메탄올만 이용하여 성장하는 Methylobacillus의 분리 및 특성 (A Methylobacillus Isolate Growing Only on Methanol)

  • An obligate methanol-oxidizing bacterium, Methylobacillus sp. strain SK1, which grows only on methanol was isolated from soil. The isolate was nonmotile Gram-negtive rod. It does not have internal membrane system. The colonies were small, whitish-yellow, and smooth. The guanine plus cytosine content of the DNA was 48 mol%. Cellular fatty acids consisted predominantly of large amounts of straight-chain saturated $C_{16:0}$ acid and unsaturated $C_{16:1}$ acid. The major ubiquinone was Q-8, and Q-10 was present as minor component. The cell was obligately aerobic and exhibited catalase, but no oxidase, activity. Poly-.betha.-hydroxybutyrate, endospores, or cysts were not observed. the isolate could grow only on methanol in mineral medium. Growth factors were not required. The isolate was unable to use methane, formaldehyde, formate, methylamine, and several other organic compounds tested as a sole source of carbon and energy. Growth was optimal at 35.deg.C and pH 7.5. It could not grow at 42.deg.C. The doubling time was 1.2h at 30.deg.C when grown with 1.0%(v/v) methanol. The growth was not affected by antibiotics inhibiting cell wall synthesis and carbon monoxide but was completely suppressed by those inhibiting protein synthesis. Methanol was found to be assimilated through the ribulose monophosphate pathway. Cytochromes of b-, c-, and o- types were found. Cell-free extracts contained a phenazine methosulfate-linked methanol dehydrogenase activity, which required ammonium ions as an activator. Cells harvested after the late exponential phase seemed to contain blue protein.ein.

가미지황탕(加味地黃湯), 가미사군자탕(加味四君子湯) 및 가미군자지황탕(加味君子地黃湯)의 항종양활성(抗腫瘍活性) (Study on Antitumor Activity of Kamisagoonjatang, Kamijihwangtang and Kamigoonjajihwangtang)

  • To evaluate the antitumor activity of Kamisagoonjatang(KST), Kami-jihwangtang (KJT) and Kamigoonjajihwangtang(KKJT), studies were done experimentally. The results were obtained as follows: 1. In cytotoxicity against B16-F10, HT1080, SNU, and L1210, con-centration inhibiting cell growth up to below 50% of control was recognized at $10^{-3}g/ml$ of KKJT. 2. In cytotoxicity against A549, SK-OV-3, XF498 and HCT15, concentration inhibiting cell growth up to below 30% of control was over $400{\mu}g/ml$ of KKJT only and also over $200{\mu}g/ml$ against SK-MEL-2. 3. In Inhibitory effect on activity of DNA topoisomerase I, the $IC_{50}$ was shown $200-400{\mu}g/m{\ell}$, of KST, over $400{\mu}g/m{\ell}$ of KJT and $100-200{\mu}g/m{\ell}$ of KKJT. 4. The T/C% was 122.8 in KJT, 127.4 in KST and 158.4 in KKJ-Ttreated group in S-180 bearing ICR mice. 5. In hematological changes in S-180 bearing ICR mice, numbers of WBC were decreased significantly in KJT and KKJT treated groups as compared with control, whereas those of platelet were increased with no significance in all groups as compared with control. From above results it was concluded that KKJT could be usefully applied for the prevention and treatment of cancer.

Role of Siderophores in Biocontrol of Fusarium solani and Enhanced Growth Response of Bean by Pseudomonas fluorescens GL20

  • Plant growth-promoting Psudomonas fluorescens GL20 was isolated from a ginseng rhizosphere on chrome azurol Sagar. P. fluorescens GL20 produced a large amount of hydoxamate siderophore in an iron-deficient medium. The siderophore showed significantly high specific activity of 20.2 unit. Using an in vitro antifungal test, P. fluorescens GL20 considerably suppressed growth of phytopathogenic fungus Fusarium solani, inhibiting spore germination and germ tube elongation. In pot trials of kidney beans with P. fluorescens GL20, disease incidence was remarkably reduced up to $68{\%}$ compared with that of F. solani alone, and plant growth was also increased nearly 1.6 fold as compared to that of the untreated control, promoting elongation and development of the roots. These results indicate that the plant growth-promoting activity of P. fluorescens GL20 can play an important role in biological control of soil-borne plant disease in a rhizosphere, enhancing the growth of plants.

Antibacterial Activities of Persimmon Roots-derived Materials and 1,4-Naphthoquinone's Derivatives against Intestinal Bacteria

  • The growth-inhibiting activities of persimmon roots-derived materials against intestinal bacteria were evaluated and compared with that of 1,4-naphthoquinone as a positive control. The active constituent isolated from persimmon roots was characterized as 5-hydroxy-2-methyl-1,4-naphthoquinone using various spectroscopic analyses. Treatment with 1,4-naphthoquinone at a dose of 1.0 mg/disc strongly inhibited the growth of 6 intestinal bacteria. Furthermore, when the structure-activity relationships of 1,4-naphthoquinone's derivatives were evaluated, 5-hydroxy-2-methyl-1,4-naphthoquinone and 2-methyl-1,4-naphthoquinone were found to strongly inhibit the growth of Clostridium difficile, Clostridium perfringens, and Escherichia coli without adversely affecting the growth of Bifidobacterium adolescentis, Bifidobacterium longum, and Lactobacillus acidophilus. Additionally, 2-hydroxy-1,4-naphthoquinone and 5-hydroxy-1,4-naphthoquinone strongly inhibited the growth of C. difficile and C. perfringens, but did not inhibit the growth of E. coli. Taken together, these results indicate that persimmon roots-derived materials and some of 1,4-naphthoquinone's derivatives could be useful preventive agents against diseases caused by harmful intestinal bacteria.

  • Benzotriazole 유도체들의 정량적인 구조와 발아후 벼(Oryzae sativa L) 및 피(Echinochloa crus-galli)에 대한 생장저해 활성값$(pI_{50})$사이의 관계들이 QSAR 방법으로 연구되었다. QSAR 분석에 의하면, 자유 라디칼 파라미터$(E_R)$가 매우 중요한 요소이었으며 $pI_{50}$ 값은 파라치환기(X)의 $E_R$ 파라미터에 대하여 포물선형의 관계를 나타내었다. (3)의 생장저해 활성은 (4)보다 우세하였으며 생장저해 활성이 가장 큰 화합물은 (3b)이었고 피의 생장저해 활성에 대한 $E_R$ 파라미터의 적정값은 각각 $E_R(3)=0.52$$E_R(4)=0.15$이었다. 분자설계 결과, 치환기(X)가 전자 끌게이고 적정값 (0.52)의 $E_R$ 파라미터를 가지는 분자가 가장 활성이 클 것으로 기대되었으며 Benzotriazole 유도체들의 생장저해 활성은 광합성반응(PS-II)에서 전자전달을 효과적으로 차단하는 결과로 믿어진다.

  • The purpose of this study was to compare the growth-inhibiting effect of sodium fluoride and stannous fluoride on a cariogenic strain of Streptococcus mutans exposed to the different concentration of $SnF_2$ and NaF The result were as follows: 1. The growth rate of the Streptococcus mutans was unaffected by 75 ppmF, slowed by 150 ppmF, and immediately arrested by 300 or 600 ppmF where increments of NaF were added to actively growing 12-hour broth culture. 2. $SnF_2$ slowed the growth rate at 75 ppmF, was to bactericidal initially at 150 and 300 ppmF, and was to totally bactercidal at 600 ppmF. 3. $SnF_2$ has been shown to be more effective than NaF in inhibiting the growth activity of Streptococcus mutans. 4. The inhibitory effect of fluoride compounds on the growth of Streptococcus mutans was increased in the decreased pH at concentration of 600 ppmF.

Biological Activities of Essential Oils from Angelica tenuissima Nakai

  • The current study was conducted to evaluate the antibacterial and antioxidant activities of the essential oil fraction from the roots of Angelica tenuissima Nakai and its main components. We extracted the essential oil fraction from the roots of A. tenuissima using steam distillation and isolated its main components. Their antibacterial activities were determined by broth dilution test against food-borne pathogenic bacteria. Antioxidant activities were evaluated by DPPH-scavenging assay and reducing-power test. Also tested was their ability to inhibit the growth of two gastrointestinal cancer cell lines, Caco-2 and MKN-45. The A. tenuissima oil fraction and its main components, ligustilide and butylidene phthalide exhibited marked inhibitory effects against most of the tested antibiotic-susceptible and antibiotic-resistant bacterial strains with minimum inhibiting concentrations (MICs) from $0.21{\pm}0.08$ to $3.60{\pm}0.89mg/ml$. They also showed growth-inhibiting activity against Caco-2 and MKN-45 cells. The oil fraction showed significant antioxidant activities in DPPH radical scavenging assay and reducing-power test. Taken together, A. tenuissima essential oil could be used as a safe additive for preventing food contamination by pathogenic bacteria. Additionally, its antioxidative activity and the ability to inhibit gastrointestinal carcinoma cell lines could increase its value for functional foods and prevention of cancer.

수홍화자(水紅花子)의 항암활성(抗癌活性) 및 항전이(抗轉移) 효과(效果)에 관(關)한 연구(硏究) (Study on Antitumor Activity and Antimetastatic Effects of Polygoni Orientalis Fructus(POF))

  • To evaluate the antitumor activity and antimetastatic effects of Polygoni Orientalis Fructus(POF), studies were done experimentally. The results were obtained as follows : 1. In cytotoxicity against A549, and XF498 cell concentration inhibiting cell growth up to below 30% of control was recognized at $200{\mu}g/m{\ell}$ of POF. Also POF inhibited cell growth up to below 30% of control against SK-OV-3, SK-MEL-2, HCT15 and MCF-7 cell at $100{\mu}g/m{\ell}$. 2. The concentration inhibiting adhesion of A549 and SK-OV-3 to complex extracellular matrix up to below 70% of control was recognized at $100{\mu}g/m{\ell}$ of POF. 3. In pumonary colonization assay, a number of colonies in the lungs were decreased significantly in POF treated group as compared with control group. These results suggested that POF extracts might be usefully applied for prevention and treatment of cancer.

Chromatographically Purified Porphyran from Porphyra yezoensis Effectively Inhibits Proliferation of Human Cancer Cells

  • In this study, we isolated porphyran was isolated from the red seaweed Porphyra yezoensis and assessed in terms of in vitro anti-proliferative activity. Sequential anion-exchange and gel-filtration chromatography led to purification of 3 porphyrans of different molecular masses, which contained <$50\;{\mu}g/mL$ protein and >$10\;{\mu}g/mL$ porphyran. Crude porphyran inhibited cell growth in a dose-dependent manner (0-5 mg/mL). When HT-29 colon cancer cells and AGS gastric cancer cells were cultured with various concentrations of the purified porphyran, cancer cell growth was inhibited by 50% at a low concentration (5 or $10\;{\mu}g/mL$). Furthermore, the polysaccharide portion of the porphyran preparation, rather than the protein portion, is the most effective at inhibiting cancer cell proliferation via apoptosis, as indicated by increased caspase-3 activity. Our results indicate that purified porphyran has significant in vitro anti-proliferative activity (p<0.05).

Conditioned Media of ASC-17D Sertoli Cells Induce G1-Growth Arrest of DU145 Human Prostate Cancer Cells

  • We studied the effects of ASC-17D rat Sertoli cell-conditioned media (rSCCM) on the proliferation of the DU145 prostate cancer cells. rSCCM was prepared from ASC-17D cells cultured in DMEM/F-12 serum-free media at a nonpermissive temperature of $40^{\circ}C$, which is the condition for the high expression of c1usterin. We found that rSCCM could inhibit the proliferation of DU145 cells by arresting the cell cycle in the G1 phase in a dose-dependent manner. This growth arresting activity was abolished by boiling rSCCM for 5 min. The G1 growth-inhibiting activity of rSCCM was also detected in other prostate-originated cancer cells examined (i.e., LNCaP and PC-3) but not in other cells (ASC-17D, HepG2, SK-N-SH, and NIH3T3). Western blot analysis of partially purified growth inhibiting fractions with the clusterin antibody showed that the cytostatic factor in rSCCM was not c1usterin. This cytostatic factor was semi purified by DEAE-Sepharose, ammonium sulfate precipitation, and Phenyl-Sepharose column chromatography, and was estimated to have a molecular weight of 88 kDa by Sephacryl S-300 gel filtration.
