This study was conducted to investigate the effect of cooling systems on reproductive performance, body temperature, blood metabolites, and the intestinal microbiome in early gestating sows exposed to high ambient temperature. In total, 39 pregnant sows (Landrace × Yorkshire; 2 parities) were randomly assigned to and maintained in the following three treatment groups (13 sows per group) over days 0 to 35 of pregnancy: (i) air cooling (AC; 26.87 ± 1.23℃), (ii) water-drip cooling (WC; 28.81 ± 0.91℃), and (iii) a lack of cooling with heat stress (HS; 30.72 ± 0.70℃). Backfat thickness was measured before and after HS. Feces were collected on day 0 and 35 d of the trial for microbiome analysis, whereas blood was taken at day 35 of pregnancy and analyzed. Reproductive performance and physiological responses were identified at day 35. Respiration rate along with rectal and skin temperatures were lower (p < 0.05) in the AC group than in the HS and WC groups. Serum blood urea nitrogen values were increased (p < 0.05) in the WC group compared with those measured in the AC and HS groups. Triiodothyronine was found at greater levels (p < 0.05) in the AC than in the HS group. Reproductive performance was not affected by the cooling systems. At the phylum level, fecal pathogenic Spirochaete and Euryarchaeota were found in higher numbers (p < 0.05) in all groups after HS. Similarly, at the genus level, the amount of Treponema was greater (p < 0.05) in all groups after HS. In conclusion, our results suggest that AC or WC can ameliorate or mitigate the adverse effects of HS on the physiological parameters of pregnant sows reared under high temperatures.

Objective: This experiment was conducted to investigate the effect of insoluble cellulose supplementation to diets on the growth performance, intestinal morphology, the incidence of diarrhea, nutrients digestibility, and inflammatory responses in altering environmental conditions of animals housing. Methods: A total of 108 male pigs (Duroc×[Yorkshire×Landrace]) were randomly allocated to one of three dietary treatments (cellulose 0%, 1%, 2%) and two environmental conditions (good sanitary condition vs. poor sanitary condition) to give 6 replicate pens per treatment with three pigs per each pen at 14 days post-weaning. Results: Pigs were in good sanitary condition had higher average daily gain (p<0.01) and improved feed efficiency (p<0.05) from day 1 to 14 after weaning compared to their counterparts. The interactions were found between environmental conditions and dietary treatments (day 7: crypt depth [p<0.01], villous height to crypt depth [p<0.001]; day 14: crypt depth [p<0.001], villous to crypt ratio [p<0.01]) in ileum morphology. Crypt depth was decreased (p<0.05), and villous to crypt ratio was increased (p<0.05) only in poor sanitary conditions. Pigs exposed to the good sanitary condition had higher (p<0.05) apparent ileal digestibility (day 7, gross energy; day 14, dry matter), apparent total tract digestibility (day 14, dry matter and crude protein) compared to pigs housed in the poor sanitary condition. Meanwhile, pigs fed a diet supplemented with 2% cellulose had decreased (p<0.05) apparent ileal digestibility (day 7, dry matter; day 14, crude protein), apparent total tract digestibility (day 7, dry matter; day 14, crude protein, gross energy) compared to pigs fed a diet supplemented with 0% or 1% cellulose. Conclusion: Our results indicated that a diet supplemented with 1% cellulose increased villous to crypt ratio, however feeding a diet containing cellulose (1% or 2%) impaired nutrient digestibility for 14 day after weaning in both good sanitary and poor sanitary conditions.
Background: Obesity is a complex, medical condition causally contributing to many chronic diseases and a number of efforts have been made to find the associated markers for novel prevention and treatment of obesity. Our study was to evaluate the relationship between gut immune response and obesity and overweight with use of fecal calprotectin (FC) both in adult and children groups. Methods: Fecal samples were obtained from 74 subjects: 14 non-obese and overweight children (PN), 13 obese and overweight children (PO), 20 non-obese and overweight adults (AN), and 27 obese and overweight adults (AO). FC was measured using a commercial Legend Max quantitative enzyme-linked immunosorbent assay (BioLegend). Mann-Whitney U-test was used for statistical analysis. Results: Median FC concentration was $7.9{\mu}g/g$ (range, $1.9-28.9{\mu}g/g$) for PN, $5.0{\mu}g/g$ (range, $2.6-29.6{\mu}g/g$) for PO, $9.5{\mu}g/g$ (range, $0.8-28.9{\mu}g/g$) for AN, and $10.0{\mu}g/g$ (range, $1.6-25.6{\mu}g/g$) for AO, respectively. In both adults and children age groups, the FC showed no statistically significant difference between AO and AN or PO and PN. However, FC showed statistically significant difference (P<0.05) between AO and PO while not significant between AN and PN. Conclusion: FC level in AO was significantly higher than that in PO, suggestive of different pathophysiologic mechanism between children obesity and adults obesity.
Background: Korean Red Ginseng (KRG; Panax ginseng Meyer) is a widely used medicinal herb known to exert various immune modulatory functions. KRG and one of its purified components, ginsenoside Rg3, are known to possess anti-inflammatory activities. How they impact helper T cell-mediated responses is not fully explored. In this study, we attempted to evaluate the effect of KRG extract (KRGE) and ginsenoside Rg3 on Th1 cell responses. Methods: Using well-characterized T cell in vitro differentiation systems, we examined the effects of KRGE or enhanced Rg3 on the Th1-inducing cytokine production from dendritic cells (DC) and the naïve $CD4^+$ T cells differentiation to Th1 cells. Furthermore, we examined the change of Th1 cell population in the intestine after treatment of enhanced Rg3. The influence of KRGE or enhanced Rg3 on Th1 cell differentiation was evaluated by fluorescence-activated cell sorting, enzyme-linked immunosorbent assay, and quantitative real-time polymerase chain reaction. Results: KRGE significantly inhibited the production level of IL-12 from DCs and subsequent Th1 cell differentiation. Similarly, enhanced Rg3 significantly suppressed the expression of interferon gamma ($IFN{\gamma}$) and T-bet in T cells under Th1-skewing condition. Consistent with these effects in vitro, oral administration of enhanced Rg3 suppressed the frequency of Th1 cells in the Peyer's patch and lamina propria cells in vivo. Conclusion: Enhanced Rg3 negatively regulates the differentiation of Th1 cell in vitro and Th1 cell responses in the gut in vivo, providing fundamental basis for the use of this agent to treat Th1-related diseases.
Farm animals such as piglets are often affected by environmental stress, which can disturb the gut ecosystem. Antibiotics were commonly used to prevent diarrhea in weaned piglets, but this was banned by the European Union due to the development of antibiotic resistance. However, the use of probiotics instead of antibiotics may reduce the risk posed by pathogenic microorganisms and reduce the incidence of gastrointestinal diseases. Therefore, this study was conducted to investigate the effects of Lactobacillus casei Zhang on the mechanical barrier and immune function of early-weaned piglets infected using Escherichia coli K88 based on histomorphology and immunology. Fourteen-day-old weaned piglets were divided into a control group and experimental groups that were fed L. casei Zhang and infected with E. coli K88 with or without prefeeding and/or postfeeding of L. casei Zhang. The L. casei Zhang dose used was $10^7CFU/g$ diet. Jejunum segments were obtained before histological, immunohistochemical, and western blot analyses were performed. In addition, the relative mRNA expression of toll receptors and cytokines was measured. Piglets fed L. casei Zhang showed significantly increased jejunum villus height, villus height-crypt depth ratio, muscle thickness, and expression of proliferating cell nuclear antigen and tight junction proteins ZO-1 and occludin. The use of L. casei Zhang effectively reduced intestinal inflammation after infection. We found that L. casei Zhang feeding prevented the jejunum damage induced by E. coli K88, suggesting that it may be a potential alternative to antibiotics for preventing diarrhea in early-weaned piglets.
Objective: This experiment was carried out to determine the effects of different creep feed types on suckling performance and further adjustments to solid feed after weaning. Methods: A total of 24 multiparous sows and their litters were allotted to one of three treatment groups: i) provided highly digestible creep feed (Creep), ii) provided a pig weaning diet (Weaner), and iii) provided sow feed (Sow) as creep feed until weaning. After weaning, a total of 96 piglets were selected for evaluation of post-weaning performance. Results: For pre-weaning performance, the Creep treatment led to a significantly higher feed intake from 14 to 28 d (p<0.05) and higher body weight gain from 21 to 28 d than piglets that were provided other diets. However, after weaning, the Weaner treatment yielded a significantly higher feed intake and average daily gain than other treatments from 0 to 14 d after weaning (p<0.05); Creep treatment tended to generate lower villus heights in the duodenum than the other treatments (p = 0.07). Conclusion: Highly digestible creep feed improved pre-weaning performance, but feed familiarity and grain-based creep feed improved post-weaning performance.
Dietary fiber is defined as soluble and insoluble polysaccharide consisted in the plant cell wall-associated fibers naturally occurring in fruits, vegetables, and cereal products, and of isolated fibers that are added to processed foods which are also artificially modified. There are so many difference types of dietary fibers as arabinoxylan, polydextrose chicory, oligosccharide. inulin, pectin, bran, cellulose, ${\beta}$-glucan, resistant starch and some seaweed polymers as alginate. Most of them provide many biological benefits in the intestine, as lower risk for developing coronary heart disease, stroke, hypertension, diabetes, obesity and some of the gastrointestinal disease like as colon cancer. And also lowering cholesterol levels, improves glycemic and insulin sensitivity to non-diabetic and diabetic persons including immune system. Beside of many benefits, average consumers in developed and under developing countries take far less amounts of dietary fiber that international organization recommended. Adequate intake of dietary fiber is 14g/1,000kcal base using the energy guide line of 2,000kcal/day for women and 26,000 kcal/day for men, dietary intake is 28g/day of adult women and 36g/day for adult men. The mechanisms behind the reported effects of dietary fiber on metabolic health are not fully well established. It is suggested that changes in intestinal viscosity resulting mucus increasing, macro-nutrients absorption, rate of passage of large intestinal, production of short chain fatty acids by fermentation. Production of gut hormones and changes of microbiota in intestine. It is necessary to do more research in this field in the future and combined interdisciplinary works together.

The larvae and juveniles of Porocottus leptosomus belonging to the family Cottidae were collected (n = 95, 3.9-16.5 mm in body length, BL) from Busan, Korea, in March 2015. The larvae and juvenile were identified using DNA barcoding as P. leptosomus, and their morphological description was presented in detail. The yolk-sac larvae (3.9-5.6 mm BL) body was slightly compressed, the head was large, the eye was round and large, and the anus was before the middle of the body. The preflexion larvae (5.2-10.0 mm BL) body length drastically increased; caudal fin rays began to occur. The flexion larvae (9.4-11.8 mm BL) notochord flexion started; dorsal, pectoral, and anal fin rays began to occur; pelvic fin buds are seen; they possessed a pair of parietal spine; and a pair of supraocular cirri was first to develop. At 12 mm BL, the notochord was completely flexed. The larva stage (3.9-12.6 mm SL) had the stellate melanophores in the head, isthmus, gut, and tail (along to the ventral midline). During the juvenile stage (11.4-16.5 mm BL), melanophores covered the head and began to form five black bands on the side of the body. The larvae of P. leptosomus spent pelagic life, but moved to the bottom during the juvenile stage. The larvae and juveniles of P. leptosomus differ from other cottid larval fishes by body shape, melanophore head pattern, and spine development. P. leptosomus can be distinguished from Porocottus allisi by morphological development and the occurrence of larval fish: preopercular spine development, melanophore pattern, and caudal fin development.
A total of 18 sows with an average body weight (BW) of 211.6 kg (within 24 h after farrowing) were used for a 5-week experiment to determine the effects of the dietary valine : lysine (V : L) ratios on the performance and fecal score in lactating sows and litters. Sows were raised in individual pens and assigned to one of three experimental diets consisting of different V : L ratios (0.83, 0.85, and 0.88%). Sows BW was improved with the 0.85% V : L ratio compared to the 0.83 and 0.88%. However, no significant difference was observed for BW loss, average daily feed intake, back fat thickness and days to return to estrus among the three treatment groups during the experiment period. Furthermore, the growth performance of the piglets, the weaning weight, was improved in the group receiving the diet with the 0.85% V : L ratio compared to the 0.83 and 0.88%. Moreover, the average daily gain of the piglets was increased when the V : L ratio was 0.88% compared to 0.83 and 0.85%. The fecal score of the sows was better (p > 0.05) when the V : L ratio was 0.85% compared to the V : L ratio of 0.83%. In the piglets, the V : L ratios 0.88 and 0.85% had a better fecal score compared to the V : L ratio of 0.83% at day 14. In conclusion, the V : L ratio had no effect on the gut microflora shown by the fecal scores of the sows during lactation.

Ulcerative colitis (UC) is one of inflammatory bowel disease (IBD), characterized by chronic inflammatory response and dysregulation of immune function. The severity of US has been influenced by environmental factors and food habit. The immune modulatory, anti-inflammatory and steroidal medicine have been used for the treatment of UC. However, long-term administration of those medicine is accompanied with side-effect. So, it is necessary to develop the non side-effect medicine using natural product. Prebiotics influences intestinal condition and food consumption. The heredity, immunity and environmental condition are related with occurrence of UC. In recent study, UC patients had lower level of prebiotics such as Lactobacillus and Bifidobacterium compared with healthy people. Also, previous study announced that imbalance of enteric flora aggravates the severity of UC. The effectiveness of probiotics might affect colon ability and viable bacteria also could promote the proliferation of beneficial intestinal bacteria. Prebiotics, such as herbal medicine, could lead to balance of intestinal bacteria or increase beneficial bacteria. So, proper choice of herbal medicine could control the intestinal condition. This study aimed to investigate the effect of mixture of probiotics and prebiotics (synbiotics) on dextran sulfate sodium (DSS)-induced UC in vivo. The synbiotics consist of Lactobacillus buchneri, Polymnia sonchifolia and Glycine max Merr. in this study. To evaluate the effect of synbiotics, 3% DSS was administered in BALB/c mice and synbiotics was daily administered for experimental days. The administration of synbiotics regulated colon length shortening, body weight change and disease activity index effectively. Also, extract of synbiotics upregulated survival ability of Lactobacillus buchneri in gut condition. These results suggest that mixture of probiotics and prebiotics, called as synbiotics, could influence intestinal condition also regulate the colon disease. Synbiotics might be a therapeutic agent for treatment of UC.
