Intraspecific Variation in the Temperature Niche Component of the Diatom Skeletonema costatum from Korean Coastal Waters

  • Final biomass yields (peak optical density) and growth rates (divisions/day) of seven clones of Skeletonema costatum from Korean coastal waters were measured to understand their intraspecific variations in the light intensity niche component under $25^{\circ}C$ condition. Daily growth rates of 6 of 7 S. costatum, clones were maximum at 6000 lux while that of YS4, a neritic clone, was maximum at 9000 lux. The final biomass yields of 4 of the 7 S. costatum clones were maximum at the lowest light intensity of 2000 lux. Minimum final biomass yields were found at 9000 lux in all the S. costatum clones other than an estuarine clone, HDC9. The intraspecific variations of the mean growth rate and mean final biomass yield under each of the three different light intensity in terms of the coefficient of variation were not greater than 10% in any of the 7 S. costatum clones.

Improving Adherence to Growth Hormone (GH) Therapy via EasypodTM May Help Maximize the Treatment Outcome

  • In Growth Hormone (GH) therapy, suboptimal adherence is a common problem, reaching up to 82%, and there is a need for interventions to improve adherence and to maximize patients' growth potential eventually. Current studies have demonstrated the association between the rate of non-adherence and reduced height velocity. In order to maximize patients' potential to grow, an auto-injecting/recording device, such as $easypod^{TM}$, may help improve adherence and optimize the treatment effects of GH therapy. The use of $easypod^{TM}$ has contributed to high adherence rates: 87.5% and 93% in Bozzola et al.'s study and the $Easypod^{TM}$ Connect Observational study (ECOS), respectively. Improvement of adherence by $easypod^{TM}$ may lead to higher growth rates of patients receiving GH therapy. Additionally, patients' positive acceptability of $easypod^{TM}$ suggests $easypod^{TM}$ is a preferred device by patients for better adherence.

  • 본 연구에서는, 이러한 상황을 감안하여 피로시험을 위한 유사랜덤 (pseudorandom)하중 파형을 퍼스널컴퓨터 시뮬레이션에 의해 작성하는 소프트웨어를 개발함과 동시에 이를 이용하여 표면피로균열진전시험도 약간 수행하여, 변동하중하의 표면균열 진전거동에 관하여 대체적인 특성을 검토하여 보기로 하였다.

  • An experimental investigation of the fatigue through crack growth behavior under simple stepped variable loading condition has been performed using Al7075-T651. Experiments were carried out by using cantilever bending type specimens, with chevron notches on a small electro-magnetic test machine. Tensile overloads have a retarding effect on the fatigue crack growth rates, therefore tensile overloads were used for the beneficial effect on the fatigue life. While in most cases compressive overloads have only a vanishing effect on crack growth rates, some experiments with single edge crack tension specimens reveal a marked growth retardation. The stress ratios used in this investigations varies from R=0.32 to 0.81, from R=0.04 to 0.76, from R=-0.15 to 0.73, and from R=-0.33 to 0.68 and the peak load for each case was not varied. The crack growth and crack closure were measured by Kikukawa's compliance method with a strain gauge mounted on the backside of each specimens. The results obtained are as follows. When the stepped variable load was applied, the smaller the stress ration was, the larger the delayed retardation of the crack growth rate was. The fatigue crack growh rate data obtained for through cracks were plotted well against the effective stress intensity factor range from 4.0 to 20.0MP{a^{SQRT}m}. It was found that the effective stress intensity factor range ratio was related well to the opening stress intensity factor, the maximum stress intensity factor, and crack length.

  • Fatigue crack growth tests were conducted on urban railway wheel steel under mode I and mixed-mode conditions. Fatigue crack growth rates were evaluated in terms of equivalent stress intensity factor ranges, using both the extended and projected crack lengths. The equivalent stress intensity factor range with the growth rate results obtained under mode I loading conditions can be used to predict the crack growth rate under mixed-mode loading conditions. Extended crack length rather than projected crack length is appropriate for the prediction of the crack growth rate under the mixed-mode loading conditions.

Effect of salinity on growth and nutrient uptake of Ulva pertusa (Chlorophyta) from an eelgrass bed

  • The effects of salinity on various ecophysiological parameters of Ulva pertusa such as growth, nutrient uptake, photosynthetic performance and internal nutrient composition were tested. U. pertusa was collected from an eelgrass bed in a semi-protected embayment on the southwest coast of Korea. Under salinity regimes from 5 to 40 psu, the specific growth rates $(\mu)$ of U. pertusa ranged from 0.019 to $0.032\;d^{-1}$. Maximum growth rate was observed at 20 psu, and minimum at 40 psu. This species showed various uptake rates for nitrate and phosphate. Nutrient uptake was noticeably higher at intermediate salinity levels, and lower at both extremes. Salinity significantly influenced chlorophyll-$\alpha$ content and effective quantum yield. Tissue nitrogen content ranged from 1.5 to 2.9% N (dry weight), whereas tissue phosphorus ranged from 0.1 to 0.14% P (dry weight). The N : P ratio in the tissue of U. pertusa was considerably higher, ranging from 30 to 50. Increased growth at lower salinity suggests that the initial growth rate of U. pertusa is greater during the rainy season (i.e., late spring and early summer) than any other season during the year. The appearance of an Ulva bloom in eelgrass beds may be triggered by salinity more than by other environmental factors such as light and temperature.

  • Application of the relationship $da/dN=C({\Delta}K)^{m}$ is effective in the analysis of fatigue crack growth life. The values of material constant C and m have great influences on the predicted fatigue life and the relationship between fatigue crack growth rate(da/dN) and stress intensity factor range(${\Delta}K$) is effective in fatigue crack growth behavior. In this paper, fatigue crack growth behavior of the welded joints in HT60 grade TMCP(Thermo Machanical Control Process) steel have been studied. To evalute the fatigue crack growth rates of HT60 grade TMCP steel, fatigue test was performed by base metal(BM), heat affected zone(HAZ) and weld metal(WM) in TMCP steel at room temperature. We determined the relationship of $da/dN-{\Delta}K$ by correlation between C and m obtained from the Paris-Erdogan power law data supplied HT60 grade TMCP steel. The obtained results from this study indicate that fatigue crack growth rate of TMCP steel is not influenced by softening effect which occurs in the HAZ when high heat input weld is carried out. Softening effects, which affect fatigue properties. are shown that it is not affected to the fatigue growth rates significantly.

Effect of Low Adapted Temperature and Medium Composition on Growth and Erythropoietin (EPO) Production by Chinese Hamster Ovary Cells

  • Temperature and medium composition were changed with the aim of increasing growth and erythropoietin (EPO) production in EPO-producing Chinese hamster ovary (CHO) cells. We used the CHO cell line, IBE, and its derivative, CO5, which over-expresses the first two enzymes of the urea cycle, carbamoyl phosphate synthetase I (CPS I) and ornithine transcar-bamoylase (OTC). When supplements were added to the medium at $33\;^{\circ}C$, the growth of IBE and CO5 cells increased by $27\%\;and;26\%$, respectively and the maximum yield of EPO was increased by $40\%$ in both cell lines. The absolute EPO concentration in the CO5 cells was always $55{\sim}60\%$ higher than in the IBE cells. In addition, when the two cell lines were continuously cultured with supplements at $33\;^{\circ}C$ until their growth rates approached those at $37\;^{\circ}C$, the growth rates of both IBE and CO5 cells increased by $54\%$ and their maximum EPO levels increased by up to $73\%\;and\;56\%$, respectively. Therefore, the growth and EPO expression levels of CO5 cells increased 2.2-fold and 2.6-fold, respectively, compared to those of the IBE cells. These results indicate that adaptation to lower temperature as well as medium supplementation could be important for improving cell growth and EPO production.

Effects of Nitrogen Fertilizer on Growth of Indigofera pseudo-tinctoria in Kyongseodong Waste Landfill, Incheon

  • Effects of nitrogen addition on the growth of Indigofera pseudo-tinctoria (Leguminosae) in the waste landfill site was investigated. Nitrogen fertilization in the nitrogen poor soils of waste landfill may influence the growth of nitrogen fixing plants beneficially or detrimentally. When I. pseudo-tinctoria was fertilized with three different levels of nitrogen, the coverage of plants treated with 46 g N/$m^2$ and 460 g N/$m^2$ was significantly less than that of plants treated with 23 g N/$m^2$. The growth rates of plant height treated with 46 g N/$m^2$ and 460 g N/$m^2$ were significantly less than those of plants treated with 23 g N/$m^2$. The growth rates of plant diameter treated with 46 g N/$m^2$ and 460 g N/$m^2$ were significantly less than those of plants treated with 23 g N/$m^2$. Dry weights of whole plants in control sites were higher than those of all the others nitrogen treatment sites. Nodule numbers were higher in control plants than those of plants in all the other nitrogen treatment sites. It is suggested that nitrogen fertilizer addition over 23 g N/$m^2$ affect the growth of some nitrogen fixing plants, such as I. pseudo-tinctoria, negatively.

Effect of Different Seeding Rates on Seed Productivity of the "Kowinearly" Cultivar of Italian ryegrass (Lolium multiflorum Lam.) in Paddy Field

  • The present study investigated the effects of different seeding rates on growth characteristics and seed productivity of the "Kowinearly" cultivar of Italian ryegrass (Lolium multiflorum Lam.) in paddy fields. Sowing Kowinearly in paddy fields under growing rice at the rates of 20, 30, 40, and 50 kg/ha resulted in seed yields of 1.57, 1.92, 2.06, and 2.09 ton/ha, respectively. Seed yield of Kowinearly was the highest at a seeding rate of 50 kg/ha (p<0.05), at which the cultivar was able to survive in winter and the weed ratio was low. Under these conditions, most growth characteristics such as winter survival (85%), weed ratio (10%), stems per square meter (1,006) were superior than those sown at other seeding rates (p<0.05). In addition, it has been reported that the economic efficiency of the 50 kg/ha seeding rate was higher than that of the other seeding rates.
