Effect of SiO2 on yield and phenolic profile of Tartary buckwheat

  • The aim of this study was to enhance the Tartary buckwheat growth and phenolic profile by the application of $SiO_2$ on potsin the glasshouse. The liquid of $SiO_2$ were applied at three times after three weeks of sowing at ten days interval. The doses of the $SiO_2$ was low (2.5 ml/5 L water), high (10 ml in 5 L water) and control experiment was done without $SiO_2$. In this study, it is clearly shown that high dose of $SiO_2$ increased the buckwheat plant growth including, plant height, leaf length and width, stem diameter, fresh weight and number of seed per plant compared to control treatment. In the same way, the total phenolic compound (1421 mg/100 g), total flavonoid (35.1 mg/100g), rutin (3,130 mg/100g) and DPPH (82%) in plant were also increased high dose of $SiO_2$ compared to control (1,175 mg/100g, 31.9mg/100g, 860 mg/100g, 59%, respectively). Moreover, higher phenolic compound (1,421 mg/100g) and DPPH (82%) was observed in plant compared to seed (196 mg/100mg, 72 %, respectively). In the contrary, total flavonoid (36.2 mg/100g) and rutin (1,400 mg/100g) was higher in buckwheat seed compared to plant (35.1 mg/100g, 3,130 mg/100g, respectively). Finally, it is concluded that higher dose of $SiO_2$ enhance buckwheat growth and phenolic profile. Further investigation is needed to evaluate the optimum dose of $SiO_2$ according to soil conditions in the field.

  • This study was conducted to investigate effects of cooling methods on the growth and yield of tomato cv. momotaro in the glasshouse for four years from 1996 to 1999. Cooling methods were fan, fan and fogging, fan and shading(temp. control), fan and shading(radiation control), fan and shading (temp. control) with fogging. Fan, Fogging and Shading(temp. control) were operated automatically when air temperature was over 3$0^{\circ}C$. Amount of fogging was 500m1/min/100m$^2$and Droplets in a fog were 50 microns or smaller. Shading(radiation control) was operated automatically when solar radiation was over 500W/m$^2$. The growth and yield were the least in fan and shading(temp. control) method due to lack of light Intensity. Fogging method must be reconsidered for expensive equipment and maintenance expenses. As the matter stands, It is suggested to be the most considerable cooling method to increase ventilation rate with fan or use fan and shading(radiation control).

Development of Plant Regeneration and Genetic Transformation System from Shoot Apices of Sorghum bicolor (L.) Moench

  • Development of efficient plant regeneration and genetic transformation protocols (using the Particle Inflow micro-projectile Gun and the shoot-tips as target tissue) of Sorghum bicolor (L.) Moench in terms of expression of the reporter gene, $\beta$-glucuronidase(uidA) is reported here. Two Indian cultivars of sorghum were used in the study, viz. M-35-1 and CSV-15. Plant regeneration was achieved from one-week-old seedling shoot-tip explants via multiple-shoot-clumps and also somatic embryos. The multiple-shoot-clumps were produced on MS medium containing BA (0.5, 1.0 or 2.0 mg/$L^{-1}$), with biweekly subculture. Somatic embryos were directly produced on the enlarged dome shaped expansive structures that developed from shoot-tip explants (without any callus formation) when cultured on MS medium supplemented both with BA (0.5, 1.0 or 2.0 mg/$L^{-1}$) and 2,4-D (0.5 mg/$L^{-1}$). Whereas each multiple-shoot-clump was capable of regenerating more than 80 shoots via an intensive differentiation of both axillary and adventitious shoot buds, the somatic embryos were capable of 90% germination, plant conversion and regeneration. The regenerated shoots could be efficiently rooted on MS medium containing 1.0mg/$L^{-1}$ IBA and successfully transplanted to the glasshouse and grown to maturity with a survival rate of 92%. The plant regeneration efficiency of both the genotypes were similar. After the micro-projectile bombardment, expression of uidA gene was determined by scoring blue transformed cell sectors in the bombarded tissue by an in situ enzyme assay. The optimal conditions comprising a helium pressure of 2200 K Pa, the target distance of 11 cm with helium inlet fully opened and the use of osmoticum have been defined to aid our future strategies of genetic engineering in sorghum with genes for tolerance to biotic and abiotic stresses.

Effect of different fertilizer application on the dry matter production and leaf photosynthetic of Yacon(Polymnia sonchifolia) (시비수준의 차이가 야콘의 물질생산과 광합성속도에 미치는 영향)

  • This study was conducted to examine the dry matter production and leaf photosynthesis under different fertilizer application in Yacon(Polymnia sonchifolia)plants. Under field conditions, dry matter production of stable mature application plot(S-1) was much larger than non-treated control plot(CTL), but bulb dry weight of 10kg nitrogen application plot per 10a (N-10) was not differentiated from CTL. The maximum photosynthetic rates increased in Yacon plants grown at S-1 while in the rates of CTL reduced under glasshouse in the same treated pot condition. The optimum temperature for the highly photosynthetic rates is about $34{\pm}3^{\circ}C$, and the rates did decrease by stomatal resistance below 28℃. The estimated light saturation point was $1200{\mu}mol{\cdot}m^{-2}s^1$. The results of this study indicate that soil conditions were highly related to dry matter production and leaf photosynthesis rate in Yacon plants.

Varietal Variation in Germinative Energy, Capacity and Water Sensitivity for Malting Barley (맥주보리에서 발아세, 발아율 및 수감수성의 품종간 차이)

  • Forty-nine malting barley varieties were tested at 3 and 8 weeks after harvest, stored in natural glasshouse so as to investigate varietal variation of germinative energy(GE), capacity(GC) and water sensitivity(WS). The differences among GE, GC, promptness index(PI), and WS on paper at two moisture levels (4$m\ell$ and 8$m\ell$ tests) were great at 3 weeks after harvest, but small at 8 weeks after harvest. Excess water depressed the germination of dormant grains. The GE of whole varieties were excellent, but WS of only two varieties, CI 12062 and CI 15631 were bad at 8 weeks after harvest. So in the case of broad germplasm to be used for crossing materials, it is suggested that WS will be tested to screen good quality malting lines. WS was highly significantly correlated with GE, GC, and PI of 8 ml test, but was not with three factors of 4 $m\ell$ test. GE and WS were various with storage period, varieties and grain plumpness, indicated that breeding good grain-filling varieties and improving production practice for good grain-filling would be studied in the future.

  • Glasshouse experiments were conducted to evaluate nodulation, and $N_2$ fixation of 73 strains of Bradyrhizobia isolated from nodules of groundnuts and/or cowpeas grown at various location in Korea and 31 strains of cowpea Bradyrhizobia obtained from overseas sources. Of the strains tested 51.9%, 26.4%, and 21.7% were rated fully effective, partially effective, and ineffective, respectively. Eight very effective strains were selected. Nodulation, shoot dry mass, whole plant dry mass, and plant total N content were significantly correlated with each other. The highly significant correlation of total N content and plant dry mass ($r=0.98^{**}$) indicates that plant dry mass can be used as a measure of strain effectiveness in preliminary test.

  • This study was conducted to investigate the herbicidal activity against resistant biotype on sulfonylurea (SU) herbicides such as Scirpus juncoides and Monochoria vaginalis at the glasshouse and paddy rice cultivation area. In order to investigate resistance breakability against SU-resistanted annual weeds, new rice herbicides containing tefuryltrione [p-hydrophenyl pyruvate dioxygenase (HPPD) inhibitor] were tested. In both lab test and field experiment, tefuryltrione mixture, 4-HPPD inhibitor had shown excellent efficacy with a bleaching on the leaves of resistant annual weeds at early time after treatment and showed excellent persistance. Especially, tefuryltrione mixture had shown excellent controlling effect on annual and perennial SU-resistant S. juncoides in the regional field experiment. In phytotoxicity test, this tefuryltrione mixture had shown good selectivity to common rice species.

Protoplast Culture and Plant Regeneration of Rice (벼의 원형질체 배양과 식물체 재분화)

  • Embryogenic calli were induced from mature seed scutella of anther culture-derived rice variety Zhonghua 8. Cell suspension cultures were initiated from friable embryogenic calli and utilized as source material for protoplast isolation. Generally, the older and finer cell suspensions gave higher protoplast yields than younger suspension cultures. Protoplasts exhibited sustained cell division and formed microcalli when cultured in KPR medium supplemented with 0.5 mg $l^{-1}$ 2,4-D, 1.0 mg $l^{-1}$ NAA and 0.5 mg $l^{-1}$ zeatin using the agarose embedding procedure without feeder cells. Protoplast plating efficiencies ranged from 0.20 to 0.54%. Microcalli were transferred to MS medium supplemented with 2.0 mg $l^{-1}$ kinetin and 0.5mg $l^{-1}$ NAA for plant regeneration. The regeneration frequencies were 2 to 12%, depending on the cell suspension lines of Zhonghua 8. The plants were transferred to the glasshouse and were fertile.

Effect of Foliar and Root Application of Silicon Against Rice Blast Fungus in MR219 Rice Variety

  • Rice blast disease caused by Magnaporthe grisea (Hebert) Barr [teleomorph] is one of the most devastating diseases in rice plantation areas. Silicon is considered as a useful element for a large variety of plants. Rice variety MR219 was grown in the glasshouse to investigate the function of silicon in conferring resistance against blast. Silica gel was applied to soil while sodium silicate was used as foliar spray at the rates of 0, 60, 120, 180 g/5 kg soil and 0, 1, 2, 3 ml/l respectively. The treatments were arranged in a completely randomized design. Disease severity and silicon content of leaves were compared between the non-amended controls and rice plants receiving the different rates and sources of silicon. Silicon at all rates of application significantly (${\alpha}$ = 0.05) reduced the severity of disease with highest reduction (75%) recorded in treatments receiving 120 g of silica gel. SEM/EDX observations demonstrated a significant difference in weight concentration of silicon in silica cells on the leaf epidermis between silicon treated (25.79%) and non treated plants (7.87%) indicating that Si-fertilization resulted in higher deposition of Si in silica cells in comparison with non-treated plants. Application of silicon also led to a significant increase in Si contents of leaves. Contrast procedures indicated higher efficiency of silica gel in comparison to sodium silicate in almost all parameters assessed. The results suggest that mitigated levels of disease were associated with silicification and fortification of leaf epidermal cells through silicon fertilization.

Plant Regeneration from Protoplasts of Indica Rice (Indica 벼의 원형질체들로부터 식물체 재분화)

  • An efficient protocol for plant regeneration from protoplasts of the indica rice variety IR43 has been developed. The procedure involved plating of embryogenic suspension-derived protoplasts on the surface of a filter membrane overlaying agarose-embedded feeder cells. Lolium multiflorum cell suspensions were preferable to these of Oryza ridleyi as feeder cells and Lolium suspensions supported colony formation from up to 0.68% of the protoplasts, depending on the age of cell suspensions. Plant regeneration frequency was significantly improved by using maltose alone or in a 1:1(w/w) combination with sucrose as carbohydrate source and a simple dehydration treatment using a high concentration of agarose in the regeneration medium. Medium containing maltose or maltose mixed with sucrose increased the plant regeneration frequency compared with medium containing sucrose alone. The plant regeneration frequency was increased to 30.7 to 70.7% following dehydration treatment, while the non-treated controls showed a regeneration frequency of 3.1 to 30.6%. Protoplast-derived plants were transferred to the glasshouse, flowered with morphologically normal.
