Trimorphomyces papilionaceus dikaryon 으로부터 자영적인 돌연변이에 의해 분리한 이배체를 대상으로 생장속도, U. V. 광 조사 후의 생존율, U. V. 분광광도계와 Hoechst 33258 핵염색에 의한 DNA 함량등을 monokaryon 및 dikaryon 과 비교하여 결정하였다. 이배체는 dikaryon 핵의 DNA 양과 비슷하였고 monokaryon 핵의 DNA 양의 2배 정도가 되었다. Glyoxalase II 의 band 양상에서 monokayon은 한 개의 isozyme band 만을 보인 반면, 이배체와 dikaryon은 같은 위치에서 두개의 isozyme band 가 관찰되었다. 이원전개한 전기영동실험에서 dikaryon 특이 단백질은 분자량이 66.000 보다 컸으며 분자량 24,000-29,000 사이에서 이배체 특이 단백질이 존재한다.

Low soil pH causes from $H^+$ rhizotoxicity results in nutrients unavailability in the growing media, inhibits plant growth, development and reduces crop yields. The present study was carried out to reveal morpholophysiological and biochemical responses of wheat (Triticum aestivum L.) to acidity stress. Four wheat varieties viz. BARI Wheat-21, BARI Wheat-25, BARI Wheat-26 and BARI Wheat-30 were used in the study. Eight-day-old seedlings were exposed to different pH levels (3.5, 4.5, 5.5 and 6.5) of growing media. Acidity stress at any level reduced biomass, water, and chlorophyll contents in all the varieties; whereas BARI Wheat-26 showed the least damage. $H^+$ rhizotoxicity also caused oxidative stress through excess production of reactive oxygen species and methylglyoxal which increase lipid peroxidation in all the varieties but the lowest oxidative damage was observed in BARI Wheat-26 due to better performance of the antioxidant defense and glyoxalase systems. Considering the growth, physiological and biochemical attributes BARI Wheat-26 may be considered as acidity stress tolerant, among the variety examined.
Over production of methylglyoxal (MGO) a highly reactive dicarbonyl compound, has been associated in progressive diabetes with vascular complication. Therefore, we investigated whether hot water extract of Loliolus beka meat (LBM-HWE) presents a preserve effect against MGO-induced cellular damage in human umbilical vein endothelial cells (HUVECs). The LBM-HWE extract showed to inhibit MGO-induced cytotoxicity. Additionally, the LBM-HWE reduced mRNA expression of pro-inflammatory cytokines, and reduced MGO-induced advanced glycation end product (AGEs) formation. Furthermore, LBM-HWE induced glyoxalase-1 mRNA expression and reduced MGO-induced carbonyl protein formation in HUVECs. The results implicate that LBM-HWE has protective ability against MGO-induced HUVECs toxicity by preventing AGEs formation. In conclusion, LBM-HWE could be used as a potential treatment material for the prevention of vascular complications of diabetes.

Hsp31 protein is one of the members of DJ-1 superfamily proteins and has a dimeric structure of which molecular weight (MW) is 62 kDa. The mutation of DJ-1 is closely related to early onset of Parkinson's disease. Hsp31 displays $Zn^{+2}$-binding activity and was first reported to be a holding chaperone in E. coli. Its additional glyoxalase III active has recently been characterized. Moreover, an incubation at $60^{\circ}C$ induces Hsp31 protein to form a high MW oligomer (HMW) in vitro, which accomplishes an elevated holding chaperone activity. The NMR technique is elegant method to probe any local or global structural change of a protein in responses to environmental stresses (heat, pH, and metal). Although the presence of the backbone chemical shifts (bbCSs) is a prerequisite for detailed NMR analyses of the structural changes, general HSQC-based triple resonance experiments could not be used for 62 kDa Hsp31 protein. Here, we prepared the per-deuterated Hsp31 and performed the TROSY-based triple resonance experiments for the bbCSs assignment. Here, detailed processes of per-deuteration and the NMR experiments are described for other similar NMR approaches.
The effects of a herbicide butachlor[2-chloro-2', 6'-diethyl-N-(butoxymethyl) acetanilide] on the growth of the purple non-sulfur bacteria were investigated. The butachlor inhibited the growth of all species tested by 18-51%, except Rhodospirillum rubrum at concentrations of M, which would be field capacity. The photosynthetic growth rate of the species in the presence of butachlor was influenced by the nitrogen source. Cultures supplied with (NH&S04 showed a somewhat higher growth rate than those fixing dinitrogen, but they were more susceptible to butachlor (26-51%). On the contrary, butachlor enhanced the growth rate of Rhodospirillum rubrum in nitrogen gas conditions. When the culture was performed in medium with butachlor as the carbon source, the cells of fixing dinitrogen showed a higher exhaustion of butachlor than those supplemented with (N&)2S04, which exhaustion was examined by a decrease of the major absorbance at 213 nm and 260 nm. The exhaustion of butachlor as the carbon source had relation with the growth of the cells. The alkalization of culture supplemented with nitrogen gas was found in the cells treated with butachlor or untreated. The butachlor affected the carotenoid region but bacteriochlorophyll remained unaffected.
The phylogenetic relationships between Cheju native horses and Tsushima native horses were studied by protein polymorphism analyses in 16 gene loci (Trypsin inhibitor: Ti, Chymotrypsin inhibitor: CTi, Albumin: Al, Esterase: Es, Transferrin: Tf, Hemoglobin: Hb, Catalase: Cat, Esterase D: EsD, Glutamate oxaloacetate transaminase: GOT, Glyoxalase I: GLO I, Acid phosphatase: AcP, Superoxide dismutase: SOD, Lactate dehydrogenase: LDH, Hexokinase: HK, Malate dehydrogenase: MDH, Malic enzyme: ME). All allelic patterns of the protein loci, except 5 loci (SOD, LDH, HK, MDH, ME), were polymorphic in both two populations. Gene frequencies of the polymorphic loci of the population of Cheju native horses were higher than those of Tsushima native horses. Average heterozygosity in Cheju native horses was 0.375, showing higher than that of Tsushima native horses (0.304). The Da distance and gene identity of two populations were 0.108 and 0.868, respectively. The phylogenetic tree constructed by these results and those previously reported in other horse populations, consisted of three clusters. From this phylogenetic tree, it could be suggested that Cheju native horses and Tsushima native horses had diverged from the Mongolian wild horse (Equus prsewolskii).

We have previously investigated the proteome changes of rice leaves under heat stress (Lee et al. in Proteomics 2007a, 7:3369-3383), wherein a group of antioxidant proteins and heat shock proteins (HSPs) were found to be regulated differently. The present study focuses on the biochemical changes and gene expression profiles of heat shock protein and antioxidant genes in rice leaves in response to heat stress ($42^{\circ}C$) during a wide range of exposure times. The results show that hydrogen peroxide and proline contents increased significantly, suggesting an oxidative burst and osmotic imbalance under heat stress. The mRNA levels of chaperone 60, HSP70, HSP100, chloroplastic HSP26, and mitochondrial small HSP responded rapidly and showed maximum expression after 0.5 or 2 h under heat stress. Transcript levels of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and Cu-Zn superoxide dismutase (Cu-Zn SOD) showed a rapid and marked accumulation upon heat stress. While prolonged exposure to heat stress resulted in increased transcript levels of monodehydroascorbate reductase, peroxidase, glyoxalase 1, glutathione reductase, thioredoxin peroxidase, 2-Cysteine peroxiredoxin, and nucleoside diphosphate kinase 1, while the transcription of catalase was suppressed. Consistent with their changes in gene expression, the enzyme activities of APX and DHAR also increased significantly following exposure to heat stress. These results suggest that oxidative stress is usually caused by heat stress, and plants apply complex HSP- and antioxidant-mediated defense mechanisms to cope with heat stress.
