Productivity of biosurfactant (rhamnolipid) by Pseudomonas aeuginosa F722 was investigated in the several culture conditions and culture composition. Biosurfactant production by P. aeuginosa F722 was amounted to 0.78 g/l as the result of the nitrogen sources and carbon sources without investing of optimum conditions. As for that one was investigated, biosurfactant production by P. aeruginosa F722 was amounted to 1.66 g/l. Biosurfactant production increased twofold because the composition of a modified C-medium was investigated efficiently. $NE_4$Cl or $NaNO_2$ inorganic nitrogens and yeast extract or trypton organic nitrogens were effective, but others inorganic nitrogens and organic nitrogens tested were not efficient far biosurfactant production by P. aeruginosa F722. The optimum concentration of $NH_4$Cl; inorganic nitrogen and yeast extract; organic nitrogen were 0.05% and 0.1%, respectively. In various carbon sources, others with the exception of hydrophobic property substrate (n-alkane) and hydrophilic property substrate (glucose, glycol) were not found to be effective fur biosurfactant production, and 3.0% was better in yield than other concentration of glucose. This yielded C-to-N ratios between 17 and 20. In our experiment, the highest biosurfactant production by P. aeruginosa F722 were observed in 5 days cultivation, containing glucose 3.0%, $NH_4$Cl 0.05%, and yeast extract 0.1% and C-to-N ratio was 20. Optimal pH and temperature for biosurfactant production were 7.0 and $35^{\circ}C$, respectively. Under the optimal culture conditions with glucose, biosurfactant production was amounted to 1.66 g/l. Velocity of biosurfactant production and strain growth increased after nitrogen depletion. The average surface tension of 30 mN/m after the 3 days of incubation under optimal culture condition was measured by ring tensionmeter.

The aim of this study was to investigate the effect of season and location on activities of enzyme produced by psychrotrophic bacteria isolated from raw milk located in Kyunggi region of South Korea. Agar diffusion and colorimetric methods were used for the lipase and protease activities of psychrotrophic bacteria. Intensities of dark blue and transparent ring around colony were compared for activity measurement. Nutrient agar with 1% skim milk added was employed for measuing protease activity. 14 strains of Arthrobacter russicus with lipase activity and 19 strains of Chryserobacterium shigense with protease activities were found to be present. It was found that Acinetobacter genomospecies 10 (match %: 99.90) isolated from B region in fall was the most lipolytic species, whereas Serratia liquefaciens (match %: 99.39) isolated from the same region in spring was the most proteolytic species. Growth curve of Acinetobacte and Serratia liquefaciens was a typical sigmoidal form. Lipase activity increased with incubation time, but its activity began to drop at stationary to motality phase. Optimum condition for incubation time, pH and temperature for extracellular lipase from Acinetobacter genomospecies 10 (match %: 99.90) was 12 h, 8.5, and $45^{\circ}C$, respectively. Extracellular protease from Serratia liquefaciens (match %:99.39) had the same optimum incubation time and pH as extracellular lipase, but optimum temperature was $35^{\circ}C$.
The purpose of this study was to evaluate the cytotoxic effects of 6 cavity liners in vitro. Human fibroblasts were cultured in ${\alpha}$-MEM and each liner was manually mixed and filled in glass ring cylinder ($8{\times}8mm$ in diameter, in height). The cylinders filled with the liners were placed in the center of the dish (35mm in diameter) containing 3ml of ${\alpha}$-MEM. Millipore filters (pore size $0.22{\mu}m$) to simulate dentin barrier were also placed between the bottom of cylinder and the dish. Then the culture dishes were stored in 5% $CO_2$ containing incubator for 5 and 10 days at the temperature of $36.6^{\circ}C$. The results of the experiments were analyzed by counting the cells in the period of 5 and 10 days respectively, and were assessed by calculating the cell multiplication rate and the relative growth rate. The experiemntal groups and the control group were compared statistically. The results of the study were summarized as follows: 1. The cell number of Zinc oxide-eugenol was $(4.13{\pm}1.31){\times}10^4$ cells/ml at 5 days and $(4.32{\pm}1.61){\times}10^4$ cells/ml at 10 days. 2. The cell number of Cavitec was ($8.35{\pm}2.87{\times}10^4$ cells/ml and $(10.08{\pm}5.10){\times}10^4$ cells/ml at 5 and 10 days respectively. 3. The cell number of Dycal was $(13.56{\pm}3.89){\times}10^4$ cells/ml at 5 days and $(34.75{\pm}8.85){\times}10^4$ cells/ml at 10 days. 4. The cell number of life was $(11.46{\pm}3.32){\times}10^4$ cells/ml and $(21.92{\pm}6.18){\times}10^4$ cells/ml at 5 and 10 days. 5. The cell number of Base cement was $(13.73{\pm}3.73){\times}10^4$ cells/ml and $(36.68{\pm}5.20){\times}10^4$ cells/ml at 5 and 10 days. 6. The cell number of Dentin cement was $(13.58{\pm}3.90){\times}10$ cells/ml and $(66.95{\pm}24.09){\times}10$ cells/ml at 5 and 10 days. 7. The cell multiplication rate of zinc oxide-eugenol cements was significantly less than that of the calcium hydroxide and glass ionomer cement. (P < 0.05)
The purpose of this experiment was to investigate the development of new spot disease on the leaf and stem of rosemary (Rosmarinus officinalis) in commercial greenhouses at Jeonju and Namwon in Korea. Incidence of target spot on rosemary was higher at the end of the rainy season with high humidity. Those symptoms were black ring spots (3-5 mm in diameter) and withering on green leaves and stems. Conidiophores and conidia were formed on the infected tissue in moist chamber and conidia were shown as the cylindrical and oval types in chain, ranged from 55 to $275{\mu}m$ in length, and 7 to $14{\mu}m$ in width. Conidia with eight to ten pseudosepta were formed on the conidiapore. The optimum growth temperature of isolates was $30^{\circ}C$ on the PDA medium under the dark condition. In the pathogenesis test, the target spot and withering symptoms were appeared on the leaves and stems 3 days after inoculation showing similar symptoms compared to those of in nature. The same fungus was re-isolated from infected lesion, indicating that Corynespora cassiicola caused leaf target spot and twig blight on rosemary. The rDNA ITS nucleotide sequences of the pure cultured isolate from the diseased area on rosemary showed 100% similarity to the sequences of C. cassiicola available in the GenBank database (JQ595296, JQ595297, FJ852715 and AY238606). Therefore, we report that the target spot of leaves and stems in rosemary was caused by C. cassiicola.
We observed the physical and chemical properties of a soil on pine mushroom picking areas where were located in the most upper and lower parts showing a comparative climatic characteristics in Korea. The slope gradients within the investigation areas which were divided into 100 quadrates of $1m^2$ ranged from $5.7{\sim}8.6{\beta}$ to $24{\sim}22.7{\beta}$ (left to right) and $4.5{\sim}6.8{\beta}$ to $13.5{\sim}17.8{\beta}$ (top to bottom) for Ponghwa and Gansung, respectively. The amount of clay and thickness of organic matter were significantly decreased with increasing slope gradient, resulting in decrease of the soil moisture content around a fairly ring-colony of Tricholoma matsutake which was observed under the relatively thicker organic matter layer beyond 3 cm depth. Soil pHswere weak acid and average EC was $0.44dS\;m^{-1}$ in both areas. The cations were in the order of Fe K > Na > Mg > Ca and Fe > K > Na > Ca > Mg for the upper(Gansung) and the lower (Ponghwa) part. And the amount of Fe was approximately $80dS\;m^{-1}$ or greater in the pine mushroom picking soil. From this, we could assume that the growth of the pine mushroom was closely related not only with iron but also soil moisture content.
The X-linked inhibitor of apoptosis (XIAP) is a member of a novel family of inhibitors of apoptosis and has several BIR domains (BIR1, BIR2, and BIR3) and a carboxy-terminal RING zinc-finger. Since suppressionof apoptosis is fundamentally important for carcinogenesis and tumor growth, we investigated the expression and function of XIAP in DEN-induced carcinogenesis using rat model. Wistar rats were injected intraperitoneally with DEN at a dose of 50 mg/kg in twice a week for 12 weeks (Group II) and 16 weeks (Group III) followed by the recovery periods, respectively. The evaluation of DEN-induced carcinogenesis carried out the blood, RT-PCR, histopathological and western blot analysis. The level of blood chemistry including GOT/GPT, albumin, and total bilirubin were significantly exchanged comparing to control and Group I/Group II. The expression of albumin and collagen mRNA were significantly exchanged (P<0.05) in both groups. In addition, AFP mRNA expression decreased more after recovery periods than Group II. XIAP was expressed constitutively in normal rat liver as well as DEN-induced Groups I and Group II. In addition, XIAP expression increased more in Group I with 4 weeks recovery periods than Group I. However, XIAP expression shown to increase in Group lI, otherwise, it was decreased in Group II with 10 weeks repair periods. Taken together, these results suggest the alteration of XIAP expression could be involved in hepatocellular carcinogenesis.

In this study, we have prepared mesophase pitch from pyrolysis fuel oil (PFO) by heterogeneous reforming process. This process was conducted by direct fluorination at various temperature and followed by the heat treatment at $390^{\circ}C$. The reformed pitch was then investigated by softening point analysis, elemental analysis, fourier-transform infrared spectroscopy, high resolution X-ray diffraction and polarization microscope analysis. Carbon contents of reformed pitch increased according to increasing the reaction temperature of fluorination, while oxygen, nitrogen and sulfur contents were completely eliminated. As the fluorination temperature increased, the creation, growth, coalescence and alignment process of mesophase spheres were observed. Also the interlayer spacing of carbon hexagonal planar structure decreased, while its crystalline size increased. In addition, aromatic ring compound contents increased by the condensation polymerization of aliphatic compound. These results can be attributed to the radical reactivity of the fluorine increased as the reaction temperature increased. It was considered that the fluorination reaction could help PFO to generate aromatic compounds, via promoting polymerization by radical reaction.
The antimicrobial effects of ten isothiocyanates (ITCs) present in cruciferous vegetables and radish root hydrolysate were investigated against pathogenic bacteria from olive flounder. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were measured against two gram-positive bacterial strains (Streptococcus parauberis, S. iniae) and four gram-negative bacterial strains (Edwardsiella tarda, Vibrio ichthyoenteri, V. harveyi, Photobacterium damselae) by using a broth microdilution technique. The antibacterial activity of ITCs was in the order sulforaphane > sulforaphene > phenylethyl ITC > erucin > benzyl ITC > iberin > I3C > allyl ITC > phenyl ITC > hexyl ITC. The susceptibility of fish pathogens to ITCs was in the order of V. harveyi > E. tarda > P. damselae > S. parauberis > S. iniae > V. ichthyoenteri. Antimicrobial activity (MIC) of radish root hydrolysate was 0.250 mg/mL against S. iniae, 0.438 mg/mL against S. parauberis, and 0.500 mg/mL against both E. tarda and V. harveyi. The aliphatic ITCs were potent inhibitors of the growth of fish pathogens, followed by aromatic ITCs and indolyl ITC. The presence of a double bond in the chemical structure of ITCs decreased antibacterial activity, while ITCs with a thiol (-S-) group and a longer carbon chain increased antibacterial activity. These results suggest that ITCs have strong antibacterial activities and may be useful in the prevention of fish pathogens.
Mechanical valve is one of the most widely used implantable artificial organs of which the reliability is so important that its failure means the death of patient. Therefore early noninvasive detection is essentially required, though mechanical valve failure with thrombosis is the most common. The objective of this paper is to detect the thrombosis formation by spectral analysis and neural network. Using microphone and amplifier, we measured the sound from the mechanical valve which is attached to the pneumatic ventricular assist device. The sound was sampled by A/D converter(DaqBook 100) and the periodogram is the main algorithm for obtaining spectrum. We made the thrombosis models using pellethane and silicon and they are thrombosis model on the valvular disk, around the sewing ring and fibrous tissue growth across the orifice of valve. The performance of the measurment system was tested firstly using 1 KHz sinusoidal wave. The measurement system detected well 1KHz spectrum as expected. The spectrum of normal and 5 kinds of thrombotic valve were obtained and primary and secondary peak appeared in each spectrum waveform. We find that the secondary peak changes according to the thrombosis model. So to distinguish the secondary peak of normal and thrombotic valve quantatively, 3 layer back propagation neural network, which contains 7, 000 input node, 20 hidden layer and 1 output was employed The trained neural network can distinguish normal and valve with more than 90% probability. As a conclusion, the noninvasive monitoring of implanted mechanical valve is possible by analysing the acoustical spectrum using neural network algorithm and this method will be applied to the performance evaluation of other implantable artificial organs.

Purpose: E-cadherin is an adhesion molecule essential for tight connection between cells, forming the cadherin/catenin complex. Truncated $\beta$-catenin disrupts the interaction between E-cadherin and $\alpha$-catenin, leading to the loss of intercellular adhesion. Met protein, the hepatocyte growth factor receptor, plays important roles in signal transduction. We investigated the relationships between the expressions of E-cadherin, $\beta$-catenin, and c-met protein and the clinicopathological and prognostic parameters in gastric adenocarcinomas. Materials and Methods: The patterns of E-cadherin, $\beta$-catenin, and c-met protein expression were studied using immunohistochemistry in formalin-fixed, paraffin-embedded archival tissues from 76 surgically resected gastric adenocarcinomas. Results: Increased expressions of E-cadherin, $\beta$-catenin, and c-met were more significantly correlated in early gastric cancers (EGC) than in advanced gastric cancers (AGC) (P=0.002, P=0.003 and P=0.026). The positive immunoreactivities of all three markers were markedly lower in signet ring-cell type and poorly differentiated type lesions than in intestinal-type lesions. Decreased expression of the $\beta$-catenin protein correlated well with increased tumor invasion depth (P=0.039), and increased lymph node metastasis correlated well with reduced expression of c-met (P=0.046). Conclusion: In gastric cancers, reduced expressions of the E-cadherin, $\beta$-catenin, and c-met proteins may play some role in poorer tumor differentiation, deeper tumor invasion, and increased lymph node metastasis. Also, the c-met gene is thought to play a specific role in the mechanism of the yet unknown catenin action.
