Studies on the Chemical Structure of the New Polysaccharide C - (The New Polysaccharides of Gum Tragacanth. II) - (Tragacanth gum 의 신다당류(新多糖類) C 의 화학구조(化學構造) - Tragacanth gum의 신다당류(新多糖類)에 관(關)한 연구(硏究) 제2보(第二報) -)

  • The polysaccharide C prepared from gum tragacanth powder (U. S. P. grade) by the precipitation method with 85% ethanol was a neutral polysaccharide, $[{\alpha}]^{30}_D-72.2$. The polysaccharide C consisted of L-rhamnose, D-xylose, L-arabinose and D-galactose in the molar ratio 2:1:17:9 (Table 1, 2, 3, ). The polysaccharide C was methylated with dimethylsulphate and 40% NaOH, and Purdies regent. The hydrolyzate of fully methlated product ($[{\alpha}]^{22}_D-102$ in chloroform, the methoxy content 40.6%) was composed of 2, 3, 5-tri-O-methyl-L-arabofuranose (I), 3,4-di-O-methyl-L-rhamnopyranose (II), 2,3-di-O-methyl-D-xylose (III), 2,3,4-tri-O-methyl-D-galactopyranose (IV), 2,4-di-O-methyl-L-arabopyranose (?), 2,4-di-O-methyl-D-galactose(VI), 2-O-methyl-D-arabinose (VII), and L-arabopyranose(VIII) (Table 4, 5, and Fig. 4). The first partial hydrolysis (A) of the polysaccharide C with 0.05N-HCl for 4.5 hours at $80-85^{\circ}C$ released only L-arabinose: the second hydrolysis (B) with 0.1N-HCl for 5 hours at $80-85^{\circ}C$, L-arabinose and D-galactose; and the third hydrolysis (C) with 0.3N-HCl at $90-95^{\circ}C$ in sealed tube, L-rhamnose, D-xylose, L-arabinose and D-galactose. From the unhydrolyzate A' were found L-rhamnose, D-xylose, L-arabinose, and D-galactose; from B' L-rhamnose, d-xylose, L-arabinose and D-galactose; and from C' D-xylose and D-galactose respectively (Table 6). The periodate consumption and formic acid production of the polysaccharide C were measured at various time intervals. After 120 hours periodat was consumed by 1.23 mole per $C_5H_8O_4$ and formic acid was produced 0.78 mole per $C_5H_8O_4$ (Table 7). Although a definite chemical structure for this polysaccharide C may not be formulated, experimental data, especially, from methylation, partial hydrolysie and determination of its molar ratio, and periodate analysis showed that the polysaccharide C is a highly branched polysaccharide and would be constructed of galactoaraban as a main chain residue and L-arabofuranose, D-galactopyranosyl $(1{\rightarrow}1)$-L-arabofuranose, D-xylopyranosyl $(1{\rightarrow}2)$-L-rhamnopyranosyl $(1{\rightarrow}1)$-L-arabofuranose, and L-rhamnopyranosyl $(1{\rightarrow}1)$-arabofuranose, and D-galactopyranosyl-$(1{\rightarrow}2)$-L-arabopyranosyl-$(1{\rightarrow}1)$-I-arabofuranose as a branch chain or end group (page 21).

Evaluation of Grooving Corrosion and Electrochemical Properties of H2S Containing Oil/Gas Transportation Pipes Manufactured by Electric Resistance Welding

  • Electrical Resistance Welding (ERW) on a longitudinal seam-welded pipe has been extensively used in oil and gas pipelines. It is well known that the weld zone commonly suffers from grooving corrosion in ERW pipes. In this paper, the grooving corrosion performances of API X65 grade non-sour service (steel-A) and API X70 grade sour gas resistant (steel-B) steel electrical resistance welding pipelines were evaluated. The microstructure of the bondline is composed of coarse polygonal ferrite grains and several elongated pearlites. The elongated pattern is mainly concentrated in the center of the welded area. The grooving corrosion test and electrochemical polarization test were conducted to study the corrosion behavior of the given materials. A V-shaped corrosion groove was found at the center of the fusion zone in both the steel-A and steel-B ERW pipes, as the corrosion rate of the bondlines is higher than that of the base metal. Furthermore, the higher volume fraction of pearlite at the bondline was responsible for the higher corrosion rate at the bondline of both types of steel.

  • In this paper, a new competitive learning algorithm called dynamic competitive learning (DCL) is presented. DCL is a supervised learning mehtod that dynamically generates output neuraons and nitializes weight vectors from training patterns. It introduces a new parameter called LOG (limit of garde) to decide whether or not an output neuron is created. In other words, if there exist some neurons in the province of LOG that classify the input vector correctly, then DCL adjusts the weight vector for the neuraon which has the minimum grade. Otherwise, it produces a new output neuron using the given input vector. It is largely learning is not limited only to the winner and the output neurons are dynamically generated int he trining process. In addition, the proposed algorithm has a small number of parameters. Which are easy to be determined and applied to the real problems. Experimental results for patterns recognition of remote sensing data and handwritten numeral data indicate the superiority of dCL in comparison to the conventional competitive learning methods.

  • A tentative hydrogeologic and hydrodispersive study was carried out to evaluate the groundwater pollution potential at a selected site by utilizing empirical assessment methodologies in an advanced stage of quantitative computer aided assessment. The upper most aquifer is defind as saturated overburden and weathered zone including the upper part of highly fractured rock. Representative hydraulic conductivity and storativity of the uppermost aquifer are estimated at 2.88 E-6 m/s and 0.09, respectively. Also calculated Darcian and average linear velocity of groundwater along the major pathway are 0.011 m/d and 0.12 m/d with average hydraulic gradient of 4.6% in the site. The results of empirical assessment methodologies indicate that 1) DRASTIC depicts that the site is situated on non-sensitive and non-vulnerable area. 2) Legrand numerical rating system shows that the probability of contamination and degree of acceptability are classed to "Maybe-Improbable, and Probable Acceptable and Marginally Unacceptable" with situation grade of "B". 3)Waste soil-site interaction matrix assessment categorizes that the study site is located on "Class-8 Site".

Quality Characteristics, Changes in Physiochemical Properties and Functional Properties of Camembert Cheese Containing Red Ginseng Powder

  • Effects of quality, physicochemical properties and antioxidants in Camembert cheese added with red ginseng powder (RGP) were investigated. Cheese samples were prepared with 0.05%, 0.10%, 0.15% and 0.20% RGP. and then monitored during ripening at $14^{\circ}C$ for 28 d. The pH of the RGP amended treatment groups increased during the ripening period relative to the control (p<0.05). Moreover, the 1,2-Diphenyl-1-picrylhydrazyle (DPPH) was highest in the 0.15% RGP group from 21 d to 28 d. $ABTS^+$ radical scavenging activity was increased just like DPPH as the ripening period passed, 0.10% treatment was highest at from 7 d to 21 d. 0.15% RGP was contents of ginsenosides : 10,999.7 ppm. The Free fatty acids (FFA), controls with 0.15% treatment, while the total fat (TF) and monounsaturated fat (MuSF) were higher in the control than the 0.15% RGP group (p<0.05). The total free amino acid (FAA) was increased in the control, and 0.15% RGP, and control was highest at then 0.15% RGP. The samples had average contents of fat and protein were 29% and 18-20% respectively. Additionally, the $L^*$ value decreased, while the $a^*$ and $b^*$ values increased as the amount of RGP added increased. Sensory evaluation revealed that texture and total acceptability were higher in the control group at 12 d. Although the addition of RGP did not exert a better effect on the ripening of the camembert cheese, but the ripening grade was similar to that of the common camembert cheese, and the additional function of the cheese was reinforced. Functional cheese could be developed.

Lap Splice Length of Glass Fiber Reinforced Polymer (GFRP) Reinforcing Bars with Different Surface Design (외피형태에 따른 GFRP 보강근의 겹침 이음길이)

  • The lap splice lengths of deformed steel reinforcing bars and GFRP bars with two different to surface type were experimentally compared using beam specimens. The purpose was to evaluate the length required of the GFRP bar to develop strength equivalent to the conventional steel reinforcing bar. The main test variable was the lap splice length. Two different GFRP bar surfaces were tested: (1) spiral-type GFRP bars and (2) sand coated GFRP bars. For the conventional steel bars (SD400 grade), strength over 400 MPa in tension was reached using the lap splice length of $30d_b$. Splice failure was observed in the specimen with the lap splice length of $20d_b$. For the spiral-type and sand coated GFRP bars, the tensile strength developed in the GFRP bars decreased with decreasing splice lengths. Development of the cracks on beam surfaces was clearly visible for the beams reinforced with the GFRP bars. Mid-span deflections, however, were significantly smaller than the comparable beams with conventional steel bars indicating potential ductility problem.

Development of Tobacco Ripeness Grading Meter Using the Color Sensor (칼라센서를 이용한 담배 완숙도의 식별장치 개발)

  • A tobacco ripeness grading meter was designed and constructed using the color sensor, its performance was evaluated. A degree of ripeness grading of a leaf is very closely related to the measured tobacco leaf color. Measuring the small amount of the reflectance precisely depends on the apparatus including color sensor, light source, detector sensitivity, and geometric characteristics of appratus. To analyze and minimize the variational effects, experiments to select the proper condition were performed. Because of the combined effect mentioned above, the system has some variation on its response. Basis on the results of the experiments, prototype was developed and interfaced to a computer system. The main components of prototype included a tungsten lamp as a light source, Amorphous full color sensor with three filters, regulated D.C. power supply, OP - AMP(741 TC) for amplification, AR - B3001 board for interfacing to a computer with analog to digital conversion, and a compatible IBM PC XT computer. The experimental results of the developed ripeness tobacco leaf measurement system are summarized as following: [1] The output readings of ripeness grade meter for tobacco leaf, which is based on harvesting time, showed the apparent difference in variety of different quality. It was considered suitable that three filters(red, green, blue) in Amorphous full color sensor could be used in four different ripeness degree measurement of tobacco leaf. [2] The output readings of ripeness grade meter for tobacco leaf, which is based on government procurement, showed apparent difference in variety of different quality. Tobacco leaf varieties to stalk position are divided into tips, leaf, cutters, and primings, It is considered suitable that only red filter in the sensor could be used to classify the grade of tobacco leaf within the same kind tobacco stalk. However, the ripeness grade meter was not adequate to classify all the tobacco grades in the four different tobacco leaves.

Expression of Serum and Muscle Endocrine Factors at Antemortem and Postmortem Periods and Their Relationship with Pig Carcass Grade

  • Carcass weight and backfat thickness are primary yield grading factors. Insulin-like growth factor (IGF)-I/-II, transforming growth factor $\beta$1 (TGF-$\beta$1), and epidermal growth factor (EGF) regulate the proliferation and differentiation of cells including adipocytes. Also, interleukin (IL)-2/-6, cortisol, and dehydroepiandrosterone-sulfate (DHEA-S) are known to be related to muscle growth and fat depth. However, the relationships between endocrine factors and carcass grade have not been studied. Therefore, this study aimed to measure the concentrations of endocrine factors in serum and muscle, and to investigate the relationship of endocrine factors with carcass grade. A total of 60 crossbred gilts (Duroc${\times}$Yorkshire${\times}$Landrace) were used. Blood from the jugular vein was collected at antemortem (7 days before slaughter) and postmortem periods, and M. Longissimus was collected at 45 min and 24 h after slaughter. The concentrations of IGF-I/-II, EGF, TGF-$\beta$1, IL-2/-6, cortisol and DHEA-S were analyzed by radioimmunoassay (RIA) or enzyme-linked immunosorbent assay (ELISA). In general, IGF and EGF concentrations in serum and muscle of grade A carcasses were found to be higher than those of grade C carcasses at antemortem and postmortem periods, whereas the pattern of TGF-$\beta$1 concentration was reversed. In particular, the concentrations of muscle IGF-I (24 h postmortem) and serum TGF-$\beta$1 (antemortem) were significantly different between grades A and C (p<0.05). The present results indicate that serum and muscle growth factors affect carcass weight and backfat thickness, and indirectly suggest the possibility that carcass grade could be predicted by expression of serum and/or muscle growth factors.

Effects of Bovine Somatotropin (bST) Administration Combined with Controlled Internal Drug Release (CIDR) on Embryo Quality and Pregnancy of Hanwoo (Korean Native Beef Cattle) during Commercial Embryo Transfer Program

  • Effects of recombinant bovine somatotropin (bST) on plasma hormonal concentration, embryo quality, and pregnancy rate were examined during the superovulation and synchronization treatment in donor and recipient cows. Hanwoos (Korean native beef cattle) were treated with controlled internal drug release (CIDR) combined with bST (CIDR+bST) or without bST (CIDR) as donor cows. The embryos recovered from donors were transferred into Holstein recipient heifers treated with bST (CIDR+bST) or without bST (CIDR) for synchronization. The correlation between IGF-I and P4 showed a positive pattern in the CIDR+bST group (r=0.44, p<0.01), but a negative pattern was shown in the CIDR group (r = -0.59, p<0.02) at day 7 of estrous cycles. Although the number of recovered, transferable, and degenerated embryos was not different, quantities of grade 1 (excellent) embryos in CIDR+bST group were significantly higher than those of the CIDR group (p<0.01). The pregnancy rate was higher in the CIDR+bST recipient group compared to CIDR group (p<0.05), when the embryos were recovered from the donors treated with CIDR. However, the pregnancy was maintained highly in both recipient groups, when the embryos were produced by CIDR+bST treated donors. It can be concluded that bST administration combined with CIDR is an effective method for superovulation and synchronization treatment to stabilize plasma hormonal levels, to obtain excellent quality of embryos, and to get higher pregnancy rate.

Influence of polymer-coated slow-release urea on total tract apparent digestibility, ruminal fermentation and performance of Nellore steers

  • Objective: Two experiments were performed to evaluate the effects of coated slow-release urea on nutrient digestion, ruminal fermentation, nitrogen utilization, blood glucose and urea concentration (Exp 1), and average daily gain (ADG; Exp 2) of steers. Methods: Exp 1: Eight ruminally fistulated steers [$503{\pm}28.5kg$ body weight (BW)] were distributed into a d $4{\times}4$ Latin square design and assigned to treatments: control (CON), feed grade urea (U2), polymer-coated slow-release urea A (SRA2), and polymer-coated slow-release urea B (SRB2). Dietary urea sources were set at 20 g/kg DM. Exp 2: 84 steers ($350.5{\pm}26.5kg$ initial BW) were distributed to treatments: CON, FGU at 10 or 20 g/kg diet DM (U1 and U2, respectively), coated SRA2 at 10 or 20 g/kg diet DM (SRA1 and SRA2, respectively), and coated SRB at 10 or 20 g/kg diet DM (SRB1 and SRB2, respectively). Results: Exp 1: Urea treatments (U2+SRA2+SRB2) decreased (7.4%, p = 0.03) the DM intake and increased (11.4%, p<0.01) crude protein digestibility. Coated slow-release urea (SRA2+-SRB2) showed similar nutrient digestibility compwared to feed grade urea (FGU). However, steers fed SRB2 had higher (p = 0.02) DM digestibility compared to those fed SRA2. Urea sources did not affect ruminal fermentation when compared to CON. Although, coated slow-release urea showed lower (p = 0.01) concentration of $NH_3-N$ (-10.4%) and acetate to propionate ratio than U2. Coated slow-release urea showed lower (p = 0.02) urinary N and blood urea concentration compared to FGU. Exp 2: Urea sources decreased (p = 0.01) the ADG in relation to CON. Animals fed urea sources at 10 g/kg DM showed higher (12.33%, p = 0.01) ADG compared to those fed urea at 20 g/kg DM. Conclusion: Feeding urea decreased the nutrient intake without largely affected the nutrient digestibility. In addition, polymer-coated slow-release urea sources decreased ruminal ammonia concentration and increased ruminal propionate production. Urea at 20 g/kg DM, regardless of source, decreased ADG compared both to CON and diets with urea at 10 g/kg DM.
