Characteristics for Reinforcement Corrosion and Chloride Ion Diffusion of High Volume Fly Ash Concrete (하이볼륨 플라이애시 콘크리트의 철근부식 및 염소이온 확산 특성)

  • The purpose of this experimental research is to evaluate the resistance of reinforcement corrosion and chloride ion penetration of high volume fly ash (HVFA) concrete. For this purpose, concrete test specimens were made for various strength level and replacement ratio of fly ash, and then compressive strength and diffusion coefficient for chloride ion of them were measured for 28, 91 and 182 days, respectively. Also, corrosion monitoring by half cell potential method was carried out for the made lollypop concrete test specimens to detect the time of corrosion initiation for reinforcement in concrete. As a result, it was observed from the test results that compressive strength of HVFA concrete was decreased with increasing replacement ratio of fly ash but long-term resistance against reinforcement corrosion and chloride ion penetration of that was increased.

Enhanced Sialylation of Recombinant Erythropoietin in CHO Cells by Human Glycosyltransferase Expression

  • Sialylation, the attachment of sialic acid residues to a protein, can affect the biological activity and in vivo circulatory half-life of glycoproteins. Human ${\alpha}2$,3-sialyltransferase (${\alpha}2$,3-ST) and ${\beta}1$,4-galactosyltransferase (${\beta}1$,4-GT) are responsible for terminal sialylation and galactosylation, respectively. Enhanced sialylation of human erythropoietin (EPO) by the expression of ${\alpha}2$,3-ST and ${\beta}1$,4-GT was achieved using recombinant Chinese hamster ovary (CHO) cells (EC1). The sialic acid content and sialylation of N-glycans were evaluated by HPLC. When ${\alpha}2$,3-ST was expressed in CHO cells (EC1-ST2), the sialic acid content (moles of sialic acid/mole of EPO) increased from 6.7 to 7.5. In addition, the amount of trisialylated glycans increased from 17.3% to 26.1 %. When ${\alpha}2$,3-ST and ${\beta}1$,4-GT were coexpressed in CHO cells (EC1-GTST15), the degree of sialylation was greater than that in EC1-ST2 cells. In the case of EC1-GTST15 cells, the sialic acid content increased to 8.2 and the proportion of trisialylated glycans was markedly increased from 17.3% to 35.5%. Interestingly, the amount of asialoglycans decreased only in the case of GTST15 cells (21.4% to 14.2%). These results show that coexpression of ${\alpha}2$,3-ST and ${\beta}1$,4-GT is more effective than the expression of ${\alpha}2$,3-ST alone. Coexpression of ${\alpha}2$,3-ST and ${\beta}1$,4-GT did not affect CHO cell growth and metabolism or EPO production. Thus, coexpression of ${\alpha}2$,3-ST and ${\beta}1$,4-GT may be beneficial for producing therapeutic glycoproteins with enhanced sialylation in CHO cells.

Glycosylation Enhances the Physicochemical Properties of Caffeic Acid Phenethyl Ester

  • In this study, we synthesized a glycosylated derivative of caffeic acid phenethyl ester (CAPE) using the amylosucrase from Deinococcus geothermalis with sucrose as a substrate and examined its solubility, chemical stability, and anti-inflammatory activity. Nuclear magnetic resonance spectroscopy showed that the resulting glycosylated CAPE (G-CAPE) was the new compound caffeic acid phenethyl ester-4-O-${\alpha}-{\small{D}}$-glucopyranoside. G-CAPE was 770 times more soluble than CAPE and highly stable in Dulbecco's modified Eagle's medium and buffered solutions, as estimated by its half-life. The glycosylation of CAPE did not significantly affect its anti-inflammatory activity, which was assessed by examining lipopolysaccharide-induced nitric oxide production and using a nuclear factor erythroid 2-related factor 2 reporter assay. Furthermore, a cellular uptake experiment using high-performance liquid chromatography analysis of the cell-free extracts of RAW 264.7 cells demonstrated that G-CAPE was gradually converted to CAPE within the cells. These results demonstrate that the glycosylation of CAPE increases its bioavailability by helping to protect this vital molecule from chemical or enzymatic oxidation, indicating that G-CAPE is a promising candidate for prodrug therapy.

The systematic sampling for inferring the survey indices of Korean groundfish stocks

  • The Korean bottom trawl survey has been deployed on a regular basis for about the last decade as part of groundfish stock assessments. The regularity indicates that they sample groundfish once per grid cell whose sides are half of one latitude and that of one longitude, respectively, and whose inside is furthermore divided into nine nested grids. Unless they have a special reason (e.g., running into a rocky bottom), their sample location is at the center grid of the nine nested grids. Given data collected by the survey, we intended to show how to appropriately estimate not only the survey index of a fish stock but also its uncertainty. For the regularity reason, we applied the systematic sampling theory for the above purposes and compared its results with a reference, which was based on the simple random sampling. When using the survey data about 11 fish stocks, collected by the spring and fall surveys in 2014, the survey indices of those stocks estimated under the systematic sampling were overall more precise than those under the simple random sampling. In estimates of the survey indices in number, the standard errors of those estimates under the systematic sampling were reduced from those under the simple random sampling by 0.23~27.44%, while in estimates of the survey indices in weight, they decreased by 0.04~31.97%. In bias of the estimates, the systematic sampling was the same as the simple random sampling. Our paper is first in formally showing how to apply the systematic sampling theory to the actual data collected by the Korean bottom trawl surveys.

The experimental study for hemodynamic changes in the heart-lung preparatio by autoperfusion (자가관류법에 의한 체외심폐의 혈역학적 변동에 관한 실험적 연구)

  • The experimental study for extracorporeal preservation of the heart-lung preparation by autoperfusion system was performed in 10 dogs. Under intravenous Pentothal endotracheal anesthesia bilateral thoracotomies were performed. A 24F cannula connected to a plastic reservoir bag located 100 cm above the level of the heart was introduced into the aortic arch. Left subclavian, innominate artery, and descending aorta were ligated and divided. Both vena cavae were ligated and divided after the bag was half filled with blood. A 24F catheter inserted into right atrium and connected to the plastic bag in order to keep constant the preload. The thoracic trachea was intubated and the lungs were ventilated. The heart-lung preparations were removed en bloc and floated in a $34^{\circ}C$ bath of Hartmann solution. The preparations were observed for from 2 hours to 8 hours, with the average of 5.2 hours. Hemodynamic and hematologic variables were measured during preharvest and autoperfusion. The pH revealed severe respiratory alkalosis due to very low $PaCO_2$ during autoperfusion ; $PaO_2$ remained constant for 130-140 mmHg; $A-aDO_2$ increased markedly. The static inspiratory pressure [SIP] at late autoperfusion [6hr] increased significantly as compared with at early autoperfusion [2hr]. There was no difference between white blood cell counts from right atrium and those of left atrium. Heart rates remained constant for 110-120/min; cardiac outputs maintained to approximately 0.6L/min; mean aortic pressures, 75 mmHg; mean pulmonary arterial pressures, 15-18 mmHg; mean right atrial pressures, 9-13 mmHg; mean left atrial pressures, 12 mmHg lower than those of right atrium. Serum Na maintained with normal range during autoperfusion; K increased significantly; Ca decreased progressively. Hemoglobin and hematocrit decreased significantly during autoperfusion. The study demonstrated that stable hemodynamics could be maintained throughout the experiment and the preparation of the lung seemed to be inadequate, especially after 3-4 hours, such as high $A-aDO_2$, increased SIP, and scattered atelectasis and edema in their gross appearances.

Effect of the characteristics of buoy on the holding power of trapnet (부이의 특성이 통발어구의 고정력에 미치는 영향)

  • In this paper, numerical modeling is conducted to analyze the tension of an anchor line by varying the size and drag coefficient of a buoy when the trapnet is influenced by the wave and the current simultaneously. A mass-spring model was used to analyze the behavior of trapnet underwater under the influence of waves and current. In the simulation of numerical model, wave height of 3, 4, 5 and 6 m, a period of 4.4 s, and the flow speed of 0.7 m/s were used for the wave and current condition. The drag coefficients of buoy were 0.8, 0.4 and 0.2, respectively. The size of buoy was 100, 50 and 25% based on the cylindrical buoy ($0.0311m^3$) used for swimming crab trap. The drag coefficient of the trapnet, the main model for numerical analysis, was obtained by a circular water channel experiment using a 6-component load cell. As a result of the simulation, the tension of the anchor line decreased proportional to buoy's drag coefficient and size; the higher the wave height, the greater the decrease rate of the tension. When the buoy drag coefficient and size decreased to one fourth, the tension of the anchor line decreased to a half and the tension of the anchor line was lower than the holding power of the anchor even at 6 m of wave height. Therefore, reducing the buoy drag coefficient and size appropriately reduces the trapnet load from the wave, which also reduces the possibility of trapnet loss.

Influence of freezing upon the cooking time and eating quality of beans (동결이 두류의 cooking time과 맛에 미치는 영향)

  • The effect of freezing on cooking times, tastes and microstructures of beans were examined. Freezing was effective in shortening of cooking time and improving of the taste: while the cooking time was reduced to one-half by freezing, textures, flavors and overall acceptabilities of black bean and soybean were improved by freezing. A high correlation was found between sensory texture and Instron puncture force, and sensory texture was predictable from puncture force using equation. The microstructure of cotyledonary cells of soybean was characterized with thick cell wall and no difference was observed between frozen and non-frozen soybean. But the spherosome enclosing the protein body was destructed by freezing.

Improvement of semiconductor contact hole filling of Copper by ionized cluster beam deposition technique (이온화클러스터빔 증착법에 의한 구리 박막의 반도체 접촉구 메움 향상에 관한 연구)

  • A study to improve filling of semiconductor contact holes by enhancement of the directionality of the source beams has been undertaken. The collimation of source beams was improved by the ionized cluster beam deposition technique with modification of the cell geometry. The collimation tested with neutral beam was excellent. But, the Cu flims were grown in a columnar mode due to the lack of surface mobilit of the impinged clusters. A shadow effect also caused cleavage and consequent discontinuity at the steos as films grow. By applying acceleration voltage, the columnar growth in a contact hole of 0.5 $\mu$m diameter and 1 $\mu$m height disappeared and considerable coverage at the side wall of the contacts as well as perfect bottom coverage were observed. These are all due to the assistants of the accelerated ionized clusters with high kinetic energy. Thus we demonstrated that the ICB deposition technique can be used to completely fill sub-half-micron contact holes with high aspect ratio.

RF 스퍼터링법을 이용한 리튬이차전지용 Li[Li0.2Mn0.54Co0.13Ni0.13]O2 양극박막의 제조 및 전기적 특성

  • 최근 전기, 전자, 반도체 산업의 발전으로 전 고상 박막리튬전지는 초소형, 초경량의 마이크로 소자의 구현을 위한 고밀도 에너지원으로 각광받고 있다. 현재 양극박막은 대부분LCO(LiCoO2)계열이 이용되고 있으나, 코발트는 높은 가격과 인체 유해성 뿐만 아니라 상대적으로 낮은 용량(~140 mAh/g)등의 단점을 갖고 있어 향후 보다 고용량의 양극박막이 요구된다. 3원계 양극활물질 LiMO2(M=Co,Ni,Mn,etc.)은 우수한 충방전 효율 과 열적 안정성 뿐 아니라 277mAh/g의 높은 이론용량을 갖고 있어 고용량 양극박막으로의 적용시 고용량 박막이차전지 제작이 가능하다. 본 연구에서는 전 고상 박막 전지의 구현을 위하여 RF 스퍼터링법을 사용하여 Li[Li0.2Mn0.54Co0.13Ni0.13]O2 박막을 증착하였다. Li/MnCoNi의 몰 비율을 변화시켜 높은 전기화학적 특성을 갖는 분말을 합성하여 제조한 타겟으로 Pt/TiO2/SiO2/Si 기판위에 RF 스퍼터법을 이용하여 박막을 성장시켰다. 박막 증착 시 가스의 비율은 Ar:O2=3:1로 하고 증착 압력의 조절(0.005~0.02 torr)을 통하여 박막의 두께와 표면 특성을 조절하며 성장시켰다. 또한 박막을 다양한 온도에서($400{\sim}550^{\circ}C$) 열처리하여 결정화도와 전기화학적 특성을 측정하였다. 증착 된 박막의 구조적 특성은 X-ray diffraction(XRD) 과 scanning electron microscopy(SEM)로 관찰되었다. 박막의 전기화학적 특성 평가를 위하여 Cyclic voltammatry를 측정하여 가역성의 정도를 확인하고 WBC3000 battery cycler를 이용한 half-cell 테스트를 통하여 박막의 용량을 평가하였다.

A Study of Accelerated Corrosion Test and Chloride Penetration Analysis with Artificial Seawater Immersion Condition (인공해수 침지조건에 따른 부식촉진시험과 염화물침투해석에 대한 연구)

  • Steel reinforcement buried in concrete structure in submerged zone does not easily become corroded due to lack of dissolved oxygen. For that reason, accelerated corrosion test in submerged state is performed with an electrochemical method, which is not suitable for actual corrosion mechanism and makes it difficult to find relevance with long-term behavior. In this study, accelerated corrosion test was performed with the temperature and chloride concentration as main variables in order to establish a method for accelerated corrosion test in submerged zone. Corrosion was determined by the result of reinforcement corrosion monitoring based on galvanic potential measurement and half-cell potential method. The accelerated corrosion test result showed that temperature had the most dominant influence. To determine the chloride content, chloride concentration by depth in the test sample was measured. With the same conditions, chloride penetration interpretation was performed by DuCOM, a FEM durability interpretation program. Also, a test was performed to measure dissolved oxygen according to soaking conditions of artificial seawater, which was used for verifying the validity of the accelerated corrosion test result.
