Study on the Relationship between Biliary Secretion and Cyclic Nucleotides (담즙분비와 Cyclic nucleotides간의 상호관계에 관한 연구)

  • Bile formation is a complex process comprised of three separate physiologic mechanism operating at two anatomical sites. At present time, it was known that at least two processes are responsible for total canalicular secretion at the bile canaliculus. One of the processes is bile salt-dependent secretion (BSDS) hypothesis that the active transport of bile salts from plasma to bile provided a primary stimulus for bile formation: the osmotic effect of actively transported bile acid was responsible for the movement of water and ions into bile. The other process is bile salt-independent secretion (ESIS), which is unrelated to bile salt secretion at the canaliculus and which may involve the active transport of sodium. The third process for bile formation involves the biliary ductal epithelium. Secretin-stimulated bile characteristically contained bicarbonate in high concentration. Therefor, it was suggested that secretin stimulated water and bicarbonate secretion from the biliary ductules. One the other hand, it was found that a large amounts of cAMP was present in canine bile but no apparent relationship between bile salt secretion and cAMP content in dog bile. However, bile flow studies in human have demonstrated that secretin and glucagon increase bile cAMP secretion as does secretin in baboons. Secretin increases baboon bile duct mucosal cAMP levels in addition to bile CAMP levels suggesting that in that species secretin-stimulated bile flow may be cAMP mediated. It has been postulated that glucagon and theophylline which increase the bile salt-independent secretion in dogs might act through an increased in liver cAMP content. In a few studies, the possible role of cAMP on bile formation has teen tested by administration of an exogenous derivative of cAMP, dibutyryl cAMP. In the rat, DB cAMP did not modify bile flow, but injection of DB cAMP in the dog promoted an increase in the bile salt-independent secretion. Because of these contradictory results, this study was carried out to examine the relationship between cyclic nucleotides and bile flow due to various bile salts as well as secretin or theophylline. Experiments were performed in rabbits with anesthesia produced by the injection of seconal(30 mg/kg). Rabbits had the cystic duct ligated and the proximal end of the divided common duct cannulated with an appropriately sized polyethylene catheter. A similar catheter was placed into the inferior vena cava for administration of drugs. Bile was collected for determination of cyclic nucleotides and total cholate in 15 min. intervals for a few hours. The results are summerized as followings. 1) Administrations of taurocholic acid or chenodeoxycholic acid increased significantly the concentrations of cAMP and cGMP in bile of rabbits. 2) Concentration of cAMP in bile during the continuous infusion of ursodeoxycholic acid, was remarkedly increased in accordance with the increase of bile flow, while on the contrary concentration of cGMP in bile was decreased significantly. 3) Dehydrocholic acid and deoxycholic acid significantly increased bile flow, total cholate output and cyclic nucleotides in bile. 4) Only cAMP concentration in bile was significantly increased from control value by secretin, while theophylline increased cAMP as well as cGMP in rabbit bile. 5) In addition, the administration of secretin to taurocholic acid-stimulated bile flow increased cAMP while theophylline produced the increases of cAMP and cGMP in bile. 6) The administration of insulin to taurocholic acid-stimulated bile flow decreased cAMP concentration, while on the contrary cGMP was remarkedly increased in rabbit bile.

Physicochemical Properties of Commercial Sweet Potato Starches (시판 고구마전분의 이화학적 특성)

  • Physicochemical properties of commercial sweet potato starches manufactured by 7 different companies were investigated in comparison with corn and potato starches. Crude ash and protein content varied from 0.36 to 1.02%, and from 0.04 to 0.14% based on dry weight, respectively. The protein contents were relatively smaller than that of corn or potato starch. But whiteness of the sweet potato starches was less than that of corn or potato starch. Mean diameter of the sweet potato starch granules varied from 14.23 to $21.08\;{\mu}m$ depending on the company and all sweet potato starches showed bimodal size distributions. Pasting viscosity measured by Rapid Viscoanalyzer(RVA) also showed variations among the starches of different companies. The starch from D company in Korea had the lowest pasting temperature$(74.00^{\circ}C)$ whereas the starch from a phillippine company(P) did the highest one$(80.35^{\circ}C)$. The peak viscosity of sweet potato starches was higher than that of corn starch but lower than that of potato starch. The D company starch also showed the highest peak viscosity(2283 cp) among the starches tested. Paste breakdown by hot shearing ranged from 524 cp (S company) to 1279 cp (HL company). Textural properties of the starch gels appeared significantly different among the starches of different manufacturers. The greatest hardness of the gel was $137.90\;g_{f}$ at 1 day storage whereas the lowest value was $31.53\;g_{f}$. Except the starches from 2 companies (P and S), the sweet potato starches formed very soft and weak gels. P or S company starches formed the gels similar to potato starch. Syneresis by freeze-thawing treatments appeared less for sweet potato starch gels than that for corn starch gels, but greater than that for potato starch gel. The overall properties of the sweet potato starches varied by the manufacturing companies, and ranged between those of corn and potato starches.

Research on the Effect of the Control Methods of Irrigation Water on the Growth and Yield of Paddy Rice. (한발기에 있어서 용수관리 방법이 수도생육과 그 수량에 미치는 영향에 관한 연구)

  • This experiment was made to determienthe effect of various soil moisture contents in simulated drought conditions on different stages of rice growth. The drought conditions were developed at such three rice-growing stages as transplanting, immediately after transplanting and young ear forming. Three different lengths of drought periods, which are ten days, twenty days and thirty days, were applied for each growing stage of rice. The rice variety used this experiment is Nong-rim 29. This experiment was conducted at the university farm of the Kon-Kuk University during the period of $1968{\sim}1970$. Three reprications for each of 12 treatments and split plot design were employed in this study. Bottomless wood square boxes, $1^m{\times}1^m{\times}1^m$, were burried in the test plot and box top was covered with poloyethylene sheets to avoid natural rainfall drops. Standard plots were irrigated continuously with a water depth of 40mm/day and those of drought treatments were irrigated continuously up to the beginning of the planned drought period, and they were irrigated again with a depth of 40mm/day up to the maturing stage of rice. Other methods for rice raising followed those methods developed by the Field Crops Experiment Station of the Office of Rural Development. During this experiments, climatic conditions in regard to rainfalls, sunshine hours, and temperatures were observed. According to this observation, those values measured deviate slightly from the annual means. However the growing condition of rice plants were normal. The pH value of irritation water is nearly neutral, and soils in the test plots are relatively fertile, being similar to ordinary paddy soils. Analysis of variances for number of stalks, plan-height, ear sprouting date, length of stalks, ear length, number of ears per plant, fertility, grain weitght, weight of plant, and yield were carried out. The variances for plant height, ear sprouting date, length of stalk ear length, and yield has statistical significance under drought treatments applied at three different growing stages. The variance showing the effect of lengths of drought period is highly significant for all the treatments studied except that of grain weight. The interaction between drought periods and drought treatments at different growing stages is significant for plant height, stalk length, ear length, number of ears, fertility and yield, these results indicated that droughts at different growing stages have influence on plant height, ear length, yield, and length of drought period also has strong influence on all factors studied except grain weight. The combination of drought treatments at different rice growing stages and lengths of drought periods has different effects on various agronomic characteristics, including yield. Plant height under drought treatment practiced at transplanting stage is the lowest, and drought treatment applied immediately after transplanting resulted in the least number of stalks. The effect of different lengths of drought periods on plant height and number of stalks depends signis ficantly on increasing days of drought. Ear sprouting date tends to be delayed for one or two days undedrought treatments at transplanting period and with increasing days of drought. Better yield is secured in drought treatment applied immediately after transplanting. Adverse effect war observed when drought treatment was applied at ear forming period. These effects may be attributed to the alternation of irrigation and drought causing vigorous root activity. In general, yield linearly decreases as the length of the drought period increases. The results obtained in this study demonstrate that, in order to mimimize damage due to drought, and, to save irrigation water, paddy fields, immediately after transplanting, may be not irrigated, since there is sufficient moisture in the soil, and that sufficient irrigation water should be applied again in the ear forming stage of rice plant.

Effect of Processing Additives on Vulcanization and Properties of EPDM Rubber (EPDM 고무의 첨가제에 따른 가류 및 물성에 미치는 영향 연구)

  • Effects of three different types of dispersions and flow improving additives composed with fatty acid esters, fatty acid metal salts and amide compound on the vulcanization and the mechanical properties properties of rubber compounds of EPDM and carbon black as fillers. were investigated using Mooney viscometer, moving die rheometer, hardness tester, and universal test machine. The aging characteristics of vulcanized EPDM compounds were also investigated. The Mooney viscosity measured at $125^{\circ}C$ showed a tendency to decrease in the order of amide type> metal salt type > ester type additive. Scorch time showed little or no difference with the addition of ester or metal salt type additives, but the amide type additive shortened a scorch time more than one minute. Rheological measurement data obtained at $160^{\circ}C$ showed that the vulcanization time was faster for metal salt type and amide type additive systems. Delta torque values of EPDM compound increased with metal salt type and amide type additives, but slightly decreased with ester type additive. The tensile strength of the EPDM compound was greatly improved when an ester type additive was added, but the amide type or metal salt type additive had no significant effect. The elongation was significantly improved for metal salt type additive, while the rest were not significantly affected. The tear strength of the EPDM compounds increased with the addition of all kinds of additives, and it increased remarkably in the case of metal salt type additive. Hardness of the EPDM compounds was nearly same value regardless of additive types. The thermal aging of the EPDM blend at $100^{\circ}C$ for 24 h showed little change in the case of metal salt type or amide type additive, but the elongation tends to decrease by 10-20% for all EPDM compounds containing additives.

Effects of Replacing Dried Skim Milk With Wheat Gluten and Spray Dried Porcine Protein on Growth Performance and Digestibility of Nutrients in Nursery Pigs

  • Three experiments were conducted to determine the nutritional value of wheat gluten (WG) and spray-dried porcine plasma (SDPP) in diets for nursery pigs. In Exp. 1, 120 weanling pigs (5.7 kg avg initial BW) were used in a 35-d growth assay. Treatments for d 0 to 14 were: 1) dried skim milk (DSM)-dried whey-SBM based control; 2) WG to replace the protein from DSM; 3) SDPP; and 4) WG-SDPP (50:50 blend on a protein basis) to replace the protein from DSM. From d 14 to 35, all pigs were fed a common corn-SBM-whey-based diet. For d 0 to 14, there were no differences in ADG, ADFI, and gain/feed (p>0.11). However, for d 14 to 35, pigs fed diets with WG had greater gain/feed than those fed SDPP (p<0.05), and pigs fed diets with the WG-SDPP blend had greater ADG than pigs fed diets with WG or SDPP alone (p<0.07). In a second experiment, 60 weanling pigs (5.1 kg avg initial BW) were used in a 28-d growth assay. All pigs were fed the WG-SDPP diet fed in Exp. 1 for d 0 to 14, and changed to experimental diets for d 14 to 28. Treatments were: 1) the whey-SBM-based diet used for d 14 to 28 in Exp. 1; or 2) a whey-SBM based diet with 3% added SDPP. There were no differences in ADG, ADFI, gain/feed, or apparent digestibilities of DM and N among treatments for d 14 to 28 or overall (p>0.14). In a third experiment, 150 weanling pigs (5.6 kg avg initial BW) were used in a 32-d growth assay to determine the optimal blend of WG and SDPP for use after weaning. The SDPP was added as 8% of the control diet, and WG was substituted on a protein basis to yield the desired SDPP:WG blends. Treatments were (d 0 to 14): 1) SDPP; 2) 75% SDPP and 25% WG; 3) 50% SDPP and 50% WG; 4) 25% SDPP and 75% WG; and 5) WG. As in Exp. 1, all pigs were switched to a common corn-SBM-whey-based diet for d 14 to 32. For d 0 to 14, ADG and ADFI increased as replacement of the SDPP was increased up to 50% and decreased when more of the SDPP was removed from the diet (quadratic effects, p<0.004 and 0.02, respectively). Apparent digestibilities of DM and N (at d 13) were not affected by treatments (p>0.18). For d 14 to 32, treatments did not affect ADG (p>0.2), although there were quadratic responses in ADFI (p<0.04), with pigs fed the 50:50 blend suggested the greatest intake of feed. For the overall experimental period (d 0 to 32), ADG, ADFI, and gain/feed increased as WG was used to replace as much as 50% of the SDPP (quadratic effects p<0.04, 0.02, and 0.06, respectively). In conclusion, WG can successfully replace up to 50% of the SDPP in a complex nursery diet, when SDPP is included at the 8% level. There is no advantage to keeping SDPP in the diet after Phase I (d 0 to 14).

Predictive indicators of coronary artery complications in Kawasaki disease (가와사키 병 환아에서 관상동맥 합병증의 예측인자)

  • Purpose : Kawasaki disease—the most common cause of acquired heart disease in children—incidence is increasing yearly. Therefore, we evaluated the predictive indicators of coronary complications of Kawasaki disease based on clinical and laboratory data. Methods : Between January 2005 and March 2008, of the 201 children with Kawasaki disease treated at the Gil Hospital of Gachon University of Medicine and Science, 51 had coronary artery lesions (Group II) and 150 had no lesions (Group I). The reasons for coronary artery lesions were deduced from the clinical and laboratory data. Results : Analysis of the 2 groups revealed that fever duration and days of fever after and before initial intravenous gammaglobulin (IVIG) treatment were significantly longer in Group 2 than in Group I. IVIG infusions were statistically higher in Group II than in Group I. As per the laboratory data, C-reactive protein (CRP) value was significantly higher in Group II. Collectively, >10 days of fever duration, >48 h of fever duration after, and >10 days of fever before IVIG treatment increased the risk of coronary artery lesions 6-, 5-, and 3.5-fold, respectively. Furthermore, additional IVIG courses and higher CRP level increased the risk of coronary artery lesions 4-fold and 2-3-fold, respectively. Conclusion : The following 3 factors were responsible for increased risk of coronary artery lesions in children with Kawasaki disease: fever duration and days of fever after and before IVIG treatment. To identifythe predictive indicators of coronary complications, it is necessary to further elucidate the relationship between well-known forecasting factors.

Seasonal and Spatial Distribution of Soft-bottom Polychaetesin Jinju Bay of the Southern Coast of Korea (진주만에서 저서 다모류의 시 · 공간 분포)

  • Seasonal quantitative van Veen grab sampling was conducted to characterize the composition and structure of the benthic polychaete community inhabiting the shellfish farming ground of a coastal bay system of Jiniu Bay (Korea). A total of 132 polychaete species were identified and the polychaetes accounted for about $80\%$ of overall abundance of benthic animals. There was little significant seasonal difference in densities (abundances) of polychaetes, Maximum biomass was obseued in summer (August) and minimum value was recorded in winter (February) and spring (May). Conversely, diversity and richness were lowest in summer, indicating a seasonal variability in the polychaetous community structure, The cluster analysis indicated that such a seasonal variability resulted mainly from the appearance of a few small, r-selected opportunists in spring and the tubiculous species of the family Maldanidae in summer. On the other hand, several indicator species for the organically enriched environments such as Capitelia capitata, Notoniashs Jatericeus and hmbrineris sp. showed high densities during all the study period. Density and biomass of univariate measures of community structure were significantly lower in the arkshell-farming ground of the southern area than in the non-farming sites of the bay, A similar general tendency was also found in the spatial distributions of species diversity and richness. Principal component analysis revealed the existence of different groups of benthic assemblages between the arkshell-farming ground and non-farming sites, The lack of colonization of r-selected opportunists and/or tubiculous species in the former ground seemed to contribute to the spatial differences in the composition and structure of the polychaetous communities. Although finer granulometric composition and high sulfide concentration in sediments of the arkshell-farming ground and low salinity in the northern area were likely to account for parts of the differences, other environmental variables observed were unlikely. The spatial distribution of polychaetes in Jiniu Bay may be rather closely related to the sedimentary disturbance by selection of shells for harvesting in spring.

Optimization of Pre-treatment of Tropical Crop Oil by Sulfuric Acid and Bio-diesel Production (황산을 이용한 열대작물 오일의 전처리 반응 최적화 및 바이오디젤 생산)

  • In this study, the feasibility of using vegetable oil extracted from tropical crop seed as a biodiesel feedstock was investigated by producing biodiesel and analysing the quality parameters as a transport fuel. In order to produce biodiesel efficiently, two step reaction process(pre-treatment and transesterificaion) was required because the tropical crop oil have a high content of free fatty acids. To determine the suitable acid catalyst for the pre-esterification, three kinds of acid catalysts were tested and sulfuric acid was identified as the best catalyst. After constructing the experimental matrix based on RSM and analysing the statistical data, the optimal pre-treatment conditions were determined to be 26.7% of methanol and 0.982% of sulfuric acid. Trans-esterification experiments of the pre-esterified oil based on RSM were carried out, then discovered 1.24% of KOH catalyst and 22.76% of methanol as the optimal trans-esterification conditions. However, the quantity of KOH was higher than the previously established KOH concentration of our team. So, we carried out supplemental experiment to determine the quantity of catalyst and methanol. As a result, the optimal transesterification conditions were determined to be 0.8% of KOH and 16.13% of methanol. After trans-esterification of tropical crop oil, the produced biodiesel could meet the major quality standard specifications; 100.8% of FAME, 0.45 mgKOH/g of acid value, 0.00% of water, 0.04% of total glycerol, $4.041mm^2/s$ of kinematic viscosity(at $40^{\circ}C$).

  • Thirty Bradyrhizobium japonicum isolates (10 strains per each soil) from 1 uncultivated [Sangnam(Soil 1), Milyang]- and 2 cultivated [Dong(Soil 2)and Chinbuk(Soil 3), Changweon] upland soils in Yeongnam area were evaluated on their symbiotic effectiveness to soybean [Glycin max (L.)] cv. Korean Jangbaekkong and American Clark and examined on their serological diversity. The results obtained were summarized as follows : 1. On symbiotic effectiveness of B. japonicum with plant genotypes, isolates showed a relatively high value of nodule mass in Jangbaekkong cv. and of shoot dry weight and total nitrogen in Clark cv. demonstrating the order of Soil 1> Soil 2> Soil 3 isolates. 2. Among 30 B. japonicum isolates, YCK 141 showed the best effectiveness on mean nitrogen fixation of two cultivars. 3. Thirty indigenous B. japonicum showed 6 types of serological diversities in the immunoblot analysis which were present in various proportions at Soil 2(5) and Soil 3(5) except Soil 1 where all isolates fell into the YCK 117 serogroup. And their distribution order was serotype YCK 117( 12 strains) > USDA 1l0(5strains), USDA 123(5 strains) > YCK 150(4 strains) > YCK 141(3 strains) > YCK 226(1 strain). 4. Especially, 10 isolates from Soil 1, an uncultivated orchard, showed a very homologous pattern in not only effectiveness but serological distribution. It seemed to indicate that the isolates were typically affected by numerous physical and environmental factors of the soil.

Growth kinetics and chlorine resistance of heterotrophic bacteria isolated from young biofilms formed on a model drinking water distribution system (모델 상수관망에 형성된 초기 생물막에서 분리한 종속영양세균의 생장 동역학 및 염소 내성)

  • The present work quantified the growth of young biofilm in a model distribution system that was fed with chlorinated drinking water at a hydraulic retention time of 2 h. Bacterial biofilms grew on the surface of polyvinyl chloride (PVC) slides at a specific growth rate of $0.14{\pm}0.09day^{-1}$ for total bacteria and $0.16{\pm}0.08day^{-1}$ for heterotrophic bacteria, reaching $3.1{\times}10^4cells/cm^2$ and $6.6{\times}10^3CFU/cm^2$ after 10 days, respectively. The specific growth rates of biofilm-forming bacteria were found to be much higher than those of bulk-phase bacteria, suggesting that biofilm bacteria account for a major part of the bacterial production in this model system. Biofilm isolates exhibited characteristic kinetic properties, as determined by ${\mu}_{max}$ and $K_S$ values using the Monod model, in a defined growth medium containing various amounts of acetate. The lowest ${\mu}_{max}$ value was observed in bacterial species belonging to the genus Methylobacterium, and their slow growth seemed to confer high resistance to chlorine treatment (0.5 mg/L for 10 min). $K_S$ values (inversely related to substrate affinity) of Sphingomonas were two orders of magnitude lower for acetate carbon than those of other isolates. The Sphingomonas isolates may have obligate-oligotrophic characteristics, since the lower $K_S$ values allow them to thrive under nutrient-deficient conditions. These results provide a better understanding and control of multi-species bacterial biofilms that develop within days in a drinking water distribution system.
