Effect of glucose addition to xylose medium on xylitol production was investigated by using Candida parapsilosis ATCC 21019 mutant. With increasing the ratio of glucose to xylose in total amount of 50 g/l as glucose and/or xylose, xylitol production was decreased but ethanol and glycerol production were increased. Ethanol and glycerol concentration were maxmum in 10 g/l of xylose and 40 g/l of glucose medium as 21.5 g/l and 3.6 g/l, respecti- vely. No xylitol was formed in glucose medium without xylose because xylitol could be not produced from glucose. With increasing the ratio of glucose to xylose, the activity of xylose reductase which converted xylose to xylitol were decreased. The activities of xylitol dehydrogeiiase which converted xylitol to xylulose and then cell materials were found to be constant regardless of the ratio of glucose to xylose. This results indicated that glucose addition to xylose medium on cell growth was not affected. In order to prevent the inhibitory effect of glucose on xylitol production, glucose in a fermentor was fed with low concentration and then ethanol and glycerol was critically decreased and the xylitol yield from xylose of the culture with glucose feeding was recovered the almost same as that with only 50 g/l of xylose. However, the xylitol yield from total sugars (xylose and glucose) was decreased and glucose was not contributed to xylitol production. Therefore, the fermentation at high concentration of xylose without glucose was carried out. A final xylitol concentration of 242 g/l which corresponding 80.7% of xylitol yield was obtained from 300 g/l of xylose for 273 hours.
Objectives : The objective of this study is to observe the effect of Saryuktangkamibang, Saryuktang with Liriopis Tuber, Lycii Fructus, Schizandrae Fructus, and Bombyx Batryticatus, on the experimental diabetes. Methods : In order to induce diabetes experimentally, injected streptozotocin to the vein in the tail of rats and then treated oral administration of Saryuktangkamibang water extracts. In the measurement of the variation levels of glucose, ALP, creatinine, and BUN concentration for each concentration levels for serum (0.2g/100g, 0.4g/l00g, 0.6g/100g), concentration level of glucose significantly decreased in 0.4g/l00g concentration level of Saryuktangkamibang. With this 0.4g/l00g concentration level of Saryuktangkamibang, the following conclusion was derived from the measurement of the serum levels of glucose. ALP, AST, ALT, creatinine, and BUN concentration for the injection of each 4th, 11th, and 18th day. Results : 1. In the measurement for each concentrations, the glucose concentration level in the serum significantly decreased on the 9th day in the Saryuktangkamibang-dosed group. 2. The creatinine and BUN concentration level in the serum significantly decreased on the 9th day in the Saryuktangkamibang-dosed group to which 0.6g/100g injected. 3. In the measurement for each dosage dates, the glucose concentration level in the serum significantly decreased on the 11th day in the Saryuktangkamibang-dosed group to which 0.6g/100g injected. 4. The AST concentration level in the serum significantly decreased on the 11th day in the Saryuktangkamibang-dosed group. Conclusion : Saryuktangkamibang, Saruktang with Liriopis Tuber, Lycii Fructus, Schizandrae Fructus, and Bombyx Batryticatus, has an effect of hypoglycemia, with no hepato-renal dysfunction. For the experimental results above, in that nephrotoxicity significantly decreased on the 0.6g/100g concentration, another research such as study for effect in other ranges and study for effect of dosage term except the concentration range of this study is necessary. Also dosage of appropriate concentration of Saryuktangkamibang is thought to provide valuable aid to cure on diabetes.
The effect of dissolved oxygen concentration on the metabolism of glucose in Pseudomonas putida BM014 was investigated. Glucose was completely converted to 2-ketogluconate via extracellular oxidative pathway and then taken up for cell growth under the condition of sufficient dissolved oxygen concentration. On the other hand, oxygen limitation below dissolved oxygen tension (DOT) value of 20% of air saturation caused the shift of glucose metabolism from the extracellular oxidative pathway to the intracellular phosphorylative pathway. Specific activities of hexokinase and gluconate kinase in intracellular phosphorylation pathway decreased as the DOT increased, while 2-ketogluconokinase activity in extracellular oxidative pathway increased under the same condition. This result can be usefully applied to microbial transformation of glucose to 2-ketogluconate, the synthetic precursor for iso-vitamine C, with almost 100% yield via extracellular oxidation by simple DOT control.
Metformin is a biguanide antihyperglycemic agent often used for the treatment of non-insulin dependent diabetics (NIDDM). In this study, the pharmacokinetics and pharmacodynamics of metformin were investigated in Korean healthy volunteers during a fasting state for over 10 h. In order to evaluate the amount of glucose-lowering effect of metformin, the plasma concentrations of glucose were measured for a period of 10 h followed by the administration of metformin (oral 500 mg) or placebo. In addition, the concentration of metformin in blood samples was determined by HPLC assay for the drug. All volunteers were consumed with 12 g of white sugar 10 minutes after drug intake to maintain initial plasma glucose concentration. The time courses of the plasma concentration of metformin and the glucose-lowering effect were analyzed by nonlinear regression analysis. The estimated $C_{max}$, $T_{max}$, $CL_{t}$/F (apparent clearance), V/F(apparent volume of distribution), and half-life of metformin were 1.42$\{pm}$0.07 $\mu\textrm{g}$/mL, 2.59$\{pm}$0.18h, 66.12$\{pm}$4.6 L/h, 26.63 L, and 1.54 h respectively. Since a significant counterclock-wise hysteresis was found for the metformin concentration in the plasma-effect relationship, indirect response model was used to evaluate pharmacodynamic parameters for metformin. The mean concentration at half-maximum inhibition $IC_{50}$, $k_{in}$, $k_{out}$ were 2.26 $\mu\textrm{g}$/mL, 83.26 $H^{-1}$, and 0.68 $H^{-1}$, respectively. Therefore, the pharmacokinetic-pharmacodynamic model may be useful in the description for the relationship between plasma concentration of metformin and its glucose-lowering effect.
To elucidate the effect of ginseng saponin on adrenaline-induced hyperglycemia and hyperlipidemia, ginseng saponin was administered before and after administration of adrenaline, and concentration of glucose, triglyceride(TG), and free fatty acid (FFA) as well as lipase activity in plasma was determined. 1) Glucose concentration was slightly increased by administration of ginseng saponin at 1 hour before and 10 minutes after adrenaline administration, but decreased by administration of ginseng saponin at 7 and 4 hours before adrenaline administration. TG and FFA concentration was also greatly inhibited by administration of ginseng saponin in advance. 2) When gineseng saponin 3, 10, 30mg/kg were administered 7 hours before adrenaline administration, glucose and TG concentration as well as lipase activity were inhibited in proportion to the dose of ginseng saponin, but FFA concentration was slightly inhibited. 3) It was suggested that protopanaxatriol group have potentiating effect on adrenaline induced hyperglycemia but protopanaxadiol group have inhibitory effect. And ginseng saponin seems to have strong inhibitory action on TG mobilization into blood and stimulatory effect on esterification of FFA in liver and adipose tissue.
Polyacrylonitrile diagnostic membranes were prepared to measure blood glucose level of diabetics. Final absorbances at 680 nm through activated polyacrylonitrile membranes were measured at various concentration of glucose in plasma. The end-point results of K/S values obtained from the absorbances had a linear relationship toward the glucose concentration. The effect of temperature on the measurements of glucose concentration was studied. The stability of polyacrlonitrile diagnostic membranes was examined at RH 80%.
The production of nitric oxide (NO) and the expression of inducible nitric oxide synthase (iNOS) are known to be modulated by a variety of factors. Recent study showed that endotoxin-induced NO synthesis and iNOS expression were greatly enhanced by elevation of extracellular glucose concentration in murine macrophages. Although this was suggested to be due to the activation of protein kinase C (PKC) via sorbitol pathway, there was lack of evidence for this speculation. This study was performed to delineate the underlying intracellular mechanisms of glucose-enhancing effect on endotoxin-induced NO production in Raw264.7 macrophages. The levels of NO release induced by lipopolysaccharide (LPS) significantly increased by the treatment of glucose in a concentration dependent manner and also, this effect was observed in LPS-preprimed cells. Concurrent incubation of cells with PKC inhibitors, H-7 or chelerythrine, and LPS resulted in the diminution of NO production regardless of glucose concentration but this was not in the case of LPS-prepriming, that is, chelerythrine showed a minimal effect on the glucose- enhancing effect. PMA, a PKC activator, did not show any significant effect on glucose-associated NO production. Modulation of sorbitol pathway with zopolrestat, an aldose reductase inhibitor, did not affect LPS-induced NO production and iNOS expression under high glucose condition. And also, sodium pyruvate, which is expected to normalize cytosolic $NADH/NAD^+$ ratio, did not show any significant effect at concentrations of up to 10 mM. Glucosamine marginally increased the endotoxin-induced nitrite release in both control and high glucose treated group. 6-diazo-5-oxonorleucine (L-DON) and azaserine, glutamine: fructose- 6-phosphate amidotransferase (GFAT) inhibitors, significantly diminished the augmentation effect of high glucose on endotoxin-induced NO production. On the other hand, negative modulation of GFAT inhibitors was not reversed by the treatment of glucosamine, suggesting the minimal involvement, if any, of glucosamine pathway in glucose-enhancing effect. In summary, these results strongly suggest that the hexosamine biosynthesis pathway and the activation of PKC via sorbitol pathway do not contribute to the augmenting effect of high glucose on endotoxin induced NO production in macrophage-like Raw264.7 cells.

The effect of glucose concentration on the production of erythritol by Trichosporon sp. was mainly studied. The specific growth rate and production rate of erythritol gave the highest values of $0.23 h^{-1}\; and\; 4.2 g/\ell/h,\; respectively,\; on\; 100 g \; glucose/\ell$ of medium. The conversion yield of erythritol during the exponential phase and the stationary phase was constantly maintained at 19% and 51 %, respectively, while the glucose concentration in the medium varied from 100 g/$\ell$ to 400 g/$\ell$. The maximum overall erythritol conversion yield of 47% was obtained when the glucose concentration in the medium was 400 g/$\ell$. It corresponded to a 74% increase compared with the 100 g/$\ell$ glucose medium. The diauxy growth of this microbe was also observed. It grew exponentially consuming glucose, then after the second lag phase, biomass slowly increased using glycerol and erythritol.
Atherosclerotic vascular disease is a major cause of the increased morbidity and mortality assciated with diabetes mellitus. The prominent role of nutrition in hypercholesteolemia and atherosclerosis is generally accepted. Diet is a key element in the management of diabetes (type I-IDDM), yet the appropriate diet for patient with diabetes mellitus is not well known. Dietary protein has been shown to have a significant effect on plasma cholesterol levels in both experimental animals and humans. The present experiment was designed to determine the effect of the dietary protein level(20% vs 60%) on plasma glucose concentration, lipids profile, insulin and glucagon levels from non-diabetic and streptozotocin-induced diabetic rats. Results showed that a high protein diet decreased triglyceride concentration in diabetic rats. Also diabetic rats fed a high protein diet were hypocholesterolemic than rats fed a control diet. There were no effects by level of protein on fasting blood glucose concentration and insulin/glucagon ratio. Results from the present study suggest that a high protein diet may be beneficial to control pasma lipids in chemically-induced diabetic rats.

This study was carried out to investigate the motor activity and glucose transport and metabolism of Gaewool-Whadam-Jian(GWJ) in rat gastro-intestinal tract. The motor activity of the rat gastro-intestinal tract has been investigated by means of measuring barium sulfate passage degrees. Atropine treatment significantly delayed barium sulfate transit, and GWJ pretreatment increased intestinal motor activity, but not significant. GWJ administration showed no toxicity to kidney and liver. Transport and metabolism of glucose were studied in everted sac of rat small intestine with incubation under several conditions. The transport and metabolism of glucose were greater at jejunum than ileum. So, everted jejunum of rat were used to study the effect of GWJ. When GWJ were treated, the concentration of glucose were higher than untreated group. This result was thought to be influenced by the glucose in GWJ. When 2, 4 dinitrophenol and phlorizin were treated, the transport and metabolism of glucose were decreased, but GWJ treated together, the concentration of glucose in serosal solution increased. Gastric juice secretion and total acidity significantly decreased by administration of GWJ through duodenum region. The mechanism of effect of GWJ was still unidentified, Dut through continuous investigation, the effect of GWJ should be investigated.
