Bacteriological Characteristics of Unidentified Vibrio sp., Hemolysin Producer Isolated from Brackish Water -2. Bacteriological Characteristics of Vibrio sp. E10 Similar to Vibrio mimicus- (기수에서 분리된 용혈독소를 생산하는 미분류 Vibrio sp.의 세균학적 특징 -2. Vibrio mimicus와 유사한 Vibrio sp. E10의 세균학적 특성-)

  • A hemolysin producing bacterial strain which belong to Vibrio species was isolated from the Kum River estuary. In the process of identification, the strain did not show characteristics of known Vibrio species; thus, the strain was designated as Vibrio sp, E10 (V. kunsan) tentatively and further identification study was carried out by comparing its bacteriological characteristics. Morphologically Vibrio sp, E10 was comma shaped rod with a polar flagellium. Clear hemolysis zones were observed with the strain against human and sheep blood agar. Hemollytic toxicity was confirmed by strong vascular Permeability and fatal toxicity against mouse was also observed. Therefore the strain was a pathogenic vibrio. Growth conditions for Vibrio sp. E10 were ranged salinity of 0$\~$$4.5\%$, pH of 6.2$\~$9.2, temperature of 14$\~$42$^{\circ}C$, respectively, 16S rDNA partial sequence of Vibrio sp, E10 showed $99\%$ homology with dozens of V. cholerae species including V, cholerae El Tor N16961 and V, snmisnfus ATCC 33653T. This strain belonged to Proteobacteria; gamma subdivision; Vibrionacea: Vibrio. But, among knorn Vibrio species no identical styains were found when using automatic bacteria identification system ($MicroLog^{TM}$system, release 4.0, Biolog Inc., USA) which evaluated the ability of metabolizing 95 kinds of carbon and nitrogen sources. Vibrio sp, E10 showed 18 and 11 different responses as compared to V. mimicus and V, cholerae, respectively.

Identification and Characterization of Paraconiothyrium brasiliense from Garden Plant Pachysandra terminalis (가든식물 수호초(Pachysandra terminalis)로부터 Paraconiothyrium brasiliense의 분리 및 동정)

  • A fungal isolate DUCC5000 from a garden plant Pachysandra terminalis was identified as Paraconiothyrium brasiliense based on the results of morphological and molecular studies. The fungus formed brown to black conidiomata of (0.2-0.7)-2(-3.5) mm singly or as a group on PDA. Conidia measured $2-5{\times}1.8-3{\mu}m$ in size, hyaline, ellipsoid to short-cylindrical, and rounded at both ends. The internal transcribed spacer (ITS) DNA of the isolate shared 100% nucleotide sequence homology with those of known P. brasiliense isolates. Phylogenetic tree inferred from the ITS sequence analysis showed that the DUCC5000 isolate formed a clade with known isolates of P. brasiliense. The fungal mycelia grew better on oatmeal agar than on MEA and PDA. On PDA media under various pH conditions, fungal mycelial growth was observed at pH 9. Colony morphology of the fungus tended to alter depending on the kinds of nutrient media and pH condition. On chromagenic media, the fungus demonstrated its ability to produce extracellular enzymes including amyalse, avicelase, ${\beta}$-glucosidase, protease, and xylanase. However, in pathogenicity testing, no disease symptoms were observed on the leaves of P. terminalis. This strain is the first report on P. terminalis in Korea.

Development of Efficient Screening Method for Resistant Cabbage and Broccoli to Plasmodiophora brassicae (양배추 및 브로콜리 뿌리혹병에 대한 효율적인 저항성 검정 방법 확립)

  • Clubroot caused by Plasmodiophora brassicae Woron. is one of the most important diseases in Brassica crops worldwide. To establish more simple and reliable screening method for resistant cabbage and broccoli to P. brassicae, the development of clubroot on the plants according to inoculum concentration and incubation period after inoculating with the pathogen was investigated using P. brassicae GN1 isolate (race 9). To facilitate and acquire precise result of resistance screening of cabbage and broccoli to clubroot, 14-day-old seedlings were inoculated by drenching roots with the spore suspension of P. brassicae to give inoculum density of $2.5{\times}10^9$ spores/pot. To develop the disease, the inoculated seedlings were incubated in a growth chamber at $20^{\circ}C$ for 3 days, and then cultivated in a greenhouse ($20{\pm}5^{\circ}C$) for five weeks. Under the optimum conditions, 16 cabbage and 17 broccoli cultivars were tested for resistance to four field isolates (GN1, GN2, GS and YC) of P. brassicae collected from four regions in Korea. Among them, some cabbage and broccoli cultivars showed different resistance response to three isolates (GN1, GN2 and GS) determined as race 9 by using the differential varieties of Williams. On the other hand, all the tested cultivars were highly susceptible to YC isolate (race 2). The results suggest that this method is efficient screening method of cabbage and broccoli for resistance to P. brassicae.

Development of an Efficient Bioassay Method for Testing Resistance to Bacterial Soft Rot of Radish (효과적인 무 무름병 저항성 검정법 개발)

  • Pectobacterium carotovorum subsp. carotovorum (Pcc) causes bacterial soft rot on a wide range of crops worldwide, especially in countries with warm and humid climates. This study was conducted to establish an efficient screening method for resistant cultivars of radish (Raphanus sativus) to bacterial soft rot. Resistance degrees of 60 commercial radish cultivars to the Pcc KACC 10421 isolate were investigated. For further study, six radish cultivars (Awooriwoldong, YR Championyeolmu, Jeonmuhumu, Bitgoeunyeolmu, Sunbongaltari, Housecheongok) showing different level of resistance to the bacterium were selected. The development of bacterial soft rot on the cultivars was tested according to several conditions such as incubation temperature, seedling stage of radish, inoculum concentration to develop the disease. On the basis of the results, we suggest that an efficient screening method for resistant radish to Pcc is to inoculate twenty-day-old seedlings with a bacterial suspension of Pcc at a concentration of $8{\times}10^5cfu/ml$ and then to cultivate the plants in a growth room at $25^{\circ}C$ and 80% RH with 12-hour light per day.

  • The streaming potential generated by the electrokinetic flow within electric double layer of charged microchannel is applied to determine the zeta potential of hollow-fiber membrane pore by using the general Helmholtz-Smoluchowski equation. The streaming potential is know to provide a useful real-time information on the surface property and the interaction between pore and particles in actual situations and physicochemical conditions. The influence of physicochemical parameters upon the filtration with hollow-fibers has been examined with an in-situ and simultaneously monitoring the streaming potential as well as permeate flux. In particular, the present study examined an experimental method to identify the effect of cake layer which can vary according to the axial position of a hollow-fiber and the progress of membrane fouling by measuring the position-dependent streaming potential. As the latex concentration increases, the permeate flux decreased but the streaming potential increased. The growth of cake layer has been mire developed with increasing latex concentration, however, the effect of surface charges of latexes deposited on the membrane surface leads to increase the streaming potential. With increasing ionic concentration of KCI, both the permeate flux and the streaming potential decrease. The increase of ionic concentration provides a compact cake layer due to the shrinkage of Debye length and the decreased streaming potential results from the weakened ionic flows owing to a thin diffusive double layer.

  • This study was conducted to investigate the effect of oxadiazon, molinate, thiobencarb before seeding on control of red rice and barnyardgrass in water-seeded rice. High application rate plot among oxadiazon treatment plots could observe phytotoxicity symptoms depending on field conditions, but these injury recovered gradually with time. Molinate and thiobencarb application plots at the concentration of 225~400, 210~420g ai/l0a respectively were not observed phytotoxicity. Control of red rice was different according to kinds of herbicides and application rates. Oxadiazon showed higher control performance at the concentration of more than 60g ai/10a. Control effect of molinate and thiobencarb against red rice was enhanced with the increase of application rate, and both herbicides showed satisfactory effect at more than 300g ai/10a. Control of barnyardgrass showed up to 90~100% in all tested herbicides. There was no significant yield reduction by oxadiazon, molinate, and thiobencarb application before seeding in all tested field. In the pot experiment, crop injury, seedling stand, and early growth were more advantageous at time of drainge after one day after seeding than flooding until rooting.

Isolation and Optimal Culture Conditions of Brevibacillus sp. KMU-391 against Black Root Pathogens Caused by Didymella bryoniae (덩굴마름병원균인 Didymella bryoniae의 생물학적 방제를 위한 길항세균의 분리 및 특성)

  • We isolated a bacterium which produces antifungal substances from the Sanktpeterburg soils at Russia. The iso-lated strain was identified as Brevibacillus sp. and shown a strong antifungal activity on plant pathogenic fungi. Brevibacillus sp. KMU-391 produced maximum level of antifungal substances under incubation aerobically at $30^{\circ}C$ for 48 hours in trypticase soybean broth containing 1.0% sucrose and 1.0% polypeptone at 180 rpm and initiated pH adjusted to 7.0. Precipitate of culture broth by $30{\sim}60%$ ammonium sulfate precipitation exhibited strong antifungal activity against Didymella bryoniae by dry cell weight. Butanol extract of cultured broth also shown fungal growth inhibitory activity against Botrytis cinerea KACC 40573, Botrytis fabae KACC 40962, Colletotrichum gloeosporioides KACC 40804, Colletotrichum orbiculare KACC 40808, Didymella bryoniae KACC 40669, Fusarium graminearum KACC 41040, Fusarium oxysporum KACC 40037, Fusarium oxysporum KACC 40052, Fusarium oxysporum f, sp. radicis-Iycopersici KACC 40537, Fusarium oxysporum KACC 40902, Monosporascus cannonballus KACC 40940, Phytophthora camvibora KACC 40160, Rhizoctonia solani AG-1(IA) KACC 40101, Rhizoctonia solani AG-4 KACC 40142, and Scleotinia scleotiorum KACC 41065 by agar diffusion method.

Studies on the Biological Treatment of Waste Water from Acetaldehyde Plant (아세트 알데히드(특수산업) 폐수의 생물학적 처리)

  • In order to establish the biological treatment system which can be used for treatment of waste aster from acetaldehyde plant, it was investigated optimum nutrient requirements and growth conditions by mixed culture of Micrococcus roseus AW-6, Micrococcus luteus AW-22, Microbacterium lacticum AW-38 and Microbacterium laevaniformans AW-41 as well as the effect of coagulants and neutralization reagents. Also, it was carried out the continuous culture as well as batch culture to treat the waste water by mixed culture of these strains. The COD removal rate was reached to maximum state for 96hrs culture at pH7.0 and $30^{\circ}C$ NaOH as the neutralization reagents was the most effective, but the coagulants had no effect on the COD remonal rate and the optimum dilution times for treatment were 10 fold. The COD removal rate was also increased by supplimenting 200 ppm $NH_{2}NO_{3}$, 50 ppm $KH_{2}PO_{4}$, 15 ppm $CaCl_{2}$ and 1 ppm $MgSO_{4} \cdot 7H_{2}O $ as additional nutrients. The removal rate coefficient $K_{1}$ on the batch culture was $4.5\times 10^{-6}$, and the detention time for BOD removal rate of 85% was approximately 45hrs. The COD of waste water was reduced to 15% of its initial value by the continuous culture. The COD and BOD of the effluents were to be about 60 ppm and 40 ppm, respectively, and final pH was 7.0.

Synthesis and Conductive Properties of Li1+xAlxTi2-x(PO4)3 (x = 0, 0.3, 0.5) by Sol-Gel Method (Sol-Gel법에 의한 Li1+xAlxTi2-x(PO4)3 (x = 0, 0.3, 0.5)의 합성 및 전도특성)

  • $Li_{1+x}Al_xTi_{2-x}(PO_4)_3$(LATP) is a promising solid electrolyte for all-solid-state Li ion batteries. In this study, LATP is prepared through a sol-gel method using relatively the inexpensive reagents $TiCl_4$. The thermal behavior, structural characteristics, fractured surface morphology, ion conductivity, and activation energy of the LATP sintered bodies are investigated by TG-DTA, X-ray diffraction, FE-SEM, and by an impedance method. A gelation powder was calcined at $500^{\circ}C$. A single crystalline phase of the $LiTi_2(PO_4)_3$(LTP) system was obtained at a calcination temperature above $650^{\circ}C$. The obtained powder was pelletized and sintered at $900^{\circ}C$ and $1000^{\circ}C$. The LTP sintered at $900{\sim}1000^{\circ}C$ for 6 h had a relatively low apparent density of 75~80%. The LATP(x = 0.3) pellet sintered at $900^{\circ}C$ for 6 h was denser than those sintered under other conditions and showed the highest ion conductivity of $4.50{\times}10^{-5}$ S/cm at room temperature. However, the ion conductivity of LATP (x = 0.3) sintered at $1000^{\circ}C$ decreased to $1.81{\times}10^{-5}$ S/cm, leading to Li volatilization and abnormal grain growth. For LATP sintered at $900^{\circ}C$ for 6 h, x = 0.3 shows the lowest activation energy of 0.42 eV in the temperature range of room temperature to $300^{\circ}C$.

'Asian Flame' an Asiatic Hybrid Lily with Unspotted Red Petals (무반점 적색 아시아틱 나리 '아시안플레임' 육성)

  • An Asiatic lily cultivar 'Asian Flame' was bred in 2007 at National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA) of Korea. The crossing was conducted between female parent A94-34 ('Montreux' ${\times}$ 'Anabelle') and male parent 'Fire' by stigmatic pollination at Suwon in 1996. First selection was done and tentatively named as A98-98 in 1998. After in vitro multiplication and bulb production of A98-98 line, growth and flowering characteristic tests has been conducted from 2005 to 2007. The evaluation of characteristics and preference were surveyed at a lily flower show of NIHHS in 2007 in Taean. The new cultivar 'Asian Flame' flowers in middle of June and grows to an average of 122.7 cm in height. Flowers bloom upward-facing, red (RHS 45B) petals. Year-round flowering is possible by storage of the bulb under $-1.5^{\circ}C$ conditions. For forcing culture, it is necessary to add calcium to the fertilizer or remove side scales of large bulb to prevent leaf scorch.
