Study on the Effect of Ginseng on the cyclic AMP Content in the Rat Hearts (인삼(人蔘)이 백서심장(白鼠心臟)의 cyclic AMP함량(含量) 변화(變化)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • It was previously reported from our laboratory that the rate of deterioration of the force of contraction was slower in heart from Panax ginseng extract treated rats. The study carried out to elucidate its mechanism of the action on hearts. The cyclic AMP content in the rat hearts was measured by the method of radioimmunoassay techniques. Panax ginseng extract (100mg/kg/day) was administered orally to male Sprague-Dawley rats weighing 150g to 250g for 1 week and after 24 hrs the hearts were isolated and the cyclic AMP content in the fresh heart was assayed. The difference in cyclic AMP content between the rats treated with Panax ginseng extracts and normal rats was not significant. Panax ginseng extract(l00mg/kg/day) was administered orally to the rats for I week and after 24 hrs the hearts were isolated and perfused with Krebs-Henseleit buffer (pH7.4) for 90min. The cyclic AMP content in the both treated and normal rats was not also significantly different. On the other hand, when total ginseng saponin (50mg/kg/day) was administered orally to rats for 1 week and after 24 hrs, the isolated hearts were perfused with Krebs Henseleit solution for 32min, the cyclic AMP content in total ginseng soponin treated hearts was decreased by 18.7% compared to normal rats. It was also observed that when isolated hearts were perfused with total ginseng saponin $(10^{-4}g/ml)$ for 12 min after 30 min equilibration period, the cyclic AMP content in total ginseng saponin treated hearts was decreased by 23.7% compared to normal rats. Isolated hearts were perfused with ginseng saponins $(10^{-4}g/ml)$ or with Krebs-Henseleit solution alone for 10min and subsequently with dl-isoproterenol $(1/2{\times}10^{-6}M)$ until the positive inotropic effect of isoproterenol was initiated. The cyclic AMP content in each rat hearts treated with total ginseng saponin, or with ginsenoside $Rb_1$, or with Krebs-Henseleit solution alone were increased by 35.5%, 42.4%, 47.5%, respectively, compared to normal rats.

Effects of Korean red ginseng and herb extracts mixture (KTNG0345) on procollagen biosynthesis and matrix metalloproteinase-1 activity in human dermal fibroblast (홍삼 생약 복합물(KTNG0345)이 섬유아세포의 Procollagen 생합성과 Matrix metalloproteinase-1(MMP-1) 활성에 미치는 영향)

  • Skin wrinkles are associated with collagen synthesis and matrix metalloproteinase-I (MMP-1) activity. This study was carried out to select optimum ratio of 3 herbs in skin health food for anti-wrinkle. Human dermal fibroblast cell was incubated with experimental samples, which were Korean red ginseng ethanol extracts (ER), Torilis fructus water extracts (WT), Corni fructus water extracts (WC) and their mixtures (WM1, WM3). And then we determined effects on collagen biosynthesis, MMP-1 activity and SOD activity in human dermal fibroblast cell. In control group, collagen biosynthesis was amounted at 474.8 ng/ml and 533.9 ng/ml, 539.3 ng/ml, 514.1 ng/ml in ER, WT and WC respectively. Furthermore, WM3 (KTNG0345) was increased to 561.45 ng/ml. MMP-1 activity of ER, WT, WC, WM1 were determined to 31.9 ng/ml, 32.85 ng/ml, 32.0 ng/ml, 31.3 ng/ml and WM3 (KTNG0345) was decreased to 28.85 ng/ml. In addition, the experimental samples showed a antioxidative activities. From this results, we conclude that Korean red ginseng ethanol extracts, Torilis fructus water extracts, Corni fructus water extracts and their mixtures have a anti-wrinkle effect and WM3 (KTNG0345) may be regarded as an optimum composition for synergic effect producing. The standardized components of KTNG0345, ginsenoside-$Rb_1$, torilin and loganin were identified at 10.85 mg/g, 0.128 mg/g and 3.92 mg/g respectively.

EFFECTS OF GINSENG COMPONENTS ON RODENTICIDE VACOR-INDUCED DIABETES MELLITUS IN RATS (인삼성분이 살서제(Vacor)로 유발시킨 쥐의 당뇨에 미치는 영향)

  • It is now well established that the rodenticide Vacor (N-3-pyridyl-mehtyl-N'-p-nitropheny-lurea) causes a hyperglycemia in human and rats. It is also reported that there are some components (DPG-3) in ginseng radix which cause hypoglycemic effect on alloxan diabetic mice. In the present study, attempts were made to demonstrate in Vacor-poisoned rats the hypo-glycemic activity of red ginseng component(RGC), which was extracted by Kimura's DPG-3 extraction procedure and found to be effective for lowering a hyperglycemia in alloxan-diabetic rats. Vacor in a dose of $LD_{50}$ (10mg/kg) produced a glucose intolerance with a paradoxical moderate increase in blood immunoreactive insulin and derangement in glucose metabolism of epididymal adipocytes in rats. Although RGC (20mg/kg, i.p.) did not exert any significant influence on a hyperglycemia induced by large lethal doses (25mg/kg) of Vacor ingestion, it improved the LDso Vacor-induced glucose intolerance and caused a further increase in blood insulin levels in Vacor-poisoned rats. The administration of RGC (20mg/kg, i.p.) normalized Vacor-induced depression of glucose metabolism and lipogenesis in the epididymal adipocytes with an improvement of reduced responses to insulin of adipocytes from Vacor-poisoned rats. These results suggest that some red ginsneng components contained in RGC fraction normalize the depressed peripheral glucose unitlization and insulin response and eventually lead to an improvement of abnormal glucose tolerance developed in rats poisoned with small doses of Vacor.
