Characterization of the Folding Structure of 3'-end of Lactate Dehydrogenase A-mRNA Isolated from Hormone Stimulated Rat $C_{6}$ Glioma cell culture (홀몬으로 처리된 쥐의 $C_{6}$ glioma 세포배양으로부터 분리된 낙산탈수소 효소 A-mRNA의 3'-말단의 2차 구조)

  • Rat liver LDH A-cDNA has been isolated from a .lambda.gt11-rat lover cDNA library and partially characterized. The size of the isolated rat liver LDH A-cDNA if shown to be 1.6Kb and restriction enzyme sites for the rat liver LDH A-cDNA are also mapped. 682-nucleotide sequence coding for 3'-end of rat liver LDH A-cDNA has been analyzed and compared to the nucleotide sequence of the same region of rat $C_6$-glioma cell LDH A-cDNA which has been cloned from the hormonally stimulated cell cultures. The result shows that 177 nucleotide sequences coding for the C-terminal 59-amino acids are identical but 505 nucleotide sequences of 3'-nontranslated region of the two LSH A-cDNA exhibit characteristic differences in thier nucleotide sequences. Computer analysis for the folding structures for 3'-end 400 nucleotide sequences of the two LDH A-cDNA shows a possibility implying that the two LDH A-mRNAs isolated from different tissues of rats may have different half life and therefore their translational efficiency may be different. It has been previously demonstrated that isoproterenol stimulated rat $C_6$ -glioma cell cultures produce LDH A-mRNA showing 2 to 3-fold longer half life in comparison to that of noninduced LHD A-mRNA. The result therefore support for the idea that hormonally stimulated rat $C_6$-glioma cells may produce LDH A-mRNA containing different nucleotide sequences at the 3'-end nontranslated region by which the hormonally induced LDH A-mRNA could have more stable secondary mRAN structure in comparison to that of noninduced LDH A-mRNA.

Design of the Wideband Microwave Absorber for X-band Applications (X-대역 응용을 위한 광대역 전파 흡수체 설계)

  • In this paper, a wideband microwave absorber for X-band(8~12 GHz) applications is proposed. The structure of the proposed absorber unit cell consists of a resonator with a slot and slit, a backing ground plate, and a Taconic RF-30(${\varepsilon}_r=3$, $tan{\delta}=0.0014$) substrate with a dimension of $8.5{\times}8.5{\times}0.5mm^3$. The proposed absorber has a dual resonance at 9.83 and 10.37 GHz. To demonstrate the operating principle of the proposed absorber structure at each resonance frequency, the simulated current distributions on the unit cell are analyzed. To verify the performance of the proposed absorber, a prototype absorber was fabricated with a planar array of $20{\times}20$ unit cells. The measured results exhibit two absorptivity peaks stronger than 99 % and full-width at half-maximum(FWHM) bandwidth of 1.1 GHz(9.51~10.61 GHz).

Cell-Based IL-15:IL-15Rα Secreting Vaccine as an Effective Therapy for CT26 Colon Cancer in Mice

  • Interleukin (IL)-15 is an essential immune-modulator with high potential for use in cancer treatment. Natural IL-15 has a low biological potency because of its short half-life and difficulties in mass-production. IL-15Rα, a member of the IL-15 receptor complex, is famous for its high affinity to IL-15 and its ability to lengthen the half-life of IL-15. We have double-transfected IL-15 and its truncated receptor IL-15Rα into CT26 colon cancer cells to target them for intracellular assembly. The secreted IL-15:IL-15Rα complexes were confirmed in ELISA and Co-IP experiments. IL-15:IL-15Rα secreting clones showed a higher anti-tumor effect than IL-15 secreting clones. Furthermore, we also evaluated the vaccine and therapeutic efficacy of the whole cancer-cell vaccine using mitomycin C (MMC)-treated IL-15:IL-15Rα secreting CT26 clones. Three sets of experiments were evaluated; (1) therapeutics, (2) vaccination, and (3) long-term protection. Wild-type CT26-bearing mice treated with a single dose of MMC-inactivated secreted IL-15:IL-15Rα clones prolonged survival compared to the control group. Survival of MMC-inactivated IL-15:IL-15Rα clone-vaccinated mice (without any further adjuvant) exceeded up to 100%. This protection effect even lasted for at least three months after the immunization. Secreted IL-15:IL-15Rα clones challenging trigger anti-tumor response via CD4+ T, CD8+ T, and natural killer (NK) cell-dependent cytotoxicity. Our result suggested that cell-based vaccine secreting IL-15:IL-15Rα, may offer the new tools for immunotherapy to treat cancer.

A Study on Oxygen Reduction Reaction of PtM Electrocatalysts Synthesized on Graphene for Proton Exchange Membrane Fuel Cell (고분자전해질연료전지를 위한 그래핀 기반 PtM 촉매들의 산소환원반응성 연구)

  • In this research, we investigate electrical performance and electrochemical properties of graphene supported Pt (Pt/G) and PtM (M = Ni and Y) alloy catalysts (PtM/Gs) that are synthesized by modified polyol method. With the PtM/Gs that are adopted for oxygen reduction reaction (ORR) as cathode of proton exchange membrane fuel cells (PEMFCs), their catalytic activity and ORR performance and electrical performance are estimated and compared with one another. Their particle size, particle distribution and electrochemically active surface (EAS) area are measured by TEM and cyclic voltammetry (CV), respectively. On the other hand, regarding ORR activity and electrical performance of the catalysts, (i) linear sweeping voltammetry by rotating disk electrode and rotating ring-disk electrode and (ii) PEMFC single cell tests are used. The TEM and CV measurements demonstrate particle size and EAS of PtM/Gs are compatible with those of Pt/G. In case of PtNi/G, its half-wave potential, kinetic current density, transferred electron number per oxygen molecule and $H_2O_2$ production % are excellent. Based on data obtained by half-cell test, when PEMFC singlecell tests are carried out, current density measured at 0.6V and maximum power density of the PEMFC single cell employing PtNi/G are better than those employing Pt/G. Conclusively, PtNi/Gs synthesized by modified polyol shows better ORR catalytic activity and PEMFC performance than other catalysts.

Flow Cytometric Analysis of Endothelial Cell Viability in Arterial Allograft (동종동맥판 혈관내피세포의 생육성 평가에 관한 연구)

  • Arterial allografts have known advantages over prosthetic vascular conduit for treatment of heart valvular disease, congenital heart disease and aortic disease. Cell viability may play a role in determining the longterm outcome of allografts. Endothelial cell is one important part in determining the allograft viability. To evaluate the viability of endothelial cells using current allograft preservation technique, porcine heart valve leaflets and arterial wall were subjected to collagenase digestion. Single endothelial cell suspension was labeled with GSA-PITC(Griffonia simplicifolia agglutininfluorescein isothiocyan te), a vascular, endothelial cell specific marker. The cell suspension was washed and incubated with Pl(Propidium iodide), which does not bind with viable cells, Endothelial cell viability was evaluated by calculating the percentage of GSA-FITC(+) and Pl(-) group using flowcytometric analysis. Allografts were treated with $4^{\circ}C$ antibiotic solo!ion for 24 hours for sterilization. After this, half of allografts were stored in $4^{\circ}C$ RPMI 1640 with HEPES buffer culture medium with 10% fetal bovine serum for 1 to 14 days(Group I). Another half of allografts were cryopreserved with a currently used technique (Group II). During the procurement and sterilization of arterial allografts, 22.8% and 24.4% of endothelial cell viability declined, respectively. In Group I, 11.9% of endothelial cell viability declined further steadily during 14 days of storage. In Group II, 13.7% of endothelial cell viability declined. These results show that largest loss of endothelial cell viability occurs during the nitial process. After 14 days of arterial allograft storage under $4^{\circ}C$ nutrient medium or cryopreservation, about 40% of endothelial cell viability is maintained. There were no differences between the endothelial cell viability from aortic valve leaflet, pulmonic valve leaflets, aortic wall and pulmonic wall.

  • Statement of problem. Confusion about the relationship of surface characteristics of implant to osteoblast cell attachment. Purpose. This study attempted to bone cell attachment to the implant surface which was modified by heat. Material and methods. Commercially pure titanium grade 2, $4{\times}4mm$ sheet 40 pieces were treated for 10 minutes with ultrasonic cleaner with methylethyl ketone, ethanol, deionized distilled water, and half of the specimen 20 pieces were heat treated in $980^{\circ}C$ for 15 minutes. All 40 specimens were autoclaves. Total 6 dishes were prepared, 3 dishes were for control group, and the other 3 dishes were for heat treatment. In fourth day, cell account was done. Conculsion. The change of surface characterization by heat treatment could affect the cell attachment in the early stage however, the change of surface characterization would not be prolonged.

  • A simplified resistor network model for electrical and mass transport in anode-supported planar solid oxide fuel cell (SOFC) was constructed in order to investigate the effect of interconnect rib geometry on the cell performance. For accurate potential calculation, activation and concentration over-potentials at the electrode/electrolyte interfaces were fully considered in this calculation. When contact resistance was not considered, the optimum interconnect rib length were calculated to be $0.1{\sim}0.2$ mm for 2 mm half unit cell for given operation conditions and properties. However, with realistic contact resistance, the interconnect rib length should be increased to provide larger contact area and thus to obtain better performance.

  • The higher mode cutoff frequencies in Crawford TEM cells computed by the Galerkin method(GM) describe in this paper. The authors also report the half mode boundaries to solve not only the cut-off frequencies of symmetric TEM cells and those of asymmetric TEM cells. It is shown that the measured resonant frequencies of the present symmetric TEM cells and a designed asymmetric TEM cell are agreed with the calculated results.

  • In this paper, the performance of Pt/$WO_3$ and Pt-$WO_3$ electrodes was studied for the direct methanol fuel cell. The characteristics of Pt/$WO_3$ electrode which was prepared by using electrodeposition method was tested with half-cell experiment. The characteristics of Pt-$WO_3$/C electrode which was Prepared by using freeze-drying method was tested with a single cell experiment. The performance of DMFC single cell which was prepared by Pt-$WO_3/C$ and Pt/C showed a current density of $32mA/cm^2$ at $110^{\circ}C$ & 0.3V(0.5mg Pt/$cm^2$).

Ultrastructural Study on the Cellular Compatibility in Self-Parasiting Cuscuta australis (자기기생하는 실새삼(Cuscuta australis)에서 세포 화합성에 관한 미세구조 연구)

  • Cellular compatibility in the self-parasitism of Cuscuta australis R. Brown was studied at the ultrastructural level. The front cells of the haustorium penetrated the host stems independently grew within the host tissues and transformed into elongate, filamentous hyphae. Each hyphal cells contained a large nucleus and dense cytoplasm with abundant cell organelles. Multilamellar structures were contained in the cytoplasm and cell walls of the penetrating hyphal cells. When the hyphal cells did not yet invade the host cells, the middle lamella and the fused cellulosic cell walls of the two partners at the host-parasite interface were preserved well. As the invasion of the parasitic hyphal cells progressed, however, the middle lamella was not found at the interface and the host cell walls and plasma membranes were partially broken down. A hyphal cell penetrated deeply into the host cell had a more darkly stained cytoplasm with numerous of cell organelles. In the host cells attacked by the hyphal cells the limiting membranes of plastids were broken down and several vesicles were arrayed near the cell walls. No plasmodesmatal connections between the host and parasite cell walls were found; however, half-plasmodesmata were observed frequently on the side of the hyphal cell walls. These results suggested that the compatibility response in the self-parasitism of Cuscuta was expressed by cell walls, not by plasmodesmata, between the host and the parasite cells.
