Immunization of broiler chicks deprived food and water with live Newcastle disease vaccine(LaSota strain) by drinking water

  • To confirm the effect of food and water deprivation prior to Newcastle disease(ND) virus vaccination, three hundred chicks were divided into five groups with three replications. ND vaccine were sprayed to at 1 -day old chicks at commercial hatchery. Secondary and third vaccination was conducted at 2-week old and 24-day old chicks by LaSota strain. Control was conventionally vaccinated without withdrawing the food and water before or after vaccination. In group 2(G2) and 3(G3), LaSota strain was vaccinated to chicks before and after fasting the food and water for 3 and 2 hours, respectively. Group 4(G4) has the same fasting time of group 2, but supplemented the skim milk in vaccin dilution water. In group 5(G5), skim milk was added into group 3. Weight gain, feed intake and feed conversion were weekly measured for 5 weeks. Blood was collected from wing vein at 24 and 35 days of age. Each serum antibody level were measured by hemagglutination inhibition(HI) test. The average weight gain, feed intake, feed conversion of all group were not significantly different. Weight gain of each groups was 1910.30(control), 1875.28(G2), 1952.12(G4) and 1896.05(G5), respectively. Feed intake of all group was recorded at 3160.67(control), 3167.07(G2), 3189.48(G3), 3157.85(G4) and 3178.16(G5), respectively. The feed conversion of each groups was 1.655(control), 1.688(G2), 1.633(G3), 1.699(G4) and 1.676(G5), respectively. The HI titer of G4 was $ 5.50{\Pm}$1.40 and significantly higher than the other groups (p<0.05)(control : $4.36{\Pm}$1.87 , G2 : $5.18{\Pm}$2.14, G3 : $4.51{\Pm}$2.19, G : $5.28{\Pm}$1.58 at 35 days old. The results of this experiment indicated that two or three hours of fasting time before or after vaccination would be able to show the higher antibody level against ND virus.

일본뇌염 백신 접종후 항 일본뇌염 항체의 생성율과 지속적인 면역반응에 대한 연구 (Studies on the Duration of Immunity and Production of Antibody following Immunization with Inactivated Killed Japanese Encephalitis Vaccine)

  • 목 적 : 국내에서 사용되고 일본뇌염 바이러스 백신에 대한 야외 실험을 실시하여 국내 백신 제조에 사용되는 Nakayama-NIH주의 국내 접종자에 대한 항체 생성율 및 중화 항체의 지속율을 조사하고, 일본뇌염 접종 스케줄의 재조정 및 국내 면역 집단의 민감도 조사 등을 통하여 방역 대책의 기초 자료로 활용하고자 하였다. 방 법 : 일본뇌염 백신의 면역효과 및 항체지속 기간을 알아보고자 1994년부터 계속 사업으로 강화군에 소재하고 있는 K 초등학교 학생들을 대상으로 조사를 실시하였다. 대상자들의 혈액을 시기에 따라 채혈하여 hemagglutination inhibition test (HI) 및 plaque reduction neutralization test (PRNT)를 실시하여 항체가를 조사하였다. 결 과 : 총 조사 대상자 213명은 이미 기초 접종이 끝난 자들로서 HI 항체가는 '95년 4월 백신 면역전에 이미 63.4%가 1:10 이상의 항체가를 가지고 있었으며 중화항체가는 전원이 1:20 이상의 항체가를 가지고 있었다. '95년 백신 접종 후 12개월이 지난 1996년 4월까지 HI 항체가 1:20 이상이 55.7%이며, 중화항체가는 전원이 1:20 이상을 보유하고 있었다. 이들에게 1996년 4월에 재접종 후 6개월째에는 HI 항체가가 1:10 이상이 69.7%로 증가하였고 G.M.T값은 1:11.6에서 1:13.23, 중화 항체의 G.M.T는 1:275.7에서 1:348.1로 증가하였다. 결 론 :위의 실험 결과에 따르면 현재 국내에서 사용중인 불활화 일본뇌염 백신은 자연계 에서 일본뇌염 바이러스가 공격하는 $10^5$ $LD_{50}$를 막을 수 있는 중화항체가 1:20을 기본면역 1회로 1년 이상 유지하고 있었으며, 충분한 boosting 효과를 보이고 있었다. 따라서 일본뇌염 백신의 추가면역은 기본접종후 개인의 상태에 따라 2~3년 간격의 추가접종이 적절하다 고 생각된다.

한국 소아의 Mumps 바이러스 항체보유에 관한 혈청학적 진단방법의 비교 (A Comparative Study of Serologic Methods for Detection of Mumps Antibody in Korean Children)

  • Mumps is an extremly common infectious disease affecting predominantly young children hut it is not a severe disease in terms of mortality. One hundred and two sera from infants of 3 different groups which are vaccinated, unvaccinated and unknown were detected to mumps antibody. The tests used were Complement Fixation(CF) test, Single Radial Hemolysis(SRH) test, Hemagglutination Inhibition(HI) test, Enzyme Linked Immunosorbent Immunoglobulin G(ELISA IgG) test, Enzyme Linked Immunosorbent Immunoglobulin M(ELISA IgM) test. 1. The rate of positivity for mumps antibody in 102 sera wera 89.16%(74/83) by Hl test, 68.83%(53/77) by ELISA IgG test, 64.58%(62/96) by SRH test, 63.24%(43/68) by ELISA IgM test and 50.00%(49/98) by CF test. 2. The rate of positivity by 5 tests for 55 sera turned out to be very similar with above results respectively. 3. The correlation coefficients(r) between ELISA IgG test ant H1 test, ELISA IgG test and ELISA IgM test were 0.34(P<0.0l) and 0.31(P<0.02), respectively. 4. The percentage of apparently natural infection of mumps seemed to be 65.15%(43/66) in infants. 5. Seroconversion rate of mumps by vaccination were 90.91%(10/11). 6. Among the 53 infants who were tested with ELISA IgG 15 were below 15 months age of(28.30%) and this percentage may be taken as a suggestion that mumps vaccination should be given earlier than present practice. 7. ELISA IgG test was found very sensitive and recommendable method for large scale screening for the presence of antibody to mumps.

팽이버섯으로부터 Lectin의 정제와 특성 (Purification and Characterization of the Lectins from Mushroom Flammulina velutipes)

  • 식용버섯인 팽이버섯(Flammulina velutipes)으로부터 적혈구 응집능력을 갖는 렉틴(flammulina velutipes lectin, FVL) FVL-1과 FVL-2을 정제하였다. FVL-1과 FVL-2의 분자량은 각각 10.6 kDa과 37 kDa으로 추정되었다. FVL-2는 사람적혈구의 경우 모든 혈액형에서 응집현상을 나타내었으나, FVL-1은 O형의 적혈구에 대하여 높은 상대활성도를 나타내었다. 렉틴의 hemagglutination inhibition test 결과 FVL-1과 FVL-2는 fetuin, bovine submaxillary mucin, asialofetuin, porcine stomach mucin에 의해 적혈구 응집력이 저해되었다. FVL-1은 1.56 mM $Cu^{2+}$에서 적혈구 응집력이 저해되었으며, $Mg^{2+}$, $Mn^{2+}$, $Ca^{2+}$, $Fe^{2+}$ 등에서도 저해되었다. 반면 FVL-2는 금속 이온에 대한 저해가 나타나지 않았다. pH 안정성에 있어서 FVL-1은 pH $5{\sim}11$에서 안정하였으며, FVL-2는 pH 4까지의 범위에서 안정하였다. 온도 안정성에 있어서는 FVL-1과 FVL-2 각각 $55^{\circ}C$, $45^{\circ}C$까지 안정하였다. FVL-1은 단백질이었으며 FVL-2는 당 단백질이었다.

국내(國內) 경주마(競走馬)에서의 Getah 바이러스 분리(分離) 동정(同定) (Isolation of Getah virus from racehorse in Korea)

  • Getah virus is known as a causative agent of recognized febrile illness of horses characterized by fever, rash and edema. A serological survey indicated that hemagglutination inhibition antibody against Getah virus was detected in 34% of 464 racehorses from Korean Horse Affairs Association and 57% of 262 ponies from Cheju island, respectively. Several field strains of Getah virus isolated were from the racehorse that have been shown fever and febrile signs in 1989. The field isolates produced cytopathic effect in Vero, MA-104, BHK-21 cell cultures. Especially, they multiplied to the highest titer($10^6TCID_{50}/0.1ml$) in Vero cell cultures. When day-old mice were inoculated with field isolates by the intracerebral route, they showed a typical paralysis sign and died within seven days after inoculation. The guinea pig exhibited skin rash and edema, and died with neural signs after inoculation with the field isolates. In the cross neutralization test and indirect immunofuorescent assay, the field isolates were proved to be closely related to the Sakai strain of Getah virus antigenically.

  • It has long been believed for the presence of infectious coryza affecting serious economic loss in domestic poultry industry. However, the etiologic agent has not been isolated until quite recently. From 1979, several strains of Haemophilus-like organism were isolated from chickens with symptoms similar to infectious coryza, and their colonial morphology, growth requirement, biochemical properties and pathogenicity were assessed. In addition, serological properties of the isolates by cross hemagglutination inhibition test was also investigated. The results indicated that all the isolates were identified as Haemophilus gallinarum which had similar characteristics to the reference strains.

종계군(種鷄群)의 Newcastle Disease에 대(對)한 면역상태(免疫狀態)에 관(關)한 연구(硏究) (Immune Status of Breeding Hens Against Newcastle Disease)

  • The present study was undertaken to observe the immune status of breeding hens and laying hens against Newcastle disease (ND). The methods of extraction of hemagglutination inhibition (HI) antibody from egg yolk, the detection of HI antibody in egg albumen and the correlation between HI antibody titers in maternal sera and egg yolks were discussed. For the purposes of these experiments, 9 flocks of breeding hens and 16 flocks of laying hens immunized against Newcastle disease virus were investigated. The vaccination program of tested flocks was 3-3-3 or 4-4-4 in general. The results obtained are summerized as follows: Freezing-thawing was the best method far antibody extraction from egg yolk for HI test. The HI antibody against NDV was found in egg albumen (geometric mean, 4.5), but lower than that found in egg yolk (32.1). The geometric mean of HI antibody titers of egg yolks (84.1) was higher than that of maternal sera (68.4) and day-old chicken sera (25.3). There was correlation between HI antibody titers of maternal sera(Y) and those of egg yolks(X). The coefficient correlation was r=0.63, and the line of regression of Y on X was $\hat{Y}$=35.91+0.35X.

특정(特定) 전염성(傳染性) 병인체(病因體)에 대한 국내(國內) 종계(種鷄)의 항체(抗體) 보유상황(保有狀況) (Prevalence of Serum Antibodies in Breeder Chickens against Specific Avian Pathogens)

  • Incidence of avian infectious diseases in breeder chickens was followed serologically. Serum samples were collected during the period of 1978~1979 from breeders throughout country and tested for the presence of antibodies against Salmonella pullorum(SP), Mycoplasma gallisepticum(MG), Avian Infectious Bronchitis virus(AIBV), Infectious Bursal Disease virus(IBDV) and Egg Drop Syndrome Virus(EDS). The tests used serum plate agglutination for SP and MG, immuno-diffusion for AIBV and IBDV and hemagglutination-inhibition test for EDS virus. The results are summarized as follows: 1. Individuals and Hocks incidence rate of Avian Infectious Bronchitis virus were 16.9% and 55.3%. 2. Individuals and Hocks incidence rate of Infectious Bursal Disease virus were 50.1% and 66.4%. 3. Individuals and Hocks incidence rate of sal. pullorum were 17.2% and 65.9%. 4. Individuals and flocks incidence rate of M. gallisepticum were 36.2% and 63.2%. 5. Individuals and flocks incidence rate of Egg Drop Syndrome (BC 14) virus were 14.1% and 46.3%.

Serologic monitoring of animal welfare-oriented laying-hen farms in South Korea

  • As animal welfare issue becomes important, the European Union bans conventional cages for laying hens from 2012. So the alternative housing systems like floor pens, aviaries or free range systems have been suggested. From 2011 to 2014, we monitored 20 welfare-oriented laying hen farms in South Korea to figure out serological status of major viral diseases. During this period, total 3,219 blood samples were collected from the randomly selected chickens to test and evaluate the hemagglutination inhibition titers for low pathogenic avian influenza, Newcastle disease and egg drop syndrome '76. A total of 2,926 blood samples were tested through enzyme linked immunosorbent assay (ELISA) to assess the serological status of infectious bronchitis (IB). The distribution of ELISA titers for IB was various from almost 0 to 20,000 through the all weeks of age. Also, the antibody coefficient of variation for most of the diseases in this study was higher than those of typical cage layers. As this study was the first surveillance for major avian viral diseases of the animal welfare-oriented farms in South Korea, the results obtained from this study will help to determine what information and resources are needed to maintain better biosecurity and to improve the health and welfare of laying hen flocks.

닭와포자충 감염이 닭의 뉴캣슬병 예방접종에 대한 면역억제 효과 (Immunosuppressive effect of Cryptosporidium baileyi infection on vaccination against Newcastle disease in chicks)

  • 닭와포자충 감염이 다른 병원체에 대한 닭의 체액성 면역에 미치는 영향을 규명하기 위한 연구의 일환으로 2일령 SPF 병아리에 $5{\;}{\times}10^5$의 닭와포자충 오오시스트를 한 번에 경구투여한 다음 4일과 21일 두 번에 걸척 뉴캣슬병 불활화 (사독) 백신을 접종하였다. 오오시스트 접종 후 2주부터 1주 간격으로 13주까지 채혈하여 혈구응집억제반응으로 ND HI $log_2$ 역가를 경시적으로 측정하여 대조군과 비교, 검토하였다. 일반적으로 뉴캣슬병 바이러스에 대한 Hl가는 전 실험기간을 통하여 대조군에 비하여 실험군이 상당히 낮았으며 (p<0.01), 보다 빨리 음전하는 경향이었다. 백신의 보강주사 후 역가는 점점 높아져서 2주 ($5.00(\;}{\pm}{\;}0$; 비감염 대조군)와 4주 ($3.88{\;}{\pm}{\;}0.6658$; 감염 실험군)에 최고치에 이른 다음 점점 낮아졌다. 한편, 분변 내의 오오시스트 배설 양상은 통상적인 감염례와 같았다. 이로 미루어 보아 병아리가 닭와포자충에 감염되면 건강한 병아리에 비하여 뉴캣슬병 바이러스에 대한 면역억제 현상이 일어나 감수성이 증가할 수 있을 것으로 생각된다.
