Spawning, Hatching and Larval Growth of Red and Black Clownfish Amphiprion melanopus (Red and Black Clownfish Amphiprion melanopus의 산란, 부화 및 자어성장)

  • We studied spawning, hatching and larval growth of Amphiprion melanopus under laboratory conditions. Each time a fish spawned about 440 to 650 eggs. The eggs were hatched 9 days later. Nest colour of the eggs changed from clear yellow orange to greyish violet with processing of embryogenesis. With regard to spawning according to the substrates, the fish prefered red clay pot and ceramic pot to volcanic scoria and live rock. Hatching rate (%) of the eggs on the former substrates was significantly higher than that on the latter. The number of eggs varied with the food stuff supplied to the fish. The fish fed Tetra bits (commercial diet), meat of short necked clams and mussels, and astaxanthin showed significantly higher fecundity with 638 eggs than that fed only Tetra bits (477 eggs) or Tetra bits with meat of shellfish without astaxanthin ($549{\sim}553\;eggs$). Higher temperature ranging from 24 to $30^{\circ}C$ induced shorter incubation period and smaller total length, but higher hatching rate. The optimum salinity and L:D cycle with $60\;{\mu}mol$ photon $m^{-2}s^{-1}$ for hatching were 33 psu and 16:8, respectively. The newly hatched larvae about 4.6 mm in total length grew to 7.5 mm in one week, 18.8 mm in a month and 37.9 mm in 70 days after hatching.

  • We examined the effect of salinity and water temperature on hatching and survival rates of fertilized eggs of hybrid grouper (Epinephelus fuscoguttatus ♀ ${\times}$ E. lanceolatus ♂) at different developmental stages, determining optimal conditions for their long-distance transportation. Deformities and hatching rates of fertilized grouper eggs were observed at salinities of 24, 27, 30, 33, 36, and 39 psu. The optimal salinity was determined to be 36 psu, with a survival rate of $70.0{\pm}2.0%$. Transportation experiments at 36 psu were conducted at water temperatures of 21, 24, 27, and $30^{\circ}C$, different developmental stages such as morula, 5-myomere, and tail beating for hatching and survival rates. The optimal water temperature and developmental stage for transporatation were $30^{\circ}C$ and tail beating stage and those hatching rates were $50.6{\pm}1.9%$ and $86.3{\pm}1.3%$, respectively. At $21^{\circ}C$, the survival rate by transportation water temperature was highest ($73.1{\pm}10.6%$), but the hatching rate ($17.1{\pm}3.1%$) was lowest. Therefore, the hybrid grouper fertilized eggs (E. fuscoguttatus ♀ ${\times}$ E. lanceolatus ♂) can be most efficiently produced under long-distance transportation conditions during the tail beating stage and at a water temperature of $30^{\circ}C$.

  • Implantation rates remain low following human in vitro fertilization (IVF). Suboptimal culture conditions may limit the ability of embryos to hatch as blastocysts, and artificial opening of the zona pellucida has been proposed as a means to promote subsequent hatching (assisted hatching). In this study, assisted hatching (AH) by zona drilling using acidic Tyrode's solution was performed in 320 patients, due to their age of more than 38 years (group A), the thick zona pellucida (group Z; $ZP\geq0.18{\mu}m$), and failures in implantation more than 3 times in previous IVF-ET trial (group P). This study was designed firstly, to study the effects of AH on the outcomes of IVF-ET according to the indications and secondly, to verify the appropriate application of AH. The results were as follows; 1. There was no difference in pregnancy rate between AH group (26.6%) and non-AH group (26.5%). 2. Assisted hatching (AH) showed significantly higher pregnancy rate of the patients with thick zona pellucid a than those of the patients with age factor and with the history of repeated implantation failure. But in the patients with age factor only, AH resulted in higher pregnancy rate. 3. Interestingly, the patients with complex factors including zona factor (Z: 33.9%; ZA: 30.4%; ZP: 31.6%; ZAP: 21.4%) showed higher pregnancy rates than other complex factors excluding zona factor (A: 24.4%, P: 0%; AP: 10.8%). From these results, AH is more helpful to the patients with thick zona pellucida rather than patients with older age and/or previous repeated implantation failure.

Yolk resorption, onset of feeding and survival potential of larvae of red spotted grouper, Epinephelus akaara (붉바리 자어의 난황흡수 및 첫 먹이 섭취시기와 관련한 생존 특성)

  • Yolk resorotion and commencement of external feeding in larvae of red spotted grouper, Epinepheus akaara were studied to understand the high mortality of larvae. Consumption of yolk was higher than that of oil globule for the first 24hs after hatching. The resorption rates of yolk and oil globule increased with the increase of temperature. The time yolk was comsumed up to 99% was 84hs after hatching in 23~$25^{\circ}C$,72hs at ~$27^{\circ}C$, and 60hs in 29~~$31^{\circ}C$, while the time for oil globule was 96hs in 23~$25^{\circ}C$, 84hs at $27^{\circ}C$, and 60hs in 29~$31^{\circ}C$. The relationship between mouth openning time (Y) and water temperature (X) was repersented as a linear regression, Y=-2.53X+118.4, (r2=0.89). In 72~96hs after hatching, upper jaw sizes of the larvae reached around 0.114 mm. By the time the larvae comsumed most of the yolk and oil globule, and started external feeding. The relationship between upper jaw length (Y) and total length (X) was represented as a linear regression, Y=0.139X-0.165, (r2=0.88). Rotifer was not found in the guts of larvae before 72hs after hatching at $28^{\circ}C$, when larvae resorbed their yolk and oil globule over 99%. But, at 96hs after hatching, about 33% of the larvae had the rotifer in their guts. Delayed feeding experiments revealed that the earlier food supply within 84hs after hatching showed the better survival rates for the larvae. The larvae fed at 96hs after hatching died all within 120hs.

Osteological Development of Larvae and Juveniles of Sea Raven, Hemitripterus villosus in Coastal Waters off Yeosu (여수 연안산 삼세기(Hemitripterus villosus) 자치어의 골격발달)

  • Hemitripterus villosus, a promising aquaculture fish species, is facing declining stocks. This study aims to provide normative standards for skeletal development to address persistent skeletal deformities in farmed fish. Specimens utilized in the study underwent artificial insemination with captured fish, and the resulting larvae and fry were preserved in a formalin solution. The skeletal ossification process commenced immediately after hatching, affecting the parasphenoid, premaxillary, maxillary, and dentary structures at an average total length of 13.65±0.71 mm (n=5). By sixty-five days post-hatching, ossification extended to the ethmoid and supraorbital, completing the head's development at an average total length of 21.24±0.50 mm (n=5). Clavicle ossification occurred at seven days post-hatching, corresponding to an average total length of 14.61±0.52 mm (n=5). At forty-four days post-hatching, the ossification of 4 actinosts took place, completing the shoulder girdle, with an average total length of 18.15±0.61 mm (n=5). Vertebral ossification initiated at ten days post-hatching, with an average total length of 14.80±0.65 mm (n=5). By fifty-four days post-hatching, 39 vertebral columns were ossified, reaching an average total length of 18.67±0.54 mm (n=5). Vertebral development was complete at sixty days post-hatching, with an average total length of 20.25±0.45 mm (n=5). This study sheds light on the skeletal development of H. villosus, providing valuable standards and fundamental data for understanding skeletal deformities in this species.

Induced Morphological Changes in Larval Rock Bream, Oplegnathus fasciatus, under Starvation

  • Morphological changes in the reared rock bream, Oplegnathus fasciatus, from hatching to six days after hatching were examined during the early growth stage under starvation. All the larvae died within five days when feeding was delayed for three days after hatching. These results imply that initial larval food should be supplied within two days of hatching. Changes in the pectoral angle and the ratios of eye height to head height, gut height to standard length, and gut height to myotome height in the rock bream are alternative indicators for the identification of starving fish. These indicators might prove useful in evaluating the successful transition from endogenous to exogenous feeding in this species.

  • An assessment of Marsh crocodile's (Crocodylus palustris LESSON, 1831) habitat, its feasibility in supporting the total population, feeding, hatching difficulty and variations in egg and clutch size of Marsh crocodile were carried out in Bangabandhu Sheikh Mujib Safari Park for one year (July 2010 to June 2011). Major problem identified was the breeding failure, i.e. till now no hatching were found. Mainly two local weather variables (average temperature and humidity) were identified as the most obstructing parameters. Higher temperature (average $37.03^{\circ}C$) and Lower Humidity (average 75.33%) in comparison with standard range of incubation temperature and humidity for Marsh crocodile were seems to be vital reason for the failure of hatching. Understanding of the identified problem of this paper can direct the wildlife conservationist and the policy maker to take proper action for conserving the Marsh Crocodile in Bangladesh.

  • This experiments were carried out to know hatching power on treatments of cold temperature and hydrochioric acid Iron Late January to Early February. Used silkworm variety was Jam 103${\times}$ Jam 104 laying in Spring. The results may be summarized as follows: 1. The resting of silkworm eggs activated to Late January, to longer cold treatment was, the more effects of activating increased completely. So activated eggs were possible to incubation. 2. How to the light or short hours of hycrochloric acid were compare with heavy or long each other, it was inclined that the former seems to be worse than the latter. 3. On the interaction of cold treatments and hydrochloric acid, in case of short cold treatment, hatching power was better effect all treatments of hydrochloric acid than control, but no difference between specific gravity or treatment hours. On tile contrary, in case of more longer of cold treatments, hatching power depended not only upon the specific gravity, but also upon the treatment hours.

Effect of Microsporidian Infection on Reproductive Potentiality on Mulberry Silkworm, Bombyx mori L. (Lepidoptera: Bombycidae) in Different Seasons

  • Infection of pathogenic microsporidia, Nosema bombycis and Nosema mylitta (Chakrabarti and Manna, 2006) decreased egg production, fecundity, hatching % and increased sterile eggs in heavily infected mulberry silkmoth, Bombyx mori L. On an average a disease free moth laid upto 442.67 eggs with high hatching % (99.53) and less sterile eggs ($0.47{\sim}2.00%$). While an infected moth laid less number of eggs ($7.00{\sim}412.00$) with low hatching % ($32.437{\sim}98.643$) and high sterile eggs ($2.143{\sim}129.571$). Fecundity of disease free laying was highest (468.714) during season-1 then gradually decreased during season- 2 (414.000) to season- 3 (404.285). But fecundity of an infected laying was highest during season-2 and hatched eggs were lowest during season-2. Higher inoculums concentration of N. mylitta infected to 5th stage larva of mulberry silkworm drastically decreased the fecundity in season - 3 and lower inoculums concentration of N. bombycis decreased the fecundity in season-1 and 3. Season-3 was most effective season to decrease the fecundity and increase sterile eggs when both temperature and humidity were fluctuated from the optimum level.

Immunohistochemical study on insulin, glucagon and somatostatin immunoreactive cells of the pancreas of the duck(Anas platyrhynchos platyrhyncos, Linne) (청둥오리 췌장의 insulin, glucagon 및 somatostatin 면역반응세포에 관한 연구)

  • The distributions and relative frequencies of insulin-, glucagon-, and somatostatin-immunoreactive cells in the pancreas of the duck(Anas platyrhynchos platyrhyncos, Linne) were investigated immunohistochemically on 23 days of incubation, at hatching, 1 week, 2 weeks, 3 weeks, 5 weeks, 6 weeks, 7 weeks, 9 weeks, 10 weeks, and 32 weeks after hatching. In the duck pancreas on 23 days of incubation and at hatching, mammalian type islets(mixed type) were only observed, thereafter three type's islets(mamalian, A and B type's islets) were identified. Insulin-immunoreactive cells were detected in central region of the islets, while glucagon- and somatostatin-immunoreactive cells were detected in marginal region of light(B type) or mammalian type islets, and in central region of dark islets(A type). Insulin-, and somatostatin-immunoreactive cells were also detected in the exocrine regions. In this region the insulin-immunoreactive cells were detected from 23 days of incubation to 6 weeks, however not detected after 7 weeks. At hatching the relative numbers of somatostatin-immunoreactive cells were more frequent than those of other groups, and then decreased with ages.
