Effect of the Extruded Ginseng on Antioxidant Activity (압출 성형 인삼의 항산화 활성 증강 효과)

  • This study was conducted in order to evaluate the antioxidant activity of extruded ginseng in different extracted fractions. Each of the fractions obtained from extruded ginseng and ginseng (control) were extracted with 80% ethanol, and then the lipophilic components were removed with ether while the hydrophilic components were separated with water-saturated butanol. Each of the 80% ethanol/butanol/water layers were collected and evaporated to acquire samples for tests of saponin content and antioxidant activity. The antioxidant activity of extruded ginseng fractions and ginseng fractions were determined via the oxygen radical absorbance capacity (ORAC) assay. Overall, the extruded ginseng samples harbored saponin contents of 2.2 (Rg1), 2.3 (Re), 1.2 (Rc), 1.3 (Rb2), and 2.2 (Rd) times that measured in the ginseng prior to extrusion. Antioxidant capacity was also higher, not only in the 80% ethanol/butanol which harbor a significant quantity of saponin, but also in the water fractions, which harbor relatively low quantities of saponin as compared to the control samples.   • Recently, we isolated a toxic substance named 'toxohormone-L' from ascites fluid of patients with various malignant tumors. The toxohormone-L stimulated lipolysis in rat adipocytes and induced anorexia in rats. Both the lipolytic and the anorexigenic actions of toxohormone-L were found to be inhibited by ginsenoside $Rb_2$ in Korean red ginseng. Isolated rat adipocytes are well known to possess opposite pathways of lipid metabolism: lipolysis and lipogenesis. Both of the metabolism respond to various biologically active substances such as epinephrine, ACTH and insulin. Epinephrine and ACTH stimulate lipolysis and insulin accelerates lipogenesis. Recently. Korean red ginseng powder was found to contain adenosine and an acidic substance which inhibited epinephrine-induced lipolysis and stimulated insulin-mediated lipogenesis from added glucose. The chemical structure of this acidic substance is determined to be pyro-glutamic acid (Pyro-Glu), Pyro-Glu exhibits selective modulations toward the opposite metabolic pathways in rat adipocyte; It inhibits the lipolysis but stimulates the lipogenesis. We call these substances (adenosine, Pyro-Glu) 'selective modulators' or 'insulin-like substances'. Based on these results, physiological significances of these substances in Korean red ginseng were discussed.

Physicochemical quality characteristics of hot water extracts of processed ginseng based on different heat treatments (열처리 방법에 따른 가공 인삼 열수추출물의 이화학적 품질 특성)

  • The present study was carried out to investigate the physicochemical properties of hot water extracts of red ginseng powder prepared by two-stage hot air drying method using steamed ginseng and steaming liquid for 2.5 h under high-temperature and high-pressure autoclave condition. The total polyphenols, total flavonoids, total sugar, acid polysaccharides and crude saponin in hot water extracts from red ginseng powder were analyzed and determined, and the flavor components of ginseng were measured using color difference meter and an electronic tongue. The total polyphenol, total flavonoid, total polysaccharide, and acid polysaccharide of the red ginseng hot water extract obtained by autoclaving (ARG) were 9.06 mg GAE/g, 3.38 mg NE/g, 35.22 g/100 g, and 10.90 g/100 g, respectively. The final contents of the total polyphenols, total flavonoids, crude saponin were higher than those determined using other red ginseng methods; the time required for steamed red ginseng production reduced. The total ginsenoside content of ginseng including Rb1 was 10.69 mg/g, which is the lowest ARG. The processing conditions affected the conversion to ginsenosides unique to red ginseng. Red ginseng and steaming liquid obtained from the autoclave are expected to be in need for non-food materials and products as well as foods by improving the flavor components through conversion of red ginseng components into low molecular weight.

Development of Fermentation Process of Ginseng Leaf Extraction Probiotic Strain and Characterization of Product Quality (프로바이오틱 균주에 의한 인삼 잎 추출물 발효공정 확립 및 생성물의 품질 특성분석)

  • This study was carried out to investigate extraction efficiency by microwave for extraction of pesticide residues and the bioconversion of ginsenosides of ginseng leaf by using various lactic acid bacteria in order to promote the utilization of ginseng leaf. The hexane extraction by microwave of tolclofos-methyl and azoxystrobin in ginseng leaf was efficient. The optimal condition for extraction of tolclofos-methyl and azoxystrobin in ginseng leaf by microwave was 50 to 95 watts of power supply, 3 minutes of extraction.The gisenosides Rg1 and Rb1 contents have decreased, while the Rh1, Rg3, Rk1 and Rh2 have increased due to fermentation. The ginsenosides Rg3 of the fermented ginseng leaf has increased and the contents were $70.62{\sim}77.61{\mu}g/g$(control $2.77{\mu}g/g$). The total phenolic acid content and electron donating ability of the ginseng leaf have totally decreased after 7 days of fermentation. The total phenolic acid contents of the fermented ginseng leaf with various lactic acid bacteria did not show any tendency as different strains.

Anti-inflammatory activity and toxicity of the compound K produced by bioconversion (생물전환에 의해 생성된 Compound K의 항염증 및 독성 효과)

  • Compound K (20-O-β-(D-glucopyranosyl)-20(S)-protopanaxadiol) is an active ingredient of ginsenosides. Compound K has been known to produce from biotransformation by β-glucosidase action of human intestinal microbes after oral admistration of ginseng. We have investigated the cytotoxicity of compound K obtained from bio-converted ginseng extract. As a result, compound K showed no significant cytotoxicity in the concentration of 0.001 to 1 ㎍/mL and inhibited the production of TNF-α, MCP-1, IL-6 and NO in RAW 264.7 cells induced by LPS inflamation. In the same concentration, HaCaT cells induced by inflammation with TNF-α and IFN-γ decreased IL-8 production due to compound K treatment. In the brine shrimp lethality assay, the LC50 of compound K was 0.37 mg/mL indicating some toxicity, but the bioconverted product containing 35% compound K showed relatively low toxicity with an LC50 of 0.87 mg/mL. These results suggest that the compound K enriched extract is a potential functional material for acne relief cosmetic products.

The Analysis of Growth Characteristics and Ginsenoside Contents Wild-simulated Ginseng (Panax ginseng C. A. Meyer) with Different Years of Rusty Roots (적변에 따른 연근별 산양삼 생육특성과 진세노사이드 함량 분석)

  • 본 연구에서는 7년근과 13년근 산양삼을 선정하여 적변 유무에 따라 분류하고 적변에 따른 산양삼의 생육특성과 진세노사이드 함량을 조사하기 위해 수행되었다. 산양삼 재배지 9개소의 산양삼 지하부의 생육특성을 분석한 결과, 7년근과 13년근 산양삼의 생육특성은 적변에 따른 유의적인 차이는 확인되지 않았다. 적변에 따른 연근별 산양삼의 진세노사이드 함량을 비교 분석한 결과, 7년근 산양삼은 Rb1과 Rg1은 진세노사이드 함량이 일반삼에 비해 적변삼에서 높았고, 진세노사이드 Rc, Rd, Re, Rg2는 적변삼에 비해 일반삼에서 유의적으로 높은 것을 확인하였으나 적변에 따른 13년근 산양삼의 진세노사이드 함량 유의적인 차이는 확인할 수 없었다. 산양삼 생육특성과 진세노사이드 함량 간의 상관관계를 분석한 결과, 일반삼의 경우 진세노사이드 Rb2, Rc, Rd, Rf, Rg1이 산양삼의 지하부 길이와 유의적인 정의 상관관계를 보였고, 적변삼은 진세노사이드 Rd가 지하부 길이와는 유의적인 정의 상관관계, 주근직경과는 유의적인 부의 상관관계를 보였다. 본 연구에서는 7년근과 13년근 모두 적변에 따른 생육특성의 차이는 확인되지 않았고, 진세노사이드 함량은 7년근에서는 적변에 따라 유의적인 차이를 보였지만 13년근에서는 유의적인 차이가 없는 것을 확인하였다. 임가에서 산양삼은 7년근에 비해 13년근이 보다 고가에 거래되는 만큼 본 연구의 결과를 통해 적변삼에 대한 소비자들의 인식을 개선하여 산양삼 재배 농가의 소득 증대에 도움을 주고, 향후 산양삼의 품질규격을 정립하는 데 있어 유용한 정보를 제공할 수 있을 것으로 사료된다.

Hepatoprotective Effects of Ginseng Intestinal Metabolite IH-901 on Chemical-Induced Hepatic Damage

  • Hepatoprotective effects of white ginseng extract (WGE), and IH-901 (20-O-${\beta}$-D-glucopyranosyl-20(S)-protopanaxadiol) derived from intestinal metabolite of ginsenoside $Rb_1$ were studied using two experimental animal models with chemical-induced hepatic damage. Administration of WGE (200 and 500 mg/kg) and IH-901 (0.01, 0.05, and 0.1 mM/kg) significantly decreased aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in acute hepatitic mice induced by $CCl_4$. Administration of WGE (l00 mg/kg) and IH-901 (0.02, 0.04, and 0.08 mM/kg) significantly decreased AST and ALT levels in acute hepatitic rats induced by D-galactosamine. AST and ALT levels of IH-901 groups decreased. These results suggested WGE and IH-901 may have protective effects against chemical-induced hepatic damage.

Effects of Extraction Temperature and Time on Saponin Content and Quality in Raw Ginseng (Panax ginseng) Water Extract (수삼의 추출 온도 및 시간이 물 추출액의 사포닌 함량 및 품질에 미치는 영향)

  • In this study, raw ginseng water extract solutions were analyzed to set up the functional saponin content and quality optimization condition. The highest saponin content among the total raw ginseng water extracts was $74.6\;mg/100\;m{\ell}$ which was extracted at $75^{\circ}C$ for 24 hours. In addition, the saponin content decreased according to the increased extraction temperature and time. The highest total content of $Rb_2$ and Re was $19.9\;mg/100\;m{\ell}$ at $75^{\circ}C$ for 12 hours which decreased according to the increased extracted temperature and time. The highest prosapogenin ($Rg_2\;+\;Rg_3\;+\;Rh_1$) content among the total raw ginseng water extracts was $28.6\;mg/100\;m{\ell}$ which was extracted at $85^{\circ}C$ for 36 hours. The reducing sugar content, sweetness and turbidity were increased according to the increased extraction temperature and time. But pH were decreased according to the increased extracted time.

Antidepressant Effect of the Subchronic Administration of the Methanolic Extract of Wild-ginseng and Cultivated-ginseng in Mice Tail Suspension Test (산삼과 인삼 메탄올 추출물 아만성 복용의 Mice Tail Suspension Test에서의 항우울 효과에 대한 비교연구)

  • Objectives : The antidepressant effect of the subchronic administration of the methanolic extract of wild ginseng(WG) was investigated compared with that of cultivated ginseng(CG, panax ginseng) extract. Methods : To assess the antidepressant effect of the ginseng extracts, tail suspension test(TST) was executed in mice after daily administration of WG or CG extract for five consecutive days. Results : The WG extract at daily dose of 600mg/kg significantly reduced the total duration of immobility in the TST, whereas there was no significant reduction at daily dose of 300mg/kg WG and 600mg/kg CG. There were no individual differences between experimental groups in open field test (OFT) to evaluate psychostimulant effects of WG or CG extract. In the high performance liquid chromatography(HPLC) analysis of the extracts, it was found that WG included four times more ginsenoside Rg1 and Re, three times more Rf, and six times more Rb1 and Rc than CG. Conclusions : It is suggested that WG extract has stronger antidepressant effect than CG extract, which means it includes more antidepressant compounds than CG.

Study on the Effect of Ginseng on the cyclic AMP Content in the Rat Hearts (인삼(人蔘)이 백서심장(白鼠心臟)의 cyclic AMP함량(含量) 변화(變化)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • It was previously reported from our laboratory that the rate of deterioration of the force of contraction was slower in heart from Panax ginseng extract treated rats. The study carried out to elucidate its mechanism of the action on hearts. The cyclic AMP content in the rat hearts was measured by the method of radioimmunoassay techniques. Panax ginseng extract (100mg/kg/day) was administered orally to male Sprague-Dawley rats weighing 150g to 250g for 1 week and after 24 hrs the hearts were isolated and the cyclic AMP content in the fresh heart was assayed. The difference in cyclic AMP content between the rats treated with Panax ginseng extracts and normal rats was not significant. Panax ginseng extract(l00mg/kg/day) was administered orally to the rats for I week and after 24 hrs the hearts were isolated and perfused with Krebs-Henseleit buffer (pH7.4) for 90min. The cyclic AMP content in the both treated and normal rats was not also significantly different. On the other hand, when total ginseng saponin (50mg/kg/day) was administered orally to rats for 1 week and after 24 hrs, the isolated hearts were perfused with Krebs Henseleit solution for 32min, the cyclic AMP content in total ginseng soponin treated hearts was decreased by 18.7% compared to normal rats. It was also observed that when isolated hearts were perfused with total ginseng saponin $(10^{-4}g/ml)$ for 12 min after 30 min equilibration period, the cyclic AMP content in total ginseng saponin treated hearts was decreased by 23.7% compared to normal rats. Isolated hearts were perfused with ginseng saponins $(10^{-4}g/ml)$ or with Krebs-Henseleit solution alone for 10min and subsequently with dl-isoproterenol $(1/2{\times}10^{-6}M)$ until the positive inotropic effect of isoproterenol was initiated. The cyclic AMP content in each rat hearts treated with total ginseng saponin, or with ginsenoside $Rb_1$, or with Krebs-Henseleit solution alone were increased by 35.5%, 42.4%, 47.5%, respectively, compared to normal rats.
