Factors Affecting the Production of In Vitro Plants from the Nodal Pieces of Chinese Yam (Dioscorea Opposita Thunb)

  • This study was carried out to establish The regeneration of healthy seedlings from the nodal segment culture of Chinese yam (Dioscorea opposita cv. Danma), cultivated in Korea. Different explants such as leaves, petioles, roots and nodal pieces, excised from the in vitro grown seedlings of Chinese yam, were cultured on MS medium supplemented with various combinations of growth regulators. All the growth regulators used induced plantlet regeneration from the nodal segments at a high frequency, while there was no induction of shoot or callus from leaf, petiole or root tissues. The medium supplemented with 0.01mg/L NAA, 0.5mg/L BA, 0.5-1.0mg/L kinetin and without plant growth regulator was effective for shoot development of buds from the nodal segment culture. The concentration of BA and NAA was an important factor in the bud induction of buds from the nodal segments of Chinese yam. Nodal segments cultured on the medium containing 1.0mg/L NAA and 0.5-1.0mg/L BA gave the best response to bud formation. The addition of GA$_3$ to the culture medium suppressed shoot induction and growth, while it increased microtuber formation. The shoot growth and microtuber formation were also affected by medium strength and solidity. The MS basal medium containing 1 g/L gelrite was suitable for microtuber formation from the nodal segment of Chinese yam.

중금속 오염배지에서 식물성장증진 근권미생물에 의한 식용 피 발아율과 유식물 성장 증진 (Improved Germination and Seedling Growth of Echinochloa crus-galli var. frumentacea in Heavy Metal Contaminated Medium by Inoculation of a multiple-Plant Growth Promoting Rhizobacterium (m-PGPR))

  • Positive effect of multiple-PGPR (Plan Growth Promoting Rhizobacteria), isolated from heavy metal contaminated soil, on the germination of Barnyard grass (Echinochloa crus-galli var. frumentacea) was quantitatively estimated in 5 heavy metal (Cd, As, Ni, Cu, and Pb) contaminated liquid medium. The $EC_{50}$ value for respective heavy metal was estimated by TSK (Trimmed Speraman-Karber) model based on germination rate. The results showed overall increase in $EC_{50}$ with PGPR inoculation. The $EC_{50}$ value increased 1.4% from 96.0 mg/L (control) to 97.4 mg/L (PGPR-treated) in As contaminated medium. In Ni contaminated medium, the $EC_{50}$ value increased 31.9% from 148.0 mg/L (control) to 195.2 mg/L (PGPR-treated), while the $EC_{50}$ showed 4.8% increase from 63.4 mg/L (control) to 66.5 mg/L (PGPR-treated) in Cu medium. Overall seedling growth was stronger in the PGPR treated seeds than that in the control, but positive effect on seedling growth was not conspicuous. At effective concentration of 100 mg/L, the average seedling length of the PGPR treatment in As, Cd, Cu, and Ni medium, respectively, was 1.13, 0.14, 0.40, and 0.06 cm longer than that in the control. However, the increase of seedling growth was statistically insignificant (p < 0.05). These results suggest that inoculation of the isolated-PGPR exerts positive effects on seed germination by reducing heavy metal toxicity and can be an effective tool for application of phytoremediation on heavy metal contaminated soils.

Suspension Culture of Gardenia jasminoides Ellis Cell for Production of Yellow Pigment

  • Gardenia callus was induced in MS medium containing $10{\;}{\mu}M$ of 2,4 diphenoxy acetic acid (2,4-D), $1{\;}{\mu}M$ kinetin, and 3% sucrose in the dark. $B_5$ medium was identified to be the most adequate medium for cell growth. Indole-3-acetic acid (IAA) was better growth regulator than 2,4-D not only for cell growth but slso for carotenoid production. Ligt also played a critical role on synthesis of carotenoid. Gardenia cells grown in $B_5$ medium could utilize a polysaccharide, soluble starch, as a carbon source. The cell growth was stimulated in $B_5$ medium fortified with 0.2% yeast extract. The optimum pH for cell growth was 5.7. High density cultures can be maintained by increasing inoculum size and medium concentration accordingly. Specific growth rate and mass doubling time were 0.095 $day^{-1}$ and 7.3 days, respectively. The cell immobilized in alginate tends to formulate more enlarged vacuoles containing yellow pigment compared with those of suspended cell. Carotenoid content of immobilized cell was about $264.4{\;}{\mu}g/g$ fresh weight (F.W.) corresponding twice of the content of suspended cell ($112.08{\;}{\mu}g/g$ F.W.). The color of gardenia cell was shifted from yellow to red when carbohydrase-secreting fungus, Trichoderma reesei, was co-cultivated with gardenia cells.

  • 본 연구는 한국 중소기업의 수출 증대와 고용 증대에 양(+)의 영향을 미치는 요인들에 대한 연구 흐름의 깊이를 심화시키기 위해 외부 기술 연구개발 협력 네트워크의 다양성이 중소기업의 수출 증대와 고용 증대에 미치는 영향을 연구 모형화 하였으며, 대한민국의 2,200개의 중소기업으로부터 수집된 데이터를 사용하여 연구 모형을 실증적으로 검증 하였다. 본 연구는 이러한 실증 검증 결과를 통해 다음과 같은 2개의 연구 결과를 조명한다. 첫째, 중소기업의 외부 기술 연구개발 협력 네트워크의 다양성은 기술 개발로 인한 고용 증대에 유의한 양(+)의 영향을 미친다. 둘째, 중소기업의 기술 개발로 인한 수출 증대는 기술 개발로 인한 고용 증대가 외부 기술 연구개발 협력 네트워크의 다양성에 의해 받는 양(+)의 영향을 부분 매개한다.

Interaction Between time of Nodal Explant Collection and Growth Regulators Determines the Efficiency of Morus alba Micropropagation

  • The hormonal requirement suiting micropropagation of Morus alba during any season throughout the year was studied. Sprouting frequency from axillary buds of M. alba was greatly influenced by the time of explant collection, the highest value was achieved when nodal explants were collected at the end of bud dormancy period (late in March) and cultured on Murashige and Skoog (MS) medium supplemented with low concentration (0.5 mg/L) of BAP, kinetin or IBA (85-68%). In addition, they showed higher axillary bud sprouting on growth-regulators-free medium (49%) than others collected in autumn or winter and cultured on medium supplemented with various growth regulators (47-48%). Regardless of that period, young explants with greenish buds collected in summer exhibiting high sprouting frequency (66%) on MS medium supplemented with 0.5 mg/L kinetin and 0.5 mg/L GA3. Shoot multiplication via adventitious bud formation was achieved when the nodal explants were cultured on MS medium supplemented with 2 mg/L BAP and 0.2 mg/L IBA. Further multiplication via nodal explants of in vitro grown shoots was obtained on MS medium supplemented with 0.5 mglL BAP and 0.5 mg/L GA3. While half strength MS medium supplemented with low concentration (0.5 mg/L) of IBA, IAA or 2,4-D stimulated adventitious root formation, IBA was the best. After transfer the plantlets to the soil, acclimatization for three weeks was essential prerequisite for survival in high frequency (92%). Peroxidase activity is related to break of bud dormancy where maximum enzyme activity was detected when the lateral buds were induced to commence growth under field condition (early in spring) or in vitro.

하늘타리 형질전환근의 생장 및 Trichosanthin의 생합성을 위한 최적화 (Optimal Culture Conditions for Transformed Root Growth and Trichosanthin Formation in Trichosanthes kirilowii Max.)

  • 하늘타리 (Trichosanthes kirilowii)로부터 조직배양 기법을 이용하여 약리작용을 나타내는 생리활성물질인 trichosantin을 생산하기 위한 연구로 Agrobacterium rhizogenes ATCC15834 의 감염을 통하여 기내 배양된 유묘의 잎절편으로부터 형질전환 모상근을 유도하였다. 유도된 모상근 clones으로부터 성장속도와 분지화 정도가 빠르며 단백질 합성능이 가장 우수한 TR-03 clones을 선발하였으며, 이를 사용하여 생체량과 배지내 가용성 단백질의 생산을 최대로 높일 수 있는 최적 배양조건을 조사하였다. TR-03 clone을 4% sucrose가 첨가된 MS 배지에 초기 접종농도 2 g (생중량)을 접종하여 100 rpm으로 배양시 생중량과 배지내 가용성 총단백질의 함량이 플라스크당 약 2.4 g (건중량)과 $28.3ug/\ell$으로 최대치를 나타내었다. 한편, 배지내 가용성 단백질에 대한 RIP활성 검정에서 배양 4주 후 21.3 unit로 최대치를 보였다.

  • We researched effects of growth on initial inoculum size, liquid volume, and medium feeding rate etc. Cell suspension inoculated at low cell concentrations showed a typical growth reduction, whereas root cultures showed an improvement in growth. In this paper, Hairy roots showed high growth rate at 0.4 % inoculum size and 100 mL liquid volume in 250 mL flask cultures.

Production of Antihypertensive Angiotensin I-Converting Enzyme Inhibitor-Enriched Edible Yeast Using Gugija (Lycium chinesis Mill)

  • To produce bioactive compound enriched yeast using medicinal Gugiga (Lycium chinensis Mill), several edible Saccharomyces species were cultured in Gugija extracts added yeast extract, peptone and dextrose medium (GE - YEPD medium) at $30^{\circ}C$ for 24 hr, and their growth were determined. Growth of Saccharomyces cerevisiae K-7 and Sacchromyces cerevisiae ACTC 7904 were better than those of the other yeasts. Two yeasts were selected and then determined their some physiological functionalities after cultivated the yeasts in the GE - YEPD medium and compared those grown on YEPD medium. Antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of S. cerevisiae K-7 grown on GE - YEPD medium was about 20% higher than that grown on YEPD medium. Superoxide dismutase-like activity of S. cerevisiae ACTC 7904 was also about 12% more high. However, the other physiological functionalities were almost same or lower. Optimal addition concentration of Gugija extract was 10%, and maximally growth and ACE inhibitory activity of S. cerevisiae K-7 were shown when the strain was cultured in 10% Gugija extracts containing YEPD medium at $30^{\circ}C$ for 12 hr.

Effects of Varying Nutritional and Cultural Conditions on Growth of the Ectomycorrhizal Fungus Pisolithus tinctorius SMF

  • The culture conditions and nutritional requirements for enhanced mycelial growth of the ectomycorrhizal fungus P. tinctorius SMF were determined in flask scale experiments. Optimum culture conditions for growth of P. tinctorius SMF in a further modified Melin-Norkrans broth were as follows; temperature 25~$27^{\circ}C$, agitation 120 rpm, and pH 4.0. P. tinctorius SMF utilized various carbon sources including monosaccharides, disaccharides, and polysaccharides. D-Glucose and mannitol were respectively the first and second most suitable carbon sources for mycelial growth. With D-Glucose as the principal carbon source, supplementation of modified Melin-Norkrans (MMN) broth with Lysine (800 mg/l), Glutamic Acid (500 mg/l), or Proline (50 mg/l) enhanced mycelial yields 63%, 34%, and 22% respectively as compared to growth in medium lacking amino acids. ThiaminㆍHCl+biotin+pyridoxine supplementation also enhanced growth. As compared to mycelial growth in the MMN medium, growth of P. tinctorius SMF was enhanced 120% in MMN broth when the carbon/nitrogen ratio was 25/1 in citrate buffer at pH 4.5, and growth was 50% greater in MMN broth of carbon/nitrogen ratio with a 10/1~20/1 without using the buffer. Standard conditions established for growth of P. tinctorius SMF in MMN broth were 25~$27^{\circ}C$, agitation 120 rpm, buffered to pH 4.0 with citrate, in MMN medium containing 10 g/l D-glucose supplemented with 800 mg/l lysine. In this medium the carbon/nitrogen ratio was 20/1~25/1, and the maximal mycelial yield ($Y_{x/s}$ ) was 0.472 (4.72 mg/ml) after 7 days of incubation, as compared to 0.214 (2.14 mg/ml), when the fungus was grown in standard MMN broth.

환경조건에 따른 Aphanizomenon flos-aquae (Cyanophyceae) 균주의 성장 반응 및 독소 생성 (Response of Growth and Toxigenicity to Varying Temperature and Nutrient Conditions in Aphanizomenon flos-aquae (Cyanophyceae))

  • The purpose of this study is to investigate growth response and toxigenicity under various temperature and nutritional conditions, in order to understand the physioecological characteristics of Aphanizomenon flos-aquae, which is a bloom-forming cyanobacterium in the Nakdong River. The strain was inoculated into media under combinations of four temperatures (4, 12, 21, $30^{\circ}C$) and three nutrients (modified CB medium, P-depleted CB medium, N-depleted CB medium) for 28 days. The algae-inhibition tests were performed to assess the potential allelopathic effects of the strains' filtrates on the growth of four algae strains (Microcystis aeruginosa, Aulacoseria ambigua f. spiralis, Aphanizomenon flos-aquae, Scenedesmus obliquus). Toxin production of a strain was measured by Enzyme-Linked ImmunoSolbent Assay (ELISA). The optimal growth temperature (Topt) of strains was $19.9^{\circ}C$ ($18.3-21.2^{\circ}C$), and the temperature range for growth was from $-0.3^{\circ}C$ to $34.3^{\circ}C$. Specific growth rate (${\mu}$) in modified CB medium varied from 0.10 to $0.16day^{-1}$, and the maximum growth rate (${\mu}_{max}$) was $0.17day^{-1}$. Although growth curves under N-existed and N-depleted conditions were almost the same, growth under N-depleted condition was relatively slowed (${\mu}=0.09$ to $0.14day^{-1}$), with a decreased maximum cell density. However, growth under the P-depleted condition was restricted for all temperatures, Two stains of Aphanizomenon flos-aquae were confirmed as not producing toxins, because saxitoxin and cylindrospermopsin were not detected by ELISA. The exudates or filtrates from the Aphanizomenon flos-aquae (DGUC003) resulted in significant inhibition of algal growth on the Aulacoseira ambigua f. spiralis (DGUD001) and Aphanizomenon flos-aquae (DGUC001) (p < 0.01).
