當歸補血湯加味方이 모발탈락 및 성장에 미치는 영향 (Effect of DanGuiBoHyulTangGami-Bang on the Alopecia and Hair Growth Stimulation)

  • We made an experiment if the extracts of DanGuiBoHyulTangGami-Bang(DBTG) and 15 kinds of the medical herbs used the materials of DBTG were effective on the hair formation palpation and the falling out of hair, and came to the following conclusions. 1. The extracts of Paeonia lactiflora, Cuscuta chinensis and Angelica tenuissima of DBTG consisted of the 15 kinds of the medical herbs kept the activity of 5${\alpha}$-reductase type Ⅱ from being active 75.3$\%$, 63.8$\%$. 75.5$\%$. 2. The hair growth index, 1.6(control group 0.8) of the extracts of DBTG bas a little effect on the hair growth palpation and that of Rubus coreanus 1.8(control group 0.4) was the most effective one of the medical herbs, and Paeonia lactiflora 2.3(control group 1.7) and Vitex rotundifolia 2.3(control group 1.5) showed the effect on hair formation palpation. 3. The hair growth period couldn't be extended by DBTG in this experimental stage. 4. The 15 kinds of constitution medicines of DBTG didn't have effects in dermal papilla cells DNA increase, IGF- I, KGF, HGF the revelation of a gene heredity, the protein synthesis of the hair follicle tissues. 5. All of the 15 kinds of constitution medicines of DBTG didn't have the antibacterial activity in Paper disc rule. 6. The results from the test of a radical scavenging activity of the 15 kinds of constitution medicines of DBTG showed that the extracts of Paeoria lactiflora, Scutellaria baicalensis, Rubus coreanus have the superior antioxidant activity in the concentration of 0.01$\%$ and 0.001$\%$ 7. In the formation controlled experiment, Vitex rotundifolia (70.6$\%$), Scutellaria baicalensis (47.1$\%$, Saposhnikovia (44.8$\%$) of the 15 kinds of constitution medicines of DBTG in the 50㎍/㎖ concentration controled NO forming and Vitex rotundifolia (12.7$\%$) controled NO forming in the 5㎍/㎖ concentration in order. 8. MTT(lC/50) of the extracts of Rehmannia glutinosa, Paeonia lactiflora, Scutellaria baicalensis, Lycium chinense, Rubus coreanus of the 15 kinds of constitution medicines of DBIG was more than 500㎍/㎖ and had the least cell virulence.

  • Backgrounds : Androgenetic alopecia is a relatively common disorder, but its precise mechanism is not elucidated. There are two commercial drugs approved by FDA. One(finasteride) has an inhibition activity of 5$\alpha$-reductase(type 2) and the other(minoxidil) has a vasodilation activity. Objectives : A verified herbal remedy for baldness is needed for medicinal treatment or preventing alopecia, which could be demonstrated by alopecia-related in vitro & in vivo tests Methods : On the basis of oriental pharmacognosy, we classified many herbal medicines into four groups (malnutrition, aging, alopecia and gray hair) according to its effect. The mitosis induction of hairy dermal papillae cell and the metabolic inhibition for type 2 $5{\alpha}-reductase$ were tested with five herbal extracts. Also, five herbal extracts were added to the normal essence formulation (HHRHG0202-80) in ranges of 0.1~0.3%, which was applied two-mouse models to validate each hair growing activity in vivo. Results : Stimulation of follicular papillae cell proliferation was observed in treatment of three herbal extracts (Glycyrrhizae radix:159.7%, Corni fructus : 144.7%, and Coicis semen : 136.6%) at a dose of $10\mu\textrm{g}/ml$. Three herbal extracts (Biotae semen. Glycyrrhizae radix and Coicis semen) showed inhibitory activity for $5{\alpha}-reductase$(type 2) at 93.18%, 73.36% and 47.6%, respectively at the same dose. We observed the enhancement of hair growth activity in C57b1/6 mouse and the inhibition of alopecia in AGA mouse after topical administration of the hair essence. Conclusions : Hair essence product, which contains five medicinal plants, would be used for the remedy for male pattern baldness (MPB) and the other alopecia diseases.

The Success of Thread-embedding Therapy in Generating Hair Re-growth in Mice Points to Its Possibly Having a Similar Effect in Humans

  • Objectives: Recently, thread-embedding therapy (TET) has been widely applied in Korean medicine for cosmetic purposes such as reducing skin wrinkles. An inserted thread was reported to have induced continuous stimulation, followed by support for connective tissue regeneration. However, the potential role of TET in hair-growth has not yet been reported. Methods: We designed this study to evaluate whether TET has a hair-growth-promoting effect. C57 black 6 (C57BL/6) mice were divided into three groups: normal saline-treated, minoxidil-treated, and thread-embedded groups. Normal saline or 5% minoxidil was topically sprayed on the dorsal skin of the mice once a day for 16 days. Medical threads were embedded into the dorsal skin of the mice in a single application. Hair growth activity was evaluated by using dermoscopic and microscopic observations. Sections of the dorsal skin were stained with hematoxylin and eosin. Expressions of bromodeoxyuridine (BrdU), proliferating cell nuclear antigen (PCNA), fibroblast growth factor-7 (FGF-7), and fibroblast growth factor-5 (FGF-5) were detected by using immunohistochemical staining. A reverse transcription-polymerase chain reaction (RT-PCR) analysis was adopted to measure the messenger RNA (mRNA) expressions of FGF-7 and FGF-5. Results: TET enhanced anagen development in the hair follicles of C57BL/6 mice. The expressions of BrdU and PCNA, both of which imply active cellular proliferation, were increased by using TET. Moreover, TET increased the expression of FGF-7, an anagen-inducing growth factor, while decreasing the expression of FGF-5, an anagen-cessation growth factor, both at the protein and the mRNA levels. Conclusion: TET enhanced hair re-growth in C57BL/6 mice. TET regulated the expressions of anagen-associated growth factors and activated the proliferation of hair follicular cells in depilated skin lesions. Considering its long-lasting effect, TET may be a good alternative therapeutic for the treatment of alopecia.

In vitro 및 in vivo에서 검은콩 추출물의 육모 효과 (Hair Growth Promoting Effect of Black Soybean Extract In Vitro and In Vivo)

  • 본 연구에서는 검은콩의 육모 촉진 효과를 평가하기 위하여 검은콩 추출물을 이용하여 모유두 세포의 성장 촉진과 모세혈관 확장에 미치는 효과를 평가하였으며, C57BL/6 마우스 모델을 이용하여 검은콩 추출물의 경구 섭취가 모발 성장에 미치는 효과를 2% minoxidil과 비교하여 평가하였다. 그 결과 검은콩 추출물이 모유두 세포의 증식을 촉진하고 모세혈관을 확장 시키는 효과가 있으며, 우수한 성장기 모낭 유도 촉진 효과를 통해 모발의 성장을 촉진하는 것을 확인하였다. 또한, 혈액의 항산화능을 증가시키고 혈중 중성 지질을 감소하는 효과를 가져 혈액을 맑게 하고 혈액 순환에 도움을 주어 육모 및 모발 건강 증진에 기여할 수 있음을 확인하였다. 검은콩 추출물의 우수한 육모 효과를 확신하기 위해서는 사람을 대상으로 6개월 이상의 인체 효능 평가와 검은콩 추출물 내의 유효 성분을 규명하는 연구가 후속으로 진행되어야 할 것이다. 그러나 본 연구는 민간에서 널리 알려진 검은콩의 육모 촉진 효과를 체계적으로 평가하였다는 것에 큰 의의가 있으며, 이를 통하여 검은콩 추출물이 혈액 순환 개선 및 모발 성장 주기 조절을 통해 모발 건강 개선 및 육모 촉진제로써 이용될 수 있다고 기대할 수 있다.

ER71/ETV2 Promotes Hair Regeneration from Chemotherapeutic Drug-Induced Hair Loss by Enhancing Angiogenesis

  • Chemotherapy-induced alopecia and hair loss can be stressful in patients with cancer. The hair grows back, but sometimes the hair tends to stay thin. Therefore, understanding mechanisms regulating hair regeneration may improve the management of chemotherapy-induced alopecia. Previous studies have revealed that chemotherapeutic agents induce a hair follicle vascular injury. As hair growth is associated with micro-vessel regeneration, we postulated that the stimulation of angiogenesis might enhance hair regeneration. In particular, mice treated with 5-fluorouracil (5-FU) showed delayed anagen initiation and reduced capillary density when compared with untreated controls, suggesting that the retardation of anagen initiation by 5-FU treatment may be attributed to the loss of perifollicular micro-vessels. We investigated whether the ETS transcription factor ETV2 (aka ER71), critical for vascular development and regeneration, can promote angiogenesis and hair regrowth in a 5-FU-induced alopecia mouse model. Tie2-Cre; Etv2 conditional knockout (CKO) mice, which lack Etv2 in endothelial cells, presented similar hair regrowth rates as the control mice after depilation. Following 5-FU treatment, Tie2-Cre; Etv2 CKO mice revealed a significant reduction in capillary density, anagen induction, and hair restoration when compared with controls. Mice receiving lentiviral Etv2 injection after 5-FU treatment showed significantly improved anagen induction and hair regrowth. Two-photon laser scanning microscopy revealed that enforced Etv2 expression restored normal vessel morphology after 5-FU mediated vessel injury. Our data suggest that vessel regeneration strategies may improve hair regrowth after chemotherapeutic treatment.

발모효과를 가지는 한방복합처방단 (Medicinal Herbal Complex Extract with Potential for Hair Growth-Promoting Activity)

  • 탈모방지 및 발모효과를 가지는 소재를 개발하기 위해 한의학에서 전통적으로 사용되는 23가지 한방소재를 선정하여 한방복합처방단을 개발하였다. 한방복합처방단을 구성하는 한방소재들은 예로부터 전통적으로 발모 및 탈모방지, 흰머리방지, 염증 치료 및 혈액순환개선 효과를 가진 것으로 알려져 있는 당귀, 보골지, 측백엽, 한련초, 구기자, 복분자, 상백피, 숙지황, 여정실, 적하수오, 흑지마, 고삼, 백지, 익모초, 단삼, 도인, 몰약, 감국, 유향, 인삼, 천궁, 합환피, 현호색 등이다. 또한, 한방복합처방단의 발모효과를 확인하기 위해 in vitro와 in vivo 평가모델을 이용하여 모발성장 및 촉진에 미치는 영향을 실험하였다. In vitro 상에서는 모유두세포, 각질형성세포 및 섬유아세포의 증식을 확인하였다. 또한, 흰머리방지 효과와 관련하여 멜라노마 세포에서의 멜라닌 합성능력을 확인하였다. 한방복합처방단의 in vitro 상에서의 육모효과는 C57BL/6 마우스를 이용한 in vivo 상에서도 확인하였다. 연구 결과 한방복합처방단은 $50{\mu}g/mL$의 농도에서 모유두 세포의 증식을 175 %까지, 섬유아세포인 NIH3T3 세포의 증식은 120 %까지 증가시켰으며, $20{\mu}g/mL$의 농도에서 각질형성세포인 HaCaT 세포의 증식을 133 %까지 증가시켰다. 멜라닌 합성의 경우, $50{\mu}g/mL$의 농도에서 154 %까지 증가시켰다. 또한, C57BL/6 마우스를 이용한 육모효과에 있어서는 한방복합 처방단 처리 4주 후 98 % 이상의 육모효과를 나타내는 것을 확인하였다. 이상의 결과로 볼 때 본 연구에서 개발한 한방복합처방단은 모발의 성장 촉진에 유용하게 활용될 수 있는 처방으로 사료된다.

적하수오 에탄올 추출물의 melanin 합성 촉진효과 (Promotive Effect of Polygonum multiflorum radix Ethanol Extract on Melanogenesis)

  • 모발의 색은 melanin의 종류와 양에 의해 결정 된다. Melanocyte는 주로 자외선 자극에 의하여 L-tyrosine으로부터 멜라닌을 합성한다. 활성산소종은 백발의 진행과정에 중요한 역할을 하고 있다. 적하수오는 백발화되는 노화과정을 억제한다고 보고되고 있다. 본 연구의 목적은 흑모발생과 연관된 melanin 합성에 대한 적하수오 에탄올 추출물(PMEE)의 효능을 조사한 것이다. 항산화 실험에서 PMEE는 DPPH radical을 감소시켰고 환원력을 증가시켜 PMEE는 백발과정에 관여하는 활성산소종을 소거할 수 있다고 볼 수 있다. PMEE는 농도 의존적으로 세포생존능을 감소시켰다. 더욱이 melanin의 생성에 대한 PMEE의 효과는 DOPA assay와 tyrosinase 활성측정에 의하여 결정되었다. PMEE는 tyrosinase 활성을 증가시켰으며 melanin 합성을 촉진시켰다. 뿐만 아니라 Western blot 분석을 이용하여 tyrosinase, TRP-1, TRP-2 및 MITF 뿐만 아니라 SOD-3와 catalase와 같은 항산화 효소의 단백질 발현을 조사하였다. PMEE의 항산화 활성의 증가로 인하여 SOD-3와 catalase의 발현 수준은 감소된 것으로 나타났다. 특히, PMEE는 tyrosinase와 TRP-2의 발현 수준은 증가시켰다. 이러한 결과들은 PMEE가 백발화 된 모발에서 흑발로 변하는 것과 관련된 melanin 합성을 촉진할 수 있다는 것을 암시하고 있다.

Myristoleic Acid Promotes Anagen Signaling by Autophagy through Activating Wnt/β-Catenin and ERK Pathways in Dermal Papilla Cells

  • Alopecia is a distressing condition caused by the dysregulation of anagen, catagen, and telogen in the hair cycle. Dermal papilla cells (DPCs) regulate the hair cycle and play important roles in hair growth and regeneration. Myristoleic acid (MA) increases Wnt reporter activity in DPCs. However, the action mechanisms of MA on the stimulation of anagen signaling in DPCs is not known. In this study, we evaluated the effects of MA on anagen-activating signaling pathways in DPCs. MA significantly increased DPC proliferation and stimulated the G2/M phase, accompanied by increasing cyclin A, Cdc2, and cyclin B1. To elucidate the mechanism by which MA promotes DPC proliferation, we evaluated the effect of MA on autophagy and intracellular pathways. MA induced autophagosome formation by decreasing the levels of the phospho-mammalian target of rapamycin (phospho-mTOR) and increasing autophagy-related 7 (Atg7) and microtubule-associated protein 1A/1B-light chain 3II (LC3II). MA also increased the phosphorylation levels of Wnt/β-catenin proteins, such as GSK3β (Ser9) and β-catenin (Ser552 and Ser675). Treatment with XAV939, an inhibitor of the Wnt/β-catenin pathway, attenuated the MA-induced increase in β-catenin nuclear translocation. Moreover, XAV939 reduced MA-induced effects on cell cycle progression, autophagy, and DPC proliferation. On the other hand, MA increased the levels of phospho (Thr202/Tyr204)-extracellular signal regulated kinases (ERK). MA-induced ERK phosphorylation led to changes in the expression levels of Cdc2, Atg7 and LC3II, as well as DPC proliferation. Our results suggest that MA promotes anagen signaling via autophagy and cell cycle progression by activating the Wnt/β-catenin and ERK pathways in DPCs.

  • 진단이 지연되었거나 적절하게 치료되지 않은 선천성 부신 과형성증을 가진 남아에서 장기간 고농도의 남성호르몬에 노출될 경우 처음에는 가성 성조숙증이었다가 이후 진성 성조숙증으로 전환될 수 있으므로 선천성 부신 과형성증의 조기진단과 치료가 필요하다. 저자들은 가성 성조숙증에서 진성 성조숙증으로 전환된 선천성 부신 과형성증을 가진 남아에서 hydrocortisone과 GnRH 유도체를 사용하여 큰 부작용 없이 2차 성징의 퇴축을 가져온 1례를 경험하고 문헌고찰과 함께 보고하는 바이다.
