Halobacterium halobium 의 생육조건 및 Protease 에 관한 연구 (A Study on Growth Condition and Proteolytic Enzyme of Halobacterium halobium)

  • In salt-preserved foods of every kinds, it was examined the growth condition of halophilic bacteria that induced a change of colour, taste, nutritive substance, a production condition of enzyme and a character of crude enzyme. Used bacteria is H. halobium a kind of extremely halophilic bacteria, and the required of optimum culture needed a quite long time of crude enzyme production is 168 hours. Optimum pH is about 7-7.5, so the traditional food of such neutrality pH as soybean paste and soy sauce particularly come into trouble because the growth can flourish in neutrality or alkaliescence, and the crude enzyme also appeared that best activation between pH 6 and pH 8. The optimum temperature is about 37$^{\circ}C$, the optimum temperature of enzyme is about 40 $^{\circ}C$ and the temperature stability is settled for 15 minutes and it is completely inactivated at 10 minutes. In the influence of each metal ion, Fe++ and Mn++ a stimulated the growth of H.halobium and the activation of enzyme, Cu++ and Zn++ were identified that made the growth and the activation of enzyme inhibit.

  • Halobacterium Halobium에서 항우울제의 부분 몰 부피와 몇 가지 지질의 부유물은 과량의 부피 팽창 계를 사용하여 $25^{\circ}C$에서 측정되었다. 일반적인 항우울제 인 Fluoxetine의 효능은 오랫동안 지질 용해도와 관련이 있는 것으로 알려져 왔다. Halobacterium Halobium의 보라색 막에서 유일한 막 단백질 인 소포의 변성은 280nm에서 흡수 변화에 의해 연구되었고 과도한 부피 팽창 계로 330nm에서 형광 변화를 보였다. 확산 교환에 의한 점성 중합체 용액의 1H NMR 분석은 측정을 위한 중요한 단계로, Halobacteriun Halobium에서 Fluoxetine의 부분 몰 부피와 입자 크기는 양성으로 측정되었다. 항우울제는 낮은 사기와 우울증에 근거하여 다양한 인지 및 정신 증상을 유발하는 질병을 예방하여 일일 수행 능력을 저하시키는 걸로 나타났다.

  • Bacteriorhodopsin in the purple membrane (PM) of halobacteria has recently been attracting much attention to be used as a component of molecular electron device and optical computers. In order to increase the productivity of bacteriorhodopsin in high cell density cultures of Halobacterium halobium R1, an internal membrane cell-retention bioreactor system was employed. As a result, the production of cell mass at OD660 of 12 and of bacteriorhodopsin at 125-130 mg/L were obtained using the internal membrane bioreactor system at a dilution rate of 0.066 hr-1. The productivity achieved by the internal membrane system (0.7 mg/L$.$hr) was 3.5-fold higher than that obtained by the corresponding batch cultivations (0.2 mg/L$.$hr).

  • The excess volumes of mixing of benzyl alcohol, ethyl alcohol, halothane, and procaine in vesicle and suspensions of several lipids have been determined at $25^{\circ}C$ using a excess volume dilatometer. The potency of general anesthetics has long been known to be correlated with lipid solubility. Denaturations of the bacteriorhodopsin, which is a sole membrane protein in the purple membrane of Halobacteriun Halobium, were studied by UV/Vis absorption changes. The excess volumes of mixing of benzyl alcohol and procaine in egg lecithin were all found to be negative and this result was confirmed as Miller's supposition.

Denaturations of the vesicle, which is a sole membrane protein in the purple membrane of Halobacteriun Halobium, were studied by absorption changes at 280 nm and fluorescence changes at 330 nm and excess volume dilatometer. The particle size analysis of viscous polymer solutions by diffusional interchange is the key step by measurement. The excess volume of mixing in chitosan was found to be negative, whereas them of purple membrane, Halobacteriun Halobium and red membrane were positive in benzyl alcohol and halothane. This result was confirmed as Miller's supposition.

  • Halobacterium halobium에서 추출한 Bacteriorhodopsin(bR)을 고분자 물질에 첨가해 박막화하였다. 이 단백질 박막 소자의 photodynamic특성을 이용하여 광 switching system을 구성하였다. 광 switch system은 pump광을 이용해 B상태와 M상태에서 축퇴4광파 혼합방식으로 구성했다. Red, blue, violet 광원을 사용하여 B상태와 M상태 격자를 형성하고 on-off되는 위상공액신호를 측정했다. (중략)

산성 pH가 박테리오로돕신의 분광학적 성질에 미치는 효과 (The Effect of Acidic pH on the Spectral Properties of Bacteriorhodopsin)

  • Halobacterium halobium으로부터 분리 정제된 퍼플멤브레인을 7.5% 폴리아크릴아미드겔에 혼합시켰다. 이 겔을 사용하여 pH 변화에 따른 흡수 스펙트라와 원편광 이색성스펙트라를 얻었다. pH 7.0에서 보여준 이들 스펙트라의 성질들은 수용액내에 부유하고 있는 퍼플멤브레인으로부터 얻은 것들과 동일하였다. pH 2.7에서는 최대흡광도를 605nm에서 나타내었으며 pH 0.8에서는 565nm에서 보여주었다. 광에 노출되지 않았던 겔의 경우는 광에 노출되었던 겔과는 달리 등흡광점을 보여주었다. pH 2.7과 pH 0.8에서 측정한 원편광 이색성스펙트라는 pH 7.0에서 보여준 bilobed형을 유지하였으며 UV영역에서 분자편광도나 스펙트라의 모양도 pH에 따라 큰 영향을 받지 않았다. pH 2.7에서 생성된 bR605acid 가 퍼플멤브레인의 정상 광화학 순환기 중간 생성물인 $O^{640}$와 유사한 성질을 가진종이 아닐까 추측된다.

  • Dry orderly oriented purple membrane from Halobacterium halobium was obtained by a new technique of preparation. This oriented purple membrane film was very stable, nearly permanently, and showed long term reproducibility with respect to its photochemical behavior. In addition, we have investigated the photooptical properties in terms of the M$_{412}$ intermediate of the bacteriorhodopsin photocycle with respect to the humidity of the film. The relative optical density, i.e. its apparent concentration of the M$_{412}$ intermediate was decreased with the humidity increase as a function of the intensity of the exciting flash within our experimental range. It is suggested that the bound water molecules play an important role in the structure of the bacteriorhodopsin.
