Combined Effects of Sex Hormones and Dietary Oils on Lipid Peroxidation

  • The effects of orchildectomy with/without testosterone replacement in wale rats, and ovarectomy with estrogen replacement in female rats on lipid peroxidation were studied in male and female rats fed with diets fortified with 20% w/w, soybean oil or palm oil for 4 months. Serum, liver and heart homogenates were assayed for malonaldehyde and conjugated diene levels. Orchidectomy was found to reduce levels of lipid peroxidation products in the serum, liver and heart. Testosterone replacement did not increase the lipid peroxidation products to levels in the non-orchildectomised rats, while estrogen did not influence lipid peroxidation significantly. Palm oil decreased, but soybean oil increased lipid peroxidation in the liver and heart of both the castrated and sex hormone-replaced male and female rats.

  • This study was designed to investigate the effects of methionine(Met) on the activities of heart lipid peroxidation related enzymes in ethanol administrated rats. Male Sprague-Dawley rats were fed on diets containing one of the three levels of Met(0%, 0.3%, 0.9% of kg diet) and ethanol(2.5g/kg of body weight) was administrated as 25v/v% ethanol to ethanol treated groups orally. The rats were sacrificed after 5 and 10 weeks of feeding. Xanthine oxidase(XO) and catalase activities increased with ethanol administration and those activities were higher n Met excessive and deficiency group than those of Met normal group at 5 and 10 weeks dieting. Superoxide dismutase(SOD) activity in heart decreased significantly in Met deficiency and Met excessive group as compared to that of control. Glutathione peroxidase(GSH-Px) activity in heart significantly decreased in Met deficiency group as compared to that of Met excessive and normal group. Glutathione S-transferase(GST) activity of heart tissue significantly increased by ethanol administration. Glutathione(GSH) content in heart decreased with ethanol administration and shwoed no significant differences with Met levels. Ethanol administration increased the content of lipid peroxide(LPO).

Effect of Rosiglitazone on Myocardial Ischemia-Reperfusion Injury in Rat Heart

  • This study was undertaken to evaluate whether peroxisome proliferator-activated-receptor-gamma $(PPAR-{\gamma})$ agonist-rosiglitazone (ROSI) induces postischemic functional recovery in Langendorf heart model. Hearts isolated from normal rats were subjected to 20 min of normoxia or 25 min zero-flow ischemia followed by 50 min reperfusion. In this acute protocol, ROSI $(20\;{\mu}g/ml)$ administered 10 min before ischemia had no effect on hemodynamic cardiac function, but had protective effect on lipid peroxidation in in vitro experiments. In chronic protocol in which ROSI was given by daily gavage (4 mg/kg) for three consecutive days, ROSI could not prevent the hemodynamic alteration on cardiac performance, but has protective effect on the activity of superoxide dismutase (SOD). There was no significant difference in the contents of reduced glutathione (GSH) and catalase activity between ischemia-reperfusion (IR) and ROSI treated IR hearts. Although ROSI had no effect on hemodynamic factor, it had effect on antioxidant activity. Our results indicate that ROSI provides partial beneficial effects by inhibiting lipid peroxidation and/or recovering normal level of SOD activity in the ischemic reperfused heart.

  • To investigate effects of dietary fish oil and vitamin E level on the tissue levels of vitamin E and vitamin A and to see which tissue is sensitive to lipid peroxidizability, male Sprague-Dawley rats were fed experimental diets composed of either menhaden oil or soybean oil nad either low(equivalent to 17 mg $\alpha$-tocopherol) or high (equivalent to 140mg $\alpha$-tocopherol) vitamin E level for 4 weeks. Palsma TBARS per mg lipid was significantly elevated in rats fed fish oil with low vitamin E level compared to soybean oil-fed rats. TBARS levels of liver, heart, kidney and liver microsomes were also increased by feeding fish oil with low vitamin E level. Plasma TBARS level was significantly correlated with TBARS levels of liver, heart, kidney and liver microsome. Plasma vitamin E level of groups with vitamin E supplementation was elevated significantly as compared to the those without vitamin E supplementation, whereas vitamin E levels of liver, heart and kidney were not changed significantly. Plasma TBARS was negatively correlated with plasma vitamin E(r=0.5763, P<0.001) and A(r=-0.4523, P<0.01) and seems to be a good indicator of in vivo lipid peroxidative stress.

  • This paper presents the effect of the supramolecular complex of GA (Glycyrrhizic acid) and Mt(menthol) and GA: Mt (4: 1) obtained on their basis can restore functional dysfunction of the liver mitochondria in alloxan diabetes, ie, inhibit lipid peroxidation. The hypoglycemic activity and mitochondrial membrane stabilizing properties of the supramolecular compound GA: Mt (4: 1) in alloxan diabetes were more pronounced than those of menthol, GA and its GK: Mt (2: 1) and GA: Mt (9: 1) compounds. According to the results obtained, the concentration of GA did not affect the peroxidation of lipid membranes of the liver mitochondria. However, a concentration of 15 μM of GA was found to reduce LPO (lipid peroxidation) formed by the effect of Fe2+ / ascorbate on the mitochondrial membrane by 58.0 ± 5.0% relative to control. 생쥐에서 N-Nitrosodiethylamine에 의한 산화성 스트레스에 대한 Lutein의 항산화효과 (Antioxidant Effect of Lutein on N-Nitrosodiethylamine-induced Oxidative Stress in Mice)

  • To investigate the antioxidant effect of lutein on N-nitrosodimethylamine (NDEA)-induced oxidative stress in mice, we measured lipid peroxidation, superoxide dismutase (SOD) and catalase of various tissues. Body weight was almost similar in lutein and control groups during 3 weeks. NDEA increased significantly the activities of typical marker enzymes of liver function (AST, ALT and ALP) in both groups. However, the increase of plasma aminotransferase activity significantly decreased in lutein group. Lipid peroxidation and SOD in various tissues, such as heart, lung, liver, kidney, spleen and plasma were significantly increased by NDEA, which were significantly reduced by lutein at a dose of 50 mg/kg. Catalase activity decreased significantly in control and lutein groups treated with NDEA, the effect being less in lutein group. Lesser effect on SOD and catalase in NDEA-treated lutein group indicates the improvement of protective mechanisms by lutein. Thus, it can be concluded from the present study that lutein can offer a useful protection against NDEA-induced oxidative stress.

  • ln an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen See radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde(MDA) level and activities of catalase and superoxide dismutase in the heart of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260gm were used. Experimental groups consist of Control group (=breathing with air), HBO group(=exposed to hyperbaric oxygen(HBO, 3ATA, 100%) after air breath), CO group(=exposed to CO(3,970 ppm) after air breath), CO-Air group(=exposed to CO after air breath followed by air breath) and CO-HBO group(=exposed to CO after ai. breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of Co-induced cardiotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the heart of the rats exposed to carbon monoxide.

스트렙토조토신-당뇨쥐에서 지질과산화 및 항산화계의 경시적 변화 (Changes in in vivo Lipid Peroxidation and Antioxidant Defense System in Streptozotocin-Induced Diabetic Rats: a Time Course Study)

  • This study was carried out to examine a part of the mechanism for the etiology of diabetic complications. Thirty normal and forty streptozotocin(STZ)-induced diabetic rats were used as the animal models. The animals were sacrificed at the time points of 3 days, 1,2,4 and 6 weeks after STZ-injection and a time course changes in the concentrations of thiobarbituric acid-reactive substances(TBARS) in blood, urine, and tissues, along with the levels of conjugated dienes in tissues were measured as indices of in vivo lipid peroxidation. The activities of antioxidant enzymes, catalase, superoxide dismutase, glutathione peroxidase and the levels of blood retinol and alpha-tocopherol were also measured. The diabetic rats maintained a slightly higher plasma TBARS level throughout the experiment. The urinary TBARS level was significantly higher in diabetic group and gradually increased with time. Concentrations of TBARS in liver, heart, and kidney tissues from diabetic animals were higher than those from the normal group. An increase of conjugated dienes was also observed in the all tissues examined. The kidney tissue of diabetic animals revealed more significant lipid peroxidation state than any other organ tissues. The activities of hepatic antioxidant enzymes such as catalase, superoxide dismutase, glutathione peroxidase were higher in diabetic animals compared to the control ones and increased with the duration of diabetes mellitus. The plasma levels of vitamin A and E were loser in diabetic animals than in normal controls throughout the experimental period. The level of vitamin E in diabetic animals was significantly decreased with the duration of the disease. The results of this study suggest that an effective regimen to suppress the adverse changes in lipid peroxidation and antioxidant defense system is required from the early stage of the disease to prevent the development of diabetic complications.   • We evaluated the effects of butterbur (Petasites japonicus Maxim) addition to the diet on lipid profiles and antioxidant biomarkers such as total glutathionine, TBARS value, carbonyl value, GPx, GR, SOD and paraoxonase activity in the plasma or liver of mice. The distribution of body fat deposition, total cholesterol (TC) contents, and atherogenic index in the plasma were significantly decreased in the butterbur group. The levels of GSH and the activity of GR and SOD were significantly higher in the liver of the butterbur group than in that of the control group. Lipid oxidation of the liver and kidney and protein oxidation of the liver and heart were decreased in the butterbur group. Additionally, the DNA damage, as determined using the comet assay (single cell gel assay) with alkaline electrophoresis and as quantified by measuring the tail length (TL), was decreased in the butterbur group. The results of the present study showed that a diet with added butterbur exerts degenerative disease-protective effects on oxidative DNA damage and lipid peroxidation.

  • This study was aimed to evaluate the anti-stress effect of danchisoyosan on the rats stressed by immobilization. The experimental animals were immobilized in the stress box($5{\times}5{\times}20cm$) for 12 hours in a day during 3 days, and administered $500mg/5m{\ell}/g$ of Danchisoyosan extract for 14 days before stress. There were measured the change of body weight and organ weight under immobilized-stress. The norepinephrine, epinephrine, dopamine, serotonine contents were measured by HPLC method in rat brain. There were measured the GOT, GPT contents in serum and tissue lipid peroxidation in the brain, liver, spleen, adrenalgland, pancreas, testes, thymus, heart. The following results were obtained: 1. The change of organ weight was significantly lower in control than normal group. Sample group inhibited decreased weight from stress comparing to control group. 2. Lipid peroxidation in the liver was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 3. Lipid peroxidation in the kidney was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 4. GPT contents in serum was significantly higher in control than normal group. Sample up shows significant decrease comparing to control group. 5. Dopamine contents in the brain was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 6. Serotonine contents in the brain was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group.
