Isolation and Purification of Fibrinolytic Enzyme of Edible Mushroom, Sarcodon aspratus(Berk.)S. Ito (능이버섯으로부터 Fibrin 분해활성이 있는 단백질의 분리 및 정제)

  • To isolate and purify fibrinolytic active substance from Sarcodon aspratus(N $H_4$)$_2$S $O_4$ precipitation, DE52 anion exchange column chromatography, Sephacryl-S 200gel filtration chromatography and Mono S cation FPLC were carried out and the characterizations of the purified enzyme were investigated. The bound active fraction on DE52 anion exchange column chromatography were eluted with 0.2 M NaCI and the fibrionlytic enzyme was purified after following Sephacryl-S200 gel fitration chromatography and Mono S cation EPLC. The specific activity of purified enzyme was 55.2 U/mg protein and increased 11.3 fold comparing crude extract and the yield was 49.5%. 12% SDS-PAGE electrophoresis and gel filtration chromatography revealed that Sarcodon aspratus fibrionloytic enzyme was highly purified and had 29.300 Da molecular weight. Enzyme activity of the purified fibrinolytic enzyme from Sarcodon aspratus was increased on higher pH and was stable until pH 10.5. On temperature dependent stability, the enzyme activity was decrease sharply but remained 25% relative activity on 8$0^{\circ}C$. This enzyme activity was inhibited by heavy metal ion, C $U^{2+}$ and $Co^{3+}$ with 68% and 38%, respectively. And also, the enzyme activity was inhibited with $Ca^{2+}$ chelator EDTA and serine protease inhibitor PMSF. These results from this study suggested that the fibrinolycit enzyme from Sarcodon aspratus is a serine protease and the enzyme activity was increased by $Ca^{2+}$ or $Mg^{2+}$ ion.n.ion.n.

Purification and Characterization of Lipase from Acinetobacter sp. B2 Isolated from Oil­contaminated Soil (유류오염지역에서 분리한 Acinetobacter sp. B2로부터의 Lipase 정제 및 특성)

  • Three hundreds thirty two bacterial colonies which were able to degrade crude oil were isolated from soil sam­ples that were contaminated with oil in Daejeon area. Among them, one bacterial strain was selected for this study based on its higher oil degrading ability, and this selected bacterial strain was identified as Acinetobactor sp. B2 through physiological-biochemical tests and analysis of its 16S rRNA sequence. Acinetobactor sp. B2 was able to utilize various carbohydrates but did not utilize trehalose and mannitol as a sole carbon source. Acinetobactor sp. B2 showed a weak resistance to antibiotics such as kanamycin, streptomycin, tetracycline and spectinomycin, but showed a high resistance up to mg/ml unit to heavy metals such as Ba, Li, Mn, AI, Cr and Pb. The optimal growth temperature of Acinetobactor sp. B2 was $30^{\circ}C.$ The lipase produced by Acinetobactor sp. B2 was purified by ammonium sulfate precipitation, DEAE-Toyopearl 650M ion exchange chromatography and Sephadex gel filtration chromatography. Its molecular mass was about 60 kDa and condition for the optimal activity was observed at $40^{\circ}C$ and pH 10, respectively. The activation energy of lipase for the hydrolysis of p­nitrophenyl palmitate was 2.7 kcal/mol in the temperature range of 4 to $37^{\circ}C,$ and the enzyme was unstable at the temperature higher than $60^{\circ}C.$ The Michaelis constant $(K_m)\;and\;V_{max}$ for p-nitrophenyl palmitate were 21.8 uM and $270.3\;{\mu}M\;min^{-1}mg^{-1},$ respectively. This enzyme was strongly inhibited by 10 mM $Cd^{2+},\;Co^{2+},\;Fe^{2+},\;Hg^{2+},$ EDTA and 2-Mercaptoethalol.

Analysis of the Controlling Factors of an Urban-type Landslide at Hwangryeong Mountain Based on Tree Growth Patterns and Geomorphology (부산 황령산에서의 수목 성장 및 지형 특성을 이용한 도시 산사태의 발생원인 분석)

  • We investigated the causes and characteristics of a landslide at Hwangryeong Mountain, Busan, based on aerial photos, annual precipitation data, rock fracture patterns, and geomorphic features using GIS Software, and a statistical analysis of tilted trees. The analyzed slope shows evidence of a previous slope failure event and the possibility of future failures. Although the NW-SE trending slope was relatively stable until 1975, a large-scale slope failure occurred between 1975 and 1985 due to complex factors, including favorably oriented geologic structures, human activity, and heavy rain. This indicates that a detailed study of geologic structures, slope stability, and rainfall characteristics is important for slope cuttings that could be a major factor and cause of urban landsliding events. The statistic analysis of tilted trees shows a slow progressive creeping type of mass wasting with rock falls oblique to the dip of the slope, with the slope having moved towards the west since 1985. A concentration of tree tilting has developed on the northwestern part of the slope, which could reach critical levels in the future. The analysis of deformed trees is a useful tool for understanding landslides and for predicting and preventing future landslide events.

A Study on the Waste Treatment from a Nuclear Fuel Powder Conversion Plant (핵연료 분말제조 공정에서 발생하는 폐액의 처리에 관한 연구)

  • Treating methods and characteristics of waste from a nuclear fuel powder conversion plant were studied. To recovery or treat a trace uranium in liquid waste, the ammonium uranyl carbonate(AUC) filtrate must be heated for $CO_2$ expelling, essentially. Uranium content of final treated waste solution from fuel powder processes for a heavy water reactor(HWR) could be lowered to 1 ppm by the lime treatment after the ammonium di-uranate(ADU) precipitation by simple heating. Otherwise, in case of the waste from fuel powder processes for a pressurized light water reactor(PWR), it is result in 0.8 ppm as a form of uranium peroxide such as $UO_4{\cdot}2NH_4F$ compounds. Optimum condition was found at $101^{\circ}C$ by the simple heating method in case of HWR powder process waste. And in case of PWR powder process waste, optimum condition could be obtained by precipitating with adding hydrogen peroxide and adjusting at pH 9.5 with ammonia gas at $60^{\circ}C$ after heating the waste In order to expelling $CO_2$. As the characteristics of recovered uranium compounds, median particle size of ADU was increased with pH increasing in case of HWP waste. Also, in case of uranium proxide compound recovered from PWR waste, the property of $U_3O_8$ power obtained after thermal treatment in air atmosphere was similar to that of the powder prepared from AUC conversion plant.

The Treatment of Heavy Metal Hydroxides by Crossflow-Microfiltration (정밀여과에 의한 중금속수산화물의 처리)

  • In the treatment of the wastewater containing metals($Cu^{2+}$, $Zn^{2+}$, $Ni^{2+}$, $Cr^{3+}$) by using batch precipitation and flocculation followed by membrane filtration, permeate flux and removal efficiency were investigated according to by the effect of pH and coagulants, and the type of membranes used and pore size. It was found that it is most effective to use $0.45{\mu}m$-polysulfone membrane and coagulant(PAC) at the conditions of the pH of 10.0~10.5 for the case of copper containing wastewater, $0.1{\mu}m$-PVDF membrane and coagulant(PAC) at the conditions of the pH of 10.0~10.5 for the case of zinc containing wastewater, $0.1{\mu}m$-PVDF membrane and coagulant at the conditions of the pH of 11.0~11.5 for the case of nickel containing wastewater, $0.2{\mu}m$ membrane and coagulant at the conditions of the pH of 8.0~8.5 for the case of chromic containing wastewater, and $0.2{\mu}m{\sim}0.45{\mu}m$ membrane and coagulant at the conditions of the pH of 11.0~11.5 for the case mixture wastewater. The permeate flux could higher as to be used coagulants except for the case of copper containing wastewater.

Problems of lake water management in Korea (한국의 호수 수질관리의 문제점)

  • In Korea most of annual rainfall is concentrated in several episodic heavy rains during the season of summer monsoon and typhoon. Because of uneven rainfall distribution many dams have been constructed in order to secure water supply in dry seasons. The Han River system has the most dams among Korean rivers, and the river is a series of dams now. Reservoirs need different strategy of water quality control from river water. Autochthonous organic matter and phosphorus should be the major target to be controlled in lakes. In this Paper some problems are discussed that makes efforts of water quality improvement ineffective in lakes of Korea, even after the substantial investment to wastewater treatment facilities.1) Phosphorus is the key factor controlling eutrophication of lakes and the reduction ofphosphors should be the major target of water treatment. However, water quality management strategy in Korea is still stream-oriented, and focused on BOD removal from sewage. Phosphorus removal efficiency remains as low as 10-30%, because biological treatment is adopted for both secondary treatment and advanced treatment. The standard for TP concentration of the sewage treatment plant effluent is 6 mgP/l in most of regions, and 2 mg/l in enforced region near metropolitan water intake point. TP in the effluents of sewage treatment plants are usually 1-2 mg/1, and most of plants meet the effluent regulation without a further phosphorus removal process. The generous TP standard for effluents discourages further efforts to improve phosphorus removal efficiency of sewage treatment. Considering that TP standard for the effluent is below 0.1 mg/l in some countries, it should be amended to below 0.1 mg/l in Korea, especially in the watershed of large lakes.2) Urban runoff and combined sewer overflow are not treated, even though their total loading into lakes can be comparable to municipal sewage discharges on dry days. Chemical coagulation and rapid settling might be the solution to urban runoff in regard of intermittent operation on only rainy days.3) Aggregated precipitation in Korea that is concentrated on several episodic heavyrains per year causes a large amount of nonpoint source pollution loading into lakes. It makes the treatment of nonpoint source discharge by methods of other countries of even rain pattern, such as retention pond or artificial wetland, impractical in Korea.4) The application rate of fertilizers in Korea is ten times as high as the average ofOECD countries. The total manure discharge from animal farming is thought to be over the capacity of soil treatment in Korea. Even though large portion of manure is composted for organic fertilizer, a lot of nutrients and organic matter emanates from organic compost. The reduction of application rate and discharge rate of phosphorus from agricultural fields should be encouraged by incentives and regulations.5) There is a lot of vegetable fields with high slopes in the upstream region of the HanRiver. Soil erosion is severe due to high slopes, and fertilizer is discharged in the form of adsorbed phosphorus on clay surface. The reduction of soil erosion in the upland area should be the major preventive policy for eutrophication. Uplands of high slope must be recovered to forest, and eroded gullies should be reformed into grass-buffered natural streams which are wider and resistant to bank erosion.

Study on the Distribution Characteristics of Storm Damage Area : The Case of Gyeonggi-do (수해지 분포 특성에 관한 연구 : 경기도 사례를 중심으로)

  • The main purpose of this study is to address flooding resilient land use management strategy based on the distributional characteristics of storm damage areas in Gyeonggi-do. The employed methods are 1) Exploratory Spatial Data Analysis (ESDA) to understand the spatial patterns of storm damage areas occurred from 2005 to 2009, 2) Local Indicator of Spatial Association (LISA) to examine spatial autocorrelation existed in storm damage areas for the year of 2009. The results show that 1) crop land damage is very sensitive to heavy precipitation, 2) damaged buildings are found in all over the Gyeonggi areas, but relatively more damages are in the regions closed to the City of Seoul, 3) damaged roads-bridges, streams, and reaches are found in mostly rural areas, 4) building and crop land damage occurs mostly in lowlands with different spatial patterns. These findings imply that 1) it will be useful to consider the average distances and slopes of damaged building and crop lands from streams for the decision making of land use management strategy, 2) further management efforts are required in the north, east, and south regions of Gyeonggi areas to prevent roads-bridge, stream, and reach damages, 3) the present land use pattern needs to be carefully investigated by considering the damage clustered areas for the year of 2009 based on watershed and municipality boundaries.

A Study on Rainfall-Pattern Analysis for determination of Design flow in small watershed (소유역의 설계유량 산정을 위한 강우현상 분석에 관한 연구)

  • The rainfall pattern analysis on time distribution characteristics of rainfall rates in important in determination of design flow for hydraulic structures, particularly in urban area drainage network system design. The historical data from about 400 storm samples during 31 years in Seoul have been used to investigate the time distribution of 5-minute rainfall in the warm season. Time distribution relations have been deveolped for heavy stroms over 20mm in total rainfall and represented by relation percentage of total storm rainfall to percentage of total storm time and grouping the data according to the quartile in which rainfall was heaviest. And also time distribution presented in probability terms to provide quantitative information on inter-strom variability. The resulted time distribution relations are applicable to construction of rainfall hyetograph of design storm for determination of design flow hydrograph and identification of rainfall pattern at given watershed area. They can be used in conjuction with informations on spatstorm models for hydrologic applications. It was found that second-quartile storms occurred most frequently and fourth-quartile storms most infrequently. The time distribution characteristics resulted in this study have been presented in graphic forms such as time distribution curves with probability in cumulative percent of storm-time and precipitation, and selected histograms for first, second, third, and fourth quartile stroms.

Adsorption Characteristics of Copper Ion onto a Bentonite (벤토나이트에 의한 구리이온의 흡착특성)

  • Bentonite has been considered as a liner material to restrict the release of hazardous heavy metals from the landfill. The adsorption of copper onto a domestic bentonite was studied to provide the adsorption isotherm and the effect of solution chemistry and temperature. The copper adsorption was fitted well to a Freundlich isotherm, in which Freundlich constants and correlation coefficient were calculated to be $K_F=1.18$, n=1.65, and $r^2=0.97$, respectively. The distribution coefficients ($K_d$) for the adsorption of copper decreased with increasing initial copper concentration. The $K_d$ increased with increasing the pH of solution, and drastically increased at pH > 5.3 because of precipitation of most copper species. As the ion strength of $Na^+$ in solution increased the $K_d$ decreased, while it increased with increasing the concentration of $SO_4{^{2-}}$ in solution. An increase in the temperature of experimental solution decreased the $K_d$ values.

Estimating Worst Case Flood and Inundation Damages under Climate Change

  • To generate information that contributes to climate change risk management, it is important to perform a precise assessment on the impact in diverse aspects. Considering this academic necessity, Japanese government launched continuous research project for the climate change impact assessment, and one of the representative project is Program for Risk Information on Climate Change (Sousei Program), Theme D; Precise Impact Assessment on Climate Change (FY2012 ~ FY2016). In this research program, quantitative impact assessments have been doing from a variety of perspectives including natural hazards, water resources, and ecosystems and biodiversity. Especially for the natural hazards aspect, a comprehensive impact assessment has been carried out with the worst-case scenario of typhoons, which cause the most serious weather-related damage in Japan, concerning the frequency and scale of the typhoons as well as accompanying disasters by heavy rainfall, strong winds, high tides, high waves, and landslides. In this presentation, a framework of comprehensive impact assessment with the worst-case scenario under the climate change condition is introduced based on a case study of Theme D in Sousei program There are approx. 25 typhoons annually and around 10 of those approach or make landfall in Japan. The number of typhoons may not change increase in the future, but it is known that a small alteration in the path of a typhoon can have an extremely large impact on the amount of rain and wind Japan receives, and as a result, cause immense damage. Specifically, it is important to assess the impact of a complex disaster including precipitation, strong winds, river overflows, and high tide inundation, simulating how different the damage of Isewan Typhoon (T5915) in 1959 would have been if the typhoon had taken a different path, or how powerful or how much damage it would cause if Isewan Typhoon occurs again in the future when the sea surface water temperature has risen due to climate changes (Pseudo global warming experiment). The research group also predict and assess how the frequency of "100-years return period" disasters and worst-case damage will change in the coming century. As a final goal in this research activity, the natural disaster impact assessment will extend not only Japan but also major rivers in Southeast Asia, with a special focus on floods and inundations.
