Histopathological changes on the testis by Ivermectin toxicity (Ivermectin투여가 고환에 미치는 영향에 관한 병리조직학적 관찰)

  • To know the effect of Ivermectin(IVM) toxicity in testis, histopathologic changes as well as clinical signs were observed in experimental animals including dogs by the subcutaneous injection with 3-50mg/kg of IVM. Clinically, it was observed to have depression and ataxia in all groups whereas tremor and coma in mice, rats and guinea pigs, coma in hamsters and rabbits, and tremor and salivation in dogs were shown. The clinical signs were different by the dosage of IVM, species and individuals in all animals. Susceptibility to IVM was most sensitive in dogs, especially in a Tosa dog and this was susceptible in mice, hamsters and rabbits, guinea pigs and rats in order. Microscopical observation revealed that the seminiferous tubules of testis had decreased thickness of germinal epithelium due to the necrosis and desquamation of the spermatids and spermatocytes. The progressive pattern by the times of administration showed vacuolar formation between the layer of spermatids and spermatogonia due to the marked necrosis of spermatocytes and the presence of multinucleated giant cells derived from spermatid throughout the seminiferous tubules of testis. Only a layer of spermatogonia, a few spermatogonia, and Sertoli cells wore observed with atrophied wavelike basement membrane in the seminiferous tubules of testis. Necrotic germinal cells, sloughed immature spermatids and spermatocytes were present in the lumen of epididymis and ductus deferens. Microscopical observation showed different susceptibility to IVM with clinical observation in which it was also most sensitive in dogs, especially in a Tosa dog and this was susceptible in rabbits and guinea pigs, hamsters, rats and mice in order. It was considered that IVM affects mainly spermatocyte or spermatid stage in the spermatogenesis and disturbs their developing beyond these stage.

  • Three morphological types of the genus Metagonimus were found encysted in the dace, Tribolodon tacganowskii, caught from the Sumjin River, Korea. They include Metagonimus Yokogawa type (M. yokogawai) , Miyata type, and Koga type according to Saito's classification. The metacercariae were experimentally fed to rats and hamsters and a total of 1,624 Metagonimus worms were recovered from their small intestine. The number of each type was 820 for Yokogawa type, 688 for Miyata type, 46 for Koga type, and 70 undetermined. The major differential keys between Yokogawa and Miyata types were in the position of two testes, distribution of uterine tubules, and size of the eggs, and the keys between Miyata and Koga types were in the position of two testes, distribution of vitelline follicles, and size of the eggs. The validity of Miyata type as a specific level should be retained until more convincing evidences are obtained. Koga type is regarded as a synonym of M. takahashii. The present results reveal that there are 3 morphological types of Metagonimus whose larvae are encysted in the dace, T. tacganowskii, from the Sumjin River. The Effect of Irradiation on the Expression of PCNA and Apoptosis Induction during the DMBA Induced Carcinogenesis in Hamster Buccal Pouch (DMBA 유도 햄스터 협낭암 발생과정에서 방사선 조사가 증식세포핵항원(PCNA)의 발현과 apoptosis 유발에 미치는 영향)

  • Purpose : This study was carried out to investigate the effect of irradiation on the expression of proliferating cell nuclear antigen (PCNA) and apoptosis induction during the carcinogenesis in hamster buccal pouch. Materials and methods: Three months old Syrian golden hamsters were divided into control and 2 experimental groups. Hamsters in control group were left untreated on buccal pouchs. Twenty four hamsters were treated with 0.5% DMBA tri-weekly on the right buccal pouch. Forty eight hamsters were treated with 0.5% DMBA tri-weekly and irradiated with the dose of 5 Gy and 10 Gy at 6, 9, 12, 15 weeks after DMBA application. Resected buccal pouches were sectioned and examined for potential expression pattern of PCNA and apoptosis. Results : The PCNA index was increased with the stages of buccal pouch epithelium carcinogenesis except the hyperplasia stage in control group (p<0.05). The irradiation did not effect on the PCNA index in the dysplasia and the carcinoma in situ stage, but in the hyperplasia stage, the PCNA index was increased with 10 Gy radiation and decreased in the carcinoma stage (p<0.05). The apoptotic index was significantly decreased from the carcinoma in situ stage and the lowest in the carcinoma stage. The apoptotic index was significantly decreased in the hyperplasia and dysplasia stage with the 5 Gy irradiation and significantly increased only in the carcinoma stage with the 10 Gy irradiation (p<0.05). Conclusion: The PCNA and apoptotic index were varied according to the irradiation period and dosage in each carcinogenesis stage.

  • Effects of 5 and 10% dietary supplement tablets containing green tea, buckwheat, and grape leaf extracts on atherosclerosis, and plasma and hepatic lipid compositions in hamsters fed antherogenic diet [modified AIN-76A containing 10% coconut oil (w/w), 0.05% cholesterol] were examined. Compared to atherogenic diet only groups, addition of supplement to atherogenic diet-fed group significantly down-regulated triglyceride and total cholesterol levels in plasma and liver. F1B hamsters fed atherogenic diet had greater foam cell accumulation compared to control and those fed atherogenic diet with addition of supplement. Lipid metabolisms of plasma and liver increased with addition of supplement dose-dependently, whereas aortic lipid accumulation decreased. These results indicate tablets supplemented with green tea, buckwheat, and grape leaf extracts have potential to prevent cardiovascular diseases in golden Syrian hamsters.

Immunohistochemical detection of antigens in tissue sections of experimentally infected hamsters (Leptospira canicola 실험적 감염햄스타의 조직절편내 면역조직화학적 균체항원검출)

  • The present study was done to detect Leptospira canicola antigens in paraffin sections from experimentally infected hamsters with PAP method. The anti-Leptospira canicola serum used as first antibody was prepared by immunizing rabbits with Leptospira canicola. Positive reactions were detected in interstitium, tubular epithelial cells and glomerulus of the kidney, and in the hepatic sinusoid. With PAP method, leptospiral antigens were detected at the 48 hours after bacterial infection, but isolation, silver stain, and dark field microscopic examination of the samples did 48 hours later than PAP method. The results suggested that PAP method is considered as a reliable tool for confirmative diagnosis of this disease.

The Potential Regressive Role of Syzygium aromaticum on the Reproduction of Male Golden Hamsters

  • The flower buds of Syzygium aromaticum (clove) have been used as traditional medicine for the treatment of male sexual disorders in Asian countries. Recently, there are some reports about the effects of the clove on reproductive activities in mammals. Therefore, its effect on testicular function was examined in male golden hamsters whose reproductive activity is inhibited by photoperiod such as winter climate. The male animals were given by daily oral administrations (56 consecutive days) in three doses (4 mg, 20 mg, and 100 mg/kg BW) of the alcoholic extract of the clove. Generally lower dose (4 mg) of the extract continued to keep the reproductive activities of testes. The both middle and high doses (20 mg and 100 mg) of the extract completely inhibited the testicular activity in some animals. Taken together, these results suggest a possible biphasic action of alcoholic extract of Syzygium aromaticum flower bud on testicular function.

Evaluation of Irritating Potential of Newly Developed Toothpaste in the Hamster Oral Mucous Membrane

  • Oral mucous membrane test using Syrian hamsters was performed to evaluate the reliability as a model system for the assessment of the potentially irritating substances intended for the mucous membranes, and to determine the irritating potential of a new emulsion-type formulated toothpaste. After test substances were implanted into the cheek pouches of hamsters with diluents (20 mg/kg) under pento-barbital sodium anesthesia, we made the comparison in irritation between emulsion-type and dispersion-type of triclosan (TCS) formulations in the range of 0.2% to 0.3%. The emulsion-type formulations using non-ionic surfactant showed less mucosal lesion than other commercial toothpastes with 0.3% TCS, or dispersion-type ones. However, no significant difference in irritation was detected between 0.2% and 0.3% TCS. We report that this hamster cheek pouch method could be a reliable approach for the evaluation slight difference in the irritating potentials of cosmetics and hygiene products intended for the lips or other mucous membranes, and this method showed that the new emulsion-type formulation significantly lowered the TCS-induced toxicity, compared with other commercial toothpastes.

  • To elucidate pathogenicity to hamster of encephalomyocarditis virus $K_3$ strain that was isolated in Korea from the swine with reproductive failures, adult male syrian hamsters were experimentally infected intraperitoneally with the virus at $10^{7.0}\;TCID_{50}/0.1ml$ and pathological and immunohistochemical studies were performed. The results obtained through the experiment were as follows. 1. Clinical signs such as depression, unkempt hair and bilateral parlysis of hind limbs were observed. 2. At necropsy, mild congestion was observed in the cerebrum, liver, kidney and lung, and atrophy was evident in testis. 3. In microscopic observation, degeneration and necrosis of the nervous cells and perivascular mononuclear cell infiltration were manifested in central nerve system, and various degrees of degeneration and necrosis of parenchymal cells were detected in pancreas, lacrimal gland, liver, kidney and testis. 4. In immunohistochemical observation, strong positive reactions were observed in degenerated parenchymal cell of testis, and weak positive reactions, in hepatocytes.

Timing of Fertilization In Vitro of Follicular Oocytes by Bull Spermatozoa Preincubated in the Uteri Isolated fom Estrous Hamsters (햄스터 적출자궁에서 전배양한 소 정자의 난포란에의 침입시기 및 체외수정 초기상에 관한 연구)

  • The cattle follicular oocytes matured for 26~28h in culture condition were examined at 4, 5, 6, 7, 8, 10, 12, 14, 16 and 18h after insemination with bull spermatozoa preincubated for 4.5h in the uter isolated from estrous hamsters. After further culture with spermatozoa for 4~18 h, 73~89% of the total oocytes had matured to the second metaphase. None of the follicular oocytes matured in culture, were fertilized 5h after insemination. But when the oocytes were examined at 6, 8, 10, 14 and 18h after insemination, 60, 73, 82, 80 and 87% of oocytes were fertilized, respectively. The majority of the fertilized oocytes had enlarged sperm head at 6h after insemination and a part of the fertilized oocytes begun to develop from enlarged sperm head to male pronuclear stage at 8h after insemination, and most of them developed to male and female pronuclear stage at 10h after insemination. The results suggest that the penetration of spermatozoa into the oocytes may occur earlier than 6h after insemination and development of their pronuclear stage may occur at 8h after insemination.
