Encephalitic listeriosis in a Korean native goat (Capra hircus) (흑염소의 뇌염형 리스테리아증 발생 예)

  • This paper describes a naturally occurring case of meningoencephalitis associated with Listeria monocytogenes in a 4 month-old Korean native goat. The goat was raised in a herd consisting of seventy goats, and fed on straw and leftover vegetables including onions, cauliflower, and bean sprouts. The clinical symptoms of the goat were depression, in appetence, anorexia, circling, and recumbency before death. No conspicuous gross lesions were found except consolidation of the lung. Histopathologically, severe multifocal microabscesses and perivascular cuffings were observed in the brain stem and mid brain. In addition, mild nonsuppurative meningitis in the cerebrum were seen in this case. The organism isolated from the brain stem was confirmed as L. monocytogenes using the biochemical tests and PCR assay. Based on clinical signs, histopathology, and bacterial isolation; we confirmed that this case was a typical encephalitic listeriosis.

Seroprevalence of Coxiella burnetii in native Korean goats (Capra hircus coreanae) in Gyeongbuk province, Korea (경북지역 재래염소의 Coxiella burnetii 항체보유율)

  • Q fever is a zoonotic disease caused by the obligate intracellular bacterium Coxiella (C.) burnetii and affects wild and domestic animals worldwide. The aim of this study was to evaluate the seroprevalence of C. burnetii in native Korean goat (Capra hircus coreanae) in Gyeongbuk province, Korea, using ELISA. A total of 256 goat blood samples from 56 farms in Gyeongbuk province were collected between May 2012 and March 2013. Among them, 22 (8.6%) samples from 10 (17.9%) farms were seropositive for C. burnetii by ELISA. According to regional analysis, the seroprevalences among goat farms in eastern, western, southern, and northern areas of Gyeongbuk province were 0%, 18.2%, 36.8%, and 6.3%, respectively, showing the highest seroprevalence in the southern region. Among 22 counties in Gyeongbuk province, 10 (45.5%) counties had one or more farms positive to C. burnetii antibody. Accordingly, the seroprevalence of C. burnetii in high-risk humans and animals are constantly demanded by regional investigation.

  • Animal clones derived from somatic cells have been successfully produced in a variety of mammalian species such as sheep, cattle, mice, goats, pigs, cat and rabbits. However, there are still many unsolved problems in the present cloning technology. Somatic cell nuclear transfer has shown several developmental aberrancies including high rate of abortion in early gestation and increased perinatal death. These developmental failures of cloned embryos may arise from abnormal reprogramming of donor genome and/or incomplete cloning procedure. We have found that overall genomic methylation status of cloned bovine embryos is quite different from that of normal embryos in various genomic regions, suggesting that the developmental failures of cloned embryos may be due to incomplete reprogramming of donor genomic DNA. Many of the advances in understanding the molecular events for reprogramming of donor genome will more clarify the developmental defects of cloned embryos.

Effect of Dextrose, NaCl and Temperature on the RBC Sedimentation Rates in Goat (산양적혈구(山羊赤血球) 침강속도(沈降速度)에 미치는 온도(溫度), NaCl 및 Dextrose에 대하여)

  • Eight Korean native black goats were used, $10m{\ell}$ of blood was collected from the Jugular vein into heparinized tubes a week interval. The heparinized blood was centrifuged for separation to blood plasma and corpuscles. The hematocrit, per centage of blood that is red blood cells, was reschuffuled of 10 %, 20 %, 30 %, 40 % and 50 % using blood plasma, 0.9 % NaCl solution and 5.4 % dextrose solution. The sedimentation rates of red blood cell obtained at $7^{\circ}C{\pm}1^{\circ}C$ and $27^{\circ}C{\pm}1^{\circ}C$ are summarized as follows. 1) The sedimentation rates of red blood cell were more increased by lower PCV%, i.e. there was a reverse relationship between the sedimentation rates and PCV% at any condition of these experiments. 2) The RBC were sedimented the most quickly in the NaCl solution and slower in the plasma compare with the dextrose solution at the same PCV%. 3) There was no temperature effect on the sedimentation rates between the two groups of $7^{\circ}C{\pm}1^{\circ}C$ and $27^{\circ}C{\pm}1^{\circ}C$(at PCV 20% and 10%), even though the temperature difference is $20^{\circ}C$.

Recent advances in dairy goat products

  • Goat population world-wide is increasing, and the dairy goat sector is developing accordingly. Although the new technology applied to the goat industry is being introduced slowly because the weight of traditional subsector in the dairy sector, considerable advances have been made in the last decade. Present review focuses on the emerging topics in the dairy goat sector. Research and development of traditional and new dairy goat products are reviewed, including the new research in the use of goat milk in infant formula. The research in alternatives to brine, production of skimmed goat cheeses and the use of different modified atmosphere packaging are also addressed. Special attention is given to antibiotic residues and their determination in goat milk. Functional foods for human benefits are a trending topic. Health properties recently discovered in dairy goat products are included in the paper, with special attention to the antioxidant activity. The dual-purpose use of goats by humankind is affecting the way of how new technology is being incorporated in the dairy goat sector and will certainly affect the future development of dairy goat products.

  • Bovine borreliosis, caused by Borrelia theileri which is transmitted via hard tick bites, is associated with mild clinical symptoms, such as fever, lethargy, hemoglobinuria, anorexia, and anemia. Borrelia theileri infects various animals, such as cattle, deer, horses, goats, sheep, and wild ruminants, in Africa, Australia, and South America. Notably, no case of B. theileri infection has been reported in Korean cattle to date. In this study, 101 blood samples were collected from a Korean indigenous cattle breed, among which 1.98% tested positive for B. theileri via nested PCR. The obtained sequences exhibited high homology with B. theileri strains identified in other regions. Phylogenetic analysis of 16S rRNA confirmed the B. theileri group affiliation; however, flagellin B sequences exhibited divergence, potentially due to regional evolutionary differences. This study provides the first molecular confirmation of B. theileri infection in Korean livestock. Further isolation and nucleotide sequence analyses are necessary to better understand the presence of B. theileri strains in cows in Korea.

Effects of Activation Treatments and Culture Condition on In Vitro Development of Caprine In Vivo and In Vitro Oocytes (재래산양의 체내 및 체외유래 난자의 활성화 처리방법 및 배양조건이 단위발생란의 체외발달에 미치는 영향)

  • This study was conducted to examine whether activation treatments, source of oocytes and culture conditions affect in vitro developmental ability of caprine oocytes. Mature Korean native goats were pretreated with intravaginal CIDR for 10 days. The goats were then treated with a single intramuscular injection of 1,000 IU PMSG on Day 8 or twice daily injection of a total of 70 mg FSH for 3 days from Day 8 of CIDR insertion for superovulation. All the goats were injected with 10 mg PGF/sub 2a/ on Day 8 and 400 IU hCG on Day 10 of CIDR. Oocytes were surgically collected by oviduct flushing(in vivo maturation) or direct follicle aspiration(in vitro maturation) through mid-ventral incision at 35 h after hCG injection. Fifteen to twenty oocytes were placed in TCM-199 medium containing 25 mM Hepes and hormones under mineral oil at 39℃ in a humudified atmosphere of 5% CO₂ in air for 22 to 24 h. After maturation, the oocytes were activated by electric stimulation or ionomycin + 6-DMAP. The activated oocytes were then cultured in M16, TCM-199 and mSOF media supplemented with proteins at 39℃ for 6 to 7 days. Activation treatments did not affect cleavage of the oocytes. The cleavage rates were 64.1% (41/64) in oocytes activated by electric stimulation and 76.5% (218/285) in oocytes activated by ionomycin + 6-DMAP. The proportion of development to blastocyst was 15.6% (34/218) in oocytes activated by ionomycin + 6-DMAP, but activation by electric stimulation did not support embryos developed beyond morula stage. There were no differences in the cleavage rates of activated oocytes experiencing in vivo (86.8%, 66/76) and in vitro maturation (69.0%, 127/184). However, the development rate to blastocyst stage was significantly (P<0.05) higher for oocytes matured in vivo (50.0%, 33/66) compared to in vitro (0.8%, 1/127). Culture conditions did not affect the cleavage of -activated oocytes. The cleavage rates were 51.6% (49/95) in M16, 64.3% (18/28) in TCM-199 and 81.0% (145/179) in mSOF, respectively. By contrast, the development rate of activated oocytes to stage was greater (P<0.05) for oocytes cultured in mSOF medium (23.4%, 34/145) than in M16 or TCM-199 (0.0%). Our results suggest that source of oocytes and culture conditions are major factors affecting in vitro development of caprine parthenogenetic oocytes.

Histochemical study on the tracheal development in fetuses and neonates of Korean native goats (한국재래산양의 태아 및 신생아 기관의 조직발달에 관한 조직화학적 연구)

  • The present study was performed to investigate the patterns of appearance of lectin in trachea of fetuses of 60, 90 and 120 days old and neonates of Korean native goat. Carbohydrate markers were used in histochemistry for the determination of the lectin by staining of avidin-biotin-peroxidase complex(ABC), and the markers consisted of biotin-labeled concanavalin A(Con A), dolichos biflorus agglutinin(DBA), rincinus communis agglutinin(RCA-I), ulexeuropalus communis agglutinin(UEA) and wheat germ agglutinin(WGA). 1. The Con A-binding reactions appeared moderately on the apical surface of the tracheal epithelia in 60 days old fetuses, and the reactions were similar on the tracheal epithelia and glands in 90 and 120 days old fetuses and neonates. 2. Reaction of the DBA appeared as the strongest meanwhile the DBA-binding reactions were determined strongly on the apical surface of the tracheal epithelia in the 60 days old fetuses. Reaction for the DBA on the tracheal epithelia and glands of 90 and 120 days old fetuses and neonates were in same manner. 3. The RCA-I-binding reactions appeared very strongly on the apical surface of the tracheal epithelia in 60 and 90 days old fetuses. Reaction to the RCA-1 appeared moderately on certain apical surface of tracheal epithelia and glands in 120 days old fetuses and neonates. 4. No reactions provoked for the UEA in trachea of 60 days old fetuses and neonates, but the UEA-binding reactions appeared moderately in the tracheal epithelia of 90 days old fetuses and weakly in 120 days old fetuses. 5. The WGA-binding reactions appeared very strongly on the apical surface of the tracheal epithelia in 60 and 90 days old fetuses, and moreover, the reactions were determined on the luminal surface of the tracheal gland in 90 days old fetuses. On the other hand, goblet cells of the tracheal epithelia and glands in neonates reacted moderately to the WGA.

MINERAL CHARACTERISTICS OF RAW GOAT MILK FROM THE REGION OF GOIÂNIA, BRAZIL.

  • 432 samples of raw goat milk were analyzed for sodium, calcium, potassium, chloride, and ashes content. The samples came from 16 pure breed goats, 2 to 4 years of age kept in semi-feedlots. They were basically fed soy bran, cotton bran, triturated Guandu hay and disintegrated corn with its straw and corncob (according to requirements for due preservation and production). The mean values found were: sodium, $45{\pm}10.2mg%$ (24.5-65.6 mg%); potassium, $206{\pm}44.7mg%$ (116.6-295 mg%); calcium, $111{\pm}22mg%$ (66.5-155.4 mg %); ashes, $0.82{\pm}0.06mg%$ (0.68-0.95%); chloride, $235{\pm}39mg%$ (157-313 mg%). Results were related to: a) breed, the Brown and the Saanen revealed superior values for calcium and ashes, b) climate, which in the rain season led to higher contents of sodium, potassium, and calcium when compared to the local dry season, c) time of lactation which increasingly affected the contents of calcium, sodium and ashes, and d) the time between milking, when higher concentration of chloride and ashes were mostly concentrated in the evening and calcium in the morning. These results have important implications in the handling and management of the goat herd.

Brucellosis in sheep and goat of Bogra and Mymensingh districts of Bangladesh

  • Brucellosis is the most important bacterial disease of livestock in Bangladesh. The present study was conducted to determine the seroprevalence of brucellosis in goat and sheep in Mymensingh and Bogra districts of Bangladesh using slow agglutination test and Rose Bengal test as screening test and indirect enzyme linked immunosorbent assay as confirmatory test. Questionnaire based data on age, gender, area, client's complaint, number of animals in herds, disease history, reproductive problems such as abnormal uterine discharge, abortion or previous abortion in sheep and goat and their reproductive diseases were recorded. A total of 200 sera samples were collected from 80 sheep and 120 goats. The prevalence of brucellosis in goat was 2.50% and 1.25% in sheep. Positive reactors were only detected in female of both goat and sheep. In this study, there existed a significant association among abortion and the prevalence of brucellosis (p < 0.01). The prevalence of brucellosis in sheep and goat in Bangladesh is not negligible, and it is therefore worth considering the adoption of preventive measures.
