Pharmacological Studies on Root Bark Extract of Aralia elata -General Pharmac \logical Action on the Butanol Fraction- (두릅나무 근피 추출물의 약물학적 연구 - 두릅나무 추출물의 Butanol분획의 일반약리작용 -)

  • In the previous report, it was described that the butanol fraction of the root bark extract of Aralia elata had potent antigastritic and antiulcerative effects in rats, In this pharmacological profile, the fraction revealed low acute toxicities in mice and rats, no central nervous system activities, moderate analgesic action, weak inhibitory action in carrageenin edema of rats and no direct effects on isolated smooth muscles of rats and guinea-pigs. Slight lowering of blood pressure as given i.v. in rabbit was observed.

  • A porcine parvovirus inactivated vaccine was prepared and inoculated to 7 piglets and also 8 guinea-pigs, and their serum antibodies were titrated. Twenty-two field serum samples of unvaccinated sows were also tested by SN and HI methods. It was observed that SN test was superior over HI test. Therefore, it is suggested that the SN test could well be used in the detection of serum antibody for PPV in vaccinated pigs.

Action of Rodgersia podophylla Root Extracts on Melanin Biosynthesis in Skin (도깨비 부채 뿌리 추출물의 피부 과다색소침착에 미치는 영향)

  • In the previous study, we reported the inhibitory effects of Rodgersia podophylla root extract on tyrosinase activity and melanin production in melan-a cells. However, mechanism of the inhibitory activity and in vivo assay were not yet examined. This study performed the examination of the effects of Rodgersia podophylla root extract on protein expression and in vivo depigmenting activity using melan-a cells and brown guinea pigs. As the results of western immunoblotting analysis, treatment of Rodgersia podophylla root extract reduced tyrosinase expression rates in 10 and 100 ppm concentrations, dose dependently. Moreover, Rodgersia podophylla root extract exhibited depigmenting activity on UV-B induced hyperpigmentation in brown guinea pig skin. These results suggested that Rodgersia podophylla root extract could act as whitening agent for the skin via not only direct tyrosinase activity inhibition but also reducing of tyrosinase expression.

  • We have studied about the safety, immunity and protective potency in swine and experimental am mais of two inactivated vaccine produced with NYJ-1-87 strain of ADV that was isolated in Korea. Result obtained through the experiments were summarized as follows. 1. The safe potency of ADV antigens inactivated with BEI and formaline to mouse & guinea pig was on the whole good, but protective potency rates of those to challenge with ADV were 60-75% without the differences to two antigens. 2. Safety, immunity & protective potency of ADV antigens inactivated with BEI and formaline to swine were on the whole excellent, except for a mild increase of rectal temperature in some pigs after challenge with ADV. 3. When virus excretion of the experimental groups after challenge with ADV was examed by swabbing of nasal, all pigs of control gorup excreted virus from 2 days p.c., partially to 10 days p.c.. But in vaccinated groups, only 25-50% of all pigs of each group excreted virus during experimental periods. 4. Titers of antibodies in swine & quinea pig vaccinated with inactivated ADV antigens become increased after the 1 weeth p.i. showing the highest liters on the 4-5 weeths p.i.

Experimental Studies on the Anti-diarrhea Effects of Jisa-tang (지사탕(止瀉湯)의 항사하작용(抗瀉下作用)에 관(關)한 연구(硏究))

  • Objective : The aim of this experimental study was to investigate the anti-diarrhea effects of Jisa-tang using mice and guinea pigs. Methods : We feed Jisa-tang to mice and guinea pigs to investigate its effects for anti-diarrhea action. We observed its actions on gastrointestinal smooth muscles, on the transportability of small and large intestines, on diarrhea induced by castor oil and magnesium sulfate, and on enteropooling by castor oil and prostaglandin $E_2(PGE_2)$. Results : 1. Jisa-tang showed alleviation, depending on the density, only on the contraction of mice's gastrointestinal smooth muscles induced by histamine. 2. The transportability of the small intestine was not significantly constrained by Jisa-tang. However, the enhancement of pyridostigmine-induced transportability of he small intestine was significantly constrained in the group administered 900mg/kg of Jisa-tang (p<0.05). 3. The transportability of large intestine was significantly constrained in the group administered 1,800mg/kg of Jisa-tang. 4. Jisa-tang showed significant anti-diarrhea effects on diarrhea induced by castor oil and by $MgSO_4$ in the group administered 1,800mg/kg of Jisa-tang. 5. Significant reduction of effects of enteropooling induced by caster oil and by prostaglandin $E_2$ were observed only in the group administered 1,800mg/kg of Jisa-tang. Conclusions : We conclude that Jisa-tang has advantageous effects on drug-induced diarrhea and will contribute to the development of diarrhea treatment through further related studies.

An Ototoxic Antibiotic Gentamicin Can Increase PKA-caveolin-1 Signaling Pathway in Differentiated Vestibular Cell Line (UB/UE-1)

  • Caveolin proteins are mediators of cell death or the survival of injured cells, and they are inhibitors of various signaling pathways. The expression of caveolin-, which is involved in the protein kinase A (PKA) signaling pathway, was examined in the differentiated mouse vestibular cell line UB/UE-1 after gentamicin ototoxicity. Caveolae in the vestibular hair cell of healthy guinea pigs were observed through an electron microscope. UB/UE-1 cells were cultured at 95% $CO_2$ with 5% $O_2$ at $33^{\circ}C$ for 48 hours and at 95% $CO_2$ with 5% $O_2$ at $39^{\circ}C$ for 24 hours for differentiation. Cells were treated with 1 mM gentamicin, 0.02 mM H89 (PKA inhibitor), and then incubated for 24 hours. Caveolin-1 expression was examined by western blotting and PKA activity by a $PepTag^{(R)}$ assay. Caveolae were observed in the vestibular hair cells of healthy guinea pigs by electron microscopy. Caveolin-1 was expressed spontaneously in differentiated UB/UE-1 cells and increased after gentamicin treatment. PKA was also over-activated by gentamicin treatment. Both gentamicin-induced caveolin-1 expression and PKA over-activation were inhibited by H89. These results indicate that gentamicin-induced caveolin-1 expression is mediated by the PKA signaling pathway. We conclude that caveolae/ caveolin activity, induced via a PKA signaling pathway, may be one of the mechanisms of gentamicin-induced ototoxicity.

Virulence-associated plasmids of Salmonella spp. isolated from animals in Korea (동물에서 분리된 Salmonella균의 병원성 관련 Plasmid에 관한 연구)

  • This paper dealt with plasmid DNA profile in 98 Salmonella(S) isolated from pigs and cattle sources in Taegu, Gyeongbook and Gyeongnam during the period from 1984 to 1987. Also we were studied for restriction enzyme analysis of the plasmid DNA, and mouse infection, Sereny test and normal setum resistance test in guinea pig for S typhimurium and S enteritidis harbored or cured 60 megadalton(Md) plasmid and 36 Md plasmid, respectively. Of the 13 Salmonella isolated from cattle, 7 Salmonella harbored one or more plasmids and molecular sizes of the large plasmids were 60 Md for S typhimurium and 36 Md for S enteritidis. Of the 85 Salmonella isolated from pigs, 47 Salmonella were confirmed as being one or more plasmids, and all the S typimurium stains harbored 60 Md plasmid. In enzyme digestion with 8 types of restriction endonuclease for 60 Md plasmid DNA of S typhimurium, cleavage patterns were varied to enzymes, and the DNA was segmented into 4 to 15 fragments. In restriction enzyme analysis of 36 Md plasmid DNA obtained from four strains of S. enteritidis, the DNA showed the same cleavage patterns obtained with Eco RI, Hind III and Bam H I, and was segmented into 3 to 5 fragments. In virulence for mice by measuring the 50% lethal dose ($LD_{50}$), the $LD_{50}$ values obtained for 60 Md virulence-associated plasmid harbored strains of S typhimurium and 36 Md virulence-associated plasmid of S enteritidis were up to $10^4$-fold lower than the values obtained for the plasmid-cured strains of the same serotype. Only the plasmid harbored strains were resistant to the bactericidal activity of 90% guinea pig serum, and only they gave positive responses in sereny test. We suggested that their plasmid DNA might be associated with virulence for mice.

Near-Infrared Laser Stimulation of the Auditory Nerve in Guinea Pigs

  • This study has investigated the feasibility of 980-nm low-energy pulsed near-infrared laser stimulation to evoke auditory responses, as well as the effects of radiant exposure and pulse duration on auditory responses. In the experiments, a hole was drilled in the basal turn of the cochlea in guinea pigs. An optical fiber with a 980-nm pulsed infrared laser was inserted into the hole, orientating the spiral ganglion cells in the cochlea. To model deafness, the tympanic membrane was mechanically damaged. Acoustically evoked compound action potentials (ACAPs) were recorded before and after deafness, and optically evoked compound action potentials (OCAPs) were recorded after deafness. Similar spatial selectivity between optical and acoustical stimulation was found. In addition, OCAP amplitudes increased with radiant exposure, indicating a photothermal mechanism induced by optical stimulation. Furthermore, at a fixed radiant exposure, OCAP amplitudes decreased as pulse duration increased, suggesting that optical stimulation might be governed by the time duration over which the energy is delivered. Thus, the current experiments have demonstrated that a 980-nm pulsed near-infrared laser with low energy can evoke auditory neural responses similar to those evoked by acoustical stimulation. This approach could be used to develop optical cochlear implants.
