Hemoglobinemia is found in hemolytic anemia, paroxysmal nocturnal hemolobinuria and paroxysmal hemoglobinuria. Recently increasing use of extracorporeal circulation in cardiac surgery has stimulated the development of more precise methods for rapidly evaluating the formation and clearance of extracoporeal hemoglobin. A spectrophotometric method of analysis seemed to offer the best solution to the requirements. The method presented is on the basis of hemoglobin derivatives converted to cyanmethemoglobin, and the absorbance is measured in a spectrophotometer at 540 and 680 nm. The blank value ismeasured at 680 nm while the absorbance at 540 nm measures the hemoglobin derivatives. This method estimates all the hemoglobin derivatives such as oxyhemoglobin, carboxyhemoglobin, methemoglobin, sulfhemoglobin and hemialbumin. The method was tested in recovery experiments, which is given table 1. A good degree of correlation was obtained in a comparision with the method described by Crosby and Furth for non-hemolyzed serum. The spectrophotometric technique described offers many advantages in speed and simplicity over the chemical procedure.
Little work has been done on the changes in the blood picture occurring at different ages in cattle. In our country Kim (1963) and Jeong (1965) recorded the blood picture of mature Korean native cattle and, recently, Mun et al. (1974) has recorded the blood picture of Holstein cows. And a comprehensive survey of the blood picture of Korean native cattle has not been made. The object of the present investigation was to make good this deficiency, and to suggest standards for the blood picture of Korean native cattle at frequent intervals from birth to maturity. The cattle were kept under average farming conditions in this country. Observations were made at the following ages: at birth; 1, 2 and 4 days; 1, 2 and 4 weeks; 2,4,6 and 9 months; 1, 2 and 3 years of age. Blood samples were drawn from the jugular vein. Erythrocyte enumerations, concentration of hemoglobin in blood and hematocrit values were made in usual manner. Erythrocyte counts increased from $8{\times}10^6/mm^3$ during the first week to a level of $10{\times}10^6/mm^3$ at 2 months of age, showing steady state untill 6 months of age; it then fell to adult level ($8{\times}10^6/mm^3$) at 9 months of age. Concentration of hemoglobin in blood and hematocrit values were closely related to the changes of erythrocyte counts. The values increased from 10 g/100 ml and 31~35 ml/100 ml during the early life to 11 g/100 ml and 38 ml/100 ml at 2 months of age, showing steady state untill 6 months of age; these then fell to adult level (10 g/100 ml and 31~34 ml/100 ml) at 9 months of age for concentration of hemoglobin in blood and hematocrit values, respectively. Mean corpuscular volume and mean corpuscular hemoglobin showed a common pattern. The values were $42{\sim}43{\mu}m^3$ and 12.4 pg during the early life and fell a little to 6~9 months of age; these then increased to reach adult levels of $43{\mu}m^3$ and 12.6 pg for mean corpuscular volume and mean corpuscular hemoglobin, respectively. Mean corpuscular hemoglobin concentration was little affected with age.
This paper has studied the nutritional iron status of adults in the city of Taegu. The subjects of the experiment were twenty six men and women living in the city of Taegu. The iron intake was 26.1mg for men and 17.1mg for women, which were respectively 217.5% and 95% of RDA. For men heme iron was 5% and nonheme iron, 95% for women heme iron was 5.8% and nonheme iron, 94.2%. The absorption rate of dietary iron was 6.2% for men and 9.1% for women. The hemoglobin was 14.5g/dl for men and 11.9g/dl for women, and the rate of developing anemia was 8.3% in men and 57.1% in women. The serum ferritin was 57.8ng/ml with men, which was found to be normal, and 14.7ng/ml with women, which showed that 57.1% of women were in the condition of iron depletion or iron deficiency anemia. The rate of developing anemia resulting from the deficiency of serum ferritin was as high as that from the deficiency of hemoglobin. The correlation between iron intake based on the dietary record, and energy intake, vegetable protein intake and dietary fiber intake was found to be positive. So was the correlation between the iron intake, and hemoglobin, serum ferritin and urinary iron excretion. There was also a positive correlation between hemoglobin, and hematocrit and serum ferritin. The correlation between fecal iron excretion and serum ferritin was positive. Fecal iron excretion and urinary iron excretion can be complemented by iron intake but the fact should be considered that 95% of iron intake is nonheme iron, which is difficult to absorb. Based on the above-mentioned things, serum ferritin and hemoglobin can be used as the standards of measuring anemia. As women in their child bearing years show the high rate of 57.1% of developing anemia, the more thorough nutritional education of iron is required.
To elucidate the effect of gamma-irradiation on the molecular properties of hemoglobin, the secondary, tertiary structure, and the molecular weight size of the protein were examined after irradiation at 0.5, 1, 5, and 10 kGy. Gamma-irradiation of hemoglobin solutions caused the disruption of the ordered structure of the protein molecules, as well as degradation, cross-linking, and aggregation of the polypeptide chains. A SDS-PAGE study indicated that irradiation caused initial fragmentation of the proteins and subsequent aggregation due to cross-linking of the protein molecules. The effect of irradiation on the protein was more significant at lower protein concentrations. Ascorbic acid decreased the degradation and aggregation of proteins by scavenging oxygen radicals that were produced by irradiation. A circular dichroism study showed that irradiation decreased the helical content of hemoglobin with a concurrent increase of the aperiodic structure content. Fluorescence spectroscopy indicated that irradiation decreased the emission intensity that was excited at 280 nm.
This study has examined the removal of mixed soil of protein and fat by protease. Cotton and PET fabrics were soiled by spotting of hemoglobin and triolein, respectively. The soiling order and soil concentration were changed in this procedure. The soiled fabrics were aged at 130℃ for 20 minutes. Protease was added in the alcohol ethoxylate(AE) detergent solution. The removal effciency was evaluated by analysis of protein and/or fat on the fabrics before and after washing, respectively. The detergency of PET fabrics was higher than that of cotton fabrics. The removal efficiency of hemoglobin was improved by protease from cotton and PET fabrics. Especially the removal efficiency of hemoglobin was remarkably improved from cotton fabrics. With the increase of hemoglobin and triolein (1:1) mixed soil, the removal of mixed soil was increased in proportion to mixed soil content up to a certain point. but it began to decrease above the point from cotton fabrics, while it was generally increased from PET fabrics. The detergency of total mixed soil from cotton fabrics was higher in case of soiling order with triolein after hemoglobin than in case of soiling order with triolein before hemoglobin. But the soiling order was not greatly effected in the detergency of total mixed soil from PET fabrics.

Ethylene oxide is a genotoxic carcinogen with widespread uses as industrial chemical intermediate and gaseous sterilant. 2-hydroxyethylated N-terminal valine in Hb is a good biomarker for biological monitoring of ethylene oxide exposure, because of its stability. For measuring the hemoglobin adduct formed by exposure of ethylene oxide, we studied the determination of (N-2-hydroxy-ethyl)valine(HEV) in hemoglobin adduct by using GC/MS. Firstly we synthesized HEV with 2-amino-ethanol and bromoisovaleric acid(BIVA) and confirmed it with GC/MS-FID. Its fragmentations were m/z 116(base ion, M+-45) and m/z 130(M+-31). For measuring HEV with higher sensitivity, we use derivatives which were PFPITH(pentafluorophenylisothiocianate) and TBDMS (tributyldimethylsilylation) by using Edman procedure. Its fragmentation were m/z 425(M+-57), m/z 383(M+-99) and m/z 172(M+-310) by using GC/MS. We did biological monitoring for mice inhalation exposure with 400 ppm ethylene oxide. The concentrations of hemoglobin adduct were $168{\pm}3.8\;and\;512{\pm}04$(nmol g-1 globin) at 0.5 hr/day 400 ppm ethylene oxide inhalation exposure group, and $631{\pm}17\;and\;2265{\pm}9.4$(nmol g-1 globin) at 1.0 hr/day 400 ppm ethylene oxide inhalation exposure for 1 and 4 weeks, respectively. We confirmed that (N-2-hydroxy-ethyl)valine(HEV) of hemoglobin was a good biomarker for biomonitoring of ethylene oxide exposure, and can measured with derivatives such as PFPITH(pentafluorophenylisothiocianate) and TBDMS(tributyldimethylsilylation) by using GC/MS.
Purpose: Liver cirrhosis is a major cause of hospital admission and mortality among children. Understanding the factors that influence disease severity is essential for preventing and reducing mortality. This study explored the association between hemoglobin levels and liver disease severity in children with cirrhosis. Methods: This cross-sectional study included 326 children with cirrhosis admitted to Namazi Teaching Hospital between 2015 and 2020. Clinical data, Child-Turcotte-Pugh (CTP) scores, and pediatric end-stage liver disease/model for end-stage liver disease (PELD/MELD) scores were collected to assess disease severity. Anemia was defined based on age, sex, and hemoglobin levels. Results: Among the children with cirrhosis, 275 (84.4%) were anemic, with a mean age of 5.4±4.8 years. The overall mean hemoglobin level was 9.2±2.1 g/dL. A significant inverse correlation was observed between hemoglobin levels and CTP and PELD/MELD scores in children with anemia (p<0.001). Moreover, lower hemoglobin levels were associated with more higher CTP classes (p<0.001). Conclusion: According to the data analysis, a significant correlation was observed between hemoglobin level and the severity of liver disease, and hemoglobin level decreased with increasing severity of liver disease. According to CTP class, the mean hemoglobin level decreased progressively as the disease progressed. A comparison of the mean CTP scores between children with and those without anemia revealed that those with anemia had more severe disease than those without anemia.
Glycated hemoglobin ($HbA_{1c}$) has been commonly used to screen and diagnose for patients with diabetes mellitus. Here the accelerated procedure of microwave-assisted sample treatment from acid hydrolysis to enzyme digestion followed by isotope dilution liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) was optimized and applied to measure $HbA_{1c}$ in an effort to speed up analysis time. First, two signature peptides of $HbA_{1c}$ and hemoglobin $A_0$ were certified with amino acid analysis by setting optimized acid hydrolysis conditions to $150^{\circ}C$, 1.5 h and $10{\mu}M$ sample concentration in 8 M hydrochloric acid. Consequently, the accurate certified peptides above were used as calibration standards to implement the proteolytic procedure with endoproteinase Glu-C at $37^{\circ}C$, 700 W for 6 h. Compared to the traditional method, the microwave heating not only shortened dramatically sample preparation time, but also afforded comparable recovery yields. The optimized protocol and analytical conditions in this study are suitable for a primary reference method of $HbA_{1c}$ quantification with full SI-traceability and other similar proteins in complex biological samples.
The encapsulation of the Purified bovine hemoglobin with Phospholipids obtained from egg folk was performed using a rotary vacuum evaporator. The prepared Hb-containing liposome (hemosome) had good properties as artificial red blood cell. The shape and size of the hemosomes were measured by a phase contrast microscope and image analyzer. The function of the hemosome as oxygen carrier was tested by measuring oxygen saturation curve with blood gas analyzer and infusing it into rats. The prepared hemosome was 1.184 + 0.423 ${\mu}m$ in diameter and round in shape. $P_{50}$ value of the hemosome solution was 28 mmHg. The synthesized red cells seem to function as oxygen carrier, because the severely bled rats were prolonged in their life by transfusion of the hemosome solution containing bovine hemoglobin.
