Histopathological Effects of Diazinon in Rat Liver (Diazinon을 투여한 rats에 있어서 간의 병리조직학적 변화에 관한 연구)

  • The toxicity and histopathologic changes of diazinon (O, O-diethy-O-(2-isopropyl-6-methyl-4-pyrimidinyl)phosphorothioate) was investigated in rat. Rat was treated with diazinon (100 mg/kg/day) by oral administration for 12 days. The experimental results were summarized as follows. Biochemical parameters such as ALT, AST, LDH and glucose in serum were significantly increased and hematological parameters such as Hb, Hct and PLT in blood were slightly increased in treated groups. Also the activities of serum cholinesterase were very significantly decreased in treated groups. In the histopathological changes, the normal lobular architectural pattern of the liver was well preserved in all treated groups. However, vacuolation or fatty change were represent in hepatocytes. Sections of liver from rats treated with diazinon for 3 and 6 days contained slight lipid infiltration in the form of small droplets randomly distributed that were graded minimal (+) or moderate (++) compared with the respective control condition. After 9 days, there were numerous small and large vacuoles in the terminal hepatic venule and perilobular areas of many serial sections of these rats indicative of fatty infiltration which were graded moderate (++) and severe (+++). After 12 days, fatty infiltration progressed periportal tract areas and graded severe (+++) and very severe (++++) in experimental groups. Note absence of cellular necrosis or inflammation.

Effects of Subacute Oral Administration of Mancozeb on the Immunopathological Parameters and Splenocytes Proliferation in Mice (Mancozeb의 아급성 노출이 마우스의 면역병리학적 인자 및 비장세포 증식능에 미치는 영향)

  • Mancozeb, a polymeric complex of zinc and manganese salts of ethylene bisthiocarbamate (EBDC), is used widely in agriculture as fungicides, insecticides, and herbicides. Mancozeb can be occupationally and environmentally exposed to human and has been reported to induce estrogenic activity, therein it is considered as an endocrine disrupter. After female ICR mice were treated Mancozeb orally at the doses of 250, 1,000 and 1,500 mg/kg/day for consecutive 30 day, we investigated the effects of Mancozeb on the immunopathological parameters (body-, thymus-, spleen-, liver- and kidny-weight, splenic cellularity, hematological parameters) and mitogen (Con A, LPS)-induced splenocyte proliferation (SP). Liver- and kidney- weight were increased, but body- and thymus-weight, number of splenocytes and WBC were decreased, when compared with control group. When splenocytes isolated from the mice exposed to Mancozeb for 30 days were cultured in presence of mitogens, the SP against Con A was significantly and dose-dependently decreased and the SP against LPS was also slightly decreased. Our present results indicate that subacute exposure of Mancozeb to mice might show immunotoxic effect.

  • Mercury exists naturally and as a man-made contaminant. The release of processed mercury can lead to a progressive increase in the amount of atmospheric mercury, which enters the atmospheric-soil-water distribution cycles where it can remain in circulation for years. Mercury poisoning is the result of exposure to mercury or mercury compounds resulting in various toxic effects depend on its chemical form and route of exposure. The major route of human exposure to methylmercury (MeHg) is largely through eating contaminated fish, seafood, and wildlife which have been exposed to mercury through ingestion of contaminated lower organisms. MeHg toxicity is associated with nervous system damage in adults and impaired neurological development in infants and children. Ingested mercury may undergo bioaccumulation leading to progressive increases in body burdens. This review addresses the systemic pathophysiology of individual organ systems associated with mercury poisoning. Mercury has profound cellular, cardiovascular, hematological, pulmonary, renal, immunological, neurological, endocrine, reproductive, and embryonic toxicological effects.

Effectiveness of Phytogenic Feed Additive as Alternative to Bacitracin Methylene Disalicylate on Hematological Parameters, Intestinal Histomorphology and Microbial Population and Production Performance of Japanese Quails

  • This study was conducted to evaluate the effects of phytogenic additive and antibiotic growth promoter in laying Japanese quails. One hundred and sixty five quails were divided into three groups of 5 replicates and 11 quails (8 females and 3 males) in each replicate. Treatment 1 was fed control diet, treatment 2 was fed control diet supplemented with 0.05% bacitracin methylene disalicylate as antibiotic growth promoter and treatment 3 was fed control diet supplemented with 0.1% phytogenic feed additive (PFA) for two periods of 3 weeks each from 37 to 42 weeks of age. Results showed that egg production, eggshell strength, eggshell weight, villus height and villus height to crypt depth ratio were significantly (p${\leq}$0.05) increased and feed consumption, feed conversion ratio, albumen, Haugh unit, cholesterol, low-density lipoprotein, alanine transaminase, gamma glutamyltransferase, alkaline phosphatase, high-density lipoprotein, triglyceride, number of goblet cell, crypt depth and intestinal bacterial population of Coliforms, Salmonella and E. coli were significantly (p${\leq}$0.05) decreased in PFA fed group. It is concluded that addition of PFA containing phytomolecules and organic acids as main ingredients could significantly improve the production parameters and the general health of laying quails as an alternative to antibiotic growth promoters.

Oral Toxicity of Crude Antifungal Compounds Produced by Lactobacillus Plantarum AF1 and Lactobacillus Plantarum HD1 (Lactobacillus plantarum AF1과 Lactobacillus plantarum HD1이 생성한 조항균 물질의 독성평가)

  • This study investigates the acute and repeated-dose oral toxicity of crude antifungal compounds produced by Lactobacillus plantarum AF1 (Lb. plantarum AF1) and Lactobacillus plantarum HD1 (Lb. plantarum HD1) in male and female Sprague Dawley rats. In the acute toxicity study, crude antifungal compounds (500, 1,000, and 2,000 mg/kg) did not reduce mortality or produce significant changes in general behaviors or the gross appearance of external and internal organs. In the repeated-dose toxicity study, crude antifungal compounds were administered orally to rats at doses of 500, 1,000, and 2,000 mg/kg daily for 28 days. There were no test-article-related deaths, abnormal clinical signs, or body weight changes. In addition, there were no significant differences between groups treated with crude antifungal compounds and the control group in their organ weight, hematological and serum biochemical parameters, or any other factors. These results suggest that the acute or repeated-dose oral administration of crude antifungal compounds produced by Lb. plantarum AF1 plus Lb. plantarum HD1 is not toxic in male and female rats.

A Screening Tool for Identifying High-Risk Pregnant Women of Fe Deficiency Anemia : Process I (임신부의 철분 영양 잠재위험집단의 조기선별을 위한 스크리닝 도구의 개발 I)

  • Iron deficiency anemia is a worldwide public health problem relevant to unsound nutritional practice. While the prevalence of iron deficiency anemia is very common among pregnant women, appropriate nutritional service programs to improve the iron status are lacking in Korea. In an attempt to develop a nutritional screening tool to separate the high-risk subjects of iron deficiency, we carried out a nutritional survey for 115 Korean pregnant women whose gestational age ranged from 13 to 24 weeks. Each subject was interviewed with questionnaires for general characteristics and dietary habits. Food intake was measured by 24-hour recap method and 2 day record. Fasting blood was drawn for measuring hemoglobin and serum ferritin. It appeared that half of the pregnant women belonged to the anemia group and had insufficient dietary habits to provide adequate amounts of dietary iron. The first gravida and the working women had better hematological iron indicators than the second or more gravida and the housewives. It also appeared that women who had bigger family size and lower BMI in pre-pregnancy had poorer iron status. Among the food consumption habits, fruit dependent dietary habit was related to poor iron status. Sufficiently consumed green leafy vegetable and appropriate amount of food before morning sickness were positive factors of iron stares. Our results indicated that parity, BMI, current job, family size, food habits including consumption of fruits, green & yellow vegetables, and food habits before the onset of morning sickness are significant factors to contribute the Fe deficiency anemia during pregnancy.

Acute Immunotoxic Effects of Chlorpyrifos in CBA Male Mice (유기인계 Chlorpyrifos가 생쥐에 미치는 급성 면역 독성)

  • Chlorpyrifos, o,o diethyl-o-(3,5,6-trichloro-2-pyridyl) phosphorothioate, is a broad spectrum organophosphate insecticide. The use of chlorpyrifos has been increased more and more as pesticide. But the effects of chlorpyrifos on the immune alterations has not been yet observed. Therefore, we investigated the effects of chlorpyrifos on the immune alterations in CICA male mice. Chlorpyrifos was administered to mice by a single intraperitoneal injection for the purpose of observing acute effects. On the one hand to get the information on immunopathologic alterations we observed hematological values, counted total circulating leukocytes and assessed the ratio of lymphocytes and neutrophils from the peripheral blood, measured the ratio of organ/body weight and counted splenic cellularity in CBA male mice which treated chlorpyrifos intraperitoneally. But we could not find any significant immunopathologic alterations statistically by a single intraperitoneal injection. Also, the exposure of chlorpyrifos caused no significant change in the number of PFC/10$^6$ spleen cells at any three given doses. On the other hand a singte intraperitoneal injection of chlorpyrifos decreased the lymphocyte proliferation response slightly to ConA or LPS stimulation at a dose of 6 mg/kg b.w. Administrations of chlorpyrifos reduced mixed leukocyte response(MLR). MLR was decreased moderately at doses of 3mg/kg b.w. and 6mg/kg b.w. Therefore, all these findings suggest that chlorpyrifos may alter the immune functions acutely. especially by the changes of T lymphocyte activity.

An Experimental Study on Acute Toxicity of Benzo[a]pyrene (벤조피렌의 급성독성시험 연구)

  • Objectives: This study was performed to observe the toxicological changes caused by a single exposure to benzo[a]pyrene. Methods: Based on the results of a preliminary study, 300 mg/kg was set as the middle dose. A highest dose of 2,000 mg/kg and a lowest dose of 50 mg/kg were selected based on GHS guidelines. Benzo[a]pyrene was orally administered once to female and male SD rats at dose levels of 50, 300, and 2,000 mg/kg(body weight). All animals were monitored daily for clinical signs and mortality over 14 days. Hematological and biochemical values were examined as well. Results: There were neither dead animals nor significant changes in body weights during the experimental period. In addition, no differences were found between the control and treated groups in clinical sign, hematology, serum biochemical, and histopathological analysis. Conclusion: Compared with the control group, we could not detect any toxic alteration in all treated groups. These studies indicate that the acute toxicity of benzo[a]pyrene is relatively low.

Effects of Fermented Liquid of Males meles with Medicinal Plants on the Menopausal Symptoms, Estradiol Content and Bone Health Indices in the Postmenopausal Women (한약재를 첨가한 오소리 발효액이 폐경기 증후군과 Estradiol 농도 및 골대사에 미치는 영향)

  • The purpose of this study was to investigated the possibility of Meles meles as an edible functional resource. To assess the effects of Meles meles fermented liquid(ML) with medicinal plants in postmenopausal women, the menopause symptoms, hematological and serum chemical variables and bone health indices were examined. Twenty five postmenopausal women participated in the study. As a result, in postmenopausal women, ML supplements were effective in reducing the incidence of menopause symptoms, such as insomnia(44.0%), sensitivity(40.0%), headache(34.0%), hot flush and decreased vaginal lubrication(20.0%) after 12 weeks. Also serum estradiol and calcitonin and calcium levels were 23.27 pg/mL, 14.88 pg/mL and 8.86 mg/dL before the ML intake. Levels were significantly increased of estradiol and calcitonin and calcium after ML intake during 12 weeks to 30.36 pg/mL, 21.61 pg/mL and 10.01 mg/dL. Osteocalcin and bone alkaline phosphatase activity were 14.36 ng/mL, 21.55 U/L before the ML intake, but according to ML intake, the levels were significantly decreased. In conclusion, ML play benefical roles in the prevention of bone loss and menopause symptoms. Howere current data are not sufficient to determine the effective doses for benefical effect as well as harmful effect and support dietary recommendation level for ML. Therefore, our results should be viewed with caution. But Meles meles fermented liquid with medicinal plants can be used as possible food resources and functional food materials, more studies are needed to identify the proper mechanism of ML.

Anti-inflammatory effects of low-level laser in burn wound models in rats

  • Objective: The anti-inflammatory effects of low-level laser in burn wound model in rats were investigated. Design: Randomized controlled trial. Methods: The rats were assigned to three experimental groups. Group I received second-degree burn wounds; Group II received dressing film and low-level laser ($1.2J/cm^2$) treatment after a burn wound; Group III received dressing film and low-level laser ($2.3J/cm^2$) treatment after a burn wound. After inducing a deep second-degree burn wound, the wound was observed every day and the burn area diameter and retraction quantification at 1, 7, and 14 days were evaluated. Low-level laser was investigated on hematological parameters after 14 days. Effects of low-level laser on the inflammatory cytokines (tumor necrosis $factor-{\alpha}$ [$TNF-{\alpha}$] and interleukin-6 [IL-6]) concentrations in the serum were evaluated using immunosorbent assay kits. Results: Group III showed a significant difference in wound size on days 7 and 14 compared to Group I (p<0.05). Group II showed a significant difference in wound size on day 14 compared to Group I (p<0.05). For wound contraction percentage, both laser therapy treatment groups showed a significant difference compared with Group I (p<0.05). There was also a significant difference in wound contraction percentage in Group III compared to Group II (p<0.05). Compared with the model control group, decreased $TNF-{\alpha}$ and IL-6 levels in the serum was observed at 14 days after burn wound induction. Conclusions: The results of this study suggest that low-level laser therapy can assist in burn wound healing, which might be associated with decreased concentrations of $TNF-{\alpha}$ and IL-6 related proinflammatory cytokines.
