가 성장함에 따라 감소하였으며 각 실험군 간에는 유의적인 차이를 보이지 않았으나, B군의 경우 과량 vitamin E 식이는 간 vitamin A농도를 감소시키는 경향으로 나타났다. Vitamin E, vitamin A, cholesterol 및 다른 여러 요소들간의 복잡한 상호관계를 알아내기 위해, age가 다른 그리고 여러 species의 동물을 이용한, 장기간의 단계적이고도 체계적인 많은 연구가 요구되어진다.

제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
