Effects of Betaine on the $CCI_4$-Induced Toxicity in Primary Cultured Rat Hepatocytes (일차 배양한 흰쥐의 간세포에서 사염화탄소로 인한 독성에 미치는 비테인의 효과)

  • Betaine, a major component of Lycii Fructus, was evaluated for its anti-hepatotoxic activity on carbon tetrachloride-induced hepatotoxicity in primary cultured rat hepatocytes. Betaine was found to attenuate carbon tetrachloride-induced hepatotoxicity both morphologically and biochemically. Typical hepatocyte necrosis due to carbon tetrachloride seemed to be reduced by 50 to 500 $\mu{M}$ of betaine under microscopical observation. The value of glutamic pyruvic transaminase released from the hepatocytes into the medium significantly decreased as betaine concentration increased. Betaine also significantly elevated the reduced activities of some enzymes, cytochrome P-450, 7-ethoxycoumarin-0-deethylase and glutathione-S-transferase, involved in xenobiotic metabolism due to carbon tetrachloride-induced hepatotoxicity. These results demonstrate a possible hepato-protective role of betaine against fatty liver that could be easily induced by carbon tetrachloride.

Chemopreventive effects of polysaccharides extract from Asterina pectinifera on HT-29 human colon adenocarcinoma cells

  • We examined the effects of polysaccharides extracted from Asterina pectinifera on the activities of quinone reductase (QR), glutathione S-transferase (GST), ornithine decarboxylase (ODC), cyclooxygenase (COX)-2 and glutathione (GSH) levels in HT-29 human colon adenocarcinoma cells. We found that the polysaccharides extract induced QR activity in a dose-dependent manner over a concentration range of $20-60\;{\mu}g/ml$ and increased GST activity as much as 1.4-fold over controls. GSH levels were increased 1.3- and 1.5-fold with the extract at 40 and $60\;{\mu}g/ml$, respectively. The activity and protein expression of ODC in 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced colon cancer cells was inhibited by the extract. The polysaccharides suppressed TPA-induced prostaglandin (PG) production. These data indicate that polysaccharides from A. pectinifera increase phase II detoxification enzyme activity and inhibit ODC and COX-2 activities in HT-29 human colon adenocarcinoma cells. Consequently, this effect may contribute to the protective effect of polysaccharides from A. pectinifera against colon cancer.

Effect of Acron Extracts on the Antioxidative Enzyme System (도토리추출물이 흰쥐의 체내 항산화효소계에 미치는 영향)

  • This study was conducted to investigate the effect of acorn extracts on the antioxidative enzyme system. Male Sprague-Dawley rats(110$\pm$10g) were fed on containing normal and high fat diets. They were orally administrated (0.02g/100g B.W) of acorn ethylacetate-extract and water-extract at the same time once a day, respectively. Net weight gain and feed efficiency ratio were increased in high fat diet groups and decreased by acorn extracts administration. The effect of acorn extracts on hepatic glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) and catalase activities did not show significancy in normal fat diet groups. GST and catalase activities and lipid peroxidation content(LPO) were significantly increased in high fat diet groups and this increment were decreased by acorn extracts administration. However GSH-Px activity and GSH content were decreased in high fat diet groups but increased by acorn extracts administration. The activities of lactate dehydrogenase(LDH), alkaline phosphatase(ALP) and aminotransferase in serum were significantly increased in high fat diet group but these increment reduced in acorn extracts administration groups. These results indicate that acorn extracts could improve the liver function and prevent the metabolic diseases by hyperlipidemia.

Oxidative stress and non-specific immune responses in juvenile black sea bream, Acanthopagrus schlegelii, exposed to waterborne zinc

  • Juvenile black sea bream, Acanthopagrus schlegelii, were exposed to waterborne zinc (Zn) at concentrations of 0, 200, and $400{\mu}g/L$, at temperatures of 18 or $26^{\circ}C$ for 4 weeks. Superoxide dismutase (SOD) activities in the liver and gill of A. schlegelii significantly increased following exposure to waterborne Zn. Significant reduction in glutathione S-transferase (GST) activity in the liver and gill was observed following exposure to waterborne Zn. Glutathione (GSH) concentrations in the liver and gill also significantly decreased following exposure to waterborne Zn. Phagocytosis and lysozyme in the plasma and kidney were significantly increased following exposure to waterborne Zn. High water temperature increased alterations in the antioxidant and immune responses. The results of the present study suggest that waterborne Zn induced significant alterations in oxidative stress, increased immune responses and high temperature that trigger Zn toxicity.

The Hepatotoxicity and Testicular Toxicity Induced by Arecoline in Mice and Protective Effects of Vitamins C and E

  • Arecoline is a major alkaloid of areca nuts which are widely chewed by southeast Asian and it manifests various toxic effects in different organs of human and animals. In this work, mature mice were treated by vitamins C plus E, arecoline, or both daily for four weeks. The results showed that arecoline significantly increased the levels of serum alkaline phosphatase (ALP), glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT) and significantly decreased the levels of reduced glutathione (GSH), glutathione-S-transferase (GST), superoxide dismutase (SOD) and catalase (CAT) in the liver tissues. Additionally, the body weight, testis weight, sperm counts, motility and normal sperms also were significantly decreased. The supplement of vitamins C and E can bring the activities of ALP and GPT to normal levels and partially restore the sperm counts compared to the arecoline-treated group but have no other positive effects. In conclusion, the vitamins C and E partially attenuated the arecoline-induced hepatotoxiciy but basically had on protective effects against the arecoline-induced testicular toxicity.

Effect of Circadian Rhythms on the Bromobenzene Metabolism in Rats (랫드에 있어서 주야 시차가 Bromobenzene 대사에 미치는 영향)

  • To investigate the circadian variation in the bromobenzene metabolism, bromobenzene(400 mg/kg body weight) was intraperitoneally administered to the rats every other day for 6 days both in the night; 24:00 and the day; 12:00. Each group of animals was sacrificed at 8hr after last injection of bromobenzene. The contents of hepatic CYP were more increased in control rats of night phase than those of day phase but in case of bromobenzene treatment there were no differences in hepatic CYP between rats of the night phase and those of day phase and the injection of prednisolon inhibited the hepatic CYP content in rats. Furthermore, the decreasing rate of hepatic glutathione contents to the control was higher in rats of day phase than those of night phase by the bromobenzene treatment. And the hepatic glutathione S-transferase activities were increased both in control and bromobenzene treated rats of the night phase than those of day phase. On the other hand, liver weight per body weight(%), hepatic lipid peroxide content, serum levels of alanine aminotransferase were more increased both in bromobenzene-treated and control rats of the night phase than those in the day phase. These results indicate that the rats of night phase may induce more accelerated formation of bromobenzene 3,4-oxide from bromobezene than those of day phase in rats.

Study on Bromobenzene Metabolism in Rats with Liver Damage (흰쥐에 있어서 간손상 정도에 따른 Bromobenzene 대사)

  • To compare the severe liver damage with the slight one on the bromobeazene metabolism in rats, the animal group described as B7 group was induced the stage of slight liver damage with 7 times bromobenzene injection every other day (400 mg/Kg body wt. i.p.), whereas B40 group was induced that of more severe liver damage with bromobeazene 40 times injection as identified with determination of serum levels of alanine aminotransferase(ALT) activity and the histopathological findings. In the present experimental animal model, the decreasing rate of glutathione(GSH) and the increasing rate of glutathione S-transferase activity to the control group were higher in B7 group than B40 group. Furthermore the single dose of bromobenzene was injected to the two groups and sacrificed at 8hr and the hepatic aniline hydroxylase(AH) activity, GSH content and GST activity were determined. The increasing rate of AH activity to the control was lower in B40 group than B7 group and the decreasing rate of GSH to the control was also lower in B40 than B7 group. Moreover, B7 group showed the increased activity of hepatic GST to the control whereas B40 group showed the decrease activity of the enzyme. And Vmax value in GST was more decreased in B40 group than B7 group.

Anti-Proliferative and Anti-Carcinogenic Enzyme-inducing Activities of Delphinidin in Hepatoma Cells

  • Delphinidin, an aglycone form of anthocyanins, was demonstrated to have anti-carcinogenic potential. The compound at $50\;{\mu}g/mL$ caused a significant increase of quinone reductase activity, an anti-carcinogenic marker enzyme, in mouse hepatoma cell lines (Hepa1c1c7 and BPRc1). Delphinidin enhanced the expression of other detoxifying or antioxidant enzymes including glutathione s-transferase, gamma-glutamylcysteine synthetase, heme oxygenase 1, and glutathione reductase. It suppressed the proliferation of murine hepatoma cells in a dose-dependent manner, with approximately $IC_{50}$ of $70\;{\mu}g/mL$. These results suggest that delphinidin might be useful for cancer prevention.

Antidiabetic and Antioxidative Effects of Lycii fructus in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 흰쥐에 대한 구기자 추출물의 혈당강하 및 항산화 작용에 미치는 효과)

  • This study was done to investigate the antidiabetic and antioxidant effects of Lycii fructus in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Lycii fructus was orally administrated once a day for 7 days. The contents of serum glucose, total cholesterol were significantly decreaed (p<0.05) in Lycii fructus treated group compared to the those of STZ-control group, also content of Triglyceride (TG), Altherogenic index (AI) were decreased, but not statistically significant. But high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were increased in Lycii fructus treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6PDH), glutathione-s-transferase(GST), superoxide dismutase(SOD) were significantly increased(p<0.05), glucose-6-phos-phatase(G-6pase)was significantly decreased(p<0.05) and The glutathione(GSH), glucokinase(GK) were increased. But not statistically significant In Lycii fructus treated group compared to the those of STZ-control group These results indicated that ethanol extract of Lycii fructus would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.

Effects of Defatted Seasme Flour on Oxidative Stress Induced by Ethanol-feeding in Rats (식이 참깨탈지박이 에탄올을 공급한 쥐에 유도된 산화 스트레스 억제효과)

  • To evaluate the effect of defatted sesame flour(DSF) on the oxidative stress of ethanol feeding in rats, Wistar male rats were divided into 4 groups of control, ethanol, DSF and DSF ethanol. Each group was sacrificed after feeding for 4 weeks and was examined by measuring the formation of 2 thiobarbituric acid reactive substance(TBARS), total cholesterol(TC) in serum, redox glutathione S transferase(GST) enzyme activity and the contents of glutathione(GSH) in the liver. The formation of TBARS in the liver after ethanol feeding was significantly increased comparing to the control, but the levels were significantly decreased by the DSF as compared to the ethanol feeding group(p<0.05). When compared to fed control diet, we found that serum TC levels were significantly lower in the DSF fed group than control group (p<0.05). The activity of hepatic GST was significantly increased by DSF as compared to the control and was decreased by ethanol feeding. On the other hand, the hepatic contents of GSH were unaffected by DSF feeding. Our findings suggest that feeding DSF may inhibit ethanol induced oxidative stress may be due to the stimulation of antioxidative activity by sesaminol glucosides in DSF.
