Effect of Feeding Basal Diet Supplemented with Mugwort Powder on the Serum Components in Growing Rat (쑥 첨가급식이 성장기 흰쥐의 혈청 성분에 미치는 영향)

  • This study was designed to observe the effects of edible mugwort and medicinal mugwort ( Ganghwa medicinal mugwort) on blood components of rats. Eighteen young rats of Sprague Dawley strain that weighted 84${\pm}$9g, were used in this study. They were fed on the basal diet(control diet). 5% edible mugwort powder(EM diet) and 5% medicinal mugwort(MM diet) for 4 weeks respectively. In proximate composition of mugworts, crude protein and crude ash contents of EM were 2% higher than those of MM. Mn, Zn and vitamin A contents of EM were 3 times higher than those of MM, but in Ca content, MM was 5 times higher than that of EM. The contents of albumin and the activity of ALT, AP amylase and CK in serum were significantly increased, but the activity of AST was significantly lowered in EM and MM diet groups. The contents of TG and the activity of LDH were towered in EM diet group and MM diet group resulted in decrease of glucose content and increase of TP content.

Effects of Artemisia iwayomogi Kitamura Ethanol Extract on Lowering Serum and Liver Lipids in Rats (흰쥐의 혈청 및 간지질 저하에 미치는 쑥 에탄올 추출물의 영향)

  • Artemisia iwayomogi(A. iwayomogi) is a major edible vegetable in Korea. A iwayomogi containing a strong bitter taste, particularly, is known to manifest an effect on cough, abdominal pain, indigestion and bleeding. In this experiment, ethanol extract of A. iwayomogi(50mg/kg body weight) was fed to growing male Sprague-Dawley rats for four weeks in order to examine the lipid lowering effect of A. iwayomogi in the live and serum. High fat diet included 10% lard, 1% cholesterol and 0.25% sodium cholate. The administration of A. iwayomogi extract decreased serum cholesterol compared with the control. At the same time, serum triglyceride(TG) was decreased to 25%. Although atherogenic index and HTR tended to decrease, phospholipid level in the serum was shown to increase by A. iwayomogi administration. At the same token, total liver lipids were decreased to 40%. The group fed both high fat and ethanol extract showed 28% decrease of liver lipids compared with the group fed only high fat diet. On the other hand, HDL cholesteol level of high fat and extract fed group was increased to 30% compared with the non-extract-fed control. Liver TG also was decreased to about 21% in the extract fed groups. Liver phospholipids, particularly, exceeded more than double of the level of high fat control. The above results indicate than A. iwayomogi exert a strong hypocholesterolemic and hypolipidemic effect which may lead to the protection from hypertention, obesity, stroke and many other circulatory diseases.

Study on Relation of Effects of Electro-acupuncture with Sex, Age, and Weight in Rats (흰쥐의 성별, 체중 및 주령에 따른 족삼리(足三里) 전침자극의 면화가 소장수송능(小腸輸送能)에 미치는 영향)

  • Electroacupuncture is the combined treatment of mechanical and electric stimuli on the needle head, on which electricity runs after acupuncturing at the acupuncture points. It is reported that its treatment effect depends on the parameter of stimulus, which is wave of stimulus, frequency, voltage, and duration of electric current. To get the effective treatment, stimulus intensity over threshold is needed. We planned to experiment whether acupuncture effect is affected by sex, age, and weight. After we conducted electric stimulus at Zusanli(ST 36) which is known to increase small intestinal motility in normal rats, we observed the effects and got the following results. Though the increased effects of small intestinal motility of electro-acupuncture at Zusanli(ST 36) did not show the difference of sex, this effect was observed to have a tendency to decrease according to weight increase without distinction of sex. But the effect of electro-acupuncture shown in 4, 5, 7 weeks old without distinction of sex was observed to have a tendency to decrease with the growing of weeks-old in the male rat group. And the significant change without specific tendency was observed in the 4, 5 weeks old female rat group. This result suggested that sex, weight, and age should be considered to get the efficient electro-acupuncture. The more experimental study should be advanced to investigate the correlation between the factors that can affect the acupuncture treatment, and, afterward, the interconnection into the clinical use.

Effect of Hydroalcoholic Extract of Ribes khorasanicum on Acute Hypertension Induced by L-NAME in Rat

  • Objectives: The aim of this study was to evaluate the effect of Ribes khorasanicum (R. khorasanicum); a plant growing in north Khorasan of Iran; on cardiovascular and stress oxidative in acute hypertension induced by N-nitro-l-arginine methyl ester (L-NAME), anitric oxide synthase inhibitor. Methods: Rats were divided into Control, L-NAME (10 mg/kg), Sodium Nitroprusside (SNP) (50 mg/kg) + L-NAME and three treated groups with R. khorasanicum (4, 12 and 24 mg/kg) groups + L-NAME. L-NAME and SNP were injected intravenously and extract intraperitoneal. In R. khorasanicum groups, L-NAME was injected 30 min after injection of the extract. Systolic blood pressure (SBP), mean arterial pressure (MAP) and heart rate (HR) were recorded continuously using power lab software. At the end of study oxidative stress parameters including of total thiol content (SH), malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) in heart and aorta of all groups were also measured. Results: In groups 4 and 24 mg/kg extract +L-NAME, there was a non-significant decrease in SBP and MAP compared to L-NAME group but dose 12 mg/kg significantly attenuate the effect of L-NAME(P < 0.05). In L-NAME group the heart and aorta tissues antioxidant enzymes levels decreased, while in treated rats these enzymes significantly increased. Conclusion: The extract of R. khorasanicum in dose 12 mg/kg show anti-hypertensive effect that is mediated by an effect on NO system or antioxidant parameters.

Effect of Methanol Extract from Cassia mimosoides var. nomame on Ischemia/Reperfusion-induced Renal Injury in Rats

  • Objectives : The purpose of this study was to determine whether the methanol extract of Cassia mimosoides var. nomame Makino, a naturally growing plant in Korea, could prevent the renal-ischemia/reperfusion injury in a rat model or not. Methods : The radical scavenging activities of the extracts, and ascorbic acid as a positive control, were measured in vitro. At one hour after an intraperitoneal injection of the extract (400 mg/kg), renal ischemia/reperfusion injury was generated by 40 min clamping of the left renal artery in rats. After renal ischemia/reperfusion and 24 hr restoration of blood circulation, the serum creatinine concentration was measured. And the extent of epithelial cell injury and apoptosis was assessed by various staining technologies. The Bax/Bcl-2 ratio and activated caspase-3 were assessed by immunohistochemistry. Results : The extract showed a slightly lower level of radical scavenging activity than that of ascorbic acid. Compared to those of the vehicle-treated group, the extract-treated group displayed a significantly smaller tubular epithelial cell injury of 54% reduction in the outer medulla region and a lower serum creatinine concentration of 50% reduction. It seems that the reduction in cellular injury is due to the attenuation of the Bax/Bcl-2 ratio, and the inhibition of caspase-3 activation by the extract of Cassia mimosoides. Conclusions : Cassia mimosoides var. nomame Makino could be a good candidate for a prophylactic agent against the ischemia/reperfusion/induced kidney injury.

Preliminary Study on Effect of Lactiplantibacillus plantarum on Osteoporosis in the Ovariectomized Rat

  • Osteoporosis is a growing global health concern primarily associated with decreased estrogen in postmenopausal women. Recently, some strains of probiotics were examined for potential anti-osteoporotic effects. This study intended to evaluate the impacts of Lactiplantibacillus plantarum MGE 3038 strain (MGE 3038) in ovariectomized rats. For this purpose, twelve weeks old female Wistar rats (n=21; 250-300 g) were divided into 3 groups; ovariectomy (OVX) group, OVX/MGE 3038 group and Sham group (control). In these groups; two went through respective OVX and one had daily MGE 3038 administration through oral gavage. Prior to 16 weeks after OVX, we collected blood samples and extracted the tibiae. We scanned the extracted tibiae by in-vivo micro-computed tomography (micro-CT) and evaluated pathology by hematoxylin and eosin (H&E) and Masson's trichrome staining. The serum levels of C-telopeptide of type I collagen (CTX), osteocalcin (OC), and the receptor activator of nuclear factor-κB ligand (RANKL) were examined. The OVX/MGE 3038 group showed increases in bone mineral density, trabecular bone volume, trabecular number, and trabecular thickness (Tb.Th), and a decrease in trabecular spacing than the OVX group. However, OVX/MGE 3038 group and control group were measurably comparable in Tb.Th. Micro-CT, H&E, and Masson's trichrome findings exhibited increased preservation and maintenance of trabecular bone structure in the OVX/MGE 3038 group in comparison to the OVX group. In serum, the levels of CTX, OC and RANKL were significantly different between the OVX and OVX/MGE 3038 groups. Taken together, L. plantarum MGE 3038 could be helpful for the treatment of osteoporosis.

A Review of Interactions between Dietary Fiber and the Gastrointestinal Microbiota and Their Consequences on Intestinal Phosphorus Metabolism in Growing Pigs

  • Dietary fiber is an inevitable component in pig diets. In non-ruminants, it may influence many physiological processes in the gastrointestinal tract (GIT) such as transit time as well as nutrient digestion and absorption. Moreover, dietary fiber is also the main substrate of intestinal bacteria. The bacterial community structure is largely susceptible to changes in the fiber content of a pig's diet. Indeed, bacterial composition in the lower GIT will adapt to the supply of high levels of dietary fiber by increased growth of bacteria with cellulolytic, pectinolytic and hemicellulolytic activities such as Ruminococcus spp., Bacteroides spp. and Clostridium spp. Furthermore, there is growing evidence for growth promotion of beneficial bacteria, such as lactobacilli and bifidobacteria, by certain types of dietary fiber in the small intestine of pigs. Studies in rats have shown that both phosphorus (P) and calcium (Ca) play an important role in the fermentative activity and growth of the intestinal microbiota. This can be attributed to the significance of P for the bacterial cell metabolism and to the buffering functions of Ca-phosphate in intestinal digesta. Moreover, under P deficient conditions, ruminal NDF degradation as well as VFA and bacterial ATP production are reduced. Similar studies in pigs are scarce but there is some evidence that dietary fiber may influence the ileal and fecal P digestibility as well as P disappearance in the large intestine, probably due to microbial P requirement for fermentation. On the other hand, fermentation of dietary fiber may improve the availability of minerals such as P and Ca which can be subsequently absorbed and/or utilized by the microbiota of the pig's large intestine.

Expression of mRNAs and Proteins of Cyclin A and LATS Genes in Ovary (Cyclin A와 LATS 유전자들의 난소 내 mRNA 및 단백질 발현에 관한 연구)

  • Despite of the importance of the primordial follicle (PMF) recruitment, factors and mechanisms for process are poorly understood. To evaluate expression and role of the follicular transition from PMF to PMF/primary follicles (PMIF) in the present study, we evaluated expression of lats1, lats2, cyclin A1, and cyclin A2 mRNA and protein, and elucidated and role of lats1-cyclin A in the follicular transition from PMF to PRIF. To analysis of differential expression in PMF and PMIF, each stage follicles were collected by day1 and day5 of immuno-compromised rats (ICR) and analyzed by real-time PCR for the genes. For localization of mRNAs and proteins of the genes, in situ hybridization and immunohistochemistry were performed. We confirmed that the lats1, lats2, cyclin A1, and cyclin A2 mRNA were more expressed in PMF than PMIF. Localization of the four genes expression were observed in nuclei of oocytes from the arrested primordial, and in the surrounding granulosa cells of the growing follicles. The mRNA expressions were gradually decreased with follicular development. From immunohistochemistry studies, Cyclin A1 protein expression were observed in oocyte cytoplasmas of early stage follicles, while observed in granulose cells and oocyte nucleoli during growing follicles. This study suggested that the presence of lats gene family might perform negatively regulation of cell proliferation by modulation of the CDC2/Cyclin A complex activity. lats-cyclin A genes in oocytes of the early stage follicles might play a role in the meiotic cell cycle arrest of the primary oocytes at the primordial follicle stage as well as the follicular growth.

Immunohistochemical study on the atretic and the growing follicles after experimental superovulation in rats I. Number of follicles by superovulation (과배란 유기된 rat 난소에 퇴축난포와 성장난포에 대한 면역조직화학적 연구 I. 동원된 난포수에 대하여)

  • This study was designed to investigate the number of the growing and mature follicles following gonadotrophin treatments for superovulation in mature rats. Eighteen mature rats (Sprague-Duwely, initially 190~230gm) were randomly alloted into 3 groups. One group was control group, another FSH-treated group was injected intramuscularly with 0.5 units of follicular stimulating hormone (FSH) / rat, and third PMS and HCG-treated group was intramuscularly injected with 20~25IU of pregnant mare serum (PMS) / rat and then at the 48 hrs later, with 20~25IU of human chorionic gonadotrophin (HCG) / rat. The uteri and ovaries of rats were collected and then were observed grossly and serial sections of paraffin embedding ovaries were stained with H-E. Number of ovarian follicles by following 3 grades of large, middle and small follicles from secondary and tertiary follicles were investigated by LM photography of preparations. Small follicles were classified as secondary follicles of preantral follicles with more than 2 layers of granulosa cells surrounding the oocyte and middle follicles were classified as secondary follicles with early signs of antral cavity or with more than one small cavity on either side of the oocytes and large follicles were classified as tertiary follicles with a single medium sized antral cavity or large well-formed antral cavity. In gross findings, the uteri were slightly swelling in FSH-treated group and markedly swelling or filled with fluid in the uterine lumen in PMS and HCG-treated group. In histological findings, the shape and size of the follicles were diverse in middle and large follicles of FSH-treated group and PMS and HCG-treated group, and proportion of atretic follicles was increased in FSH-treated group and PMS and HCG-treated group than those in control group. The uteri of FSH-treated group and PMS and HCG-treated group were hypertropied or filled with fluid in the lumens and walls of uteri. The wall tissue layers were flattened and their blood and lymph vessels were dilated. The mean number of follicle per ovary in control group were appeared to be $17.1{\pm}5.6$($14.0%{\pm}4.6%$), $37.8{\pm}9.1$($30.9{\pm}7.4%$) and $67.6{\pm}30.1$($55.2{\pm}24.6%$) respectively at large, middle and small follicles and total number of these 3 grade follicles were appeared to be $122.5{\pm}40.0$. The mean number of follicle per ovary in FSH-treated group were appeared to be $22.8{\pm}7.0$($17.4%{\pm}5.3%$), $43.4{\pm}6.6$($33.2{\pm}5.1%$) and $64.5{\pm}13.0$($49.3{\pm}9.9%$) respectively at large, middle and small follicles and total number of these 3 grade follicles were appeared to be $130.7{\pm}16.6$. The mean number of follicle per ovary in PMS and HCG-treated group were appeared to be $29.7{\pm}11.0$($16.3%{\pm}6.0%$), $61.9{\pm}17.2$($33.9{\pm}9.4%$) and $91.1{\pm}28.2$($49.9{\pm}15.4%$) respectively at large, middle and small follicles and total number of these 3 grade follicles were appeared to be $182.6{\pm}32.7$. The above findings reveal that large follicles were increased 29.8% in FSH-treated group and 73.7% in PMS and HCG-treated group than those in control group and in histologic findings, proportion of atretic follicles were more increased in ovaries with more number of more developing follicles.

Effects of Electroacupuncture's Intensity and Frequency on Transfer Capability of Small Intestine with Sex and Age in Rats (전침(電鍼)의 강도(强度)와 주파수(周波數)가 백서(白鼠)의 성별(性別)과 주령(週齡)에 따라 소장(小腸) 수송능(輸送能)에 미치는 영향(影響))

  • Backgrounds : Electroacupuncture(EA) is commonly used in treatment of various disease. Despite an extensive evidence-base guiding the selection of stimulation parameters, little methodologically robust research regarding the level of intensity and frequency in need to provide effective doses. Objectives : This study is evaluated effects of various intensities and frequencies of EA's stimulation at $ST_{36}$, Zusanli, with sex and age in rats. Methods : In this study, we measured the effects of EA treatment on transfer capability of small intestine in rats. Under anesthesia, two intensities, 1, 5mA with three different frequencies, 2, 50, 100Hz were applied to the groups divided with sex and age for 30minutes. Results : 1mA-2Hz, 5mA-2, 100Hz in male and 1mA-2, 50, 100Hz in female without distinction of age elicited an increasing trend in transfer capability of small intestine. In case of 2Hz in male, it showed the effects of EA without strength or weakness of stimulation and it need 100Hz stimulation additionally with age. In the groups of 1mA-2Hz, 5mA-2Hz in male and 1mA-2Hz, 1mA-100Hz, we elicited the decreasing trend with growing of weeks-old in male. Conclusions : With this result, it was suggested that we should consider a correlation of sex, age and EA's intensities, frequencies for the efficient EA.
