Anticancer Agent인 Methotrexate에 대한 새로운 합성 연구 (A New Synthetic Studies on Anticancer Agent Methotrexate)

  • 새로운 methotrexate 중간체인 diethyl N-[4-{[(2,4-diamino-6-yl)methyl]-amino}benzoyl]-L-glutamate(10)를 합성하기 위하여 p-nitrobenzoic acid를 chlorination한 다음 L-glutamic acid와 coupling하고 이를 esterification한 후, 환원과 methylation시켜 diethyl N-(4-methylaminobenzoyl)-L-glutamate(7)를 합성하였다. 이 화합물(7)을 DMF 존재하에서 NaH와 allyl chloride를 가하여 allylation한 다음 여기에 $IN_3$ addition 반응으로 diethyl-p-[N-(2-azido-3-iodopropyl)-N-methyl]aminobenzoyl-L-glutamate(9)를 합성하였다. 이 화합물(9)을 2,4,5,6-tetraaminopyrimidine hydrochloride와 cyclization시켜 methotrexate diethylester를 얻었다.

당귀용회환(當歸龍薈丸)의 glutamate에 의한 청신경세포(聽神經細胞) 손상(損傷) 보호효과(保護效果) (Protective Effects of Danguiyonghoihwan on Glutamate-induced Auditory Sensorineuronal Cell Death)

  • Objective : The water extract of Danguiyonghoihwan (DGYHW) has been traditionally used in treatment of tinnitus in Oriental Medicine. However, little is known about the mechanism by which DGYHW rescues auditory neuronal cells from injury damages. Therefore, in this study I effort to elucidate the mechanism of the cytoprotective effect of the DGYHW extract on glutamate-induced auditory sensorineuronal cell death. Methods : I determined the elevated cell viability by DGYHW extract on glutamate-induced auditory sensorineuronal cell death. Glutamate induced neuronal damage in oranotypic explant culture also, glutamate decreased cell viability on VOT-33 cells but pretreatment with DGYHW inhibited cell death. Results : One of the main mediator of glutamate-induced cytotoxicity was known to generation of reactive oxygen species (ROS). Pretreatment with DGYHW inhibited this ROS generation from glutamated-stimulated VOT-33 cells. Also, I identified that the ROS-induced DCF-DA green fluorescence is reduced by DGYHW pretreatment. The critical markers of apoptotic cell death were cleavages of procaspase-3 protease protein. So I checked the expression level and cleavage of procaspase-3 protease protein. Glutamate-treated VOT-33 cells were shown to have cleavage of procaspase-3 protease proteins and following reduction of expression of these proteins. But I found that pre-treatment with DGYHW protects glutamate-induced changes of biochemical marker protein, caspase-3. Conclusion : These findings indicated that DGYHW may prevent cell death from glutamate induced VOT-33 cell death by inhibiting the ROS generation and modulation of protein expressions in procaspase-3, catalase and Bcl-2.

Low Non-NMDA Receptor Current Density as Possible Protection Mechanism from Neurotoxicity of Circulating Glutamate on Subfornical Organ Neurons in Rats

  • The subfornical organ (SFO) is one of circumventricular organs characterized by the lack of a normal blood brain barrier. The SFO neurons are exposed to circulating glutamate ($60{\sim}100{\mu}M$), which may cause excitotoxicity in the central nervous system. However, it remains unclear how SFO neurons are protected from excitotoxicity caused by circulating glutamate. In this study, we compared the glutamate-induced whole cell currents in SFO neurons to those in hippocampal CA1 neurons using the patch clamp technique in brain slice. Glutamate ($100{\mu}M$) induced an inward current in both SFO and hippocampal CA1 neurons. The density of glutamate-induced current in SFO neurons was significantly smaller than that in hippocampal CA1 neurons (0.55 vs. 2.07 pA/pF, p<0.05). To further identify the subtype of the glutamate receptors involved, the whole cell currents induced by selective agonists were then compared. The current densities induced by AMPA (0.45 pA/pF) and kainate (0.83 pA/pF), non-NMDA glutamate receptor agonists in SFO neurons were also smaller than those in hippocampal CA1 neurons (2.44 pA/pF for AMPA, p<0.05; 2.34 pA/pF for kainate, p< 0.05). However, the current density by NMDA in SFO neurons was not significantly different from that of hippocampal CA1 neurons (1.58 vs. 1.47 pA/pF, p>0.05). These results demonstrate that glutamate-mediated action through non-NMDA glutamate receptors in SFO neurons is smaller than that of hippocampal CA1 neurons, suggesting a possible protection mechanism from excitotoxicity by circulating glutamate in SFO neurons.

대칠기탕(大七氣湯)이 Glutamate에 의한 C6 Glial 세포의 Apoptosis에 미치는 영향 (Effects of Daechilgi-tang on Glutamate-induced Apoptosis in C6 Glial Cells)

  • Objectives : The water extract of Daechilgi-tang(DCGT) has traditionally been used for treatment of qi stagnation(氣滯), which is considered to be one of the important causes of neuronal disease in oriental medicine. However, little is known about the mechanism by which DCGT protects neuronal cells from brain cell damages. Methods and Results : The author tested the mechanism of the cytoprotective effect of DCGT on glutamate -stimulated rat C6 glial cells. DCGT significantly protected C6 glial cells from glutamate in MTT assay. Pre-treatment of C6 glial cells with DCGT markedly inhibited the DNA fragmentation of C6 cells induced by glutamate. Glutamate increased the generation of reactive oxygen species(ROS) and intracellular calcium level in C6 glial cells. However, pre-treatment with DCGT markedly suppressed the increase of ROS generation and intracellular calcium accumulation induced by glutamate. Among apoptosis signaling mediators, DCGT markedly increased the expression level of Bcl2 in glutamate-treated cells. It also inhibited the cleavage of caspase-3 and PARP proteins by glutamate in C6 glial cells. Conclusions : These results suggest that DCGT protects brain cells from glutamate cytotoxicity through inhibition of ROS generation and activation of apoptosis signaling pathway as well as induction of the anti-oxidant system.

Mechanism of Glutamate-induced $[Ca^{2+}]i$ Increase in Substantia Gelatinosa Neurons of Juvenile Rats

  • The glutamate receptors (GluRs) are key receptors for modulatory synaptic events in the central nervous system. It has been reported that glutamate increases the intracellular $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) and induces cytotoxicity. In the present study, we investigated whether the glutamate-induced $[Ca^{2+}]_i$ increase was associated with the activation of ionotropic (iGluR) and metabotropic GluRs (mGluR) in substantia gelatinosa neurons, using spinal cord slice of juvenile rats (10${\sim}21 day). $[Ca^{2+}]_i$ was measured using conventional imaging techniques, which was combined with whole-cell patch clamp recording by incorporating fura-2 in the patch pipette. At physiological concentration of extracellular $Ca^{2+}$, the inward current and $[Ca^{2+}]_i$ increase were induced by membrane depolarization and application of glutamate. Dose-response relationship with glutamate was observed in both $Ca^{2+}$ signal and inward current. The glutamate-induced $[Ca^{2+}]_i$ increase at holding potential of -70 mV was blocked by CNQX, an AMPA receptor blocker, but not by AP-5, a NMDA receptor blocker. The glutamate-induced $[Ca^{2+}]_i$ increase in $Ca^{2+}$ free condition was not affected by iGluR blockers. A selective mGluR (group I) agonist, RS-3,5-dihydroxyphenylglycine (DHPG), induced $[Ca^{2+}]_i$ increase at holding potential of -70 mV in SG neurons. These findings suggest that the glutamate-induced $[Ca^{2+}]_i$ increase is associated with AMPA-sensitive iGluR and group I mGluR in SG neurons of rats.

Effects of L-trans-pyrrolidine-2,4-dicarboxylate, a Glutamate Uptake Inhibitor, on NMDA Receptor-mediated Calcium Influx and Extracellular Glutamate Accumulation in Cultured Cerebellar Granule Neurons

  • Glutamate uptake inhibitor, L-trans-pyrrolidine-2, 4-dicarboxylate (PDC, $20{\mu}M$) elevated basal and N-methyl-D-aspartate (NMDA, $100{\mu}M$)-induced extracellular glutamate accumulation, while it did not augment kainate $100{\mu}M$-induced glutamate accumulation in cultured cerebellar granule neurons. However, pretreatment with PDC for 1 h significantly reduced NMDA-induced glutamate accumulation, but did not affect kainate-induced response. Pretreatment with glutamate $(5{\mu}M)$ for 1 h also reduced NMDA-induced glutamate accumulation, but did not kainate-induced response. Upon a brief application (3-10 min), PDC did neither induce elevation of intracellular calcium concentration $([Ca^{2+}]_i)$ nor modulate NMDA-indLiced $[Ca^{2+}]_1$ elevation. Pretreatment with PDC for 1 h reduced NMDA-induced $[Ca^{2+}]_1$ elevation, but it did not reduce kainate-induced $[Ca^{2+}]_1$ elevation. These results suggest that glutamate concentration in synaptic clefts of neurana cells is increased by prolonged exposure (1 h) of the cells to PDC, and the accumulated glutamate subsequently induces selective desensitization of NMDA receptor.

등대(燈臺)풀 유래 Furosin의 glutamate에 의한 HT22 세포 사멸 억제 효과 (Protective effect of furosin isolated from Euphorbia helioscopia against glutamate-induced HT22 cell death)

  • Objectives: In the brain, glutamate is the most important excitable neurotransmitter in physiological and pathological conditions. However, the high level of glutamate induces neuronal cell death due to exitotoxicity and oxidative stress. The present study investigated to evaluate a possible neuroprotective effect of furosin isolated from Euphorbia helioscopia against glutamate-induced HT22 cell death. Methods: Furosin was isolated from methanol extract of Euphorbia helioscopia and examined whether it protects glutamate-induced neuronal cell death. The cell viability was determined using Ez-Cytox assay. Anti-oxidative effect of furosin was determined by DPPH scavenging activities, and the levels of intracellular reactive oxygen species (ROS) were determined by the fluorescent intensity after staining the cells with $H_2DCFDA$. To evaluate apoptotic cell death, we performed nuclear staining and image-based cytometeric analysis. Results: The cell viability was significantly increased by treatement with furosin compared with the treatment with glutamate. Furosin showed a strong DPPH radical scavenging activity ($EC50=1.83{\mu}M$) and prevented the accumulation of intra cellular ROS. Finally, the presence of 50 and $100{\mu}M$ furosin significantly the percentage of apoptotic cells compared with glutamate treatment. Conclusion: The present study found that furosin is a potent neuroprotectant against glutamate-induced oxidative stress through inhibition of apoptotic cell death induced by glutamate. Therefore, the present study suggests that furosin as a bioactive compound of E. helioscopia can be a useful source to develop a drug for the treatment of neurodegenerative diseases and acute brain injuries.

Glutamate에 의한 산화적 스트레스로부터 신경세포를 보호하는 제비꽃 추출물의 영향 (Protective Effect of Neuronal Cell on Glutamate-induced Oxidative Stress from Viola mandshurica Extracts)

  • 본 연구에서는 신경독소일 뿐만 아니라 흥분성 신경전달물질로 잘 알려져 있는 glutamate 세포독성이 산화적 손상과 관련하고 있고, 여기에 방어효과를 보이는 제비꽃 추출물에 관하여 연구하였다. MTT reduction assay를 통하여 glutamate의 세포독성을 확인하였고 ascorbic acid와 같은 대표적인 항산화제를 처리한 후 광학 현미경을 이용한 형태학적 변화를 관찰하였다. N18-RE-105 세포주에 최종 농도 20mM의 glutamate를 처리 하면 40.8% 의 생존율을 보이는데 반하여 ascorbic acid 500 ${\mu}M$ 최종농도로 처리하였을 때 85.3%의 세포 생존율을 확인할 수 있었다. 그리고 신경세포 보호효과를 가지는 제비꽃을 methanol, ethanol, acetone 추출한 뒤 MTT reduction assay를 이용하여 활성을 확인하였으며 그 중 acetone 추출물을 최종농도 50, 100 ${\mu}g/ml$를 처리 시 76.8%, 79.4%로 가장 높은 세포 생존율을 확인할 수 있었다. 이 결과는 N18-RE-105 세포주의 형태학적 변화와 LDH release assey에서도 일치하는 결과를 확인하였다.

HT22 신경세포에서 금은화 추출물에 의한 글루타메이트 유도 산화적 스트레스 및 세포사멸 억제 효과 (Inhibitory Effect of Lonicera japonica Thunb. Flower Buds against Glutamate-Induced Cytotoxicity in HT22 Hippocampal Neurons)

  • Objectives : In this study, we investigated the neuroprotective effects of ethanol extract of Lonicera japonica flower buds (EELJ) on glutamate-induced neurotoxicity in mouse hippocampus-derived neuronal HT22 cells. Methods : After analyzing the cytoprotective effect of EELJ on glutamate in HT22 cells, the inhibitory effect of apoptosis was studied using flow cytometry. In order to analyze the antioxidant efficacy of EELJ, the levels of reactive oxygen species (ROS) and glutathione (GSH) were investigated, and the effects on the activities of superoxide dismutase (SOD) and catalase (CAT) were also analyzed. Furthermore, the effect of EELJ on the expression of apoptosis regulators such as Bax and Bcl-2 in glutamate-treated HT22 cells was investigated. Results : According the current results, pretreatment with EELJ significantly reduced glutamate-induced loss of cell viability and release of lactate dehydrogenase. EELJ also markedly attenuated glutamate-induced generation of intracellular ROS, which was associated with increased levels of GSH, and activity of SOD and CAT in glutamate-stimulated HT22 cells. In addition, EELJ was strikingly inhibited glutamate-induced apoptosis in HT22 cells. Furthermore, the expression of pro-apoptotic Bax was increased and the expression of anti-apoptotic Bcl-2 was decreased in glutamate-treated HT22 cells, while in the presence of EELJ, their expressions were maintained at the control levels. Conclusions : These findings indicate that EELJ protects glutamate-induced cytotoxicity in HT22 hippocampal neurons through antioxidant activity. Therefore, although identification of biologically active substances of EELJ and re-evaluation through animal experiments is necessary, this natural substance is a promising candidate for further research in preventing and treating oxidative stress-mediated neurodegenerative diseases.

Rhodopseudomonas sphaeroides의 질소 동화작용에 미치는 simazine의 영향 (Effect of simazine on nitrogen assimilation of rhodopseudomonas sphaeroides)

  • 광합성 세균인 Rhodopseudomonus sphaeroiders의 질소고정 효소 합성 및 암모니아 동화 경로에 중요한 glutamine synthetase(GS), glutamate synthase(GOGAT) 및 glutamatc dehydrogenase(GDH) 활성에 미치는 TriazinerP 제초제인 simazine의 영향을 조사하였다. 배양중에 simazine을 처리할 경우 성장 및 in vivo 질소고정효소의 합성 및 GS의 합성이 크게 저해되었다. 그리고 정상 배지에서 배양한 Rp. sphaeroides의 GOGAT 및 GDH의 활성이 GS의 활성보다 simazinc에 의해 더 큰 저해를 받았다. 이상의 결과는 simazine이 Rp. sphacroides의 광합성 과정을 억제하여, 질소고정효소의 합성을 저해하며 이로인해 GS의 합성이 저해될 가능성이 있음을 시사한다고 하겠다.
