Background: South Korea's Act on Registration and Evaluation, etc. of Chemicals (known as K-REACH) was established to protect public health and the environment from hazardous chemicals. 4,4'-Methylenedianiline (MDA), which is used as a major intermediate in industrial polymer production and as a vulcanizing agent in South Korea, is classified as a toxic substance under the K-REACH act. Although MDA poses potential ecological risks due to industrial emissions and hazards to aquatic ecosystems, no ecological risk assessment has been conducted. Objectives: The aim of this study is to assess the ecological risk of MDA by identifying the actual exposure status based on the K-REACH act. Methods: Various toxicity data were collected to establish predicted no effect concentrations (PNECs) for water, sediment, and soil. Using the SimpleBox Korea v2.0 model with domestic release statistical data and EU emission factors, predicted environmental concentrations (PECs) were derived for ten sites, each referring to an MDA-using company. Hazard quotient (HQ) was calculated by ratio of the PECs and PNECs to characterize the ecological risk posed by MDA. To validate the results of modeling-based assessment, concentration of MDA was measured using in-site freshwater samples (two to three samples per site). Results: PNECs for water, sediment, and soil were 0.000525 mg/L, 4.36 mg/kg dw, and 0.1 mg/kg dw, respectively. HQ for surface water and sediment at several company sites exceeded 1 due to modeling data showing markedly high PEC in each environmental compartment. However, in the results of validation using in-site surface water samples, MDA was not detected. Conclusions: Through an ecological risk assessment conducted in accordance with the K-REACH act, the risk level of MDA emitted into the environmental compartments in South Korea was found to be low.
The detection of many synthetic chemicals used in industry that may pose a genetic hazard in our environment is subject of great concern at present. In this respect, the genetic toxicity of fenpropathrin ((RS)-.alpha.-cyano-3-phenoxybenzyl-2,2,3,3-tetramethyl cyclopropane carboxylate, CAS No.:39514-41-8), a pyrethroid insecticide, was evaluated in bacterial gene mutation system, chromosome aberration in mammalian cell system and in vivo micronucleus assay with rodents. In bacterial gene mutation assay, no mutagenicity of fenpropathrin (62-$5000\mug/plate$) was observed in Salmonella typhimurium TA 98, 100, 1535 and 1537 both in the absence and in the presence of S-9 metabolic activaton system. In mammalian cell system using chinese hamster lung fibroblast, no clastogenicity of fenpropathrin was also observed both in the absence and in the presence of metabolic activation system in the concentration range of $7-28\mug/ml$. And also, in vivo micronucleus assay using mouse bone marrow cells, fenpropathrin also revealed no mutagenic potential in the dose range of 27-105 mg/kg body weight of fenpropathrin (i.p.). Consequently, no mutagenic potential of fenpropathrin was observed in vitro bacterial, mammalian mutagenicity systems and in vivo micronucleus assay in the dose ranges used in this experiment.
Under the limited arable land, the enhancement of agricultural productivity is indispensable to provide the food demand which is concomitant with the rapid increase in population. From this viewpoint, the upbringing and dissemination of high-yielding varieties has been promoted continuously and several modifications in cultural practices, including heavy fertilization, dense planting, and early transplanting, also have been gradually developed. However these changes in cultivation have led to the increased outbreak of insect pests and diseases. And this unexpected results have accelerated the number and complexity of pesticides employed as well as their consumption. Even though pesticides are essential materials contributing to the steady production of agricultural crops, large scale consumption of them has given rise to several adverse impacts, such as mammalian hazard and/or environmental contamination. In this respect, recent development of new pesticides has been concentrated on 'safe pesticide', as it were, that has the highly selective properties without unfavorable side influences on other ecosystem. According to literature cited up to now, feasibilities of safe pesticide development would be summarized as two categories. One of them is the development of chemical pesticides, which include the molecular structure modification of established pesticides for increased safety and synthesis of new safe chemicals which can attack the vulnerable point of physio-ecological characteristics in insect pests and diseases. The other is the biological pesticides which comprise natural enemies and microorganisms to act selectively on confined insect pests and diseases, In addition, improvement of physico-chemical properties of available pesticide formulations would be one of the desirable means for safe pesticide development in view of efficacy enhancement and minimization of hazardous properties or safe pesticide development, various approaches are feasible and needed to study, however, long period and much financial outlay are necessary to develop a new item. And under the present situation in Korea, there are many difficulties for performing research on all the possible routes. Therefore, combined pesticides by the reasonable combination of already registered resticides evaluated as the fairly safe pesticides and safe formulation based on their physico-chemical properties would be developed primarily. And many efforts would be given gradually for the development of new chemical and biological pesticides.
March 19, 1999, the renovation qf the runway of the Bo-Fai ai1field in Hua Hin, Prachubk-erikhan, Thailand, unearthed chemicals which were left over from the project "anch Hand Operation" held during the Vietnam war era. The chemical mixtures were analyzed by the US EPA, the Department oj Medical Sciences (DMSc), Ministry oj Public Health (MoPH) and the Pollution Control Department (PCD), the Ministry oj Science Technology and Environment (MOSTE) of Thailand, The samples were found to contain several defoliants used in the operation. They were 2,4-D, 2,4,5-T, Dicamba, Cocydelic acid, and Dioxins. Due to the complexity of the issue, the multiplicity of possible health effects, and the socio-economic implications for imports and exports, the Thai Society of Toxicology submitted a proposal to request World Health Organization (WHO), Geneva. The assistance is for the area of chemical safety and called for immediate action to explore the magnitude qf risk involved with Dioxins. In this paper we present our approach to health risk assessment which takes into an account the epidemiological studies of high-risk group exposed to the Ranch Hand operation. Dioxins are endocrine disruption chemicals which public concerns are developed due to presumption that a hazard exists (www.eva.gov/dioxins/html) for which current methodologies are deemed insufficient. The recent concepts of how oxidative stress toxicants may affect health end points and biomarkers of exposure of exposed individuals are discussed. The aims of this study were to develop the suitable "system software" in chemical ranking and scoring (CRS) for the food hazardous chemicals associated with environmental emission and to suggest the priority lists of food contamination by environmental-origined pollutants. Study materials were selected with reference to the priority pollutants list for environment and food management from domestic and foreign research and the number of study materials is 103 pollutants (18 heavy metals, 10 PBTs, 10 EDs, and 65 organic compounds). The Food-CRS-Korea system consisted of the environmental fate model via multimedia, transfer environment to food model, and health risk assessment by contaminated food intake. We have established that health risks of excess cancer risks, hazard quotients (HQs) by chronic toxicity and HQs by reproductive toxicity convert to score, respectively. The creditable scoring system was designed to consider uncertainty of quantitative risk assessment based on VOI (Value-Of-Information). The predictability of the Food-CRS-Korea model was evaluated by comparing the presumable values and the measured ones of the environmental media and foodstuffs. The priority lists based on emissions with background-level-correction are 15 pollutants such as arsenic, cadmium, and etc. The priority lists based on environmental monitoring date are 17 pollutants including DEHP, TCDD, and so on. Consequently, we suggested the priority lists of 13 pollutants by considering the several emission and exposure scenarios. According to the Food-CRS-Korea system, arsenics, cadmium, chromes, DEHP, leads, and nickels have high health risk rates and reliable grades.
Perfluorooctane sulfonic acid (PFOS) and Perfluorooctanoic acid (PFOA) are the principal chemicals known as perfluoroalkyl acids (PFAs). Despite the widespread use of these compounds, relatively little is known about their fate and effects. The purpose of this study was to determine the toxic effects of PFOS and PFOA on Daphnia magna. In the acute toxicity test, D. magna were exposed for 48 hours at concentrations of 0, 30, 45, 67.5, 101.25 and 151.88 mg/L PFOS, and 0, 100, 160, 225, 337.5 and 506.25 mg/L PFOA, respectively. In the case of chronic toxicity test, D. magna were exposed through water for 21 days at concentrations of 0, 0.375, 0.75, 1.5, 3 and 6 mg/L PFOS, and 0, 1.25, 2.5, 5, 10 and 20 mg/L PFOA, respectively. Acute toxicity was assessed on the basis of immobility, while chronic toxicity was assessed on the basis of fecundity. The acute toxicity test on PFOS and PFOA showed that the values of $EC_{50}$ were 50.90 mg/L and 253.47 mg/L, respectively. In the chronic test, fecundity was reduced significantly at 1.5 mg/L of PFOS and 10 mg/L of PFOA, respectively. These results indicated that PFOS is more toxic to zooplankton than PFOA, and both chemicals have some hazard demonstrates risk for acute or chronic toxicity to freshwater organism.
인류 생활 수준의 증진 및 향상을 위해 화학제품의 사용이 세계적으로 널리 확대 되고 있다. 그러나 이러한 화학제품은 많은 이점을 가지고 있는 반면에 사람과 환경 약영향을 미칠 우려가 있어서, 안전한 사용을 위한 제도를 각국에서 시행하고 있다. 2003년 UN에서 권고된 화학물질의 분류기준과 표시방법을 전세계적으로 통일하기 위한 GHS는 2008년부터 각국에서 도입하도록 권고되었다. 기존의 농약분류체계와 표시방법과 가장 많이 달라지는 것은 물질분류기준이다. 그리고, 그림문자가 표시방법이 정사각형에서 마름모형으로 변화되었고, 3종이 추가되고 1종이 폐지되었다. 따라서, 기존의 농약분류 및 표시제도와 GHS를 면밀한 비교평가와 선진국의 도입방향의 조사를 바탕으로, 우선적으로 물질분류기준변경을 하면서 유독성제품부터 부분적으로 시행하는 것이 타당할 것이다.
본 연구는 치과위생사들의 치과재료에 대한 물질안전보건자료(Material Safety Data Sheet, MSDS)의 인식도와 유해화학물질 취급 시 안전 실태에 대해 파악하고자 실시하였다. 연구는 광주광역시에 근무 중인 치과위생사 250명을 대상으로 2015년 1월부터 2월까지 조사하였고, 235명의 자료를 최종분석 대상으로 하였다. 그 결과 치과재료에 대한 MSDS 인지 정도는 연령이 높을수록 인지도가 높게 나왔으며(p<0.05), 치과병원에 근무하는 치과위생사(p<0.001)에서 유의하게 높았다. 치과재료의 유해화학물질위해성에 대한 인식도에 영향을 미치는 요인은 MSDS인식도(p<0.001)이었고, 유해화학물질 노출 시 응급처치에 대한 인식도에 영향을 미치는 요인은 유해위험성 교육을 받은 경험(p<0.001)으로 나타났다. 따라서 치과위생사들의 치과재료에 대한 MSDS 인식도와 유해화학물질 취급 시 안전 실태에 대해 파악함으로써 유해화학물질 취급 시 안전에 대한 올바른 인식 확립과 이로 인한 사고를 예방하기 위한 교육 프로그램 개발에 기초자료를 제공하고자 한다.
There are many synthetic chemicals, such as di(2-ethylhexyl) phthalate (DEHP) and dibutyl phthalate (DBP), used in chemical reaction processes in industry. The establishment of toxicity and detection of synthetic chemicals that may pose a genetic hazard in our enviornment is subjects of great concern at present DEHP, a ubiquitous phthalate plasticizer, induces a wide range of developmental and reproductive toxicities in mammals. DEHP belongs to the large diverse class of peroxisome proliferator compounds, which include herbicides, hypolipidemic drugs. DBP is a plasticizer used to products containing nitrocellulose, polyvinyl acetate, and polyvinyl chloride such as food wraps and blood bags. DBP is also used in cosmetics as a solvent and fixative for perfumes, a suspension agent for solids, an antifoamer, a skin emollient, and hair spray The present study was performed to examine patterns of gene expression in MCF-7 cells following DEHP and DBP exposure. Changes in gene expression were determined by microarray analysis using KISTCHIP-400 including 401 endocrine related genes based on public database and research papers. Of the genes analysis, we determined that genes detected by array showed a 2-fold or greater change in their expression level(increase or decrease). The results of this study demonstrate that a number of genes were differentially expressed in MCF-7 cells but these changes were not significant. Therefore, we keep going this study using microarray analysis and future studies will examine changes of gene expression on time-course and does treatment in variable cell lines.
Objectives: This study was conducted in order to improve the current understanding of rare earths(RE) and to provide supporting data for establishing occupational health policies by reviewing the toxicological data and issues caused by the use of RE compounds in various fields. Methods: To evaluate the potential toxicity of RE from the viewpoint of occupational health, we summarized extensive reviews of relevant articles in the toxicology(animals and cells), occupational health and safety, and epidemiologic literature. Results: Although occupational RE exposure occurs extensively from ore mining and refining to end users in various industrial applications, epidemiologic study has not been performed among workers up to now. Bioaccumulation and adverse effects of RE have also been mentioned in ore mining regions and nearby residences, but safety standards for each process are insufficient. Moreover, because new commercial recycling technology will soon be applied to various industries, regulation and policies are needed for preventing abuse of recycling. In the results of animal toxicity for a few REs(mostly cerium, lanthanum, and gadolinium), toxicities of liver, lung, blood, and the nervous system were identified due to oxidative stress, but study of long-term RE exposure is required. Understanding the dual effect for RE and discovery of biomarkers pose a scientific challenge in further mechanism studies. Conclusions: In the future, additional hazard evaluation based on animal experiments is required, alongside continuous research for developing analytical methods and discovering biomarkers. Finally, RE occupational health and safety management needs to be integrated into the sustainable use of these materials.
