Protection against Whole Body γ-Irradiation Induced Oxidative Stress and Clastogenic Damage in Mice by Ginger Essential Oil

  • Radioprotective effects of ginger essential oil (GEO) on mortality, body weight alteration, hematological parameters, antioxidant status and chromosomal damage were studied in irradiated mice. Regression analysis of survival data in mice exposed to radiation yielded LD50/30 as 7.12 and 10.14 Gy for control (irradiation alone) and experimental (GEO-treated irradiated) mice, respectively, with a dose reduction factor (DRF) of 1.42. In mice exposed to whole-body gamma-irradiation (6 Gy), GEO pre-treatment at 100 and 500 mg/kg b.wt (orally) significantly ameliorated decreased hematological and immunological parameters. Radiation induced reduction in intestinal tissue antioxidant enzyme levels such as superoxide dismutase, catalase, glutathione peroxidase and glutathione was also reversed following administration of GEO. Tissue architecture of small intestine which was damaged following irradiation was improved upon administration of GEO. Anticlastogenic effects of GEO were studied by micronuclei assay, chromosomal aberration and alkaline gel electrophoresis assay. GEO significantly decreased the formation of micronuclei, increased the P/N ratio, inhibited the formation of chromosomal aberrations and protected agaisnt cellular DNA damage in bone marrow cells as revealed by comet assay. These results are supportive of use of GEO as a potential radioprotective compound.

Attenuating Effects of N-acetylcysteine (NAC) against Fish Parasiticide-induced Toxicity in Carp Cyprinus carpio (잉어(Cyprinus carpio)에서 어류 구충제에 대한 N-acetylcysteine(NAC)의 독성 저감 효과)

  • This study examined whether N-acetylcysteine (NAC), a glutathione precursor, could attenuate toxic effects of three fish anti-parasitic agents, trichlorfon, hydrogen peroxide ($H_2O_2$) and formalin, all of which are known to exert side effects through free radical production. Common carp Cyprinus carpio were fed with NAC (approx. 50 mg/kg/day) for 3 consecutive days prior to anti-parasite bathing for a 24 hr period. Mortality rates were examined during this 24 hr bathing period, and selected hematological and biochemical parameters were also assessed at the termination of anti-parasite exposure. The mortality rates and plasma glucose elevations caused by all three anti-parasitics were significantly reduced by NAC pretreatment. Trichlorfon, but not $H_2O_2$ or formalin, elevated plasma levels of aspartatetransaminase (AST) and alanine-transaminase (ALT), and these elevations were attenuated by NAC. There was no change in hematocrit values in any treatment. The results provide evidence for the attenuating effects of NAC against toxicity caused by anti-parasite agents that act through free radical-producing properties. The results found in this study also suggest that NAC may be administered to fish to minimize toxicity in fish parasiticide use.

  • The effects of ethanol extracts from Asparagus cochinchinensis, Polggonatum officinale, Poria cocos, Panax ginseng and Acanthopanax sessiliflorum on the toxicity of polychlorinated biphenyls(PCBs) were examined and following results were obtained. By the administration of these extracts, PCBs intoxicated rats showed the normalizing tendency in body weights and various hematological parameters-counts of red blood cell and white blood cell, hemoglobin content and hematocrit value. These extracts ameliorated the elevated levels of serum cholesterol, total lipids, $_{8}-GOT-,\;_{8}-GPT$ and s-alkaline phosphatase activities of PCBs intoxicated rats.

Effects of a Fruit Diet and Exercise on Body Composition and Hematological Parameters according to UCP2 Genotype in Female Collegians (과일식과 걷기 운동이 UCP2 유전자 다형성 특성에 따른 여대생의 신체 조성 및 혈액 성분에 미치는 영향)

  • The objective of this study was to examine the effects of a fruit diet and exercise on body composition and hematological parameters in female collegians according to their uncoupling protein 2 (UCP2) genotypes. Nine subjects were recruited from the SahmYook University. 4 subjects carried the ID genotype, 5 the DD genotype, and none of the subjects carried the II genotype. After 10 days of the program, BMI were significantly decreased (p<0.05) in the ID genotype and the DD genotype. Mean total energy intake from fruit diet was 888.1 kcal. Fat mass and % body fat were significantly decreased (p<0.05) in subjects with the ID genotype. Hemoglobin level was significantly increased (p<0.05) in subjects with the DD genotype. This study provided evidence that fruit diet and exercise could decrease the BMI in subjects with the ID and DD genotypes and increase the hemoglobin level in those with the DD genotype.

Plasma Cholinesterase Activity in the Sprayer Occupationally Exposed to Organic Phosphate Pesticides (유기인제(有機燐劑)의 노출(露出)에 의한 혈장(血漿) Cholinesterase치(値)의 변화(變化))

  • This study was carried out to obtain the basic data for the preverition of chronic poisonig by organic phosphate pesticides. 6 sprayers who participated in the summer pest control programs of Jeonju city were chosen as the subjects in this study. Pesticides which were spread in the period of study are DDVP (1.09%), dursban (1.8%) and dibrom (1.9%). And from May 16, 1983 to Aug. 22, 1983, plasma cholinesterase activity, hematological and biochemical parameters were measured with several physical examinations for each sprayers and controls. Major findings are as following; 1. Before pesticides were sprayed, plasma cholinesterase were $7.32{\pm}1.76{\mu}M/20{\mu}l/hr$. in sprayers and $7.13{\pm}1.39{\mu}M/20{\mu}l/hr$. in the control group (p>0.05). 2. At 60th day of spraying, plasma cholinesterase activity of sprayers was $6.78{\pm}2.01{\mu}M/20{\mu}l/hr$. which is significantly decreased from the pre-exposed value (p<0.01), but plasma cholinesterease activity in the control group was not changed. 3. The specific findings which suggested to organophosphorous poisoning were not found through the period of study. 4. Hematological and bichemical parameters were not sigricantly changed in the both groups.

Biological Activities of Extracts from Crucian Carp (붕어 고음추출물의 생리활성)

  • The pharmacological effects of hydrocooked extracts of crucian carp have been investigated. All of the hydrocooked extracts showed the contractile effect on the isolated rat duodenum and decreased to a certain extent the normal blood pressure in anesthetized rat. The hydrocooked extracts also exhibited a dose-dependent relaxation on the isolated rat aorta precontracted with 50 mM KCl. All of these except for RM 60 fraction haying cytotoxic effect against MCF-7 human breast cancer cell showed neither antibacterial activity nor antitumor activity. In addition, we found that although body weight gain in the hydrocooked extracts treated group were all reduced significantly compared to control group, there were no significant changes in the hematological parameters, except that the levels of high-density lipoprotein was significantly increased. These results suggest that the hydrocooked extracts of crucian carp may contain a variety of bioactive materials.

The Effects of Toosendan fructus(TOF) treatment on Hematological and Cyto-pathological Alterations in Non-Bacterial Prostatitis Rat Model (천련자가 만성 비세균성 전립선염 Rat 모델에서 혈액 및 세포조직의 변화에 미치는 영향)

  • Objective : Although chronic non-bacterial prostatitis is a common disease, it is very difficult to treat effectively. Toosendan fructus(TOF) has been traditionally used in treatment of abdominal pain and parasite. In this study, we investigated the therapeutic effects and action mechanism of Toosendan fructus (TOF) in the rat model of non-bacterial prostatitis induced by castration and testosterone treatment. Method : Eight-month-old rats were treated with $17{\beta}$-estradiol after castration for induction of experimental non-bacterial prostatitis, which is similar to human chronic prostatitis in histophatological profiles. Toosendan fructus(TOF) and testosterone were administered as an experimental specimen and a positive control, respectively. The prostates were evaluated by histopahological parameters including the epithelial score and epithelio-stromal ratio for glandular damage. Also, the prostates were observed by hematological alterations of WBC, RBC, hemoglobin, hematocrit and platelet. Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation, the rats treated with Toosendan fructus(TOF) showed a diminished range of the tissue damage. Epithelial score was improved in Toosendan fructus(TOF) than that of the control. The epithelio-stromal ratio was lower in Toosendan fructus(TOF) when compared to that of the control. Also, the examination of bloods were not observed hematological change. Conclusions : These findings suggest that Toosendan fructus(TOF) may protect the glandular epithelial cells and may take hematological safety. We concluded that Toosendan fructus(TOF) may could be a useful remedy agents for treating the chronic non-bacterial prostatitis.

  • This study was conducted to examine the hematological factors in cultured olive flounder Paralichthys olivaceus depending on its growth stage and season. The study also aims at developing the standard hematological indicator for growth stage and season by examining total 16 parameters including whole blood (hematocrit, red blood cell and hemoglobin), biochemical (glucose, cholesterol, total protein, AST, ALT, $Na^+,\;K^+,\;Cl^-,\;Ca^{2+},\;Mg^{2+}$ and osmolarity), and endocrine (cortisol and $T_3$) factors in plasma of cultured olive flounder. The result showed a growth stage-dependent increase of $T_3$ level in olive flounder while the level of cholesterol showed an inverse correlation to fish size. For seasonal fluctuation in cultured olive flounder of the same growth stage, the highest level of Ht and RBC was observed in autumn.

Effects of Salinity Changes on Hematological Responses in Juvenile Ship Sturgeon Acipenser nudiventris

  • A 10-day trial was conducted to evaluate the salinity tolerance and hematological responses of juvenile ship sturgeon Acipenser nudiventris under different salinity levels. A total of 240 fish averaging $6.2{\pm}0.13g$ (mean${\pm}$SD) were randomly distributed into 12 tanks in groups of 20, and each tank was then randomly assigned to one of three replicates of four different salinities at 0, 4, 8 and 12‰. At the end of 10 days of experimental period, plasma protein value of fish exposed to 0‰ and 4‰ salinities was significantly higher than those of fish exposed to 8‰ and 12‰ salinities (P < 0.05). Significantly higher values for plasma cortisol and glucose were obtained in fish exposed to 12‰ salinity compared to the fish exposed to 0‰ and 4‰ salinities (P < 0.05). Plasma potassium and sodium values increased significantly by increment of salinity levels (P < 0.05). Therefore, these results indicated that in natural environments, juvenile ship sturgeon, A. nudiventris in migratory populations could be able to migrate and adjust successfully into brackish water with a salinity of up to 8‰ without any short-term hematological stress responses.

  • Objective: The aim of this study was to evaluate heat tolerance using heat tolerance indices, physiological, physical, thermographic, and hematological parameters in Santa Ines and Morada Nova sheep breeds in the Federal District, Brazil. Methods: Twenty-six adult hair sheep, one and a half years old, from two genetic groups (Santa Ines: 12 males and 4 females; Morada Nova: 7 males and 3 females) were used and data (rectal temperature, respiratory rate, heart rate, skin temperatures; hematological parameters) were collected during three consecutive days, twice a day (morning and afternoon), with a total of six repetitions. Also physical parameters (biometric measurements, skin and hair traits) and heat tolerance indices (temperature-humidity index, Iberia and Benezra) were evaluated. The analyses included analyses of variance, correlation, and principal components with a significance level of 5%. Results: The environmental indices, in general, indicate a situation of thermal discomfort for the animals during the afternoon. Breed significantly influenced (p<0.001) physiological and physical characteristics of skin, hair, biometric measurements and Iberia and Benezra heat tolerance indices. Santa Ines animals were bigger and had longer, greater number and darker hair, thicker skin, greater respiratory rate and Benezra index and lower Iberia index compared with Morada Nova breed. Conclusion: Although both breeds can be considered adapted to the environmental conditions of the region, Morada Nova breed is most suitable for farming in the Midwest region. The positive correlation found between the thermographic temperatures and physiological parameters indicates that this technique can be used to evaluate thermal comfort. Also, it has the advantage that animals do not have to be handled, which favors animal welfare.
