Studies on the Processing Properties and Interactions Between Porcine Blood Proteins and Waxy Rice Starch During Making Porcine Blood Cake

  • The physiochemical properties and interactions between porcine blood and waxy rice were determined. Addition of calcium chloride (0.15%) improved acceptability of blood cake and increased the gelatinization degree of waxy rice. The water-holding capacity of porcine blood gel (blood/water=60/40, v/v), extent of absorption and gelatinization of waxy rice, and scanning electron microscopy showed that blood protein matrix and waxy rice are competitors for holding water in the cooking procedure. Non-haem iron content increased linearly (R=0.95) when heating temperature rose. The presence of blood proteins caused increasing of peak temperature (Tp) of gelatinization in differential scanning calorimetric thermal gram, The microstnlcture of plasma proteins and haemoglobin appeared continuous changes, and interacted with surface of waxy rice flour in terms of network and mosaic form, respectively. The electrophoretic patterns revealed an interaction between plasma proteins and waxy rice glutelin and haemoglobin when heated could be found at temperatures above $60^{\circ}C$.

  • A strategy for development of a functional rice in proved with human lactoferrin and enhancement of nutrient compounds was planned. For the purposes we have cloned and characterized a human lactoferrin cDNA from human mammary gland cDNA library A endosperm storage vacuole targeting sequence and the cDNA fragment was linked to endosperm specific glutelin promoter. The fusion gene fragment was inserted into a binary vector containing MAR gene. In addition a new ${\beta}$-galactosidase gene from Bifidobacterium of human was used as a reporter gene in the vector system, Rice plants showing a high concentration of amino acids in the endosperm cells were developed by using a biochemical mutation and bred for the transformation with the binary vector system Finally we have established a transformation method for the rice endosperm cells.

  • Extracellular protein produced by Bacillus sp. $T_2-3$ was characterized for its patterns of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and sephadex G-100 filtration, spectrum of maximum absorption, composition of amino acid and solubility to solvents. The extracellular protein was composed of two kinds of protein which was little difference in molecular weight about 49,000. Maximum absorbance of the extracellular protein was showed at 230nm and main amino acids of the extracellular protein were aspartic acid, glutamic acid and alanine. Solubility of the extracellular protein was 55.8% in $H_2O$ and 28.4% in 0.4% NaOH.

  • Near full length cDNA clones encoding the rice seed storage protein, prolamine, were isolated and divided into two homology classes based on cross-hybridization and DNA sequencing analysis. These cDNA clones contain a single open reading frame encoding a putative rice prolamine precursor(M.W.=17,200) possessing atypical 14 amino acid signal peptide. Clones of these two homology classes diverge mainly by insertions/deletions of short nucleotide stretches and point mutations. The deduced primary structures of both types of prolamine polypeptides are devoid of any major tandem repetitive sequences, a feature prevalent in other cereal prolamines. No significant homology teas detected between the rice prolamine and other cereal prolamines, indicating that the rice gene evolved from a different ancestor that gave rise to other cereal prolamine genes. Developing wheat and rice endosperms were examined using ultrathin sections prepared from tissues harvested at various days after flowering. By immunocytochemical localization techniques, wheat prolamines are localized within vesicles from Golgi apparatus and in homogeneous regions of protein bodies. The involvement of the goli apparatus in the packaging of wheat prolamines into protein bodies indicates a pathway which differs from the mode of other cereal prolamines and resembles the mechanism employed for the storage of rice glutelin and legume globulins.

  • Lactoferrin is an 80-kDa iron-binding glycoprotein that is found in high concentrations in human milk. Human lactoferrin (hLF) has several beneficial biological activities including immune system modulation and antimicrobial activity. In the present study, we devolved a method of hLF expression through introducing the hLF gene construct into Oriza sativa cv. Nakdong using the Agrobacterium-mediated transformation system. The expression of the hLF gene under the control of the rice glutelin promoter was detected in the seeds of transgenic rice plants. Transformed rice plants were selected on media containing herbicide(DL-phosphinothricin) and the integration of hLF cDNA was confirmed by Southern blot analysis. The expression of the full length hLF protein from the grains of transgenic rice plants was verified by Western blot analysis. The lactoferrin expression levels in the transformed rice grains determined by enzyme-linked immunosorbant assay accounted for approximately 1.5% of total soluble protein. Taken together, these data indicate that rice grains expressing hLF can be directly incorporated into infant formula and baby food.

Protein and Amino Acid Composition of Water Cress Oenanthe stolonifera DC (미나리의 단백질의 및 아미노산 조성)

  • This study was attempted to compare the nutritive value of leaf with stem of the water cress Oenanthe stolonifera DC. in order to improve the eating habits and as a part of studying on the effective curing nutrients for the damaged liver. The contents of moisture crude proteinon the effective curing nutrients for the damaged liver. The contents of moisture crude protein crude fat and crude ash were 90.40% 2.85%, 0.42% and 0.74% in leaf while the contents of moisture crude protein crude fat and crude ash were 95.15% 0.77% 0.09%, and 0.64% of moisture crude protein crude fat and crude ash were 95.15% 0.77% 0.09% and 0.64% in stem respectively. The quantitative fractionation of proteini of both leaf and stem ranked albumin the highest content followed globulin prolamin and glutelin in order. It has been sh-own by SDS-polyacrylamide gel electrophoresis that water extractable protein of leaf 11 bands but those of stem were not detected. The scope of molecular weight for the main protein of water extractable protein of leaf was between 34.700 and 45,000. The amounts of extractive-nitrogen from leaf and stem of the water cress were 241.02mg% and 271.67mg% respec-tively. The amounts of free amino acid-nitrogen from the leaf and stem were 89.02mg% and 32.02mg% respectively. In free amino acid-nitrogen from the leaf and stem were 89.02mg% and 32.02mg% respectively. In free amino acid composition of both leaf and stem the major components were aspartic acid and glutamic acid. In total amino acid composition of water cress leaf aspartic and glutamic acid were the major components. Whereas alanine and thr-eonine were the major components in stem The assessment of water cress leaf and stem with chemical score. EAAl Rl showed that the values of stem were lower tendancy than those of leaf. Limiting amino acid of leaf was tryptophan while that of stem was lysine.

  • This study was carried out to examine the effects of phytate addition on the solubility and digestibility of the low-phytate soy protein isolate (LSPI) and high-phytate soy protein isolate (HSPI). In SDS-polyacrylamide gel electrophoresis of soy protein isolate, different patterns of proteins were observed in both HSPI and LSPI at various phytate and pH levels, suggesting that phytate may bind specifically to certain protein fractions at a particular pH. For example, proteins of M.W $1.8{\sim}3.5\;kDa$ resisted phytate binding at acidic pH. LSPI was fractionated into albumin, globulin, gliadin and glutelin, and phytate was shown to bind with difficultly to all three gliadin bands. Effects of phytate on the pepsin digestibility of soy proteins were apparent, especially in the short term digestion.

Changes of physicochemical properties of brown rice during ageing

  • The objective of this study was to determine physicochemical changes in brown rice during ageing condition. Five varieties (Haiami, Ilpum, Daecheong, Jungwon, and Dasan1) of brown rice were stored at $35^{\circ}C$ for 8 weeks. Crude protein and lipid content, seed germination rate, fat acidity, tocol content, TOYO glossiness value, pasting properties, and composition of storage proteins were measured to evaluate its quality during storage. The isomers of tocols (tocopherol and tocotrienols) were quantified using HPLC system, and the pattern of variation in rice storage proteins was examined through electrophoresis of protein extracts. Seed germination rate decreased by 2.7 times, whereas the fatty acid value dramatically increased by 4.8 times after 8 weeks of storage. Toyo glossiness value of cooked milled rice considerably affected by storage period, and the pasting properties of milled rice were also influenced by storage. The final viscosity and breakdown value increased, but setback decreased during storage. In terms of storage protein, proportion of prolamin (14.3 kDa) and globulin (26.4 kDa) increased, whereas percentage of glutelin (34-39 kDa and 21-22 kD) decreased. Furthermore, the contents of total tocol and isomers decreased in stored brown rice.

Effects of dietary protein of hog hair on the nutrients metabolism in poultry (돈모 단백질의 급여가 닭의 영양소 대사에 미치는 영향)

  • Two experiments were conducted to investigate the effect of feeding hog hair protein (HHP) on the nutritional value for poultry. In experiment 1, twenty roosters of Hanhyup-3 strain were alloted, and metabolizability of HHAA was measured. In experiment 2, forty roosters of Hanhyup-3 strain were alloted to 0, 3, 7.6 and 15.1% of HHAA treatments, 10 birds per treatment, and measured feed utilization and blood parameters. In experiment 1, no trend was found in excretion of amino acid, high in glysine and glutelin, low in valine, threonine, and methionine. HHAA metabolizability of serine, phenylalanine, alanine, and isoleucine was more higher than that of lysine, cystine, asparagine, and tyrosine. In experiment 2, as the HHAA level increased, feed intake decreased significantly in 15.1% treatment, but water intake increased significantly in 15.1% traetment. Dry matter and nitrogen metabolizability decreased in 7.6 and 15.1% treatments. Although no significant difference was found among three treatments(0, 3, and 5.7%), as the HHAA level increased, dry matter and nitrogen metabolizability decreased. Serum creatinine level was significantly increased in 15.1% treatment. In conclusion, it is considered that proper level to substitute soybean meal by HHAA was 10%.

Changes in SDS-PAGE Pattern of Mung Bean Grain Proteins During Germination (녹두 발아중 분획한 단백질의 전기영동 패턴의 변화)

  • Changes of protein contents and amino acid composition and SDS-PAGE pattern of protein of mung bean which were germinated in dark at $25^{\circ}C$ for 6 day. The total protein contents gradually decreased during germination and the contents of each fractionative soluble proteins were increased shortly after the soaking of mung bean and gradually decreased during the germination afterwards. SDS-PAGE of albumin fraction showed 18 bands, and during the germination the most of bands were diminished or disappeared. But protein bands at 24,000, 40,000, 45,000, 70,000 dalton position remained until 6th day of germination. SDS-PAGE of globulin fraction showed 6 discrete bands, and during the germination the protein band at 45,000 dalton position disappeard. But the protein bands at $14,000{\sim}25,000$ dalton position did not change during the period. SDS-PAGE of glutelin fraction showed 10 discrete bands, and during the germination the bands of $45,000{\sim}70,000$ dalton become diminished or disappeared. But the bands of 30,000, 60,000 dalton did not change throughout the germination period.
